RESUMO

Recent literature data on cryopreservation of isolated hepatocytes are summarized. The hepatocytes are considered to constitute a promising system for investigation of liver metabolic peculiarities as well as for elucidating mechanisms of low temperature effects on biological systems.

RESUMO

A method for isolating hepatocytes using the two-step liver perfusion by chelate-containing solutions is described. The hepatocytes obtained are highly inactive for the trypan blue staining test, (not less than 90%), ATP content, the rate of endogenic respiration and its stimulation by succinate, a degree of xenobiotic oxidation.

RESUMO

A study was made of the effect of normothermic incubation on the metabolic state of parenchymal liver cells, isolated by the nonenzymatic method. In the process of a 2 hour incubation in the nutritious sodium-containing medium, a high level of the energy supply system is maintained, determined by the contents of adenine nucleotides, the rate of endogenous respiration and by its stimulation by succinate.

RESUMO

The hypothermic storage of rat liver parenchymal cells in the sucrose-salt medium is studied. It is demonstrated that short-term hepatocyte storage at 4 degrees C results in a slight imbalance, that evidences for sufficient adaptive potencies of these cells to the temperature fall.

RESUMO

The activity of cytoplasmic enzymes (hexokinase, phosphorylase, glucose-6-phosphate dehydrogenase, lactate dehydrogenase) in the extracellular medium following leucocyte freezing-thawing to -30, -70, -140 or -196 degrees C was studied. The appearance of these enzymes in the freezing medium has been found to be associated with the disruption of cellular elements. The source of lactate dehydrogenase activity in the extracellular medium was shown to be due not only to disrupted leucocytes, but also to cells remained after freezing-thawing. Possible mechanisms of the release of enzyme proteins from leucocytes after freezing-thawing are discussed.
