RESUMO

The partial composition of O-antigens in Escherichia known as the causative agents of escherichioses localized in the intestine and other organs has been studied. Escherichia of groups 01, 02 and 03 are characterized by different partial composition of their O-antigens. This composition can be expressed by the formulae: 01a, 1b; 01a, 1b, 1c; 01a, 1b, 1c, 1d; 01a, 1e; 02a, 2b, 2c; 02a, 2b, 2c, 2d, 2e; 02a, 2b, 2d, 2e, 2g; 02a, 2c, 2e; 06a, 6b; 06a, 6c. No pronounced correlation between the partial composition of O-antigens within the O-group and the presence of different K- or H-antigens in the strain has been established. The scheme of adsorption has been developed and anti-Escherichia factor O-sera have been obtained. The use of these sera allows one to differentiate Escherichia strains, isolated from patients, within the O-groups according to the partial composition of their antigens, which is of both diagnostic and epidemiological importance.

A total of 373 E. coli strains, isolated in urinary tract infections from children and adults in Rostock (GDR) and Moscow (USSR), were found to belong to 63 O-groups. E. coli isolated in Rostock belonged to 36 O-groups, and in Moscow to 53 O-groups. The occurrence of E. coli serogroups in the dominating O-groups (more than 2% of the total number of strains) in Rostock (O1, O2, O4, O6, O7, O8, O9, O15, O21, O25, O75, O102) differed from that in Moscow (O1, O2, O4, O6, O8, O9, O15, O16, O17, O18, O25, O28, O73, O75) in composition and frequency, in Rostock the predominant E. coli groups being O2, O4, O6, O7, O9, O21, O75, O102, while in Moscow O1, O8, O15, O16, O17, O18, O25, O28, O73. The partial O-antigen composition of E. coli strains belonging to the same O-groups, isolated in Rostock and Moscow, was different in groups O1, O2, O125.

Algorhythm and a program for identification of bacteria of the Enterobacteriaceae family, based on Edwards and Ewing's diagnostic scheme, were worked out. Use of this program permitted to analyze different sets of abbreviated biochemical tests. To determine the genera and species of enterobacteria a minimal set of 11 tests is suggested, including indol formation, Voges-Proskauer's reaction, the presence of urease enzymes, gelatinase, lysine decraboxylase, phenylalanine deaminase, glucose fermentation (gas), or lactose, inosite, sorbit, arabinose, rhamnose. The program admits increase of both the biochemical tests, and toxonomic groups of bacteria, this permitting to consider several families. The presence of strains deviating by properties from this scheme points to the necessity of further improvement of diagnostic schemes for the enterobacteria identification.

Study of biochemical properties of 421 strains of enterobacteria permitted to refer them to the following taxonomic groups of which E. coli constituted 83,4 +/- 3.6%, Enterobactercloacae--97 +/- 3.0%, Klebsiella 4.0 +/- 2.0%. Enterobacter aerogenes, Enterobacter hafnia, Citrobacter, Proteus rettgeri, Proteus vulgaris were revealed in individual cases (0.2-1.0%). As a result of serological typing there were revealed 105 serological types of Escherichia differing by 55 O-antigen variants, 26 H-antigen variants, and the presence or absence of K-antigen. The following groups occurred most frequently-01, 02, 04, 06, 08, 09, 015, 016, 017, 018, 025, 028, and 075. There were also revealed new 0154 and 0155 groups. In 30.9 + 5,3% of cases Escherichia did not belong to the known 157 O-groups; among them 87.1 +7,2% of the strains had H-antigens which could not be referred to any of the known variants according to the international designation.

Anticoli (anti-Escherichia) plasma has been obtained from donors immunized with viable Escherichia oral vaccines, prepared from Str-d mutants of E. coli. Anticoli plasma has an therapeutic effect against diseases of Escherichia etiology.
