RESUMO

The study was undertaken to optimize the well known immunoassay of antibodies to chemical carcinogens, designed by J. Chagnaud et al., and examine the specific features of the generation of antibodies (Ab) to benzo[a]pyrene (BP) in patients with lung cancer. Ninety-two serum samples from healthy males and 80 samples from patients with lung cancer were studied. Increased efficiency was achieved by the use of bovine serum albumin instead of thyreoglobulin in the synthesis of carrier protein conjugates with BP. Better reproducibility was accomplished, by using the control serum sample with the known content of IgM, IgG, and IgA Ab to BP. Analysis of the study serum samples has established that in healthy male smokers, the count of IgA Ab was twice greater than that in non-smokers. The patients with lung cancer were found to higher levels of IgG and IgA Ab to BP (in both smokers and non-smokers) than healthy individuals. It is proposed to use the stated method to study a role of A to chemical carcinogens in the pathogenesis of malignancies.

AIM: To develop experimental model for definition of immunological images of chemical carcinogens. MATERIALS AND METHODS: The conjugates of benzo[a]pyrene, benz[a]anthracene, anthracene, chrysene and pyrene with bovine serum albumin and yeast hexokinase were synthesized. Rabbits were immunized by bovine serum albumin-hapten conjugates. Antibodies to each hapten were isolated from the serum by affinity chromatography with the hapten-yeast hexokinase-Sepharose sorbents. The binding of each hapten with each antibody was determined by competitive immunoassay. RESULTS: The immunological images of all the investigated chemical compounds were described. CONCLUSION: The model is proposed to determine the internal immunological images of anti-idiotypic monoclonal antibodies to chemical carcinogens.

Some interrelations between generation of antibodies to polycyclic aromatic hydrocarbons and immunogenic capacity of carcinoembryonic antigen were studied in patients with mammary and gastrointestinal types of cancer. IgM, IgG and IgA antibodies to polycyclic aromatic hydrocarbons were determined by a method of enzyme-linked immunosorbent assay. Immunogenic activity of carcinoembryonic antigen was evaluated by its ability to form circulating immune complexes. It was found that in patients with mammary cancer, generations of IgA to polycyclic aromatic hydrocarbons was interrelated with high immunogenic form of carcinoembryonic antigen, whereas IgG synthesis was associated with its low immunogenic form. Alternatively, patients with gastro-intestinal cancer did not exhibit any appreciable interrelation between generation of immunoglobulins and immunogenic activity of carcinoembryonic antigen.

Different classes of antibodies to polycyclic aromatic hydrocarbons (PAH) and immunogenicity of carcinoembryonic antigen (CEA) were investigated in breast cancer patients versus course. Prior to treatment, the levels of IgM, IgG and IgA antibodies to PAHs were 5.9, 7.7 and 32.5%, respectively. In cases of tumor regression, they were 13.8, 25.0 and 19.2%, while in those of progression-2.4, 2.8 and 12.8%, respectively. The immunogenicity of CEA, i.e. the power of forming circulating immune complexes, appeared to be higher in cases of tumor regression than in those of progression. CEA concentration in immune complexes exceeding 5ng/ml was observed in 24.0 and 15.1% of cases, respectively, while in whole serum, the concentration in excess of 10 ng/ml was in 8.0 and 42.4%, respectively.
