RESUMO

Methods of determination in the histological sections of the volumetric fraction of the tissue components, characterized by low content (less than 1%), uneven distribution and wide size variation are described. The measuring of the volumetric part of such a structure is carried out by two stages: in the first stage the true area of the whole section is estimated under the low magnification; in the second stage the same is made for the true area of the structure investigated. Then the volumetric density of the component in question is determined by the ratio of the area of investigated structure and that of the whole section. By this method it is identified that the normal volumetric part of the insular apparatus of the rat pancreas is on the average 0.46% in the duodenal part and 0.57% in the spleen part.

RESUMO

It has been substantiated that the use of a new synthetic drug tageflar analogous to L-enkephalin was accompanied by the depression in pancreatic enzyme activity in the blood of rats, acceleration of necrotic and lytic processes in irreversibly damaged exocrine pancreocytes in the course of experimental pancreatitis. The drug promoted microcirculation retention in the injury zone and acceleration of the arrangement and elimination of necrotic glandular tissue. The influence of tagaflar diminished the extent of perifocal necrobiotic zone and suppressed sclerotic processes and pancreatic lipomatosis.

RESUMO

The necrotic and inflammatory processes in the albino rats pancreas were studied by histological, immunomorphological and electron microscopic techniques after freezing of the pancreas. Early morphological sings of acinar cells destruction were revealed. The mechanism of secret "leakage" from zimogen granules into acinar cell cytoplasm was shown. The stages of pathologic process corresponding to different types of pancreatitis were established. The inflammatory reaction formed in the stage of parenchymal necrosis was characterised by an early activation of fibroblastic and macrophagal cells and by the delay of necrotic tissue elimination with their following calcification. The chronic pancreatitis developed on the immune basis in the intact parts of the pancreas.

Electron microscopy of the pancreas has shown after focal ethyl chloride cooling that reparative processes spreading from the borders to the epicenter of the affected parts involved (1) intracellular regeneration of partially damaged acinar cells, (2) proliferation of the epithelium of unaffected ducts, (3) proliferation of fibroblasts and capillary endothelium to form a fibrous capsule around the focus of necrosis. After sequestration of the focus of necrosis the partially damaged acinar cells of the border area underwent amitosis without cytotomy followed by the recovery of lost organelles. No acinar cells were observed to occur. Canalicular proliferates became differentiated mainly in islet cells. However, the main mass of the canalicular acinar derivatives of the border area experienced atrophy and cicatrization to form a fibrous capsule around the focus of necrosis. The compensation of the lost part of the excretory parenchyma was preceded by hypertrophy of acinar cells of undamaged segments of the pancreas.

Electron microscopy of acinar cell (AC) of the pancreas cooled to -30 degrees C with subsequent thawing has disclosed the destruction of intracellular membranes, damage to the cell organelles followed by diffusion of the zymogen granule contents to the hyaloplasm and interstice. These changes corresponded to the phase of paranecrosis (cryogenic shock of the cell), whereas the AC lost its ability to autophagocytosis. Irreversible cryodestruction provoked complete cell necrosis during 3-6 hours. The less cold-affected cells were subsequently destroyed because of additional injurious action of enzymatic cytolysis and circulatory hypoxia.
