RESUMO

Hydroxyapatite-derived eggshell biowaste (Hap-Esb) has been fabricated and developed for the electrochemical detection of uric acid (UA). The physicochemical characteristics of the Hap-Esb and modified electrodes were evaluated using a scanning electron microscope and X-ray Diffraction analysis. Utilized as UA sensors, the electrochemical behavior of modified electrodes (Hap-Esb/ZnONPs/ACE) was assessed using cyclic voltammetry (CV). The superior peak current response observed for the oxidation of UA at Hap-Esb/ZnONPs/ACE, which was 13 times higher than that of the Hap-Esb/activated carbon electrode (Hap-Esb/ACE) is attributed to the simple immobilization of Hap-Esb on zinc oxide nanoparticle-modified ACE. The UA sensor exhibited a linear range at 0.01 to 1 µM, low detection limit (0.0086 µM), and excellent stability, which surpass the existing Hap-based electrodes reported in the literature. The facile UA sensor subsequently realized is also advantaged by its simplicity, repeatability, reproducibility, and low cost, applicable for real sample analysis (human urine sample).

Sea cucumbers are marine organisms with uses in food, cosmetics, and medicine. This study aimed to identify Indonesian sea cucumbers with high antioxidant and antibacterial activities. Twenty-one sea cucumber species were used for this study. Antioxidant capacity was evaluated using the 2,2-diphenyl-ß-picrylhydrazine assay. Antibacterial activity was assessed using the disk diffusion assay, whereas the resazurin-based assay was employed to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Volatile compounds possibly related to the biological activity of sea cucumbers were analyzed via gas chromatography-mass spectrometry (GC-MS). Holothuria atra had the strongest antioxidant capacity (IC50 = 14.22 ± 0.87 µg µL-1). Stichopus vastus displayed the best antibacterial activity against Staphylococcus aureus, whereas Stichopus ocellatus extract was most potent against Vibrio cholerae. Holothuria albiventer, which controlled Bacillus subtilis most effectively while also being active against S. aureus and V. cholerae, was the optimal antimicrobial species. H. albiventer and Actinopyga echinites inhibited B. subtilis growth at 12.5 µg µL-1. The MBC tests indicated that the antibacterial activities of sea cucumbers at the MIC were bacteriostatic, rather than bactericidal, in nature. GC-MS analysis uncovered long-chain fatty acids that might be associated with the antibacterial activities of sea cucumbers.

We identified the main active, exercise performance-enhancing compounds in a hot water extract of the leather carp, Cyprinus carpio nudus, as nicotinamide and guanosine. Mice were fed casein (30 mg/ml) enriched with nicotinamide (0.1 mg/ml) and guanosine (0.05 mg/ml) once daily for a week at 10 µl/g body weight. Swimming endurance (57%) and forelimb grip strength (21%) were increased significantly. The diet had little effect on body weight. After the swimming exercise, the blood glucose and superoxide dismutase levels were significantly higher (137% and 131%, respectively) than in the saline controls. The blood lactate level was 90% of that in the controls. The estimated amount of nicotinamide in the carp fillet was 26.2 mg/kg. These results suggest that the triple combination of casein with nicotinamide and guanosine improves exercise performance and delays the onset of fatigue, supporting the traditional use of carp extract in healthcare as a tonic soup. PRACTICAL APPLICATIONS: The triple-combination of casein (30 mg/ml) + nicotinamide (0.1 mg/ml) + guanosine (0.05 mg/ml) significantly enhanced the exercise performance and anti-fatigue in mice, supporting the traditional use of carp extract in healthcare as a tonic soup.
