RESUMO

OBJECTIVE: We investigated the effects of conscious sedation by 30% nitrous oxide inhalation on the cardiac autonomic nervous system in the absence or presence of music listening. STUDY DESIGN: The power spectral analysis of heart rate variability (HRV) and cardiovascular parameters were measured in 26 healthy volunteers, who were divided into the control and N(2)O groups. RESULTS: Nitrous oxide inhalation attenuated the increase in relative low frequency (LF) power in the control group, but relative high frequency (HF) power remained constant compared to a decreased value in the control group. Nitrous oxide inhibited the increase in the LF/HF ratio. These autonomic balance shifts were observed without hemodynamic changes. Additional music during sedation did not alter HRV variables. CONCLUSIONS: Inhalational sedation with nitrous oxide shifted cardiac sympathetic-parasympathetic balance toward a parasympathetic dominance via the suppression of sympathetic activity. Music listening during sedation did not provide further relaxing effects.

The characteristics of hydrogen bond formation between trehalose and polyvinylpyrrolidone (PVP) in amorphous mixtures at different hydration states were quantitatively investigated. Amorphous trehalose-PVP mixtures were prepared by freeze-drying and equilibrated at different relative humidities (RH). Infrared (IR) spectra of the trehalose-PVP mixtures were obtained by Fourier transform IR spectroscopy,(FTIR) and the IR band corresponding to C=O groups of PVP was deconvolved into the component bands responsible for C=O groups that were free and restricted by hydrogen bonds, to estimate the degree of the trehalose-PVP interactions. The FTIR analysis indicated that approximately 80% of the C=O groups of PVP formed hydrogen bonds with trehalose in the presence of more than 3 g of trehalose per gramme of PVP, independent of the RH. IR analysis of the O--H stretching vibration of the sugar demonstrated that the presence of PVP lead to an increase in the free hydroxyl groups of trehalose that did not form hydrogen bonds at RH 0%. On the other hand, the water sorption behavior of the trehalose-PVP mixtures suggested that rehumidification diminished the effect of PVP on increasing the free OH groups. Thus a peculiar relationship may exist between Tg, RH and the composition of the mixture: The presence of PVP increased Tg at RHs 0 and above 23% but decreased Tg at 11%.

Fetal liver (FL) hematopoiesis is thought to be important for expanding the cell number during ontogeny. In order to investigate the cellular interaction molecules among FL stromal and hematopoietic cells, we established a monoclonal antibody, Ndk-10, that reacts with FL stromal cells but not with dish non-adherent cells. When Ndk-10 was added to an FL stromal and hematopoietic cell-coculture, it inhibited the survival of c-kit+ cells. The inhibitory activity of Ndk-10 was also observed in the fetal liver organ culture. The Ndk-10 recognized a 150 kD molecule in the adherent cells of FL and kidney, and the N-terminal amino acid sequence was identical to that of mouse aminopeptidase N/CD13. The peptidase activity of CD13 was inhibited by Ndk-10, and addition of its specific inhibitor resulted in the same inhibitory activity as Ndk-10. We propose that aminopeptidase N/CD13 is a critical molecule that regulates the survival of c-kit+ cells in the FL microenvironment.
