RESUMO

In French Polynesia, respiratory tract clinical isolate M26, displayed unusual phenotype and contradictory phylogenetic affiliations, suggesting a hitherto unidentified rapidly-growing Mycobacterium species. The phenotype of strain M26 was further characterized and its genome sequenced. Strain M26 genome consists in a 5,732,017-bp circular chromosome with a G + C% of 67.54%, comprising 5,500 protein-coding genes and 52 RNA genes (including two copies of the 16 S rRNA gene). One region coding for a putative prophage was also predicted. An intriguing characteristic of strain M26's genome is the large number of genes encoding polyketide synthases and nonribosomal peptide synthases. Phylogenomic analysis showed that strain M26's genome is closest to the Mycobacterium phlei genome with a 76.6% average nucleotide identity. Comparative genomics of 33 Mycobacterium genomes yielded 361 genes unique to M26 strain which functional annotation revealed 84.21% of unknown function and 3.88% encoding lipid transport and metabolism; while 48.87% of genes absent in M26 strain have unknown function, 9.5% are implicated in transcription and 19% are implicated in transport and metabolism. Strain M26's unique phenotypic and genomic characteristics indicate it is representative of a new species named "Mycobacterium massilipolynesiensis". Looking for mycobacteria in remote areas allows for the discovery of new Mycobacterium species.

Cometary nuclei consist mostly of dust and water ice. Previous observations have found nuclei to be low-density and highly porous bodies, but have only moderately constrained the range of allowed densities because of the measurement uncertainties. Here we report the precise mass, bulk density, porosity and internal structure of the nucleus of comet 67P/Churyumov-Gerasimenko on the basis of its gravity field. The mass and gravity field are derived from measured spacecraft velocity perturbations at fly-by distances between 10 and 100 kilometres. The gravitational point mass is GM = 666.2 ± 0.2 cubic metres per second squared, giving a mass M = (9,982 ± 3) × 10(9) kilograms. Together with the current estimate of the volume of the nucleus, the average bulk density of the nucleus is 533 ± 6 kilograms per cubic metre. The nucleus appears to be a low-density, highly porous (72-74 per cent) dusty body, similar to that of comet 9P/Tempel 1. The most likely composition mix has approximately four times more dust than ice by mass and two times more dust than ice by volume. We conclude that the interior of the nucleus is homogeneous and constant in density on a global scale without large voids. The high porosity seems to be an inherent property of the nucleus material.

The laboratory diagnosis of tuberculosis usually relies on culture-based isolation of the causative Mycobacterium tuberculosis bacteria. We developed and evaluated the performance of MOD9, a new blood-free derivative of the MOD4 solid medium on which we previously reported for the isolation and culture of mycobacteria. First, inoculation of Lowenstein-Jensen medium with 21 M. tuberculosis isolates at 10(5), 10(3), and 10 CFU yielded colonies in 5.7 ± 1.5 days, 7.6 ± 0.8 days, and 10.8 ± 1.7 days versus 1.5 ± 0.4 days, 3.5 ± 0.6 days, and 4.9 ± 1 days in MOD9 (P < 0.05, Student's t test). Further, the time to detectable growth of M. tuberculosis was measured on MOD9 and Lowenstein-Jensen media after duplicate inoculation of 250 clinical specimens decontaminated with 0.7% chlorhexidine. The contamination rate was 1.6% (4/250) on MOD9 versus 4.4% (11/250) on Lowenstein-Jensen medium (P = 0.11, Fisher's exact test). Chlorhexidine-MOD9 yielded 38/250 (15.2%) isolates versus 32/250 (12.8%) isolates for the chlorhexidine-LJ (P = 0.5195, Fisher's exact test). All together, eight M. tuberculosis isolates were cultured solely from chlorhexidine-MOD9, and two M. tuberculosis isolates were cultured solely from chlorhexidine-LJ. The time to detection was 9.8 ± 3.9 (range, 5 to 18) days for chlorhexidine-MOD9 versus 17.4 ± 5.9 (range, 10 to 35) days for chlorhexidine-LJ (P < 0.05, Student's t test). These data indicate the superiority of the MOD9 medium over the standard LJ medium following chlorhexidine decontamination for the recovery of M. tuberculosis.

The small and active Saturnian moon Enceladus is one of the primary targets of the Cassini mission. We determined the quadrupole gravity field of Enceladus and its hemispherical asymmetry using Doppler data from three spacecraft flybys. Our results indicate the presence of a negative mass anomaly in the south-polar region, largely compensated by a positive subsurface anomaly compatible with the presence of a regional subsurface sea at depths of 30 to 40 kilometers and extending up to south latitudes of about 50°. The estimated values for the largest quadrupole harmonic coefficients (10(6)J2 = 5435.2 ± 34.9, 10(6)C22 = 1549.8 ± 15.6, 1σ) and their ratio (J2/C22 = 3.51 ± 0.05) indicate that the body deviates mildly from hydrostatic equilibrium. The moment of inertia is around 0.335MR(2), where M is the mass and R is the radius, suggesting a differentiated body with a low-density core.

A micropropagação é uma técnica muitas vezes indicada para a multiplicação em larga escala de plantas com propriedades medicinais. Dentre elas, destaca-se a hortelã-pimenta (Mentha x Piperita L.), cujo óleo essencial é utilizado no tratamento de transtornos digestivos e respiratórios. Para otimizar o protocolo de micropropagação dessa espécie são necessários estudos, principalmente quanto à suplementação do meio de cultura para garantir a produção massal in vitro e posterior extração do óleo essencial. Nesse contexto, objetivou-se avaliar o efeito de concentrações e combinações de reguladores de crescimento vegetal na morfogênese in vitro de hortelã-pimenta. Segmentos nodais provenientes de plântulas estabelecidas in vitro foram utilizados como fonte de explante e inoculados em meio de cultura MS suplementado com 0; 2,0 e 4,0 mg L-1 de BAP (6-benzilaminopurina), 0; 0,5 e 1,5 mg L-1 de ANA (ácido naftaleno-acético) e 0; 0,5 e 1,0 mg L-1 de GA3 (ácido giberélico). O delineamento experimental adotado foi inteiramente casualizado, com os tratamentos distribuídos em esquema fatorial 3x3x3 com oito repetições. Concluiu-se que o BAP favoreceu a sobrevivência de segmentos nodais de M. x Piperita inoculados in vitro e, quando combinado ao GA3, promoveu a brotação dos explantes. Essas características, no entanto, não foram estimuladas pela adição de ANA ao meio de cultura. Conclui-se que após a multiplicação dos brotos in vitro estes devem ser transferidos para meio sem reguladores para seu desenvolvimento. Apesar dos efeitos benéficos do BAP na organogênese de M. x Piperita, elevadas concentrações deste regulador de crescimento vegetal promoveram a formação de calos.


Micropropagation is a technique used for the large-scale production of medicinal plants. Among them, peppermint (Mentha x piperita L.) may be mentioned because of the pharmacological importance of its essential oil, which is used on the treatment of digestive and respiratory disorders. Studies are needed in order to optimize the micropropagation protocol of this species, especially concerning the culture medium, to ensure the in vitro mass clonal production and to enable the future extraction of the plant essential oil. Therefore, this work aimed to evaluate the effects of concentrations and combinations of different plant growth regulators on the in vitro morphogenesis of peppermint. Nodal segments from plantlets already established in vitro were used as explants and inoculated on MS medium supplemented with 0, 2.0 and 4.0 mg L-1 of BAP (6-benzylaminopurine), 0, 0.5 and 1.5 mg L-1 of NAA (naphthalene acetic acid) and 0; 0.5 and 1.0 mg L-1 of GA3 (gibberellic acid). The experiment was in a completely randomized design, set up as a 3x3x3 factorial design with eight replicates. We concluded that BAP increases the survival rate of in vitro inoculated nodal segments of M. x piperita. In addition, its combination with GA3 stimulates explants shooting. Those aspects, however, are not promoted by the addition of NAA into the culture medium. Also, the results indicate that, after in vitro multiplication, peppermint shoots must be transferred to another medium without plant growth regulators for shoot elongation. High concentrations of BAP promote calli induction, despite having beneficial effects on the organogenesis of M. piperita.

O presente trabalho teve por objetivo estabelecer in vitro a espécie Lippia alba (Mill.) N. E. Brown e promover a aclimatização de mudas dessa espécie. Para isso, foi testada a influência de diferentes concentrações e tempos de imersão em hipoclorito de sódio na assepsia dos explantes. Segmentos nodais foram imersos em hipoclorito de sódio nas concentrações 0,4; 0,6; 0,8 e 1,0 % e nos tempos 8, 12 e 16 minutos. Após 30 dias avaliou-se a contaminação bacteriana (%), número de brotos, número de folhas, e a taxa de sobrevivência (%). A concentração de 1% de hipoclorito de sódio foi a mais eficiente no controle da contaminação. Ápices caulinares de L. alba foram estabelecidos in vitro em meio MS suplementado com diferentes doses de BAP (0; 0,5; 1,0; 1,5 mg L-1). Após 140 dias avaliou-se a contaminação (%), a taxa de sobrevivência (%), a oxidação (%) e o número de brotos. Os melhores resultados foram obtidos para a dose 1,5 mg L-1 deste regulador. Para a aclimatização foram testados quatro tipos de substratos: pó de coco + calcário (1 g L-1), Plantmax® + calcário (1 g L-1), vermiculita + calcário (1 g L-1) e pó de coco + Plantmax® + vermiculita (1:1:1) + calcário (1 g L-1). Avaliou-se a taxa de sobrevivência (%), comprimento da parte aérea (cm), comprimento da raiz (cm), massa fresca da parte aérea e da raiz (g), e a massa seca da parte aérea e da raiz (g). Os melhores resultados foram obtidos quando com o substrato comercial Plantmax®.


The present study aimed to establish the species Lippia alba (Mill.) N. E. Brown in vitro and promote the acclimatization of the seedlings of this species. Therefore, we tested the effect of different concentrations and immersiog times in sodium hypochloritetfor the disinfection of explants. Nodal segments were immersed in sodium hypochlorite at the different concentrations of ,.4; ,.6; ,.8 and ,.0% during the different times of 8, 12 and 16 minutes. After 30 days, bacterial contamination (%), number of shoots, number of leaves and survival rate (%) were evaluated. The 1% concentration of sodium hypochlorite was more effective in controlling contamination. Shoot apices of L. alba were established in vitro on MS medium supplemented with different concentrations of BAP (;, ,.5; ,.0; ,.5 mg L-1). After 140 days, we evaluated the contamination (%), survival rate (%), oxidation (%) and number of shoots. The best results were obtained when we used the dose of ,.5 mg L-1 of this regulator. For acclimatization, we tested four types of substrates: coconut powder + lime (1 g L-1), Plantmax® + lime (1 g L-1), vermiculite + lime (1 g L-1) and coconut powder + Plantmax® + vermiculite (1:1:1) + lime (1 g L-1). We evaluated the survival rate (%), shoot length (cm), root length (cm), fresh weight of shoot and root (g) and dry weight of shoot and root (g). The best results were obtained when using the substrate Plantmax®.

The Dawn spacecraft targeted 4 Vesta, believed to be a remnant intact protoplanet from the earliest epoch of solar system formation, based on analyses of howardite-eucrite-diogenite (HED) meteorites that indicate a differentiated parent body. Dawn observations reveal a giant basin at Vesta's south pole, whose excavation was sufficient to produce Vesta-family asteroids (Vestoids) and HED meteorites. The spatially resolved mineralogy of the surface reflects the composition of the HED meteorites, confirming the formation of Vesta's crust by melting of a chondritic parent body. Vesta's mass, volume, and gravitational field are consistent with a core having an average radius of 107 to 113 kilometers, indicating sufficient internal melting to segregate iron. Dawn's results confirm predictions that Vesta differentiated and support its identification as the parent body of the HEDs.

Lippia alba é uma planta de origem brasileira, pertencente à família Verbenaceae. É conhecida por combater a insônia e a asma, além de possuir ação anticonvulsivante e antifúngica. A micropropagação tem sido utilizada para multiplicação de várias espécies com propriedades medicinais, e o BAP é a citocinina sintética mais utilizada para a multiplicação in vitro. O objetivo deste trabalho foi estabelecer a concentração adequada de BAP na multiplicação in vitro de L. alba. Explantes constituídos de segmentos nodais provenientes de plântulas já estabelecidas in vitro com aproximadamente 0,5 cm foram inoculados em meio MS, suplementado com 0,0; 0,5; 1,0 e 1,5 mg L-1 de BAP em adição de 30 g L-1 de sacarose. A utilização de 1,5 mg L-1 de BAP promove a multiplicação in vitro de L. alba. A ausência deste regulador propicia maior número de folhas e o uso na concentração de 0,5 mg L-1 aumentou as massas fresca e seca de parte aérea nesta espécie.


Lippia alba is a Brazilian plant, belonging to the Verbenaceae family. It is known to combat insomnia and asthma, also has antifungal and anticonvulsant actions. Micropropagation has been used for propagation of various species with medicinal properties, and BAP is the most widely used synthetic cytokinin for in vitro multiplication. The aim of this study was to determine the appropriate concentration of BAP on in vitro multiplication of L. alba. Explants consisting of nodal segments from seedlings already established in vitro with approximately 0.5 cm were inoculated on MS medium supplemented with 0; 0.5; 1.0 and 1.5 mg L-1 of BAP and 30 g L-1 of sucrose. The use of 1.5 mg L-1 of BAP promotes the in vitro multiplication of L. alba. The absence of this regulator provides a greater number of leaves and its use at the concentration of 0.5 mg L-1 increased shoots' fresh and dry weights in this specie.

Asteroid 21 Lutetia was approached by the Rosetta spacecraft on 10 July 2010. The additional Doppler shift of the spacecraft radio signals imposed by 21 Lutetia's gravitational perturbation on the flyby trajectory were used to determine the mass of the asteroid. Calibrating and correcting for all Doppler contributions not associated with Lutetia, a least-squares fit to the residual frequency observations from 4 hours before to 6 hours after closest approach yields a mass of (1.700 ± 0.017) × 10(18) kilograms. Using the volume model of Lutetia determined by the Rosetta Optical, Spectroscopic, and Infrared Remote Imaging System (OSIRIS) camera, the bulk density, an important parameter for clues to its composition and interior, is (3.4 ± 0.3) × 10(3) kilograms per cubic meter.

O hortelã-pimenta é um hibrido triplo, Mentha x piperita, utilizado de forma medicinal no tratamento de náuseas, cólicas gastrointestinais, flatulências, cálculos biliares, icterícia, ansiedade, expectoração e expulsão de vermes intestinais. Na micropropagação, o BAP (6-benzilaminopurina) e KIN (cinetina) têm sido as fontes de citocininas mais empregadas. O objetivo deste trabalho foi avaliar o efeito de reguladores vegetais na multiplicação in vitro de hortelã-pimenta. Explantes constituídos de segmentos nodais provenientes de plântulas já estabelecidas in vitro com aproximadamente 0,5 cm foram inoculados em meio MS, suplementado com diferentes associações de BAP e KIN, em adição de 30 g L-1 de sacarose. As concentrações de citocininas utilizadas foram 0,0; 1,0 e 2,0 mg L-1 de BAP e 0,0; 0,5 e 1,0 mg L-1 de KIN. O delineamento experimental foi o inteiramente casualizado, com nove tratamentos consistindo de cinco frascos cada um, sendo que cada frasco continha quatro explantes. A utilização de 2,0 mg L-1 de BAP promove a multiplicação in vitro de hortelã-pimenta, porém, diminui o índice de sobrevivência. O uso de citocininas aumenta a massa fresca e seca dos explantes e a ausência destes reguladores propicia o alongamento nesta espécie.


Peppermint is used for medical treatments of nausea, gastrointestinal cramps, flatulence, gallstones, jaundice, anxiety, sputum and expulsion of intestinal worms. In micropropagation, BAP (6-benzylaminopurine) and KIN (kinetin) have been the most widely used sources of cytokinins. The aim of this study was to evaluate the effect of plant growth regulators in multiplication of peppermint. Explants consisting of nodal segments from seedlings already established in vitro with approximately 0,5 cm were inoculated on MS medium supplemented with different combinations of BAP and KIN and 30 g L-1 of sucrose. The concentrations of cytokinins used were 0,0; 1,0 and 2,0 mg L-1 of BAP and 0,0; 0,5 and 1,0 mg L-1 of KIN. The experiment was set up in a completely randomized design with nine treatments consisting of five bottles each, and each vial contained four explants. The use of 2,0 mg L-1 of BAP promotes the in vitro multiplication of peppermint, but decreases the survival rate. Cytokinins increase fresh and dry weight of explants and the absence of these regulators provides the elongation in this specie.

The atmosphere and ionosphere of Venus have been studied in the past by spacecraft with remote sensing or in situ techniques. These early missions, however, have left us with questions about, for example, the atmospheric structure in the transition region from the upper troposphere to the lower mesosphere (50-90 km) and the remarkably variable structure of the ionosphere. Observations become increasingly difficult within and below the global cloud deck (<50 km altitude), where strong absorption greatly limits the available investigative spectrum to a few infrared windows and the radio range. Here we report radio-sounding results from the first Venus Express Radio Science (VeRa) occultation season. We determine the fine structure in temperatures at upper cloud-deck altitudes, detect a distinct day-night temperature difference in the southern middle atmosphere, and track day-to-day changes in Venus' ionosphere.

Hyperion is Saturn's largest known irregularly shaped satellite and the only moon observed to undergo chaotic rotation. Previous work has identified Hyperion's surface as distinct from other small icy objects but left the causes unsettled. Here we report high-resolution images that reveal a unique sponge-like appearance at scales of a few kilometres. Mapping shows a high surface density of relatively well-preserved craters two to ten kilometres across. We have also determined Hyperion's size and mass, and calculated the mean density as 544 +/- 50 kg m(-3), which indicates a porosity of >40 per cent. The high porosity may enhance preservation of craters by minimizing the amount of ejecta produced or retained, and accordingly may be the crucial factor in crafting this unusual surface.

One of Titan's most intriguing attributes is its copious but featureless atmosphere. The Voyager 1 fly-by and occultation in 1980 provided the first radial survey of Titan's atmospheric pressure and temperature and evidence for the presence of strong zonal winds. It was realized that the motion of an atmospheric probe could be used to study the winds, which led to the inclusion of the Doppler Wind Experiment on the Huygens probe. Here we report a high resolution vertical profile of Titan's winds, with an estimated accuracy of better than 1 m s(-1). The zonal winds were prograde during most of the atmospheric descent, providing in situ confirmation of superrotation on Titan. A layer with surprisingly slow wind, where the velocity decreased to near zero, was detected at altitudes between 60 and 100 km. Generally weak winds (approximately 1 m s(-1)) were seen in the lowest 5 km of descent.

Exposure to toxic gases and particles or dusts while working or living in confinement animal systems pose a pulmonary health hazard. The severity of lung impairment from exposure to such environment is investigated using intratracheal instillation, intratracheal nebulization, and inhalation procedures. Ability to deliver particles with intratracheal instillation that are evenly distributed throughout the lung depends on the material used for injection. Pulmonary histopathology reflects anatomic changes following inhalation or instillation of chemicals or particles. Endobronchial saline washings of bronchioles and alveoli allow measurement of markers of pulmonary inflammation such as total nucleated cell (leukocyte) counts and those of macrophages, neutrophils and lymphocytes; TNF-alpha, and collagen concentration are used to further evaluate pulmonary response to endotoxin or dust exposure. Alveolar epithelial cells have an important role in clearing pulmonary fluid and maintaining the structure of lung tissue. After repeated exposure, damage to epithelial cells may result in their death, causing edema and collagen deposition that may lead to fibrosis.

Radio signals from Ulysses were used to probe the lo plasma torus (IPT) shortly after the spacecraft's closest approach to Jupiter. The frequencies of the two downlinks at S-band (2.3 gigahertz) and X-band (8.4 gigahertz) were recorded, differenced, and integrated in order to derive the columnar electron density of the IPT. The measurements agree qualitatively with contemporary models of the IPT based on Voyager data, but significant differences are apparent as well. The overall level of the IPT electron density is approximately the same as the prediction, implying that the amount of gas (or plasma) injected from lo is similar to that observed during the Voyager era. On the other hand, the IPT seems to be less extended out of the centrifugal equator, implying a smaller plasma temperature than predicted.
