RESUMO

The aim of this study was to evaluate the survival of a probiotic microorganism microencapsulated in cellulose acetate phthalate, added to acerola nectar. The changes in pH, Brix, organic acid content and color of the product during its shelf-life were evaluated. A total of 3 processing runs were carried out on a semi-industrial scale, each consisting of a 15-liter batch of acerola nectar with added prebiotics and a microencapsulated probiotic culture. The physicochemical characteristics of the samples remained stable throughout storage. After 30 days storage the acerola nectar samples containing microencapsulated probiotic microorganisms exhibited counts above 8 log CFU per 200 mL, within the limits set by the Brazilian regulation for functional foods. RESUMO

The purpose of the study was to investigate the rabies virus multiplication in Vero cell cultures performed on porous microcarriers, MCs (cellulose-Cytopore and gelatin-Cultispher G), which provide higher available surface area compared with solid (nonporous) MCs (DEAE-Cytodex 1). In a set of experiments performed at the same MC concentration (MCs per milliliter), cell densities regularly obtained in porous MC cultures were comparable, but almost twice as high as those in solid MC cultures. In addition, 41.1 +/- 3.9-, 35.2 +/- 2-, and 19.6 +/- 5.8-fold increases in cell concentration, relative to the initial cell number, along with maximum rabies virus titers of 6.3 +/- 0.3 x 10(4), 5 +/- 0.1 x 10(4), and 4.3 +/- 0.2 x 10(4) FFD(50)/mL were observed in Cytopore, Cultispher G, and Cytodex 1 MC cultures, respectively. When higher concentrations of MCs were employed, lower performances of virus production and MC-cell occupation (cells per MC or cells per square millimeter) were observed. Cell attachment to MCs was shown to be faster for Cytopore MCs and Cytodex 1 MCs than for Cultispher G MCs. Concerning the kinetics of cell multiplication on MCs, exponential cell growth, at similar specific cell growth rates, took place on Cytopore, Cultispher G, and Cytodex 1 MCs. In addition, cell densities as high as 2.1 +/- 0.2 x 10(6) cells/mL on Cytopore MCs, 1.8 +/- 0.1 x 10(6) cells/mL on Cultispher G MCs, and 1 +/- 0.3 x 10(6) cells/mL on Cytodex 1 MCs were regularly obtained in batch cultures. Optical as well as scanning and transmission electron microscopy studies carried out to analyze MC structure, MC cell occupation, and cell permissivity to virus infection demonstrated that there was uniform cell distribution in the external and internal areas of the MCs, suggesting an efficiency of virus synthesis. Our results indicate the usefulness of these supports for rabies virus antigen production, as well as possibilities for further optimization.

OBJECTIVE: Porphyries are metabolic disorders of the haem biosynthetic pathway. They result from excessive accumulation of neurotoxic porphyrin precursors. Unlike the majority of Inherited Metabolic Diseases, the porphyries manifest after puberty, and their occurrence in childhood is very rare. PATIENT AND METHOD: The authors report a case of acute intermittent porphyria in an 8-year-old girl, reviewing the literature on etiopathogeny, diagnosis and treatment. COMMENTS: The authors emphasize to the importance of diagnosis in these patients, since the clinical expression may be lifethreatening and requires exposures to precipitating factors, so it may be prevented.

The acetylcholinesterase activity (AChE) in the human subfornical organ (SFO) was detected by the method of Koelle and Friedenwald in 16 human brains collected between 6 and 12 hrs postmortem. The only AchE-positive structures found were neuronal cell bodies and processes, morphologically classified as stellate and fusiform neurons of large, medium and small size. Large ones prevailed in the dorsal zone. The neurons were homogeneously distributed in the rostral area of the SFO. The penetration and ramification of large blood vessels produced a decreasing neuronal density in the medium-caudal area. The architecture of the SFO in sagittal sections comprised a central zone with neurons juxtaposed to the walls of the vascular plexus, whose dendritic and axonal processes showed an intricate pattern without a special arrangement. This neuronal zone of the SFO was surrounded by a peripheral layer of neurons with axonal projections to the rostral area. This layer was thicker in the dorsal zone of the SFO, where axonal fibers "climbing-up" from the central perivascular neurons could be demonstrated. In coronal sections some neurons with prolongations of arcuate distribution connected the dorsal and ventral zones.

One hundred and sixty seven cases of pituitary adenoma were analysed using the immunocytochemical method of the Avidin-Biotin Complex (ABC), described by Hsu et al. (1981). Six pituitary anti-hormones were utilized: anti-prolactin (aPRL) at a 1:1,500 dilution; anti-growth hormone (aHGH) at a 1:4,000 dilution: anti-adrenocorticotrophic hormone (aACTH) at a 1:3,000 dilution; anti-thyrothrophic hormone (aTSH) at a 1:3,000 dilution; anti-luteinizing hormone (aLH) at a 1:1,000 dilution; and a anti-follicle-stimulating hormone (aFSH) at a 1:300 dilution. Incubation period was 14 to 16 hours at 4 degrees C. The survey of clinical, laboratory and radiological data of cases of pituitary adenomas was performed after reading the stained slides using the immunocytochemical method. Of the 167 cases of pituitary adenomas, 136 (81.4%) disclosed a positive immunoreaction to one or more anti-hormones, and the positivity index of neoplastic cells varied from 1 to 90%. The immunoreaction was positive exclusively to one anti-hormone in 80 cases (58.8%) and to two or more anti-hormones in 56 cases, and the association most frequently found was between both aPRL and aHGH. The positivity to the immunoreaction was distributed as follows: -100 cases were positive for aPRL, exclusively in 4 cases; -65 cases were positive for aHGH, exclusively in 22 cases; -31 cases were positive for aACTH, exclusively in 8 cases; -5 cases were positive for aTSH, exclusively in one case; -one patient presented an adenoma positive to aLH and another patient to aFSH.

Foram analisados 167 casos de adenomas da hipófise pelo método imuno-histoquímico utilizando o Complexo da Avidina Biotina (ABC) descrito por HSU e col. (1981). Foram usados 6 anti-hormônios hipofisários: anti-prolactina (aPRL), na diluiçäo de 1:1.500, anti-hormônio do crescimento (aHGH), na diluiçäo de 1:4.000, anti-hormônio adrenocorticotrófico (aACTH), na diluiçäo de 1:3.000, anti-hormônio tireotrófico (aTSH), na diluiçäo de trófico (aACTH), na diluiçäo de 1:3.000, anti-hormônio tireotrófico (aTSH), na diluiçäo de 1:3.000, anti-hormônio luteinizante (aLH), na diluiçäo de 1:1.000, anti-hormônio folículo estimulante (a(FSH), na diluiçäo de 1;3.000. O período de incubaçäo foi de 14 a 16 horas a 4-C. Foi realizada também a coloraçäo pelo Orange G-PAS. O levantamento dos dados clínicos, laboratoriais, e radiológicos dos casos de adenomas de hipófise foi realizado após leitura das lâminas pelo método imuno-histoquímico. Dos 167 casos de adenomas da hipófise, 136 (81,4%) mostraram imuno-reaçäo positiva a um ou mais anti-hormônios, variando o índice de positividade entre 1 e 90% das células neoplásicas. A imuno-reaçäo foi positiva exclusivamente a um anti-hormônio em 80 casos (58,8%) e para dois ou mais anti-hormônios nos 56 casos restantes (41,2%), sendo a associaçäo mais freqüentemente encontrada aquela em que a positividade ocorreu para o aPRL e o aHGH. A positividade a reaçäo imuno-histoquímica distribuiu-se da seguinte forma: 100 casos foram positivos para o aPRL, em 49 pacientes de forma isolada; 65 casos foram positivos para o aHGH, em 22 pacientes de forma isolada; 31 casos foram positivos para o aACTH, em 8 pacientes de forma isolada; 5 casos foram positivos ao aTSH, em um paciente de forma isolada; um paciente apresentou adenoma positivo ao aLH; um caso foi positivo ao aFSH

RESUMO

The difficulty in obtaining cerebrospinal fluid (CFS) in an efficient and simple manner in rats prompts us to introduce a new technique that makes use of a cannula placed in the lateral ventricle. The cannula is implanted with a stereotaxic apparatus and the CSF is collected with a glass capillary tube. The technique has proved to work well for experiments in which the CSF must be free of blood. It also permits the collection of volumes of CSF sufficient for radioimmunoassays, and may be used in chronic experiments.

The authors review the literature and report a case that illustrate the large spectrum of clinical presentation of the anterior-sacral meningocele. It is analized the presently available diagnostic procedures and it is emphasized the importance of the plain film and mielography in the correct diagnosis of this lesion.

Among 20 patients with symptoms of cerebrovascular insufficiency, dissociation between memory disturbances and cognitive functions, and PEG studies, there were 13 cases showing atrophy of the hippocampus. The neurological and psychological studies included memory functions tests, IQ, and the analysis of language and praxia. Complementary tests included standard EEG records, EEGs obtained with nasopharingeal electrodes and PEG tomography with selective filling of both temporal horns, in order to demonstrate the inner and lower surface of the temporal lobes. The volume of the Ammon horn was reduced in 13 cases. Hippocampal atrophy was unilateral in 11 patients and bilateral in 2. The fact that hippocampal atrophy was demonstrated by PEG studies in 13 out of 20 cases, appears to be highly significant. This clinico-neuroradiological correlation seems to indicate that hippocampal atrophy, following ischemia in the deep distal vertebro-basilar-posterior cerebral territory, disturbs the function of axial structures (Papez circuit or limbic system) apparently leading to the disturbances of memory functions observed in this group of patients.
