RESUMO

BACKGROUND: The diagnostic criteria for alexithymia (DCA) was proposed by Fava et al. in 1995. The purpose of this study was to examine the criterion-related validity of the DCA in a general hospital psychiatric setting. METHODS: The subjects included 126 outpatients with psychiatric disturbance. The severity of alexithymic characteristics was assessed using the DCA and Beth Israel Hospital Psychosomatic Questionnaire (BIQ). Two Japanese versions of the 26-item Toronto Alexithymia Scale (TAS-26) and Minnesota Multiphasic. Personality Inventory-2 (MMPI-2) were administered. RESULTS: The DCA had relatively high sensitivity and specificity, indicating adequate discriminant validity. The discriminant validity of the DCA was also supported by significant associations of the DCA with the TAS-26 and MMPI-2. These results indicate that when used for screening, the DCA has adequate criterion-related validity. CONCLUSIONS: The DCA may be clinically useful in a general hospital psychiatric setting.

RESUMO

We determined the most suitable conditions for measuring the somatostatin (SRIF) level by brain microdialysis and investigated its release from the hypothalamus. The relative recovery rate of SRIF was 8.4 +/- 0.5% (mean +/- SE) using a polycarbonate (PC) membrane with the push-pull method at a flow rate of 2 microliters/min. Using tubes with an internal diameter of 0.28 mm and lengths of 5, 25, 50 and 100 cm, the relative recovery rates using a PC membrane with the push method were 8.2 +/- 0.5%, 7.3 +/- 0.6%, 6.2 +/- 0.5% and 4.1 +/- 0.6%, respectively. When using tubes with an internal diameter of 0.1 mm and lengths of 5, 25, 50 and 100 cm, the relative recovery rates were 7.3 +/- 0.7%, 5.6 +/- 1.0%, 3.5 +/- 1.1% and 1.4 +/- 0.7%, respectively. The relative recovery rate was 5.2 +/- 0.5% with a polysulfone (PS-F, Fresenius) membrane, 4.5 +/- 0.4% with a PS-H (Hospal) membrane, 2.6 +/- 0.2% with an ethylenevinyl alcohol membrane (EVAL), 5.1 +/- 0.8% with a polyvinyl alcohol (PVA) membrane and 10.4 +/- 0.8% with a PS-K (Kaneka) membrane. With the push method, the extracellular SRIF level in rat pituitary was 42.8 +/- 1.8 pg/ml with a PC membrane, 23.1 +/- 2.9 pg/ml with an EVAL membrane at a flow rate of 2 microliters/min. With the push-pull method, it was 52.7 +/- 5.2 pg/ml using a PC membrane, 33.5 +/- 2.8 pg/ml using a PVA membrane and 54.4 +/- 3.2 pg/ml using a PS-K membrane.(ABSTRACT TRUNCATED AT 250 WORDS)

When a crude extract of rat pineal glands (the 1000 x g supernatant of a homogenate) was incubated with arachidonic acid, 12-hydroxy-5,8,10,14-eicosatetraenoic acid was found as a major product. The 12-lipoxygenase of rat pineal gland also reacted with linoleic and alpha-linolenic acids at 35% and 101% the rate of arachidonate 12-oxygenation, respectively. Upon Western blot analysis using polyclonal antibody against porcine leukocyte 12-lipoxygenase, the cytosol fraction of rat pineal gland showed a positive band with a molecular weight of approx. 74 kDa. A full-length cDNA for this enzyme was cloned from a cDNA library of rat pineal gland and the identity of the 12-lipoxygenase cDNA was confirmed by its expression in E. coli. The amino acid sequence of the enzyme was deduced from the nucleotide sequence of the cDNA, encoding 663 amino acids with a calculated molecular weight of 75,305. The enzyme showed 72% identity of amino acid sequence with porcine leukocyte 12-lipoxygenase and 73% with bovine tracheal 12-lipoxygenase, but only 59% with human platelet 12-lipoxygenase. Taken together, the high reactivity with C-18 fatty acids, the immunoreactivity and the amino acid homology data indicate that the rat pineal 12-lipoxygenase is more closely related to leukocyte 12-lipoxygenase than to platelet 12-lipoxygenase. Upon RNA blot analysis, by far the highest content of 12-lipoxygenase mRNA was observed in the pineal gland and negligible amounts of mRNA were detected in other parts of the brain. The predominant presence of 12-lipoxygenase mRNA in pineal gland was confirmed by in situ hybridization of rat brain. Significant amounts of 12-lipoxygenase mRNA were also detected in rat spleen, aorta, lung and leukocytes.

The levels of striatal extracellular DA, DOPAC, HVA and 5-HIAA significantly decreased in aged rats compared to young adults. The magnitude of age-related reduction was greater in dopaminergic than serotonergic substances. The levels of striatal extracellular monoamines in young adult rats showed significant diurnal variations. On the other hand, these substances of aged rats, except for HVA, did not exhibit significant diurnal variations. Enriched environment housing increased levels of striatal extracellular DOPAC in aged rats, but did not influence on the age-related changes of diurnal variations in the striatal extracellular monoamines.

We observed 24 h changes of 'extracellular' indoleamines in pineal microdialysates taken from freely moving rats under a 12 h light/12 h dark cycle. Pineal indoleamines in microdialysates were analyzed by high-performance liquid chromatography with electrochemical detection. Consequently, they exhibited marked diurnal variations. The diurnal level in the extracellular 5-HT, higher for the light and lower for the dark, was parallel to that in 5-hydroxyindoleacetic acid and diametrically opposite to that in N-acetylserotonin (NAS). The release of 5-HT into the extracellular space transiently increased for the first 2 h after the dark onset and then became lower during the dark.

RESUMO

We describe a surgical technique to implant the guide cannula for in vivo microdialysis in the rat pineal gland. This technique has the following features and advantages: (a) does not require ligation of the superior or transverse sinus, (b) minimizes bleeding from the dural veins, (c) does not disturb the sympathetic innervation originating from superior cervical ganglia, which is essential for pineal function. This new technique makes it possible to carry out chronic pineal microdialysis of freely moving rats.

Using the "variance in statistics" as an index of electroencephalogram (EEG) parameters, we observed the cortico-hippocampal EEG rhythm under a 12:12-h light-dark condition in the rat with chronically implanted electrodes for EEG recording. The above EEG variance was simply measured in real time and on line through a personal computer. It corresponded to EEG slow wave activity and expressed the process of slow wave sleep as described in the two-process model by Borbély et al. Only in the dark phase, mean power spectral density of the EEG variance had a significant peak at about 1/100 cycles/min. This 100-min rhythmicity similar to the basic rest-activity cycle in human beings was observed in rats, particularly in the dark (active) phase for nocturnal animals. We propose that this ultradian 100-min rhythm is essential for the rat to maintain the waking state dominantly over the 12-h dark period.

RESUMO

Dynamic changes of melatonin in microdialysates from the pineal gland of a freely moving rat were repeatedly determined by using on-line high-performance liquid chromatography with electrochemical detection. The detection limit for melatonin, ca. 5 pg, was well below that achieved with other systems. We observed a drastic increase of extracellular pineal melatonin during the transitional phase from the light period to the dark period. This application of microdialysis is a useful tool in the study of the physiological role of the mammalian pineal body.
