RESUMO

Probiotics contribute to the integrity of the intestinal mucosa and preventing dysbiosis caused by opportunistic pathogens, such as intestinal helminths. Bacillus cereus GM obtained from Biovicerin® was cultured to obtain spores for in vivo evaluation on experimental schistosomiasis. The assay was performed for 90 days, where all animals were infected with 50 cercariae of Schistosoma mansoni on the 15th day. Three experimental groups were formed, as follows: G1-saline solution from the 1st until the 90th day; G2-B. cereus GM (105 spores in 300 µL of sterile saline) from the 1st until the 90th day; and G3-B. cereus GM 35th day (onset of oviposition) until the 90th day. G2 showed a significant reduction of 43.4% of total worms, 48.8% of female worms and 42.5% of eggs in the liver tissue. In G3, the reduction was 25.2%, 29.1%, and 44% of the total number of worms, female worms, and eggs in the liver tissue, respectively. G2 and G3 showed a 25% (p < 0.001) and 22% (p < 0.001) reduction in AST levels, respectively, but ALT levels did not change. ALP levels were reduced by 23% (p < 0.001) in the G2 group, but not in the G3. The average volume of granulomas reduced (p < 0.0001) 65.2% and 46.3% in the liver tissue and 83.0% and 53.2% in the intestine, respectively, in groups G2 and G3. Th1 profile cytokine (IFN-γ, TNF-α, and IL-6) and IL-17 were significantly increased (p < 0.001) stimulated with B. cereus GM in groups G2 and G3. IL-4 showed significant values when the stimulus was mediated by ConA. By modulating the immune response, B. cereus GM reduced the burden of worms, improved some markers of liver function, and reduced the granulomatous inflammatory reaction in mice infected with S. mansoni, especially when administered before infection.

The present study reports the anti-mycobacterial activity of 2-hydroxy-3-(3-methyl-2-butenyl)-1,4-naphthoquinone (lapachol) as well as its influence on macrophage functions. Lapachol (L) did not induce apoptosis/necrosis of THP-1 macrophages at ≤32 µg/mL. Mycobacterium avium liquid growth was arrested by ≥32 µg/mL and intra-macrophage proliferation by ≥16 µg/mL lapachol. The main immuno-modulatory effects of lapachol observed were an up-regulation of interferon-γ-receptor 1 (IFN-γR1) and major histocompatibility complex class II (MHCII) surface expression, and a marked inhibition of IL-10 secretion. Lapachol did not affect resting, IFN-γ- or toll-like receptor 2 (TLR2)-induced levels of oxygen and nitrogen metabolism key proteins nor the TLR2-mediated secretion of TNF-α, nor induced either oxidative or endoplasmic reticulum (ER) stress. Lapachol inhibited the surface expression of the co-stimulatory molecule CD86 but not that of CD80 and CD83. The results obtained indicate that the substituted naphthoquinone lapachol exhibits an anti-mycobacterial activity that is more efficient intra- than extra-cellularly, and exerts immuno-modulatory effects some of which may enhance the capacity of the host cell to control mycobacterial growth. The immune-modulatory action of lapachol could contribute to its more efficient intra-macrophage anti-mycobacterial activity.

The chemical composition and antimicrobial activity of the Cladia aggregata (Sw.) Nyl. were evaluated. Barbatic acid, depside obtained from C. aggregata, was spectroscopically analyzed and tested, as well as organic extracts. The extracts and purified substance were tested against four Staphylococcus aureus multi-resistant strains. The structure of barbatic acid was confirmed through NMR (¹H; 13C) and elemental ® analysis. Biochromatographic assays showed action of this compound, along with other substances contained in organic extracts, suggesting a synergic action, MIC assays placed barbatic acid in the same level of inhibition to other studied lichen substances.


A composição química e a atividade antimicrobiana da Cladia aggregata (Sw.) Nyl. foram avaliadas. O ácido barbático, um depsideo obtido da C. aggregata, foi espectroscopicamente analisado e testado, bem como o extrato orgânico. o extrato e a substância purificada foram testadas contra 4 linhagens de Staphylococcus aureus multi resistentes. A estrutura do ácido barbático foi confirmada através de ensaios de CCD, CLAE, IV, RMN (¹H;13C), UV e analise elementar (r). O biocromatograma mostrou a ação deste composto junto com outras substâncias contidas no extrato orgânico, sugerindo a ação sinérgica, a CMI do ácido barbático mostrou o mesmo nível de inibição de outras substâncias liquênicas.

RESUMO

Camundongos C57Bl/10 do sexo masculino, infectados com Schistosoma mansoni foram distribuídos nos grupos misto, profilático e curativo. Cultura de Zymomonas mobilis foi administrada oralmente aos camundongos. Uma proteção de 61% foi observada no grupo curativo (p<0,05). Os estudos histopatológicos dos fígados e intestinos mostraram resultados similares.

RESUMO

C57Bl/10 male mice infected with Schistosoma mansoni were distributed into mixed, prophylactic and curative groups. A culture of Zymomonas mobilis was orally administered to mice. A 61% protection from the infection was observed in the curative group (p <0.05). Histopathological study of the livers and intestines showed similar results.
