The main objective of this study was to verify genetic trends for milk production (MY305) and age at first calving (AFC). Were also considered levels of inbreeding practiced in the Brazilian dual-purpose Guzerá population (TPOP) comparing it with the same parameters estimated for two sub-populations derived from the reference (TPOP): female donors submitted to ovum-pick up (DPOP) and in vitro embryos produced (EPOP) between 2003 and 2013. Estimated breeding values (EBV) and inbreeding coefficients (F) were regressed by the year of birth (or year of in vitro fertilization) of each animal or embryo in order to obtain annual trends for these parameters separately for each of the three populations studied. A positive quadratic (ß2 = +0.000075) effect was detected for the F values in TPOP. Both DPOP and EPOP showed positive linear coefficients (ß1), respectively, +0.00084 (P < 0.001) and +0.00024 (P > 0.05). Annual mean F for EPOP was higher than TPOP and DPOP through the time series studied. The frequency of individuals with more than 7% F was higher in DPOP. Genetic trends for AFC were -0.187 days/year (Pâ¯>â¯0.05); -0.557 days/year (Pâ¯<â¯0.05) and -1.48 days/year (Pâ¯<â¯0.05), respectively for TPOP; DPOP and EPOP. Genetic trends for MY305 were +6.75 kg/year (P < 0.001); +8.2 kg/year (P < 0.001) and +10.5 kg/year (P < 0.05), respectively for TPOP; DPOP and EPOP. For both traits analyzed, EPOP showed the highest regression coefficients, which confirms a higher selection pressure and lower generation intervals previously expected from this sub-population. Results reported in the present study suggest that mean F is increasing in the Guzerá population. Efforts for controlling inbred mating on in vitro fertilization should be considered, as the presence of a bottleneck effect seems to be getting shape on DPOP and EPOP.

Genetic improvement, without control of inbreeding, can go to loss of genetic variability, reducing the potential for genetic gains in the domestic populations. The aim of this study was to analyze the population structure and the inbreeding depression in Campolina horses. Phenotype information from 43 465 individuals was analyzed, data provided by the Campolina Breeders Association. A pedigree file containing 107 951 horses was used to connected the phenotyped individuals. The inbreeding coefficient was performed by use of the diagonal of the relationship matrix and the genealogical parameters were computed using proper softwares. The effective population size was estimated based on the rate of inbreeding and census information, and the stratification of the population was verified by the average relationship coefficient between animals born in different regions of Brazil. The effects of inbreeding on morphological traits were made by the use of inbreeding coefficient as a covariate in the model of random regression. The inbreeding coefficient increased from 1990 on, impacting effective population size and, consequently, shrinking genetic variability. The paternal inbreeding was greater than maternal, which may be attributed to the preference for inbred animals in reproduction. The average genetic relationship coefficient of animals born in different states was lower than individuals born within the same state. The increase in the inbreeding coefficient was negatively associated with all studied traits, showing the importance to avoid genetic losses in the long term. Although results do not indicate a severe narrowing of the population until the present date, the average relationship coefficient shows signs of increase, which could cause a drastic reduction in genetic variability if inbred mating is not successfully controlled in the Campolina horse population.

Nine horses were used, aged 14.5±3.3 years and weightng 531±38.7kg. The diet consisted of 1.5% of live weight (LW) in Tifton 85 hay and 0.75% of LW in concentrate for both assays. The experiment consisted of two tests of apparent digestibility using the method of total collection, before and after the odontoplasty, where the consumption time of hay and concentrate was also evaluated and the particle size of the feces was analyzed. An effect (P<0.05) of odontoplasty on the apparent digestibility coefficients of dry matter, organic matter, crude protein, neutral detergent fiber and acid detergent fiber was observed. Effect of the consumption time for the concentrate (P<0.05) was observed, but not for the forage (P<0.05). There was more retention (P<0.05) of the fecal particles in the sieves of bigger granulometry. The odontoplasty increases the digestibility of dietary nutrients and feces particle size, without changing feces quality. Animals after the procedure have shorter concentrate consumption time.(AU)

This study focused on assessing the microbiology of conveyor belts surfaces, cleaned or not with pressurized water, and verification of the influence of this process on the microbiological quality of poultry meat. A reduction on mesophilic, psychrotrophic and Enterobacteriaceae counts (P<0.05) on dry conveyor belts compared to wet ones was observed. For the chicken leg (consisting of drumstick and thigh) samples, no statistically significant differences were detected on counts of indicators of microorganisms on poultry legs (composed by drumstick and thigh). For poultry meat conducted on wet or dry conveyor belts, 99% and 86%, were positive for Listeria spp, respectively. Only one sample of chicken leg was positive for presence of L. monocytogenes. These results demonstrate that there is no need to use water for cleaning conveyor belts during processing, which allows a reduction on the use of potable water in poultry slaughterhouses without jeopardizing food safety and public health.(AU)
Objetivou-se avaliar a qualidade microbiológica das superfícies das esteiras condutoras de cortes de carne de frango, higienizadas ou não com água pressurizada, bem como verificar a influência desse processo na qualidade microbiológica de cortes de frango. Foram observadas menores contagens, com diferença estatisticamente significativa (P<0,05) entre as populações de mesófilos, psicrotróficos e enterobactérias, nas esteiras condutoras de cortes secas em relação às úmidas. Nos cortes de coxa com sobrecoxa, as médias encontradas para populações de microrganismos indicadores não apresentaram diferenças estatisticamente significativas. Nos cortes analisados conduzidos pelas esteiras seca e úmida, 99% e 86% foram positivos para o isolamento de Listeria spp., respectivamente. Apenas uma amostra de corte de coxa com sobrecoxa desossada foi positiva para a presença de L. monocytogenes. Os resultados demonstram a possibilidade do desligamento do sistema de autolimpeza das esteiras condutoras de cortes, obedecendo às questões de segurança dos alimentos e à saúde pública, o que resulta na redução do uso de água potável pelos matadouros-frigoríficos de aves.(AU)

Inbreeding has been associated with the impairment of reproductive performance in many cattle breeds. Although the usage of reproductive biotechnologies has been increasing in bovine populations, not much attention has been given to the impact of inbreeding over cow's performance on artificial reproduction. The objective of this study was to estimate the impact of inbreeding on in vitro embryo production in a Guzerá breed population. The inbreeding coefficient (F), calculated as half of the co-ancestry of the individual's parents, was used as an estimate of inbreeding. The inbreeding coefficients of the donor, sire (used on in vitro fertilization) and of the embryos were included, separately, in the proposed models either as classificatory or continuous variables (linear and quadratic effects). The percentage of non-inbred individuals (or embryos) and mean F of donors, embryos and sires were 29.38%; 35.76%; 42.86% and 1.98±2.68; 1.32±3.13; 2.08±2.79, respectively. Two different models were considered, one for oocyte production traits and other for embryo production traits. The increase of F of the donor significantly (P<0.05) impaired the number of viable oocytes (N OV), number of grade I oocytes (N GI) and number of cleaved embryos (N CLV). Moreover, the donor's F influenced the percentage of grade I oocytes (P GI), percentage of viable embryos (P EMB) and percentage of cleaved embryos that turned into embryos (P CXE). No significant (P>0.05) effects were observed for the sire (father of the embryos) inbreeding coefficient over the traits analysed. Embryo's F influenced (P<0.05) the number of viable embryos (N EMB), percentage of viable embryos (P EMB) and percentage of cleaved embryos that turn into embryos (P CXE). Results suggested that an increase in the inbreeding coefficient might impair the embryos ability to survive through challenges imposed by the in vitro environment. Submitting highly inbred Guzerá female donors to in vitro embryo production may, in the long-term, have negative implications on the number of embryos obtained per cow and increase the relative costs of the improvement programmes based on this technology. High levels of inbreeding should be avoided when selecting Guzerá female donors and planning in vitro fertilization mating.

The objectives of this study were to characterize the allelic and genotypic frequencies of polymorphisms in the µ-calpain and calpastatin genes, and to assess their association with meat tenderness and animal growth in Nellore cattle. We evaluated 605 Nellore animals at 24 months of age, on average, at slaughter. The polymorphisms were determined for the molecular markers CAPN316, CAPN530, CAPN4751, CAPN4753, and UOGACAST1. Analyses of meat tenderness at 7, 14, and 21 days of maturation were performed in samples of longissimus thoracis obtained between the 12th and 13th rib and sheared using a Warner Bratzler Shear Force. Significant effects were observed for meat tenderness at days 7, 14, and 21 of maturation for the marker CAPN4751, at day 21 for the marker CAPN4753, and at days 14 and 21 for the marker UOGCAST1. For genotypic combinations of markers, the results were significant for the combination CAPN4751/UOGCAST1 in the three maturation periods and CAPN4753/UOGCAST1 at days 14 and 21 of maturation.

Four models for in vitro embryo production traits in Guzerá cattle were compared: Gaussian (untransformed variable - LIN and transformed in logarithmic scale - LOG), Poisson (POI) and zero-inflated Poisson (ZIP). Data consisted of 5716 ovum pick-up and in vitro fertilization records performed in 1205 cows from distinct regions of Brazil. Analyzed count traits were the number of viable oocytes (NOV), number of grade I oocytes (NGI), number of degenerated oocytes (NDG), number of cleaved embryos (NCLV) and number of viable produced embryos (NEMB). Heritability varied from 0.17 (LIN) to 0.25 (POI) for NOV; 0.08 (LOG) to 0.18 (ZIP) for NGI; 0.12 (LIN) to 0.20 (POI) for NDG; 0.13 (LIN) to 0.19 (POI) for NCLV; 0.10 (LIN) to 0.20 (POI) for NEMB depending on the considered model. The estimated repeatability varied from 0.53 (LOG) to 0.63 (POI) for NOV; 0.22 (LOG) to 0.39 (ZIP) for NGI; 0.29 (LIN) to 0.42 (ZIP) for NDG; 0.42 (LIN) to 0.59 (POI) for NCLV; 0.36 (LIN) to 0.51 (POI) for NEMB. The goodness of fit, measured by deviance information criterion and mean squared residuals, suggested superiority of POI and ZIP over Gaussian models. Estimated breeding values (EBV) obtained by different models were highly correlated, varying from 0.92 for NOV (between LIN-POI) and 0.99 for NGI (between POI-ZIP). The number of coincident animals on the 10% top EBV showed lower similarities. We recommend POI and ZIP models as the most adequate for genetic analysis of in vitro embryo production traits in Guzerá cattle.

The objective of this study was to estimate variance components for oocyte and embryo production traits in Guzerá breed female donors, and investigate their associations with age at first calving (AFC). The traits analyzed were the number of viable oocytes (NOV), the number of grade I oocytes (NGI), the number of cleaved embryos (NCLV), and viable embryos produced (NEMB), and the percentages of viable oocytes (POV), grade I oocytes (PGI), cleaved embryos (PCLV), and viable embryos (PEMB). Data were obtained from 5173 ovary puncture and in vitro fertilization (IVF) sessions using 1080 Guzerá female donors of different ages, occurred from March 2005 to July 2013. Variables were log-transformed (logeX+1) prior to analysis. (Co)variance components were estimated by restricted maximum likelihood (REML), using one- and two-trait animal models. Permanent environment and IVF sire (father of the embryos) random effects were included. Estimated heritabilities for NOV, NGI, NCLV, NEMB, POV, PGI, PCLV, and PEMB were 0.19, 0.08, 0.16, 0.14, 0.04, 0.03, 0.01, and 0.07, respectively. Repeatabilities for count traits (NOV, NGI, NCLV, and NEMB) varied from 0.14 and 0.32, higher than estimated for percentage traits (POV, PGI, PCLV, and PEMB), which varied from 0.01 to 0.08. Selection for NOV may be more appropriate in breeding programs than selection for NEMB, because of its strong genetic correlation (0.68) with NEMB and its greater time- and cost-effectiveness. AFC was only weakly associated with the oocyte and embryo production traits, which indicates that there would be no effect on AFC when selecting for these traits.

The objectives of this study were to characterize and define homogenous production environments of composite beef cattle in Brazil in terms of climatic and geographic variables by using multivariate exploratory techniques; to evaluate the presence of genotype by environment interaction (GxE) for post-weaning weight gain (PWG), yearling scrotal circumference (SC), and yearling muscling (MUS). Hierarchical and nonhierarchical cluster analysis was used to group farms located in regions with similar environmental variables into clusters. Six clusters of farms were formed. The effect of sire-cluster interaction was tested by single-trait analysis. Genetic parameters were estimated by multi-trait analysis considering the same trait to be different in each cluster. The effect of sire-cluster interaction was significant (P <0.01) for PWG and MUS. Estimates of genetic correlations among clusters ranged from 0.31 to 0.93 for PWG, 0.64 to 0.89 for SC, and 0.18 to 0.80 for MUS. These results indicate the need for a genetic analysis on a regional basis or inclusion of the GxE effect in the statistical model to permit appropriate evaluation of the animals.

In the past, the focus of broiler breeding programs on yield and carcass traits improvement led to problems related to meat quality. Awareness of public concern for quality resulted in inclusion of meat quality traits in the evaluation process. Nevertheless, few genes associated with meat quality attributes are known. Previous studies mapped quantitative trait loci for weight at 35 and 42 days in a region of GGA4 flanked by the microsatellite markers, MCW0240 and LEI0063. In this region, there are 2 fibroblast growth factor binding protein (FGFBP) genes that play an important role in embryogenesis, cellular differentiation, and proliferation in chickens. The objective of this study was to identify and associate single nucleotide polymorphisms (SNPs) in FGFBP1 and FGFBP2 with performance, carcass, and meat quality in experimental and commercial chicken populations. In the commercial population, SNP g.2014G>A in FGFBP1 was associated with decreased carcass weight (P < 0.05), and SNP g.651G>A in FGFBP2 was associated with thawing loss and meat redness content (P < 0.05). Four haplotypes were constructed based on 2 SNPs and were associated with breast weight, thawing loss, and meat redness content. The diplotypes were associated with thawing loss, lightness, and redness content. The SNPs evaluated in the present study may be used as markers in poultry breeding programs to aid in improving growth and meat quality traits.

Meat quality is an important feature for the poultry industry and is associated with consumer satisfaction. The calpain 1 (CAPN1) gene is related to the tenderness process of meat post- mortem, and the calpain 3 (CAPN3) gene plays an important role in myofibrillar organization and growth. The objective of the present study was to identify polymorphisms in these genes and to determine the association between these polymorphisms and traits of economic interest in poultry. Eleven animals (F1) from an experimental poultry population at Embrapa Swine and Poultry were used to identify the polymorphisms. Four single nucleotide polymorphisms (SNPs) were found in the CAPN1 gene, and one SNP was found in the CAPN3 gene. A polymorphism from each gene was selected for genotyping in 152 chickens from the Embrapa F2 experimental population and 311 chickens from a commercial population. Polymorphism g.2554T>C (CAPN1) was associated with body weight at 35 to 42 days, thigh weight, breast weight, carcass weight, and meat lightness content. SNP g.15486C>T (CAPN3) was associated with thigh yield, thawing-cooking loss, and shear force. Results suggest the possibility of using molecular markers in CAPN1 and CAPN3 genes as a tool for performance and meat quality traits in poultry breeding programs.

Ractopamine hydrochloride (RAC) improves swine production efficiency by redirecting nutrients to favor muscle accretion rather than fat deposition. In the present study, the time-dependent effect of RAC feeding on performance, plasma urea N (PUN) concentrations, and carcass traits of finishing pigs were evaluated. In a 28-d growth study, 80 barrows (average initial BW = 69.4 ± 7.9 kg) were assigned to 1 of 5 treatments in a randomized complete block design with 8 replicate pens per treatment and 2 pigs per pen. The pigs were fed a corn-soybean meal-based diet with no added RAC (control) or 10 mg of RAC/kg fed for 7, 14, 21, or 28 d before slaughter. All diets were formulated to contain 0.88% standardized ileal digestible Lys (1.0% total Lys) and 3.23 Mcal of ME/kg. Individual pig BW and pen feed disappearance were recorded weekly to determine BW changes, ADG, ADFI, and G:F. Anterior vena cava blood samples were taken on d 28 for determination of PUN concentrations. After 28 d on trial, the pigs were slaughtered and carcass measurements made at 24 h postmortem. Overall, providing pigs with different RAC feeding durations did not affect the final BW and ADFI but resulted in a tendency (P = 0.09) for a linear increase in ADG and a linear improvement (P = 0.003) in G:F. No effect of RAC feeding was found for weekly ADFI. Weekly improvements (P < 0.05) in ADG and G:F were observed over the first 21 d of RAC feeding. However, the growth response declined (P < 0.05) in wk 4 of RAC treatment. The concentrations of PUN exhibited a quadratic decrease (P = 0.004) as the RAC feeding duration increased. Although RAC feeding did not affect any backfat measurements and carcass length, increasing the RAC feeding duration linearly increased HCW (P = 0.01), dressing percentage (P = 0.03), LM depth (P = 0.001), LM area (P < 0.001), muscle-to-fat ratio (P = 0.004), and predicted carcass lean percentage (P = 0.02). These results indicate that a greater growth rate was achieved within the first 21 d of RAC feeding whereas the magnitude of carcass response was directly dependent on the duration of RAC feeding.

Os programas de pagamento por qualidade devem ser usados como forma de incentivo à melhoria da qualidade do leite e para evitar as fraudes. O pagamento pelo requisito da gordura deve ser mantido para fins de avaliação da amostragem correta do leite e como um indicativo de possíveis fraudes na CCS do leite. Apesar do modelo experimental ter demonstrado a possibilidade da diminuição da CCS, trazendo bonificação ao produtor quanto a este componente, as análises estatísticas demonstraram que o mesmo não é viável, pois os produtores perderiam em outros componentes que também são utilizados como parâmetros para bonificação e em volume de leite, o que poderia gerar a penalização.
In order to reach the industrial profitability the industries proposed the Premium Milk Payments, where the producer would get bonus or would be punished focusing on milks composition like fat, protein and (SCC) somatic cell count. However, when there is improvement milk quality program where the producer can have bonus or being penalized there is a risk of fraud. This study aims to verify the feasibility of the experimental model to SCC fraud by subtraction portions of the supernatant of milk and evaluate the consequences of this subtraction on some components of the milk. For experimental model were used milk samples from 18 lactating cows, and milk samples subjected to two different rest period and subsequently subtracted 5,0%, 7,5% and 10,0% of supernatant of milk. The results showed that is possible to reduce SCC from the milk through the experimental model of fraud described. The statistical analyzes showed as results the best model to reduce SCC obtained by subtracting 7,5% of the supernatant by 30 minutes rest. The Premium Milk Payments should be used as an incentive to improve the quality of milk to prevent fraud. The payment requirement for fat should be kept for the purpose of assessing the correct sampling of milk and as indication of possible fraud in the SCC from milk samples. Despite the experimental model have demonstrated the possibility of decreased SCC, bringing bonus the producer on this component, the statistical analyzes show that it is not feasible, because the produces would lose in other components that are also used as a reference for subsidy and milk volume, which could be lead to penalty.

Given the important role of leptin in metabolism, we looked for a possible association of leptin and leptin receptor polymorphisms with carcass and growth traits in Nellore cattle. We examined associations of leptin and leptin receptor SNPs with ultrasound carcass (longissimus dorsi muscle area (ribeye area), backfat thickness and rump fat thickness and growth traits (weaning weight adjusted to 210 days of age, yearling weight adjusted to 550 days of age, weight gain of weaning to yearling and scrotal circumference adjusted to 550 days of age) of 2162 Bos primigenius indicus (Nellore) animals. Allele and genotypic frequencies were calculated for each marker. Allele substitution, additive and dominance effects of the polymorphisms were also evaluated. Some alleles of the molecular markers had low frequencies, lower than 1%, in the sample analyzed, although the same polymorphisms described for B. p. taurus cattle were found. Due to very low allelic frequencies, the E2JW, A59V and UASMS2 markers were not included in the analysis, because they were almost fixed. E2FB was found to be significantly associated with weight gain, ribeye area and backfat thickness. The promoter region markers, C963T and UASMS1, were also found to be significantly associated with ribeye area. T945M was significantly associated with weight gain. We conclude that the leptin and receptor gene markers would be useful for marker-assisted selection.

Our objective was to estimate Bos primigenius taurus introgression in American Zebu cattle. One hundred and four American Zebu (Nellore) cattle were submitted to mtDNA, microsatellite and satellite analysis. Twenty-three alleles were detected in microsatellite analysis, averaging 4.6 ± 1.82/locus. Variance component comparisons of microsatellite allele sizes allowed the construction of two clusters separating taurus and indicus. No significant variation was observed when indicus and taurus mtDNA were compared. Three possible genotypes of 1711b satellite DNA were identified. All European animals showed the same restriction pattern, suggesting a Zebu-specific restriction pattern. The frequencies of B. primigenius indicus-specific microsatellite alleles and 1711b satellite DNA restriction patterns lead to an estimate of 14% taurine contribution in purebred Nellore.

The effect of melatonin during in vitro maturation (IVM) on DNA damage of cumulus cells (CCs) from bovine cumulus-oocyte complexes (COCs) and embryo development was evaluated. COCs from abattoir ovaries were cultured in maturation medium (MM) with 0.5µg/ml FSH and 5.0µg/ml LH (FSH-LH); 10(-9)M melatonin (MEL) or FSH-LH+MEL (FSH-LH-MEL). After 24h of in vitro maturation, the CCs surrounding the oocyte were subjected to DNA analysis by Comet assay. After in vitro fertilization and in vitro embryo culture, the embryo development rates were evaluated on day 2 post insemination (cleavage) and days 7-8 (blastocyst). The percentage of CCs with no DNA damage was significantly superior in MEL group (37.6±2.4) than in FSH-LH-MEL (28.0±2.4) and FSH-LH (17.8±2.41) groups. Cleavage and blastocysts rates were similar among groups. Melatonin during IVM protects the CCs from DNA damage but this effect did not influence embryo development in vitro.
