RESUMO

Neuromuscular electrical stimulation (NMES) can improve physical function in different populations. NMES-related outcomes may be influenced by muscle length (i.e., joint angle), a modulator of the force generation capacity of muscle fibers. Nevertheless, to date, there is no comprehensive synthesis of the available scientific evidence regarding the optimal joint angle for maximizing the effectiveness of NMES. We performed a systematic review to investigate the effect of muscle length on NMES-induced torque, discomfort, contraction fatigue, and strength training adaptations in healthy and clinical adult populations (PROSPERO: CRD42022332965). We conducted searches across seven electronic databases: PUBMED, Web of Science, EMBASE, PEDro, BIREME, SCIELO, and Cochrane, over the period from June 2022 to October 2023, without restricting the publication year. We included cross-sectional and longitudinal studies that used NMES as an intervention or assessment tool for comparing muscle lengths in adult populations. We excluded studies on vocalization, respiratory, or pelvic floor muscles. Data extraction was performed via a standardized form to gather information on participants, interventions, and outcomes. Risk of bias was assessed using the Revised Cochrane risk-of-bias tool for cross-over trials and the Physiotherapy Evidence Database scale. Out of the 1185 articles retrieved through our search strategy, we included 36 studies in our analysis, that included 448 healthy young participants (age: 19-40 years) in order to investigate maximum evoked torque (n = 268), contraction fatigability (n = 87), discomfort (n = 82), and muscle strengthening (n = 22), as well as six participants with spinal cord injuries, and 15 healthy older participants. Meta-analyses were possible for comparing maximal evoked torque according to quadriceps muscle length through knee joint angle. At optimal muscle length 50° - 70° of knee flexion, where 0° is full extension), there was greater evoked torque during nerve stimulation compared to very short (0 - 30°) (p<0.001, CI 95%: -2.03, -1.15 for muscle belly stimulation, and -3.54, -1.16 for femoral nerve stimulation), short (31° - 49°) (p = 0.007, CI 95%: -1.58, -0.25), and long (71° - 90°) (p<0.001, CI 95%: 0.29, 1.02) muscle lengths. At long muscle lengths, NMES evoked greater torque than very short (p<0.001, CI 95%: -2.50, -0.67) and short (p = 0.04, CI 95%: -2.22, -0.06) lengths. The shortest quadriceps length generated the highest perceived discomfort for a given current amplitude. The amount of contraction fatigability was greater when muscle length allowed greater torque generation in the pre-fatigue condition. Strength gains were greater for a protocol at the optimal muscle length than for short muscle length. The quality of evidence was very high for most comparisons for evoked torque. However, further studies are necessary to achieve certainty for the other outcomes. Optimal muscle length should be considered the primary choice during NMES interventions, as it promotes higher levels of force production and may facilitate the preservation/gain in muscle force and mass, with reduced discomfort. However, a longer than optimal muscle length may also be used, due to possible muscle lengthening at high evoked tension. Thorough understanding of these physiological principles is imperative for the appropriate prescription of NMES for healthy and clinical populations.

Our study aimed to combine psychological and physical factors to explore the impact of the menstrual cycle on performance in elite academy women soccer players through weekly monitoring. Eighteen elite academy women soccer players were monitored. Players reported daily through an online anonymous survey if they were in menstruation. Players answered the Hooper Questionnaire daily, performed an Illinois Agility Test (IAT) twice a week, and rated their perceived exertion (RPE) after every training session. Tests were associated with a complete menstrual cycle reported through the online anonymous survey to determine the two weeks of the follicular phase and the two weeks of the luteal phase. Of the 18 players, 10 completed all requirements and were retained for analyses. IAT did not show significant differences throughout the menstrual cycle (p = 0.633). Fatigue (p = 0.444), Stress (p = 0.868), Sleep (p = 0.398), DOMS (delayed onset muscle soreness; p = 0.725), and Hooper Index (p = 0.403) did not show significant differences either. RPE was also comparable across the cycle (p = 0.846). Our results failed to demonstrate that hormonal variation during the menstrual cycle influenced psychological and physical markers of performance.

The aims of this study in elite youth French players were to 1) describe the anthropometric and physical characteristics of international and non-international players from U16 to U20, and 2) compare these characteristics across age categories and playing standard (international or non-international). Altogether, 1423 players from the French Rugby Federation's academies participated in a physical testing battery, part of its national young player development pathway. From seasons 2010 to 2020, players were assessed for anthropometric (body mass and height), off-field (bench press; isometric squat, vertical jump) and on-field physical characteristics (aerobic capacity: maximal aerobic speed [MAS]; speed: 10-m, 50-m sprint). A 2-way mixed model analysis of variance (ANOVA) was used to compare physical characteristics across age categories and playing standards. Two separate models were used for forwards and backs. A main statistical effect was observed for age category and playing standard (range p < 0.05 -p < 0.001). Pair-wise category comparisons showed that older players were generally taller, heavier, stronger, faster and demonstrated better aerobic qualities than younger peers. The same results were observed for INT compared with NI players while INT forwards were also taller and heavier than NI peers (range p < 0.01 -p < 0.001). Findings revealed a clear progression in anthropometric characteristics and physical qualities throughout the age development pathway in elite young French rugby players. Findings also identified certain physical qualities (strength, power and speed) necessary at younger levels to achieve international standard.

In elite-level youth soccer players, weekly training periodization is of paramount importance to plan for short- and long-term physical development. The present study investigated current practices for physical periodization strategies in elite male French academies. An online survey was completed by elite French academies strength and conditioning coaches to determine the typical weekly periodization with particular reference to daily training in relation to match day (MD) in youth soccer players. The survey attempted to characterize the importance of physical development compared to match result, and practices used (expected difficulty and content) for each training session according to duration, exercises, and objective. The frequency rates of the responses were compared using two-tailed Chi-square tests with the significance level set at p < 0.05. Fortyfive questionnaires were analyzed. Respondents indicated that their training sessions focused mainly on physical development (95.6%) rather than match result. Active recovery (34.2%) and aerobic conditioning exercises (40.8%) were primarily conducted on MD+1 and MD+2 using passing circuits and aerobic technical drills. Physical development was mostly pursued during sessions on MD-4 (38.8%) and MD-3 (37.3%). The number of large-sided games was highest on MD-3 (58.1%). On MD-2 and MD-1, a decrease in the training load was highlighted, with speed (40.4%) and tapering sessions (52.4%) mostly implemented. Intensive use of small-sided games (92.3%) and reactivity exercises was observed at MD-1 (100.0%). Our results revealed discrepancies between the physical objectives set for each day and the content implemented, which could potentially be more physically demanding than expected.

MMPs are enzymes involved in SARS-CoV-2 pathogenesis. Notably, the proteolytic activation of MMPs can occur through angiotensin II, immune cells, cytokines, and pro-oxidant agents. However, comprehensive information regarding the impact of MMPs in the different physiological systems with disease progression is not fully understood. In the current study, we review the recent biological advances in understanding the function of MMPs and examine time-course changes in MMPs during COVID-19. In addition, we explore the interplay between pre-existing comorbidities, disease severity, and MMPs. The reviewed studies showed increases in different MMP classes in the cerebrospinal fluid, lung, myocardium, peripheral blood cells, serum, and plasma in patients with COVID-19 compared to non-infected individuals. Individuals with arthritis, obesity, diabetes, hypertension, autoimmune diseases, and cancer had higher MMP levels when infected. Furthermore, this up-regulation may be associated with disease severity and the hospitalization period. Clarifying the molecular pathways and specific mechanisms that mediate MMP activity is important in developing optimized interventions to improve health and clinical outcomes during COVID-19. Furthermore, better knowledge of MMPs will likely provide possible pharmacological and non-pharmacological interventions. This relevant topic might add new concepts and implications for public health in the near future.

Weekly training loads are typically reported using absolute values and are not individualized according to competition positional demands (relative values). The aim of this study was to evaluate absolute and relative training loads and compare across playing positions during a full in-season in an elite soccer academy. 24 elite academy soccer players, categorized into five positions (CD: central defender [n = 4]; FB: full back [n = 5]; CM: central midfielder [n = 6]; WM: wide midfielder [n = 5]; FW: forward [n = 4]), were monitored using a global positioning system. Absolute training load was calculated using the total distance, the distance at moderate-speed ([15-20[ km·h-1), high-speed ([20-25[ km·h-1), sprint (> 25 km·h-1), the total number of accelerations (> 3 m·s-2) and decelerations (< -3 m·s-2). Relative training load was calculated by dividing absolute training loads by mean values from the competitive matches. Training loads were determined daily according to their distance from match day (MD). One-way ANOVAs were performed to evaluate differences between playing positions. Absolute moderate-speed distance was greater for WM compared to CD (p = 0.015, and p = 0.017), while the opposite was shown for relative values (p = 0.014, and p < 0.001) on MD-4 and MD-3, respectively. The absolute moderate-speed distance was not different between CD, FB, CM, and FW, whereas relative values were greater for CD on MD+2 and MD-4 (p < 0.05). FB and WM performed greater absolute high-speed distance than CD on MD-4 and MD-3 (p < 0.05) while no difference was highlighted for relative values.Our results demonstrated that in the present academy players, training load for CD was underestimated using absolute training loads for moderate and high-speed distances. In contrast, relative training loads highlighted WM as an underloaded position. Therefore, relative training loads are recommended as they contextualize training loads according to competitive demands and favor training individualization.

Warm-up routines include various tasks focused on the peripheral contractile properties and nervous motor command. This present study was aimed at investigating the acute effects of different warm-up routines, emphasizing either peripheral (post-activation performance enhancement, PAPE) or central (motor imagery, MI) contributions on sport-specific tasks. Eleven young female athletes took part in this cross-over, randomized, controlled trial. They underwent three experimental sessions composed of a standardized warm-up followed by 10 min of (1) rest (CONTROL), (2) maximal concentric leg press (PAPE), or (3) mental repetitions of sprint tasks (MI). Post-tests consisted of reaction time, arrowhead agility test, 20 m sprint, repeated sprint ability, and NASA-TLX fatigue questionnaire. PAPE and MI significantly enhanced the arrowhead agility test (p < 0.001 and p = 0.012, respectively) and repeated sprint ability (p = 0.002 and p = 0.035, respectively) compared to CONTROL, without any difference between PAPE and MI. The 20 m sprint time was better after PAPE as compared to MI (p = 0.005) and CONTROL (p < 0.001), without any difference between MI and CONTROL. Reaction time and the NASA-TLX questionnaire were not affected by the warm-up modalities (p > 0.05). PAPE was the most efficient to optimize warm-up due to its greater peripheral contribution that would improve muscle contractility. MI specifically improved the imagined tasks mostly by central contribution.

Background. COVID-19 patients may present sequelae, such as neuromuscular electrophysiological disorders (NED), that can be assessed using the stimulus electrodiagnostic test (SET). However, little is known about the reliability and agreement of the SET in post-COVID-19 patients.Objective. We aimed to verify the intra-inter-rater reliability and agreement of SET measurements in the rectus femoris, vastus medialis, vastus lateralis, tibialis anterior, and gastrocnemius lateralis (GL) in post-COVID-19 participants.Methods. We designed an observational prospective study to evaluate 20 (10 males and 10 females) post-COVID-19 patients, age: 44.95 ± 11.07 years, weight: 87.99 ± 19.08 kg, height: 1.69 ± 0.09 m. Two independent raters took two evaluations using the SET on selected muscles. The intra-class correlation coefficient (ICC) and 95% limits of the agreement defined the quality and magnitude of the measures.Results. For intra-rater reliability, all measurements presented correlations classified as high or very high (ICC: 0.71-1.0). For inter-rater reliability, the rheobase, chronaxie, accommodation, and accommodation index presented high or very high correlations, except for the accommodation index of the GL (ICC = 0.65), which was moderate.Conclusion. The reliability of the SET obtained by independent raters was very high, except for the GL accommodation, which presented moderate ICC. Therefore, the SET is a reliable tool for evaluating NED in post-COVID-19 patients.

Determining how the quadriceps femoris musculotendinous unit functions, according to hip and knee joint angles, may help with clinical decisions when prescribing knee extension exercises. We aimed to determine the effect of hip and knee joint angles on structure and neuromuscular functioning of all constituents of the quadriceps femoris and patellar tendon properties. Twenty young males were evaluated in four positions: seated and supine in both 20° and 60° of knee flexion (SIT20, SIT60, SUP20, and SUP60). Peak knee extension torque was determined during maximal voluntary isometric contraction (MVIC). Ultrasound imaging was used at rest and during MVIC to characterize quadriceps femoris muscle and tendon aponeurosis complex stiffness. We found that peak torque and neuromuscular efficiency were higher for SUP60 and SIT60 compared to SUP20 and SIT20 position. We found higher fascicle length and lower pennation angle in positions with the knee flexed at 60°. The tendon aponeurosis complex stiffness, tendon force, stiffness, stress, and Young's modulus seemed greater in more elongated positions (60°) than in shortened positions (20°). In conclusion, clinicians should consider positioning at 60° of knee flexion rather than 20°, regardless if seated or supine, during rehabilitation to load the musculotendinous unit enough to stimulate a cellular response.

The purpose of the present study was to characterize and compare locomotor and contact loads in U18 and U20 international rugby union competition during matches, and specifically during peak match-play phases using short rolling epochs and continuous ball-in-play (BIP) sequences. 20 international matches from French national teams were analysed in the U18 and U20 Six Nations Tournament respectively and World Rugby U20 Championship. Running loads were quantified using global positioning devices (16 Hz) and contact loads via video match analysis software. Players were split into forward (U18, n = 29; U20, n = 32) and back positional groups (U18, n = 20; U20, n = 24). Compared with U20 peers, U18 players covered a higher total distance (effect size (ES) = -0.76 ± 0.25) and at high-speeds per minute (> 4 m · s-1; ES = -0.55 ± 0.25) and performed more accelerations (ES = -0.71 ± 0.25). While a greater frequency of BIP sequences > 90 s duration was observed in U20s versus U18s match-play, U18s covered more total distance and high-speed distance (ES = -0.42 ± 0.13 and -0.33 ± 0.13 respectively) per minute during these longer sequences. During peak rolling phases shorter than 4 minutes, no clear differences existed between age categories in running activity, while U20 forwards performed more contact actions than U18 peers. The match-play loads observed in the present international U18 players suggest that they are ready to respond to the overall and peak demands observed in U20 competition. Moreover, the present information on peak activity phases can aid design of overload high-intensity conditioning sessions to respond to the running- and contact-demands identified in those competitions.

INTRODUCTION: This study aimed to apply multifractal detrended fluctuation analysis (MFDFA) to surface EMG to detect neuromuscular changes after realistic warm-up procedures that was followed by various stretching exercises. METHODS: Sixteen volunteers conducted two experimental sessions. Testing included two maximal voluntary contractions before, after a standardized warm-up, and after a stretching exercise (static or neurodynamic nerve gliding technique). EMG was registered on biceps femoris and semitendinosus muscles. EMG was analyzed using different parameters obtained from the singularity Hurst exponent function and multifractal power spectrum (both obtained from the multifractal detrended fluctuation analysis). RESULTS: The Hurst exponent, α maximum, and peak value of the multifractal spectrum significantly decreased after warm-up as compared with baseline for both biceps femoris ( P = 0.003, P = 0.006, and P = 0.003, respectively) and semitendinosus ( P = 0.006, P = 0.013 and P = 0.01, respectively) muscles. No further alteration was obtained after static or neurodynamic nerve gliding stretching as compared with post-warm-up ( P = 1.0). No significant difference was obtained for Hurst exponent range, width, and asymmetry of the multifractal spectrum ( P > 0.05). CONCLUSIONS: From the present results, EMG depicted multifractal features sensitive to detect neuromuscular changes after a warm-up procedure. An increase in multiscale complexity is revealed after warm-up without any further alteration after stretching. The multifractal spectrum depicted dominant small fluctuations that shifted toward slightly larger fluctuations that could be attributed to motor unit recruitment.

RNA-protein interactions (RPIs) are promising targets for developing new molecules of therapeutic interest. Nevertheless, challenges arise from the lack of methods and feedback between computational and experimental techniques during the drug discovery process. Here, we tackle these challenges by developing a drug screening approach that integrates chemical, structural and cellular data from both advanced computational techniques and a method to score RPIs in cells for the development of small RPI inhibitors; and we demonstrate its robustness by targeting Y-box binding protein 1 (YB-1), a messenger RNA-binding protein involved in cancer progression and resistance to chemotherapy. This approach led to the identification of 22 hits validated by molecular dynamics (MD) simulations and nuclear magnetic resonance (NMR) spectroscopy of which 11 were found to significantly interfere with the binding of messenger RNA (mRNA) to YB-1 in cells. One of our leads is an FDA-approved poly(ADP-ribose) polymerase 1 (PARP-1) inhibitor. This work shows the potential of our integrative approach and paves the way for the rational development of RPI inhibitors.

OBJECTIVE: The present study aimed to determine the association between neuromuscular function, motor function impairment, and muscle and tendon structures in individuals with spinal cord injury (SCI) compared to a control (non-disabled) population. DESIGN: A cross-sectional study with a control group. SETTING: Center of Adapted Sports Training and Special Physical Education. PARTICIPANTS: Fifteen individuals with SCI and motor function impairments participated in the study. A paired non-disabled group was recruited for comparison. INTERVENTIONS: Not applicable. OUTCOME MEASURES: Muscle (biceps brachii, rectus femoris, vastus lateralis, vastus medialis, and tibialis anterior) and tendon (quadriceps and patellar tendons) structures were assessed by ultrasound imaging (thickness, pennation angle, fascicle length, and echogenicity). Neuromuscular electrophysiological disorders were also assessed using electrodiagnosis techniques (stimulus non-responsivity and chronaxie) in the same muscles. RESULTS: Except for the biceps brachii muscle, muscle thickness, pennation angle, and fascicle length were lower (p < 0.01) while echogenicity and chronaxie were greater (p < 0.01) in SCI participants. The SCI participants had a higher prevalence of neuromuscular electrophysiological disorders for all muscles, except the biceps brachii. CONCLUSION: Neuromuscular disorders occur in association with muscle and tendon maladaptation in individuals with chronic SCI. A higher prevalence of electrophysiological disorders suggests an acquired polyneuromyopathy for muscles with motor function impairment even though the muscle was innerved, in addition to widespread muscle atrophy.

COVID-19 is associated with musculoskeletal disorders. Ultrasound is a tool to assess muscle architecture and tendon measurements, offering an idea of the proportion of the consequences of the disease, since significant changes directly reflect the reduction in the ability to produce force and, consequently, in the functionality of the patient; however, its application in post-COVID-19 infection needs to be determined. We aimed to assess the intra- and inter-rater reliability of ultrasound measures of the architecture of the vastus lateralis (VL), rectus femoris (RF), vastus medialis (VM), gastrocnemius lateralis (GL), gastrocnemius medialis (GM), soleus (SO), and tibialis anterior (TA) muscles, as well as the patellar tendon (PT) cross-sectional area (CSA) in post-COVID-19 patients. An observational, prospective study with repeated measures was designed to evaluate 20 post-COVID-19 patients, who were measured for the pennation angle (θp), fascicular length (Lf), thickness, echogenicity of muscles, CSA and echogenicity of the PT. The intra-class correlation coefficient (ICC) and 95% limits of agreement were used. The intra-rater reliability presented high or very high correlations (ICC = 0.71-1.0) for most measures, except the θp of the TA, which was classified as moderate (ICC = 0.69). Observing the inter-rater reliability, all the evaluations of the PT, thickness and echogenicity of the muscles presented high or very high correlations. For the Lf, only the RF showed as low (ICC = 0.43), for the θp, RF (ICC = 0.68), GL (ICC = 0.70) and TA (ICC = 0.71) moderate and the SO (ICC = 0.40) low. The ultrasound reliability was acceptable for the muscle architecture, muscle and tendon echogenicity, and PT CSA, despite the low reliability for the Lf and θp of the RF and SO, respectively.

French academies almost all implement reactivity drills and small-sided games (SSG) the day preceding a match (MD-1). The present study aimed to determine the physical impact of different training durations on MD-1 on the subsequent matchday performance (MD). Eleven elite U19 academy soccer players conducted three typical training sessions lasting 45 min (TS45), 60 min (TS60) and 75 min (TS75) on MD-1. During TS60, warm-up, reactivity and SSG were 10, 15 and 24 min, respectively, plus coaches' feedback or water breaks. Durations of all training components decreased by 25% for TS45 and increased by 25% for TS75. Tests were conducted on MD-4 (CONTROL) and MD before the match (TEST). Tests consisted of a counter movement jump (CMJ), 20 m sprint, Illinois agility test (IAT) and Hooper questionnaire. CONTROL values were similar over the three experimental conditions. TEST on MD revealed greater CMJ for TS45 (42.7 ± 5.1 cm) compared to TS60 (40.5 ± 5.5 cm, p = 0.032) and TS75 (40.9 ± 5.7 cm, p = 0.037). 20 m time was lower for TS45 (3.07 ± 0.10 s, p < 0.001) and TS60 (3.13 ± 0.10 s, p = 0.017) compared to TS75 (3.20 ± 0.10 s). IAT time was lower on TS45 (14.82 ± 0.49 s) compared to TS60 (15.43 ± 0.29 s, p < 0.001) and TS75 (15.24 ± 0.33 s, p = 0.006). Furthermore, the Hooper index was lower at TEST for TS45 (7.64 ± 1.50) compared to TS60 (11.00 ± 3.49, p = 0.016) and TS75 (9.73 ± 2.41, p = 0.016), indicating a better readiness level. We concluded that, as training session duration increases, performance decreases on MD. A 45 min training session including reactivity and SSG exercises is therefore recommended on MD-1.

This study aimed to apply different complexity-based methods to surface electromyography (EMG) in order to detect neuromuscular changes after realistic warm-up procedures that included stretching exercises. Sixteen volunteers conducted two experimental sessions. They were tested before, after a standardized warm-up, and after a stretching exercise (static or neuromuscular nerve gliding technique). Tests included measurements of the knee flexion torque and EMG of biceps femoris (BF) and semitendinosus (ST) muscles. EMG was analyzed using the root mean square (RMS), sample entropy (SampEn), percentage of recurrence and determinism following a recurrence quantification analysis (%Rec and %Det) and a scaling parameter from a detrended fluctuation analysis. Torque was significantly greater after warm-up as compared to baseline and after stretching. RMS was not affected by the experimental procedure. In contrast, SampEn was significantly greater after warm-up and stretching as compared to baseline values. %Rec was not modified but %Det for BF muscle was significantly greater after stretching as compared to baseline. The a scaling parameter was significantly lower after warm-up as compared to baseline for ST muscle. From the present results, complexity-based methods applied to the EMG give additional information than linear-based methods. They appeared sensitive to detect EMG complexity increases following warm-up.

Neuromuscular electrical stimulation (NMES) has been used to increase muscle strength and physical function. However, NMES induces rapid fatigue, limiting its application. To date, the effect of quadriceps femoris (QF) muscle length by knee and hip joint manipulation on NMES-induced contraction fatigability is not clear. We aimed to quantify the effects of different muscle lengths on NMES-induced contraction fatigability, fatigue index, and electromyographic (EMG) activity for QF muscle. QF maximum evoked contraction (QMEC) was applied in a 26 min protocol (10 s on; 120 s off; 12 contractions) in 20 healthy participants (24.0 ± 4.6 years old), over 4 sessions on different days to test different conditions. The tested conditions were as follows: supine with knee flexion of 60° (SUP60), seated with knee flexion of 60° (SIT60), supine with knee flexion of 20° (SUP20), and seated with knee flexion of 20° (SIT20). Contraction fatigability (torque decline assessed by maximal voluntary contraction [MVC] and during NMES), fatigue index (percentage reduction in MVC), and EMG activity (root mean square [RMS] and median frequency) of the superficial QF' constituents were assessed. After NMES, all positions except SUP20 had an absolute reduction in MVC (p < .001). Fatigue index was greater in SIT20 than in SIT60 (p < .001) and SUP20 (p = .01). There was significant torque reduction across the 12 QMEC in SUP60 and SIT60, up to 10.5% (p < .001-.005) and 9.49% (p < .001-.033), respectively. There was no torque reduction during NMES in SUP20 and SIT20. Fatigue was accompanied by an increase in RMS (p = .032) and a decrease in median frequency for SUP60 (p < .001). Median frequency increased only in the SUP20 condition (p = .021). We concluded that QF NMES-induced contraction fatigability is greater when the knee is flexed at 60° compared to 20°. In addition, a supine position promotes earlier fatigue for a 60° knee flexion, but it delays fatigue onset for a 20° knee flexion compared to the seated position. These results provide a rationale for lower limb positioning during NMES, which depends on training objectives, e.g., strengthening or task-specific functionality training.

The highly homologous protein lysine methyltransferases G9a and GLP, which catalyze mono- and dimethylation of histone H3 lysine 9 (H3K9), have been implicated in various human diseases. To investigate functions of G9a and GLP in human diseases, we and others reported several noncovalent reversible small-molecule inhibitors of G9a and GLP. Here, we report the discovery of the first-in-class G9a/GLP covalent irreversible inhibitors, 1 and 8 (MS8511), by targeting a cysteine residue at the substrate binding site. We characterized these covalent inhibitors in enzymatic, mass spectrometry based and cellular assays and using X-ray crystallography. Compared to the noncovalent G9a/GLP inhibitor UNC0642, covalent inhibitor 8 displayed improved potency in enzymatic and cellular assays. Interestingly, compound 8 also displayed potential kinetic preference for covalently modifying G9a over GLP. Collectively, compound 8 could be a useful chemical tool for studying the functional roles of G9a and GLP by covalently modifying and inhibiting these methyltransferases.

Exercise modality has been proposed to reduce the interferences between aerobic and resistance sessions during concurrent training. The aim of the study was to examine the acute effects of cycling or running sprint interval exercise on subsequent resistance training sessions. Twenty-five competitive male rugby union players were recruited. Players were tested during three conditions: CONTROL (resistance training session only), CYCLE and RUN (corresponding to a concurrent training scheme with cycling or running sprint interval exercise conducted on the morning, followed by a resistance training session). Four hours rest was proposed between the aerobic and resistance training session. Muscle performance (bar velocity during bench press and box squat, counter movement jump height) and subjective ratings (rate of perceived exertion, wellbeing) were assessed during and after aerobic or resistance training sessions. No significant difference was observed for muscle performance (vertical jump height and bar velocity). However, significant higher perceived exertion and low-value scaled subjective wellbeing were observed in RUN (7.7 ± 1.1 and 17.9 ± 4.1, respectively) as compared with the two other conditions (6.7 ± 1.5 and 21.1 ± 3.6 for CONTROL and 7.4 ± 1.1 and 20.1 ± 3.9 for CYCLE). It was concluded that the exercise modality (running or cycling) during the aerobic exercise using a sprint interval exercise did not impact the quality of the resistance session. However, subjective ratings were affected the following days. Cycling exercises might be more adequate when performing a sprint interval training session during concurrent training programs.
