RESUMO

Characterizing nanoparticles (NPs) is crucial in nanoscience due to the direct influence of their physiochemical properties on their behavior. Various experimental techniques exist to analyze the size and shape of NPs, each with advantages, limitations, proneness to uncertainty, and resource requirements. One of them is electron microscopy (EM), often considered the gold standard, which offers visualization of the primary particles. However, despite its advantages, EM can be expensive, less accessible, and difficult to apply during dynamic processes. Therefore, using EM for specific experimental conditions, such as observing dynamic processes or visualizing low-contrast particles, is challenging. This study showcases the potential of machine learning in deriving EM parameters by utilizing cost-effective and dynamic techniques such as dynamic light scattering (DLS) and UV-vis spectroscopy. Our developed model successfully predicts the size and shape parameters of gold NPs based on DLS and UV-vis results. Furthermore, we demonstrate the practicality of our model in situations in which conducting EM measurements presents a challenge: Tracking in situ the synthesis of 100 nm gold NPs.

Here we use fluorescence lifetime imaging microscopy (FLIM) to study the supramolecular organization of nanoencapsulated liposomal all-trans retinoic acid (ATRA), exploiting ATRA's intrinsic fluorescence as a source of signal and phasor transformation as a fit-free analytical approach to lifetime data. Our non-invasive method is suitable for checking for the presence of a fraction of ATRA molecules interacting with liposomal membranes. The results are validated by independent small-angle X-ray scattering (SAXS) and nano-differential scanning calorimetry (NanoDSC) measurements, probing ATRA's putative position on the membrane and effect on membrane organization. Besides the insights on the specific case-study proposed, the present results confirm the effectiveness of Phasor-FLIM analysis in elucidating the nanoscale supramolecular organization of fluorescent drugs in pharmaceutical formulations. This underscores the importance of leveraging advanced imaging techniques to deepen our understanding and optimize drugs' performance in delivery applications.

Rare-earth elements are widely used in high-end technologies, the production of permanent magnets (PMs) being one of the sectors with the greatest current demand and likely greater future demand. The combination of Nd and Dy in NdFeB PMs enhances their magnetic properties but makes their recycling more challenging. Due to the similar chemical properties of Nd and Dy, their separation is expensive and currently limited to the small scale. It is therefore crucially important to devise efficient and selective methods that can recover and then reuse those critical metals. To address these issues, a series of heptadentate Trensal-based ligands were used for the complexation of Dy3+ and Nd3+ ions, with the goal of indicating the role of coordination and solubility equilibria in the selective precipitation of Ln3+-metal complexes from multimetal non-water solutions. Specifically, for a 1:1 Nd/Dy mixture, a selective and fast precipitation of the Dy complex occurred in acetone with the Trensalp-OMe ligand at room temperature, with a concomitant enrichment of Nd in the solution phase. In acetone, complexes of Nd and Dy with Trensalp-OMe were characterized by very similar formation constants of 7.0(2) and 7.3(2), respectively. From the structural analysis of an array of Dy and Nd complexes with TrensalR ligands, we showed that Dy invariably provided complexes with coordination number (cn) of 7, whereas the larger Nd experienced an expansion of the coordination sphere by recruiting additional solvent molecules and giving a cn of >7. The significant structural differences have been identified as the main premises upon which a suitable separation strategy can be devised with these kind of ligands, as well as other preorganized polydentate ligands that can exploit the small differences in Ln3+ coordination requirements.

Controlling the size of powder particles is pivotal in the design of many pharmaceutical forms and the related manufacturing processes and plants. One of the most common techniques for particle size reduction in the process industry is powder milling, whose efficiency relates to the mechanical properties of the powder particles themselves. In this work, we first characterize the elastic and plastic responses of different pharmaceutical powders by measuring their Young modulus, the hardness, and the brittleness index via nano-indentation. Subsequently, we analyze the behavior of those powder samples during comminution via jet mill in different process conditions. Finally, the correlation between the single particle mechanical properties and the milling process results is illustrated; the possibility to build a predictive model for powder grindability, based on nano-indentation data, is critically discussed.

Physical forms of active pharmaceutical ingredients (APIs) play a crucial role in drug discovery since 85% of API molecules exhibit polymorphism and sometimes complicated phase behavior, often resulting in important differences in the respective biochemical and physical properties. Characterization and quantitation of the different forms are becoming more and more essential in the pharmaceutical industry: once these characteristics are known, it is easier to choose the best solid form for development, formulation, manufacturing, and storage. Time domain-nuclear magnetic resonance (TD-NMR) has recently been used to develop a quantitation protocol for solid mixtures, named qSRC, based on the linear combination of T1 saturation recovery curves (SRCs) collected on a bench-top instrument. Despite its potentials and ease of use, a limited number of application cases have been reported in the literature since its development and many aspects remain to be clarified for the technique to be adopted as a robust routinely industrial analytical tool. In the present work, the reliability of the qSRC approach has been studied by focusing on the role played by key experimental variables, including mixture composition, signal-to-noise ratio, and T1 differences. In silico simulations were carried out for a wide range of theoretical cases to predict the expected level of accuracy obtainable for a given sample-parameter acquisition set and to clearly define the range of applicability of the method. Results of the simulation are presented alongside a comparison with three real-case studies of commercially available APIs: piroxicam, naproxen sodium, and benzocaine.

INTRODUCTION: The aims of the PROBIT trial (clinicaltrials.gov: NCT03131284) were to prevent overweight or obesity occurring at two years of life, and improve feeding patterns during infancy. METHODS: The trial compared 252 northern Italian newborns whose paediatricians offered their parents an educational programme from the child's birth to the age of two years (intervention arm) with 216 newborns whose parents did not undergo the programme (control arm). This sample size was 80% powerful to detect, with a 0.05 α error, a 40% lower prevalence of overweight/obesity and a 57% lower prevalence of obesity in the intervention arm. At each well visit, the parents of the children in the intervention arm were given oral and written information about protective behaviours, with particular emphasis on responsive feeding. Overweight and obesity at two years of age were, respectively, defined as a body mass index of more than the 85th and the 95th percentile in accordance with the WHO growth charts. The sample size had 80% power to detect a 40% lower prevalence of overweight/obesity and a 57% lower prevalence of obesity in the intervention arm. RESULTS: At the age of two years, the prevalence of obesity in the intervention arm was 35% lower than among the controls, but the difference was not statistically significant (8.7% vs. 13.4%; p = 0.10) There was no difference in the prevalence of overweight/obesity between the groups (26.8% vs. 28.3%; p = 0.49). At the age of three months, a higher proportion of the infants in the intervention group were fed on demand (93% vs. 80%, p < 0.001). CONCLUSIONS: The PROBIT trial failed to detect a significantly lower prevalence of obesity in the intervention arm, but did improve early feeding patterns. More powerful trials and meta-analyses are required to establish whether educating newborns' parents can decrease the prevalence of early obesity.

BACKGROUND: Science curricula and teachers should emphasize evolution in a manner commensurate with its importance as a unifying concept in science. The concept of adaptation represents a first step to understand the results of natural selection. We settled an experimental project of alternative didactic to improve knowledge of organism adaptation. Students were involved and stimulated in learning processes by creative activities. To set adaptation in a historic frame, fossil records as evidence of past life and evolution were considered. RESULTS: The experimental project is schematized in nine phases: review of previous knowledge; lesson on fossils; lesson on fantastic animals; planning an imaginary world; creation of an imaginary animal; revision of the imaginary animals; adaptations of real animals; adaptations of fossil animals; and public exposition. A rubric to evaluate the student's performances is reported. The project involved professors and students of the University of Modena and Reggio Emilia and of the "G. Marconi" Secondary School of First Degree (Modena, Italy). CONCLUSION: The educational objectives of the project are in line with the National Indications of the Italian Ministry of Public Instruction: knowledge of the characteristics of living beings, the meanings of the term "adaptation", the meaning of fossils, the definition of ecosystem, and the particularity of the different biomes. At the end of the project, students will be able to grasp particular adaptations of real organisms and to deduce information about the environment in which the organism evolved. This project allows students to review previous knowledge and to form their personalities.
