BACKGROUND: Sparse research exists on predictors of element concentrations measured in deciduous teeth. OBJECTIVE: To estimate associations between maternal/child characteristics, elements measured in home tap water during pregnancy and element concentrations in the dentin of shed deciduous teeth. METHODS: Our analysis included 152 pregnant person-infant dyads followed from the second trimester through the end of the first postnatal year from the New Hampshire Birth Cohort Study. During pregnancy and early infancy, we collected dietary and sociodemographic information via surveys, measured elements in home tap water, and later collected naturally exfoliated teeth from child participants. We measured longitudinal deposition of elements in dentin using LA-ICP-MS. Multivariable linear mixed models were used to estimate associations between predictors and dentin element concentrations. RESULTS: We measured 12 elements in dentin including those previously reported (Ba, Mn, Pb, Sr, Zn) and less frequently reported (Al, As, Cd, Cu, Hg, Li, and W). A doubling of Pb or Sr concentrations in water was associated with higher dentin Pb or Sr respectively in prenatally formed [9% (95%CI: 3%, 15%); 3% (1%, 6%)] and postnatally formed [10% (2%, 19%); 6% (2%, 10%)] dentin. Formula feeding from birth to 6 weeks or 6 weeks to 4 months was associated with higher element concentrations in postnatal dentin within the given time period as compared to exclusive human milk feeding: Sr: 6 weeks: 61% (36%, 90%) and 4 months: 85% (54%, 121%); Ba: 6 weeks: 35% (3.3%, 77%) and 4 months: 42% (10%, 83%); and Li: 6 weeks: 61% (33%, 95%) and 4 months: 58% (31%, 90%). SIGNIFICANCE: These findings offer insights into predictors of dentin elements and potential confounders in exposure-health outcome relationships during critical developmental periods.

BACKGROUND: Spatial elemental analysis of deciduous tooth dentin combined with odontochronological estimates can provide an early life (in utero to ~2 years of age) history of inorganic element exposure and status. OBJECTIVE: To demonstrate the importance of data normalization to a certified reference material to enable between-study comparisons, using populations with assumed contrasting elemental exposures. METHODS: We used laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) of dentin to derive a history of elemental composition from three distinct cohort studies: a present day rural cohort, (the New Hampshire Birth Cohort Study (NHBCS; N = 154)), an historical cohort from an urban area (1958-1970), (the St. Louis Baby Tooth Study (SLBT; N = 78)), and a present-day Nigerian cohort established to study maternal HIV transmission (Dental caries and its association with Oral Microbiomes and HIV in young children-Nigeria (DOMHaIN; N = 31)). RESULTS: We report Li, Al, Mn, Cu, Zn, Sr, Ba and Pb concentrations (µg/g) and qualitatively examine As, Cd and Hg across all three cohorts. Rates of detection were highest, both overall and for each cohort individually, for Zn, Sr, Ba and Li. Zinc was detected in 100% of samples and was stably present in teeth at a concentration range of 64 - 86 µg/g. Mercury, As and Cd detection rates were the lowest, and had high variability within individual ablated spots. We found the highest concentrations of Pb in the pre- and postnatal dentin of the SLBT cohort, consistent with the prevalent use of Pb as an additive to gasoline prior to 1975. The characteristic decline in Mn after the second trimester was observed in all cohorts. IMPACT: Spatially resolved elemental analysis of deciduous teeth combined with methods for estimating crown formation times can be used to reconstruct an early-life history of elemental exposure inaccessible via other biomarkers. Quantification of data into absolute values using an external standard reference material has not been conducted since 2012, preventing comparison between studies, a common and highly informative component of epidemiology. We demonstrate, with three contrasting populations, that absolute quantification produces data with the lowest variability, compares well with available data and recommends that future tooth biomarker studies report data in this way.

Nanoscale thin film coatings and surface treatments are ubiquitous across industry, science, and engineering; imbuing specific functional or mechanical properties (such as corrosion resistance, lubricity, catalytic activity and electronic behaviour). Non-destructive nanoscale imaging of thin film coatings across large (ca. centimetre) lateral length scales, crucial to a wide range of modern industry, remains a significant technical challenge. By harnessing the unique nature of the helium atom-surface interaction, neutral helium microscopy images these surfaces without altering the sample under investigation. Since the helium atom scatters exclusively from the outermost electronic corrugation of the sample, the technique is completely surface sensitive. Furthermore, with a cross-section that is orders of magnitude larger than that of electrons, neutrons and photons, the probe particle routinely interacts with features down to the scale of surface defects and small adsorbates (including hydrogen). Here, we highlight the capacity of neutral helium microscopy for sub-resolution contrast using an advanced facet scattering model based on nanoscale features. By replicating the observed scattered helium intensities, we demonstrate that sub-resolution contrast arises from the unique surface scattering of the incident probe. Consequently, it is now possible to extract quantitative information from the helium atom image, including localised ångström-scale variations in topography.

Liver fibrosis plays a critical role in the evolution of most chronic liver diseases and is characterized by a buildup of extracellular matrix, which can progress to cirrhosis, hepatocellular carcinoma, liver failure, or death. Now, there are no noninvasive methods available to accurately assess disease activity (fibrogenesis) to sensitively detect early onset of fibrosis or to detect early response to treatment. Here, we hypothesized that extracellular allysine aldehyde (LysAld) pairs formed by collagen oxidation during active fibrosis could be a target for assessing fibrogenesis with a molecular probe. We showed that molecular magnetic resonance imaging (MRI) using an extracellular probe targeting these LysAld pairs acts as a noninvasive biomarker of fibrogenesis and demonstrated its high sensitivity and specificity in detecting fibrogenesis in toxin- and dietary-induced mouse models, a cholestasis rat model of liver fibrogenesis, and in human fibrotic liver tissues. Quantitative molecular MRI was highly correlated with fibrogenesis markers and enabled noninvasive detection of early onset fibrosis and response to antifibrotic treatment, showing high potential for clinical translation.

RESUMO

A growing body of evidence demonstrates that Atlantic-style passive margins have experienced episodes of uplift and volcanism in response to changes in mantle circulation long after cessation of rifting. Passive margins are thus an attractive archive from which to retrieve records of mantle circulation and lithospheric alteration. However, this archive remains under-utilized due to difficulty in deciphering the surficial records of passive margin tectonism and linking them to seismic velocity structure. Here we present a new approach to unraveling the tectonic history of passive margins using U-Pb dating of calcite in faults and fractures along the eastern North American margin. These ages show a 40 Myr long period of continuous fracturing and faulting from ~115 to 75 Ma followed by another episode in Mio-Pliocene time. We argue that the former event represents a response to Cretaceous lithospheric alteration whereas the latter records development of modern relief in the northern Appalachians.

RESUMO

In response to the requirements imposed by the COVID-19 pandemic in 2020, we developed a remote learning undergraduate workshop for 44 students at the University of Newcastle by embedding scanning electron microscope (SEM) images of Maratus (Peacock) spiders into the MyScope Explore environment. The workshop session had two main components: 1) to use the online MyScope Explore tool to virtually image scales with structural color and pigmented color on Maratus spiders; 2) to join a live SEM session via Zoom to image an actual Maratus spider. In previous years, the undergraduate university students attending this annual workshop would enter the Microscopy Facility at the University of Newcastle to image specimens with SEM; however, in 2020 the Microscopy Facility was closed to student visitors, and this virtual activity was developed in order to proceed with the educational event. The program was highly successful and constitutes a platform that can be used in the future by universities for teaching microscopy remotely.

A modified synthesis method for aqueous nanoparticle printing inks, based upon vacuum-assisted solvent removal, is reported. Poly(3-hexylthiophene):phenyl C61 butyric acid methyl ester nanoparticle inks were prepared via this modified miniemulsion method, leading to both an improvement in photoactive layer morphology and a substantial reduction in the ink fabrication time. A combination of UV-visible spectroscopy, photoluminescence spectroscopy and scanning transmission X-ray microscopy measurements revealed a nanoparticle morphology comprising highly intermixed donor-acceptor domains. Consistent with these measurements, dynamic mechanical thermal analysis of the nanoparticles showed a glass transition temperature (Tg) of 104 °C, rather than a pure polymer phase or pure fullerene phase Tg. Together the spectroscopy, microscopy and thermomechanical data indicate that rapid solvent removal generates a more blended nanoparticle morphology. As such, this study highlights a new experimental lever for optimising nanostructure in the photoactive layer of nanoparticulate organic photovoltaic devices by enabling highly intermixed donor-acceptor architectures to be built from customised nanoparticulate inks.

The field of taxonomy is critically important for the identification, conservation, and ecology of biological species. Modern taxonomists increasingly need to employ advanced imaging techniques to classify organisms according to their observed morphological features. Moreover, the generation of three-dimensional datasets is of growing interest; moving beyond qualitative analysis to true quantitative classification. Unfortunately, biological samples are highly vulnerable to degradation under the energetic probes often used to generate these datasets. Neutral atom beam microscopes avoid such damage due to the gentle nature of their low energy probe, but to date have not been capable of producing three-dimensional data. Here we demonstrate a means to recover the height information for samples imaged in the scanning helium microscope (SHeM) via the process of stereophotogrammetry. The extended capabilities, namely sparse three-dimensional reconstructions of features, were showcased via taxonomic studies of both flora (Arabidopsis thaliana) and fauna (Heterodontus portusjacksoni). In concert with the delicate nature of neutral helium atom beam microscopy, the stereophotogrammetry technique provides the means to derive comprehensive taxonomical data without the risk of sample degradation due to the imaging process.

Barr, M, Beaver, T, Turczyn, D, and Cornish, S. Validity and reliability of 15 Hz global positioning system units for assessing the activity profiles of university football players. J Strength Cond Res 33(5): 1371-1379, 2019-Global positioning system (GPS) units have recently become popular for monitoring and assessing the workloads of football players. Currently, there is a lack of studies examining the validity and reliability of these systems for that purpose, so the aim of the current study was to determine whether 15 Hz units (SPI HPU; GPSports, Canberra, Australia) could accurately be used to describe the physical demands of football. A series of cohort studies were conducted with Canadian university football players (n = 28). To assess the accuracy of the units' ability to measure high-velocity sprinting, 12 players performed multiple electronically timed 36.6 m sprints while wearing the units. To assess the interunit reliability, 5 players wore 2 units each during a training session. An analysis of the units' validity for measuring collisions was performed by comparing the correct number of tackles and blocks notated on video by an expert rater in 2 games with the number of collisions recorded by the units. The units were accurate for measuring high-sprinting velocities (coefficient of variation [CV] = 0.9%) and had good interunit reliability for recording distances at velocities between walking and sprinting CV (1.4-7.8%). The collision algorithm filter the accompanying software uses was found to have its best balance between precision and recall using a cut-off of 2.65g for linemen and 2.9g for nonlinemen. The devices used are effective at providing acceptably valid and reliable information to describe the physical demands of football. Position-specific locomotor zones are recommended when using GPS units with football players.

-Sprinting speed and lower-body power are considered to be key physical abilities for rugby players. A method of improving the lower-body power of athletes is simulated hypergravity. This method involves wearing a weighted vest at all times during the day for an extended period of time. There are no studies that have examined the effect of hypergravity on speed or the benefit for rugby players. An experimental group (n = 8) and a control group (n = 7) of national team rugby players took part in the study, which consisted of rugby, conditioning, speed, and strength sessions. The experimental group wore a weighted vest equating to 12% of their body mass for 8 days. All players were tested for speed and lower-body power before, 2 days after, and 9 days after the intervention. Speed testing involved the athletes completing 40-m sprints with timing lights and high-speed video cameras assessing acceleration and maximal velocity sprinting kinematics. Lower-body power was assessed using weighted countermovement jumps (CMJs). No group differences were found for sprinting speed at any point. The experimental group displayed a large decrease in acceleration ground contact time (-0.01 ± 0.005 s, d = 1.07) and a moderate increase in 15-kg CMJ velocity (0.07 ± 0.11 m·s, d = 0.71). Individual responses showed that players in the experimental group had both negative and positive speed and power responses to the training intervention. Simulated hypergravity for 8 days is likely ineffective at improving sprinting speed while undergoing standard rugby training.

Increasing lower-body strength is often considered to be important for improving the sprinting speed of rugby players. This concept was examined in a group (n = 40) of international rugby players in a 2-part study. The players were tested for body mass (BM), 1 repetition maximum power clean (PC), and front squat, as well as triple broad jump and broad jump. In addition, speed over 40 m was tested, with timing gates recording the 0- to 10-m and 30- to 40-m sections to assess acceleration and maximal velocity. Two video cameras recorded the 2 splits for later analysis of sprinting kinematics. The players were divided into a fast group (n = 20) and a slow group (n = 20) for both acceleration and maximal velocity. In the second part of the study, a group (n = 15) of players were tracked over a 1-year period to determine how changes in strength corresponded with changes in sprinting kinematics. The fast groups for both acceleration and maximal velocity showed greater levels of strength (d = 0.5-1.8), lower ground contact times (d = 0.8-2.1), and longer stride lengths (d = 0.5-1.3). There was a moderate improvement over 1 year in PC/BM (0.08 kg·kg, p = 0.008, d = 0.6), and this had a strong relationship with the change in maximal velocity stride length (r = 0.70). Acceleration stride length also had a large improvement over 1 year (0.09 m, p = 0.003, d = 0.81). Although increasing lower-body strength is likely important for increasing sprinting speed of players with low training backgrounds, it may not have the same effect with highly trained players.

Speed and sprint momentum are considered to be important physical qualities for rugby. The purpose of the study was to understand the development of these qualities in senior and junior international rugby players. In part 1 of the study, a group of senior (n = 38) and junior (n = 31) players were tested for speed over 40 m. Initial sprint velocity (ISV), maximal sprint velocity (MSV), initial sprint momentum (ISM), and maximal sprint momentum (MSM) were calculated using 10-m splits. In part 2 of the study, a group of junior (n = 12) and senior (n = 15) players were tracked over a 2-year period for body mass, ISV, MSV, ISM, and MSM. In part 1, senior backs and forwards were not found to have significantly greater ISV and MSV than junior players but were found to have greater ISM and MSM. Forwards were found to have significantly greater ISM and MSM than backs but significantly lower ISV and MSV than backs. In part 2, no significant differences were found over the 2 years between senior and junior players, but greater effect sizes for juniors were generally found when compared with seniors for improvements in ISV (d = 0.73 vs. 0.79), MSV (d = 1.09 vs. 0.68), ISM (d = 0.96 vs. 0.54), and MSM (d = 1.15 vs. 0.50). Sprint momentum is a key discriminator between senior and junior players, and large changes can be made by junior players as they transition into senior rugby. Speed appears to peak for players in their early 20s but sprint momentum appears to be more trainable.

A common suggestion is that a predetermined level of maximal leg strength is required before drop jump (DJ) training can begin. This study sought to examine the relationship between maximal squat strength (1 repetition maximum [RM]) and DJ performance in 15 female rugby players (n = 15). The subjects were tested for 1RM, countermovement jump, squat jump, and DJs from 0.24, 0.36, 0.48, 0.60, 0.72, and 0.84 m. Jump height (JH) was calculated for all jumps and relative peak eccentric force, relative peak concentric force, ground contact time (GCT), and reactive strength index were also calculated for DJs. Pearson correlations were used to examine the relationship between 1RM relative to body mass (BM) (1RM/BM) and JHs, reactive strength index, and GCT during DJs. The subjects were placed in a high strength (HS) or low strength (LS) group depending on whether or not their 1RM/BM was >1 or <1. The T-tests and 2-way analysis of variance (ANOVA) were used to compare the groups. A Fishers post hoc test was used for the ANOVA with significance set at p < 0.05. A large correlation between JH and 1RM/BM was shown at the 0.84-m dropping height (r = 0.56). A significant overall difference was found between the HS and LS groups for DJ JH with a post hoc analysis revealing a significant difference at the 0.84-m drop height (p = 0.029). It is likely beneficial for female athletes to achieve high levels of maximal leg strength if they are going to use high (>0.8-m) drop heights when performing DJs.

Here, we demonstrate the simple fabrication of a single-walled carbon nanotube (SWCNT) field emission electrode which shows excellent field emission characteristics and remarkable field emission stability without requiring posttreatment. Chemically functionalized SWCNTs were chemically attached to a silicon substrate. The chemical attachment led to vertical alignment of SWCNTs on the surface. Field emission sweeps and Fowler-Nordheim plots showed that the Si-SWCNT electrodes field emit with a low turn-on electric field of 1.5 V µm-1 and high electric field enhancement factor of 3,965. The Si-SWCNT electrodes were shown to maintain a current density of >740 µA cm-2 for 15 h with negligible change in applied voltage. The results indicate that adhesion strength between the SWCNTs and substrate is a much greater factor in field emission stability than previously reported.

We present an approach to predicting extrinsic electron resonance widths within quantum corral nanostructures based on analogies with acoustics. Established quantum mechanical methods for calculating resonance widths, such as multiple scattering theory, build up the scattering atom by atom, ignoring the structure formed by the atoms, such as walls or enclosures. Conversely, particle-in-a-box models, assuming continuous walls, have long been successful in predicting quantum corral energy levels, but not resonance widths. In acoustics, partial reflection from walls and various enclosures has long been incorporated for determining reverberation times. Pursuing an exact analogy between the local density of states of a quantum corral and the acoustic impedance of a concert hall, we show electron lifetimes in nanoscopic structures of arbitrary convex shape are well accounted for by the Sabine formula for acoustic reverberation times. This provides a particularly compact and intuitive prescription for extrinsic finite lifetimes in a particle-in-a-box with leaky walls, including quantum corral atomic walls, given single particle scattering properties.
