RESUMO

The small source sizes of third-generation synchrotron sources are ideal for the production of microbeams for diffraction studies of crystalline and non-crystalline materials. While several such facilities have been available around the world for some time now, few have been optimized for the handling of delicate soft-tissue specimens under cryogenic conditions. Here the development of a new X-ray micro-diffraction instrument at the Biophysics Collaborative Access Team beamline 18-ID at the Advanced Photon Source, and its use with newly developed cryo-diffraction techniques for soft-tissue studies, are described. The combination of the small beam sizes delivered by this instrument, the high delivered flux and successful cryo-freezing of rat-tail tendon has enabled us to record data to better than 4 Šresolution. The ability to quickly raster scan samples in the beam allows selection of ordered regions in fibrous samples for markedly improved data quality. Examples of results of experiments obtainable using this instrument are presented.

RESUMO

We have developed a compact short focal distance Bent Crystal Laue Analyzer (BCLA) for Cu speciation studies of biological systems with specific applications to cancer biology. The system provides high energy resolution and high background rejection. The system is composed of an aluminum block serving as a log spiral bender for a 15 micron thick Silicon 111 crystal and a set of soller slits. The energy resolution of the BCLA-about 14 eV at the Cu Kα line- allows resolution of the Cu Kα(1) and CuKα(2) lines. The system is easily aligned by using a set of motorized XYZ linear stages. Two operation modes are available: incident energy scans (IES) and emission energy scans (EES). IES allows scanning of the incident energy while the BCLA system is maintained at a preselected fixed position--typically CuKα(1) line. EES is used when the incident energy is fixed and the analyzer is scanned to provide the peak profile of the emission lines of Cu.

RESUMO

There is a growing interest in the biomedical community in obtaining information concerning the distribution and local chemical environment of metals in tissues and cells. Recently, biological X-ray fluorescence microscopy (XFM) has emerged as the tool of choice to address these questions. A fast-scanning high-flux X-ray microprobe, built around a recently commissioned pair of 200 mm-long Rh-coated silicon Kirkpatrick-Baez mirrors, has been constructed at BioCAT beamline 18ID at the Advanced Photon Source. The new optical system delivers a flux of 1.3 x 10(12) photons s(-1) into a minimum focal spot size of approximately 3-5 microm FWHM. A set of Si drift detectors and bent Laue crystal analyzers may be used in combination with standard ionization chambers for X-ray fluorescence measurements. BioCAT's scanning software allows fast continuous scans to be performed while acquiring and storing full multichannel analyzer spectra per pixel on-the-fly with minimal overhead time (<20 ms per pixel). Together, the high-flux X-ray microbeam and the rapid-scanning capabilities of the BioCAT beamline allow the collection of XFM and micro X-ray absorption spectroscopy (microXAS) measurements from as many as 48 tissue sections per day. This paper reports the commissioning results of the new instrument with representative XFM and microXAS results from tissue samples.

This work presents a study of elemental composition and preponderant structure of human dental calculus, as they mature in the mouth for a period of 1 year. The synchrotron radiation X-ray fluorescence technique using a white beam was employed as an analytical method. The set of samples were extracted from different dental elements of the same subject, who did not require any other clinical care. By means of analyzing the Ca/P molar ratio an estimation of the main crystallographic structure was attained, by simple comparison with stoichiometric values of the several crystalline structures that compose the calculus. The results showed a slowly progressive transformation of the initial crystalline structures (brushite) into more stable structures (hydroxyapatite), passing through octacalcium phosphate and whitlockite. The concentrations of mayor components (Ca and P) as a function of time followed a sigmoid curve. The analysis of trace element concentrations versus time indicated a null or small correlation of concentration values with the kinetics of the crystallization process.

RESUMO

The 18ID undulator beamline of the Biophysics Collaborative Access Team at the Advanced Photon Source, Argonne, IL, USA, is a high-performance instrument designed for, and dedicated to, the study of partially ordered and disordered biological materials using the techniques of small-angle X-ray scattering, fiber diffraction, and X-ray absorption spectroscopy. The beamline and associated instrumentation are described in detail and examples of the representative experimental results are presented.

RESUMO

A multiwire proportional counter was used in fluorescence X ray absorption measurements and a comparison to a Si(Li) and NaI(Tl) detectors was done. The main features of the multiwire proportional counter are its high counting rate capability (10(7)) counts x s(-1)) and large active area (6 x 6 cm2). It was shown that the MWPC is suitable for fluorescence absorption. Although the maximum capability was not reached in the present experiments, it was found that as the counting rate increase the MWPC performance became better than Si(Li) detectors and shows a similar response to the scintillator counter at medium counting rates (up to 10(5) counts x s(-1)).

RESUMO

We present here the first study of the local environment of zinc ions in biological calcium phosphates. It was suggested from in vitro studies that zinc inhibits the formation of hydroxyapatite and promotes the formation of more soluble phases, like tricalcium phosphate. Several mechanisms of zinc - calcium phosphate interaction were proposed, yielding either to the adsorption or to the incorporation of zinc ions into the phosphate structure. The results obtained here show that, under in vivo conditions, the zinc atoms are fully incorporated into the crystalline structure of the calcium phosphates.

RESUMO

The LNLS XAS beamline has been operating for external users since July 1997. Many facilities and improvements have been progressively added to it, extending the range of applications. Here, a technical description of the main beamline components is given, and results concerning important points, such as available flux at low and high energies, harmonic contamination, energy resolution and stability, are presented. Some key results are given to demonstrate the beamline performance and limitations. It is shown that the beamline can cover a large energy range, starting from the rather low energy of 2.3 keV up to 25 keV.
