The study investigated removal of hexavalent chromium Cr (VI) from aqueous solution using graphene oxide­iron oxide reinforced pectin/polyvinyl alcohol magnetic gel beads prepared through co-precipitation and freeze-drying technique. Scanning electron microscopy, X-ray photoelectron spectroscopy, Fourier-transform infrared spectroscopy, X-ray diffraction, vibrating sample magnetometer, N2 adsorption-desorption isotherm, and zeta potential are used for characterization. The surface area of magnetic gel beads calculated by BET method was determined to be 100.95 m2/g, significantly higher than that of GO and GO-Fe3O4. The optimum removal efficiency of GO-Fe3O4/Pec/PVA was assessed by batch method at variables such as pH(1-6), adsorption time(0-180 min), and temperature(25-35 °C). Accordingly, 0.2 g GO-Fe3O4/Pec/PVA dose, pH 2, contact time: 120 min at 25 °C were found to be the optimal conditions, and maximum adsorption capacity of GO, GO-Fe3O4 and GO-Fe3O4/Pec/PVA toward Cr(VI) removal was found to be 39.5, 62.5 and 78.55 mg g-1, respectively. Kinetic and isotherm studies indicate adsorption data follow pseudo-second-order kinetic and Langmuir isotherm models. Thermodynamic studies showed adsorption capacities of adsorbents decreased when temperature increased which indicated adsorption for Cr (VI) was an exothermic process. The activation energies were found to be 34.92, 26.57, and 35.23 KJ mol-1 for GO, GO-Fe3O4, and GO-Fe3O4/Pec/PVA, respectively, which illustrated adsorption of Cr(VI) onto the surface of adsorbents was a physical process. The beads exhibit excellent recoverability and reusability over five cycles. Overall, GO-Fe3O4/Pec/PVA demonstrates exceptional adsorption properties and can serve as an efficient, stable, less toxic, and magnetically separable adsorbent for removal of Cr(VI) from aqueous solution.

RESUMO

The advent of acute respiratory coronavirus disease (COVID-19) is convoyed by the shedding of the virus in stool. Although inhalation from person-to-person and aerosol/droplet transmission are the main modes of SARS-Coronavirus-2 (SARS-CoV-2) transmission, currently available evidence indicates the presence of viral RNA in the sewerage wastewater, which highlights the need for more effective corona virus treatment options. In the existing COVID-19 pandemic, a substantial percentage of cases shed SARS-CoV-2 viral RNA in their faeces. Hence the treating this sewerage wastewater with proper surveillance is essential to contain this deadly pathogen from further transmission. Since, the viral disinfectants will not be very effective on sewerage waste as organic matter, and suspended solids in water can protect viruses that adsorb to these particles. More effective methods and measures are needed to prevent this virus from spreading. This review will explore some potential methods to treat the SARS-CoV-2 infected sewerage wastewater, current research and future directions.

Overexpression of pro-inflammatory cytokines and iNOS have been found to be concomitant with several chronic inflammatory diseases and hence targeting their inhibition would be a useful therapy for inflammation. In view of this, study on discovery of natural pro-inflammatory cytokines inhibitory lead molecules from Penicillium polonicum, an endophytic fungus isolated from the fresh fruits of Piper nigrum was performed. When the culture broth extract of P. polonicum (EEPP) was subjected to LPS-induced cytokines expression (ELISA in RAW 264.7 cells), it exhibited inhibition of TNF-α, IL-6 and IL-1ß and this encouraged us to do chemical investigation on EEPP to explore the bioactive components. Four compounds isolated and characterised as 3,5-di-tert-butyl-4-hydroxy-phenyl propionic acid (1), 2,4-di-tert-butyl phenol (2), indole 3-carboxylic acid (3) and tyrosol (4) were tested for their effect on the production of TNF-α, IL-1ß and IL-6 in RAW 264.7 cells (ELISA). All the compounds exhibited a highly significant (P < 0.0001) inhibition effect, particularly against IL-1ß (IC50: 4-0.91 µM, 1-2.81 µM, 3-4.38 µM, and 2-5.54 µM). Tyrosol (4) was most active with IC50 values of 0.91, 2.67 and 4.60 µM against IL-1ß, IL-6 and TNF-α, respectively. On observing the potential activity of the compounds, two compositions C1 and C2 were prepared by mixing equimolar concentrations of compounds 1, 2, 3 & 4 (C1) and compounds 1, 2, 3, 4 & piperine (C2) in equal ratio. A synergistic effect was observed with C1 exhibiting potential suppression of IL-6 secretion (IC50 1.91 µM) and C2 against IL-1ß (IC50 5.98 µM). Also, the individual compounds and C1 were effective in controlling iNOS expressions in RAW 264.7 cells (RTPCR). Further, the in vivo performance of the compounds and compositions were studied under two in vivo inflammatory models (LPS-induced endotoxaemia and carrageenan-induced paw oedema). Compounds 1, 2, 3, 4, C1 and C2 at 50 mg/kg oral dose showed a significant control over the LPS-stimulated TNF-α, IL-1ß and IL-6 levels in plasma. C1, C2 and 1 exhibited > 50% pan-cytokine inhibition effect. Under the carrageenan-induced anti-inflammatory model, a significant reduction in the paw oedema measured in terms of the difference in the paw thickness was observed. Further, attenuation of pro-inflammatory cytokines levels following ELISA and RT-PCR experiments in the paw tissue homogenate was in agreement with paw thickness results. All compounds and C1 decreased the iNOS gene expression levels, and also the MPO activity and NO production in the paw tissue homogenate with tyrosol (4) as the most active molecule. Further, the mechanism of action was explored by testing the effect of the compounds on the expression of inflammatory markers using western blot analysis (in vitro). They were found to regulate the expression of pro-form and matured-form of IL-1ß by inhibiting NFκB. Also, the compounds reduced the translocation of the NF-κB subunit p65 to the nucleus. Thus, compounds 3,5-di-tert-butyl-4-hydroxy-phenyl propionic acid (1), 2,4-di-tert-butyl phenol (2), indole 3-carboxylic acid (3) and tyrosol (4) are reported as new natural multiple pro-inflammatory cytokines inhibitory leads. The interesting results of C1 might lay a footing for the development of a new anti-inflammatory composition.

A series of new phosphorylated derivatives of didanosine were designed, synthesized and evaluated their anticancer effects on human breast cancer cells. Their binding affinities were evaluated against aromatase enzyme and the molecular docking studies demonstrated that 9a, 9h and 9i exhibited high binding interactions than the parent molecule (ddI) and other derivatives; evaluated the aromatase enzyme inhibition. The cell viability, cell proliferation, lactate dehydrogenase showed potential anti-proliferative in dose dependent manner, these results were well correlated with hoesch stain and DNA fragmentation on MDA-MB-231 breast cancer cell lines. Cytotoxicity results disclosed that tryptophan amino acid ester substituted derivative 9i showed potential cell death against MDA-MB-231 cancer cell lines. Furthermore, compound 9i has great potential significance for further investigations (in vivo).

In recent years, diabetic retinopathy (DR) needs to be focused with the intention of developing accurate and effective approaches by accomplishing the existing challenges in the traditional models. With this objective, this paper aims to introduce an effective diagnosis system by utilizing retinal fundus images. The implementation of this diagnosis model incorporates 4 stages like (i) preprocessing, (ii) blood vessel segmentation, (iii) feature extraction, as well as (iv) classification. Originally, the median filter as well as contrast limited adaptive histogram equalization (CLAHE) help to preprocess the image. Moreover, the Fuzzy C Mean (FCM) thresholding is applied for blood vessel segmentation, which generates stochastic clustering of pixels to obtain enhanced threshold values. Further, feature extraction is accomplished by utilizing gray-level run-length matrix (GLRM), local, and morphological transformation-based features. Furthermore, a deep learning (DL) model known as convolutional neural network (CNN) is employed for the diagnosis or classification purpose. As a main novelty, this paper introduces an optimal feature selection as well as classification model. Further, the feature selection is done optimally by FireFly Migration Operator-based Monarch Butterfly Optimization (FM-MBO) which hybridized of the monarch butterfly optimization (MBO) and fire fly (FF) algorithms as the entire adopted extracted features attain higher feature length. Moreover, the proposed FM-MBO algorithm helps for optimizing the count of CNN's convolutional neurons to further improve the performance accuracy. At the end, the enhanced outcomes of the adopted diagnostic scheme are validated via a valuable comparative examination in terms of significant performance measures.

The present work aims at evaluating the in vitro biocompatibility, antibacterial activity and antioxidant capacity of the fabricated and optimized Alginate/Chitosan nanoparticles (ALG/CSNPs) and quercetin loaded Alginate/Chitosan nanoparticles (Q-ALG/CSNPs) with an improved biological efficacy on the hydrophobic flavonoid.The physicochemical properties were determined by TEM and FTIR analysis. The nanoparticles evaluated for the encapsulation of quercetin exerted % encapsulation efficiency (EE) that varied between 76 and 82.4 % and loading capacity (LC) from 31 to 46.5 %. Potential cytotoxicity of the ALG/CSNPs and Q-ALG/CSNPs upon L929 fibroblast cell line was evaluated by MTT reduction Assay and expressed as % cell viability. The in vitro antibacterial property was studied by well diffusion method against gram-positive bacteria Staphylococcus aureus (ATCC 25925) and gram-negative bacteria Escherichia coli (ATCC 25923). The inhibitory efficacy by scavenging free radical intermediates was evaluated by 1,1, diphenyl 2-picrylhydrazyl (DPPH) assay. The results of in vitro cytotoxicity showed biocompatibility towards L929 cells. Quercetin loaded Alginate/Chitosan nanoparticles inhibited the growth of microorganisms than pure quercetin. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging results have shown a high level of antioxidant property for encapsulated Quercetin in Alginate/Chitosan nanoparticles compared to free Quercetin. The findings of our study suggest that the developed ALG/CSNPs and Q-ALG/CSNPs possess the prerequisites and be proposed as a suitable system for delivering quercetin with enhanced therapeutic effectuality.

We propose a novel video sampling scheme for human action recognition in videos, using Gaussian Weighing Function. Traditionally in deep learning-based human activity recognition approaches, either a few random frames or every k t h frame of the video is considered for training the 3D CNN, where k is a small positive integer, like 4, 5, or 6. This kind of sampling reduces the volume of the input data, which speeds-up the training network and also avoids overfitting to some extent, thus enhancing the performance of the 3D CNN model. In the proposed video sampling technique, consecutive k frames of a video are aggregated into a single frame by computing a Gaussian-weighted summation of the k frames. The resulting frame preserves the information in a better way than the conventional approaches and experimentally shown to perform better. In this paper, a 3-Dimensional deep CNN is proposed to extract the spatio-temporal features and follows Long Short-Term Memory (LSTM) to recognize human actions. The proposed 3D CNN architecture is capable of handling the videos where the camera is placed at a distance from the performer. Experiments are performed with KTH, WEIZMANN, and CASIA-B Human Activity and Gait datasets, whereby it is shown to outperform state-of-the-art deep learning based techniques. We achieve 95.78%, 95.27%, and 95.27% over the KTH, WEIZMANN, and CASIA-B human action and gait recognition datasets, respectively.

Silica-supported lanthanum (III) chloride (SiO2-LaCl3·7H2O) was prepared and characterized by infrared spectroscopy, X-ray diffraction analysis, scanning electron microscope, energy-dispersive X-ray spectroscopy, thermogravimetric analysis and differential thermal analysis techniques. The catalytic activity of this silica-supported lanthanum (III) chloride was investigated in a one-pot three-component Kabachnik-Fields reaction. A library of new α-aminophosphonates was prepared employing various benzothiazole and thiadiazole amines, different substituted aldehydes and diethylphosphite under solvent-free conditions using conventional/microwave methods with good to excellent yields (85-97%). The advantages of this catalyst are that it is environmentally benign, economically inexpensive, and easy to prepare, gives high yields and high purity is less time-consuming, offers easy purification is reusable and enables products to be obtained by simple recrystallization without column chromatography.

This paper proposes an Information Bottleneck theory based filter pruning method that uses a statistical measure called Mutual Information (MI). The MI between filters and class labels, also called Relevance, is computed using the filter's activation maps and the annotations. The filters having High Relevance (HRel) are considered to be more important. Consequently, the least important filters, which have lower Mutual Information with the class labels, are pruned. Unlike the existing MI based pruning methods, the proposed method determines the significance of the filters purely based on their corresponding activation map's relationship with the class labels. Architectures such as LeNet-5, VGG-16, ResNet-56, ResNet-110 and ResNet-50 are utilized to demonstrate the efficacy of the proposed pruning method over MNIST, CIFAR-10 and ImageNet datasets. The proposed method shows the state-of-the-art pruning results for LeNet-5, VGG-16, ResNet-56, ResNet-110 and ResNet-50 architectures. In the experiments, we prune 97.98%, 84.85%, 76.89%, 76.95%, and 63.99% of Floating Point Operation (FLOP)s from LeNet-5, VGG-16, ResNet-56, ResNet-110, and ResNet-50 respectively. The proposed HRel pruning method outperforms recent state-of-the-art filter pruning methods. Even after pruning the filters from convolutional layers of LeNet-5 drastically (i.e., from 20, 50 to 2, 3, respectively), only a small accuracy drop of 0.52% is observed. Notably, for VGG-16, 94.98% parameters are reduced, only with a drop of 0.36% in top-1 accuracy. ResNet-50 has shown a 1.17% drop in the top-5 accuracy after pruning 66.42% of the FLOPs. In addition to pruning, the Information Plane dynamics of Information Bottleneck theory is analyzed for various Convolutional Neural Network architectures with the effect of pruning. SVEP1, also known as Polydom, is a large extracellular mosaic protein with functions in protein interactions and adhesion. Since Svep1 knockout animals show severe edema and lymphatic system malformations, the aim of this study is to evaluate the presence of SVEP1 variants in patients with lymphedema. We analyzed DNA from 246 lymphedema patients for variants in known lymphedema genes, 235 of whom tested negative and underwent a second testing for new candidate genes, including SVEP1, as reported here. We found three samples with rare heterozygous missense single-nucleotide variants in the SVEP1 gene. In one family, healthy members were found to carry the same variants and reported some subclinical edema. Based on our findings and a review of the literature, we propose SVEP1 as a candidate gene that should be sequenced in patients with lymphatic malformations, with or without lymphedema, in order to investigate and add evidence on its possible involvement in the development of lymphedema.

Transfer learning enables solving a specific task having limited data by using the pre-trained deep networks trained on large-scale datasets. Typically, while transferring the learned knowledge from source task to the target task, the last few layers are fine-tuned (re-trained) over the target dataset. However, these layers are originally designed for the source task that might not be suitable for the target task. In this paper, we introduce a mechanism for automatically tuning the Convolutional Neural Networks (CNN) for improved transfer learning. The pre-trained CNN layers are tuned with the knowledge from target data using Bayesian Optimization. First, we train the final layer of the base CNN model by replacing the number of neurons in the softmax layer with the number of classes involved in the target task. Next, the CNN is tuned automatically by observing the classification performance on the validation data (greedy criteria). To evaluate the performance of the proposed method, experiments are conducted on three benchmark datasets, e.g., CalTech-101, CalTech-256, and Stanford Dogs. The classification results obtained through the proposed AutoTune method outperforms the standard baseline transfer learning methods over the three datasets by achieving 95.92%, 86.54%, and 84.67% accuracy over CalTech-101, CalTech-256, and Stanford Dogs, respectively. The experimental results obtained in this study depict that tuning of the pre-trained CNN layers with the knowledge from the target dataset confesses better transfer learning ability. RESUMO

Piper longum (Indian long pepper) is known for its use as an anti inflammatory agent in Indian Ayurvedic System of medicine. Therefore, it is of interest to document the molecular docking analysis of compounds from Piper longum with COX-2 using the Autodock Vina PyRx tool. Molecular docking results show that asarinine, sesamine, fargesin, and piperlonguminine have optimal binding energy of 10, 10, -9.5 and 9.4 Kcal/mol, respectively for further consideration.

PECAM1 is a member of the immunoglobulin superfamily and is expressed in monocytes, neutrophils, macrophages and other types of immune cells as well as in endothelial cells. PECAM1 function is crucial for the development and maturation of B lymphocytes. The aim of this study was to link rare PECAM1 variants found in lymphedema patients with the development of lymphatic system malformations. Using NGS, we previously tested 246 Italian lymphedema patients for variants in 29 lymphedema-associated genes and obtained 235 negative results. We then tested these patients for variants in the PECAM1 gene. We found three probands with rare variants in PECAM1. All variants were heterozygous missense variants. In Family 1, the unaffected mother and brother of the proband were found to carry the same variant as the proband. Lymphoscintigraphy was performed to determine possible lymphatic malformations and showed that in both cases a bilateral slight reduction in the speed and lymphatic clearance of the lower limbs. PECAM1 function is important for lymphatic vasculature formation. We found variants in PECAM1 that may be associated with susceptibility to lymphedema.

SEMA3A is a semaphorin involved in cell signaling with PlexinA1 and Neuropilin-1 (NRP1) receptors and it is responsible for recruiting dendritic cells into lymphatics. Mutations in the SEMA3A gene result in abnormalities in lymphatic vessel development and maturation. We investigated the association of SEMA3A variants detected in lymphedema patients with lymphatic maturation and lymphatic system malfunction. First, we used NGS technology to sequence the SEMA3A gene in 235 lymphedema patients who carry wild type alleles for known lymphedema genes. We detected three different missense variants in three families. Bioinformatic results showed that some protein interactions could be altered by these variants. Other unaffected family members of the probands also reported different episodes of subclinical edema. We then evaluated the importance of the SEMA3A gene in the formation and maturation of lymphatic vessels. Our results determined that SEMA3A variants segregate in families with lymphatic system malformations and recommend the inclusion of SEMA3A in the gene panel for testing of patients with lymphedema.

CYP26B1 is a member of the cytochrome P450 family and is responsible for the break-down of retinoic acid for which appropriate levels are important for normal development of the cardiovascular and lymphatic systems. In a cohort of 235 patients with lymphatic malformations, we performed genetic testing for the CYP26B1 gene. These probands had previously tested negative for known lymphedema genes. We identified two heterozygous missense CY-P26B1 variants in two patients. Our bioinformatic study suggested that alterations caused by these variants have no major effect on the overall stability of CYP26B1 protein structure. Balanced levels of retinoic acid maintained by CYP26B1 are crucial for the lymphatic system. We identified that CYP26B1 could be involved in predisposition for lymphedema. We propose that CYP26B1 be further explored as a new candidate gene for genetic testing of lymphedema patients.

An electrocatalytic sensor for hydrazine using copper sulfide-ordered mesoporous carbon (CuS-OMC) is described. A facile solvothermal synthetic strategy was adopted for CuS-OMC and the ordered mesoporous carbon was obtained through nanocasting method. The synthesized CuS-OMC was characterized using microscopic and spectrochemical techniques. CuS-OMC was immobilized on GCE and evaluated for its electrochemical sensing of hydrazine using cyclic voltammetry and amperometry. CuS-OMC modified GCE exhibited better hydrazine sensing at an optimized pH 7.4 in terms of oxidation potential and current compared with that of GCE, CuS, and OMC. The observed sensing performance of CuS-OMC was attributed to the presence of Cu (I/II) in CuS dispersed in OMC which acts as an electrocatalytic center for the sensing of hydrazine. Amperometry under optimized experimental condition with an applied potential of 270 mV was employed to obtain a linear calibration plot in the range 0.25 to 40 µM (R2 = 0.9908) with a detection limit of 0.10 µM with a sensitivity of 0.915 (± 0.02) µA cm-2 µM-1. Real sample analyses were carried out by spiking of hydrazine in different water samples and the recoveries were in the range of 97 ± 2.1% (n = 3). Graphical abstract.

Trifunctional nickel tin sulfide (NixSn2xS4x) with a thiospinel-like structure composited with multiwalled carbon nanotubes (MWCNTs) (M-NixSn2xS4x) was synthesized by a facile method. The unit cell arrangement of the prepared composite was studied by density functional theory, and the theoretical interpretation satisfactorily inferred the presence of a synergistic effect between the thiospinel and MWCNTs. The high metallic conductivity and superior electrocatalytic activity of the M-NixSn2xS4x composite endow it with diverse applications. The composite shows promise as a counter electrode for dye-sensitized solar cells (efficiency of 4.67% for fluorine-doped indium tin oxide compared to 5.23% for platinum); an efficient catalyst for the hydrogen evolution reaction with good cycling stability and a low overpotential of -41 mV at a cathode current density of 10 mA cm2 and a Tafel slope of 43 mV dec-1 on a graphite sheet electrode; and an impressive capacitance material on a graphite sheet electrode alternative to expensive current collectors such as Ni foam, with a specific capacitance value of 1200 F g-1 at a current density of 1 A g-1 and a long life span of 92.6% that is retained for up to 15 000 cycles.

RESUMO

The use of "kabasura kudineer" (liquid soup made from Indian medicinal plants) for combating COVID-19 has been common in the states of Tamilnadu and Puducherry, India during the pandemic. Therefore, it is of interest to document the molecular docking analysis of IL-6 inhibitors with potential antiviral compounds from "kabasura kudineer" extract. We show the optimal binding features of gallic acid and luteolin with the Interleukin-6 protein for further consideration.

The present work reports low cost, green synthesis of Zinc oxide (ZnO) nanoparticles. The biosynthesized nanoparticles were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (ATR-FTIR). The synthesized ZnO nanoparticles were pure, predominantly spherical in shape with size ranging from 25 nm. The biosynthesized ZnO nanoparticles have been used for antibacterial and In Vitro applications. The antibacterial activity of the prepared CS/PVA/MC3-ZnO1, CS/PVA/MC3-ZnO2, and CS/PVA/MC3-ZnO3 nanocomposites samples was tested against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) due to disk diffusion method. When adding the ZnO nanoparticles both bacteria were very good inhibition zone was obtained. The transmission ZnO micrographs depicts spherical with uniform shape and good crystallinity and are composed of nanoparticles with a diameter less than 25 nm the average cube size was 100 nm. The AFM thickness of the CS/PVA/MC3-ZnO scaffold was, estimated from the AFM image, was about 10-20 nm and a roughness-like structure was observed. The FE-SEM film exhibits a scaffold exhibited porous structures. The excellent cell viability of the composite scaffolds was attributed to the good biocompatibility of the CS/PVA/MC3-ZnO3 as well as green fabrication process of the scaffolds. MTT analysis exposed that the samples did not have any toxicity. Since these positive points, these two kinds of scaffolds show appropriate properties for attachment, proliferation, and tendency to form group from L929 cells. In this work, we have prepared zinc oxide by high pressure homogenization process and the resultant zinc oxide was evaluated as fibers in CS/PVA/MC3 films.

The rationale of the present study was to isolate and identify endophytic fungi from endemic medicinal plants in Eastern Ghats and screened for antimicrobial potential of isolated fungal crude extracts. A total of 329 endophytic strains were isolated from 600 infected leaves and stem cuttings of endemic plants. The diversity and species richness was analyzed statistically and found to be higher in leaf segments than in stem segments. From isolated fungal strains, Cladosporium delicatulum was identified using molecular identification methods and selected as the most potent plumbagin-producing endophytic strain. Further the isolation and structural characterization of endophytic fungal plumbagin (5-hydroxyl-2-methylnaptalene-1,4-dione) was purified and confirmed through spectroscopy analysis. The molecular weight was determined as m/z 188 in positive mode by ESI-MS, which confirmed to be plumbagin which potentially inhibited all tested pathogens, therefore the endophytic fungal plumbagin from the current study possesses important biological activities against pathogens.
