RESUMO

The zinc content in microresected islets of rabbits, mice, rats, and guinea pigs was determined by atom-absorption spectrophotometry. The content was found to be highest in rabbits, and diabetes was induced in them by chelating agents easily. Injection of dithizone or 8-(p-toluenesulfonylamine) quinoline failed to induce diabetes mellitus in mice, rats, and guinea pigs. It is concluded that diabetogenic chelating agents are capable of producing irreversible diabetogenic affection in those beta-cells which contain a critical concentration of reactant zinc.

RESUMO

The ultrastructure of the pancreatic islets and insulin content in the beta-cells, revealed by histochemical technique after glybenclamide loading, was investigated in rats. Glybenclamide administration led to the loss in beta-cells of a greater portion of their hormone depot. Ultrastructurally, a significant development in all the organellae of the protein synthesizing apparatus was demonstrated, this testifying to a sharp rise of the hormone production. At the same time insulin released from the cells was accelerated, although no final and complete formation of beta-granules occurred, alpha-insulocytes appeared to be intact.

RESUMO

Experiments were conducted on rabbits. A study was made of the effect of administration of maninyl (glybenclamide) into the stomach in a dose of 10 mg/kg of body weight for 7 days on the blood glucose level, insulin and zinc content in the pancreatic islands, and on the "dithizone" diabetes development. Maninyl administration was accompanied by a significant glycemia reduction. The amount of deposited insulin and zinc determined histochemically was sharply reduced up to complete disappearance from the majority of beta-cells. "Dithizone" diabetes was not reproducible in animals given maninyl preliminarily: the required condition for induction of this affection was formation of zinc dithizonate in beta-cells.

RESUMO

Administration of chinoline derivatives, with a capacity to form stable complexes with metals (zinc?), was accompanied by a selective damage of the pancreatic islands with development of diabetes mellitus. Changes in the histostructure of the tissue caused by chinoline compounds of various chemical structure were unitypical. Their severity chiefly depended on the severity and duration of diabetes. Diabetes under study arising after the administration of chinoline derivatives were good models for detailed ascertaining of histostructure of the islet tissue and of the pancreatic duct in diabetes.
