RESUMO

We proposed a new method of evaluation of beta-receptor's activity on the surface of human T-lymphocytes based on the radioligand method. Optimal conditions for evaluation of specific binding to ß2-adrenoceptors of 0.5 fmol ligand per 1 million cells using [125I]-cyanopindolol were found. The possibility of using of ß2-adrenoceptor's activity assessment in clinical settings was demonstrated on human T-lymphocyte.

To study the immunological features in chronic bronchitis (CB) patients with great residual changes (GRC) after prior pulmonary tuberculosis, the authors examined two groups: 1) 40 CB patients with GRC after prior pulmonary tuberculosis (a study group) and 2) 30 CB patients without a history of pulmonary tuberculosis. The examination revealed that CB patients with GRC after prior pulmonary tuberculosis were found to have higher T lymphocytes, lower B lymphocytes on an exacerbation of the disease. The phagocytic properties of neutrophils were altered as their high phagocytic activity, decreased oxygen-dependent microbicidal activity of phagocytes with their stimulation. The humoral response remained to be slightly pronounced in CB bronchitis with GRC. The low serum levels of TBC active products may suggest the low activity of lipid peroxidation processes in the study group patients. After prior pulmonary tuberculosis, antioxidative activity preserves to be high in CB patients with GRC.

RESUMO

The influence of preparations obtained from oat and wheat seedlings (immunostimulating factors IF-1 and IF-2, respectively) on the natural resistance of mice to P. aeruginosa infection was studied. IF-1 and IF-2 were introduced intraperitoneally in a single injection in doses of 100 micrograms and 1000 micrograms per mouse 2 and 7 days prior to the inoculation of P. aeruginosa strain 8 in doses of 1 and 10 LD50. The presence of substances capable of stimulating the immunobiological reserves of the body in actively growing plants (seedlings) was shown.

RESUMO

The results obtained in the study of the dynamics of systemic and local immunity characteristics in children with prolonged and chronic dysentery under the influence of vaccinal therapy are presented. The vaccine, containing soluble antigenic complexes isolated from Shigella sonnei cells by disintegration with hydroxylamine, was introduced intrarectally in doses of 2-4 mg. The course of treatment consisted of 5-6 administrations. The vaccinal therapy resulted in an increase in the level of immunoglobulins and the titer of specific antibodies, particularly IgA, in sera and fecal filtrates. These data coincided with an increase in the number of IgA-producing cells in bioptic samples of the mucous membrane of the large intestine. The vaccinal therapy contributed to the cessation of the release of bacteria in 82.5% of the patients.

RESUMO

The oral and subcutaneous immunization of mice with the vaccine prepared from S. minnesota strain R595, chemotype Re, known as enterobacterial vaccine, was found to significantly decrease the number of shigellae in cultures obtained by the inoculation of homogenized mucosal samples taken from the large intestine of mice, the vaccine prepared from S. minnesota isogeneous strain SF 1111 with the intact structure of lipopolysaccharide had no such activity. Antisera, obtained by the immunization of rabbits with enterobacterial vaccine, contained high titers of antibodies to Re-glycolipid and were capable of decreasing the isolation rate of shigellae from homogenized mucosal samples taken from the large intestine of mice; at the same time S. flexneri were found capable of binding with antibodies to glycolipid of Re-chemotype.

RESUMO

The results of the study of systemic and local immunity in cases of prolonged and chronic dysentery in children are presented. These results indicate that a decrease in the levels of immunoglobulins and specific IgM and IgA antibodies in the blood serum and coprofiltrates was observed as the process became chronic. The titer of specific IgG antibodies in the serum remained on a relatively high level, thus indicating the presence of the active infectious process. The low level of specific IgA antibodies correlated with a decrease in IgA-producing cells in the mucosa of the large intestine, which suggested that this form of the disease was accompanied by disturbances in the mechanisms of local immunity.

RESUMO

The influence of immunization with pertussis vaccines differing in toxicity on the intensity of the formation of antibodies to heterologous antigens (S. typhi Vi-antigen) and on the resistance of the body to natural infection (S. typhimurium) was studied in mice. The toxicity of pertussis vaccines was found to be related to their capacity for changing immune response to heterologous antigen. In mice showing pronounced toxicosis the injection of pertussis vaccine resulted in a decrease in their capacity for Vi-hemagglutinin formation. The appearance of a definite degree of resistance ot S. typhimurium was observed in mice previously immunized with pertussis vaccine possessing pronounced toxic properties. Nevertheless, the appearance of enhanced resistance to infection was observed only in the animals previously immunized with a nontoxic preparation.

Opsonizing activity of guinea pig blood serum containing mercaptoethanol-resistant pertussis antibodies was studied in vitro on a model of microorganism ingestion by the mononuclears of the guinea pig peritoneal exudate. There were revealed distinct differences in the serum activity depending on the phagocytosis object. The blood serum of hyperimmunized rabbits stimulated the ingestion of Bordetella pertussis by mononuclears of guinea pigs--normal and immunized with pertussis vaccine. The blood sera of hyperimmunized guinea pigs and of mice immunized with pertussis vaccine twice displayed opsonins to B. pertussis. The blood sera of animals immunized with pertussis vaccine inhibited the staphylococcus ingestion by the peritoneal exudate mononuclears of guinea pigs, both normal and those immunized with pertussis vaccine.

Protective activity of anti-persussis rabbit and mouse sera and 19 S and 7 S fractions obtained from these sera was investigated in the test of passive protection of mice on a model of pertussis meningoencephalitis. The method of simultaneous intracerebral administration of the serum or fraction with live culture of a virulent B. pertussis strain was used. Hyperimmune rabbit serum containing mercaptoethanol-resistant agglutinins in a high titre was found to have the most pronounced protective effect. Serum of mice, collected 14 days after single immunization of the animals, did not show any protective properties. A small amount of protective activity was observed in the serum collected on the 30th day after a single administration of the vaccine. A sharp increase in the protective activity of the serum was observed after double immunization of mice. Correlation was found between the increase in the titre of agglutinins (in particular of 7 S antibodies) and the protective activity of the serum. Protective properties of 19 S and 7 S fractions isolated from immune rabbit and mouse sera by the method of gel filtration were investigated. Both fractions were found to possess protective properties, but fraction 7 S was more active than fraction 19 S.
