RESUMO

Conjugated linoleic acid (CLA), a naturally occurring anticarcinogen found in dairy products, is an intermediary product of ruminal biohydrogenation of polyunsaturated fatty acids. Our objective was to determine the effect of different dietary oils, which vary in fatty acid composition, on CLA concentrations in milk from lactating dairy cows. Twelve Holstein cows were randomly assigned to a 3 x 3 Latin square design. Dietary treatments were the addition (53 g/kg dietary dry matter) of peanut oil (high oleic acid), sunflower oil (high linoleic acid) and linseed oil (high linolenic acid). Each treatment period was 2 wk, and milk samples were collected on the last 4 d of each period. Milk yield (34.2 +/- 1.3 kg/d) and milk fat (2.25 +/- 0.06%) were not different among treatments. Milk protein during the sunflower oil treatment (mean, 3.44% protein) was significantly higher (P < 0.01) than during the other treatments. Milk fat concentration of CLA during the sunflower oil treatment was significantly different from other treatments (P < 0.001) and approximately 500% greater than typically observed when cows consume traditional diets. CLA concentrations (mg/g of milk fat) were 13.3, 24.4 and 16.7 during peanut oil, sunflower oil and linseed oil treatment, respectively. CLA concentration in milk fat can be enhanced by the addition of polyunsaturated fatty acids to the diet, especially oils high in linoleic acid.

RESUMO

The enormous cost of eliminating mouse hepatitis virus (MHV) from a mouse colony demands that a confirmed etiologic diagnosis be made to justify the necessary remedial action. We describe an outbreak of MHV in nude mice in which histopathologic findings provided a presumptive diagnosis, but results of serologic testing of affected nude mice and immunocompetent sentinels were negative. Results of transmission electron microscopy of liver specimens from affected mice were equivocal. Confirmation of the etiopathogenesis was eventually provided by reverse transcriptase-polymerase chain reaction (RT-PCR), using primers with nested sequences directed to two separate but highly conserved regions of the MHV genome. This procedure detected MHV in the liver of an affected nude mouse and in a sentinel, although in the latter a positive result was obtained only because of the increased sensitivity of nested primers used in a second round of amplification. Virus was not detected in cell lines that had been injected into the mice, and the source of the outbreak was not found. These results indicate the applicability of RT-PCR for detecting MHV in a field situation while also illustrating that conventional, complementary techniques still have an essential role in reaching a diagnosis. It is recommended that specimens should be taken for histologic examination and serologic testing, as well as for molecular studies when MHV infection is suspected.

RESUMO

An accident resulted in the deposition on an injured workman's skin surfaces, in acid-burned areas and in lacerations, of something in excess of 6 mCi 241Am. This paper describes the external decontamination procedures used, the change in americium content of the skin during the course of treatment, and some of the unusual problems encountered from the extrusion of foreign material and flaking of skin and scar tissue.

RESUMO

An injured worker, contaminated with more than 6 mCi of 241Am required special treatment and housing for 4 months. This paper is a description of the design and management of the facility in which most of the treatment and housing occurred. The problems associated with contamination control, waste handling, supplies and radiological concerns during the two-stage transfer of the patient from a controlled situation to his normal living environment are discussed in detail.

RESUMO

A computer based verification system which monitors the operation of a 4-MV linear accelerator is described. Fourteen parameters can be checked for proper alignment prior to irradiation. A training simulator was also built to minimize interference with treatment schedules and to provide technicians with experience in using the system during the developmental stages of the project.
