RESUMO

Background: Lifestyle modification is an integral aspect for the management of type 2 diabetes (T2D). However, it is difficult to ensure the accuracy of personalized lifestyle advice. The study aims to analyse the real-world effectiveness of personalized glycemic response based Diabefly-Pro digital therapeutics for better glycemic control. Methods: Data from continuous glucose monitoring (CGM) of 64 participants with T2D was analysed. All participants were provided with modified lifestyle plan based on their personalized glycemic response. The CGM data was analysed for a period of 7 days, before and after the introduction of modified lifestyle plan. Primary outcome of the study was change in time in range (TIR). Secondary outcomes of the study were change in mean blood glucose, time above range (TAR), time below range (TBR) and glucose management indicator (GMI). Results: Significant improvement in glycemic control was observed after the introduction of personalized lifestyle plan. Median reduction in mean blood glucose was from 139.5 (118.3 to 169.3) mg/dL to 122.0 (101.5 to 148.8) mg/dL (p < 0.0001). TIR and GMI improved from 70.50 (50.75 to 83.50) % to 75.00 (58.25 to 89.00) % (p = 0.0001) and 6.64 (6.13 to 7.35) % to 6.23 (5.74 to 6.86) % (p < 0.0001) respectively. TAR reduced significantly from 17.00 (4.25 to 38.0) % to 6.00 (1.25 to 26.0) % (p < 0.0001). No significant increase in TBR was observed (p = 0.198). Conclusion: Personalized glycemic response-based Diabefly-Pro digital therapeutics program was effective in achieving better glycemic control in people with T2D.

The polyfluorinated alkyl substance 6:2 fluorotelomer sulfonate (6:2 FTS) has been detected in diverse environments impacted by aqueous film-forming foams used for firefighting. In this study, a bacterial strain (J3) using 6:2 FTS as a sulfur source was isolated from landfill leachate previously exposed to polyfluoroalkyl substances in New South Wales, Australia. Strain J3 shares 99.9% similarity with the 16S rRNA gene of Dietzia aurantiaca CCUG 35676T. Genome sequencing yielded a draft genome sequence of 37 contigs with a G + C content of 69.7%. A gene cluster related to organic sulfur utilisation and assimilation was identified, that included an alkanesulfonate monooxygenase component B (ssuD), an alkanesulfonate permease protein (ssuC), an ABC transporter (ssuB), and an alkanesulfonate-binding protein (ssuA). Proteomic analyses comparing strain J3 cultures using sulfate and 6:2 FTS as sulfur source indicated that the ssu gene cluster was involved in 6:2 FTS biodegradation. Upregulated proteins included the SsuD monooxygenase, the SsuB transporter, the ABC transporter permease (SsuC), an alkanesulfonate-binding protein (SsuA), and a nitrilotriacetate monooxygenase component B. 6:2 Fluorotelomer carboxylic acid (6:2 FTCA) and 6:2 fluorotelomer unsaturated acid (6:2 FTUA) were detected as early degradation products in cultures (after 72 h) while 5:3 fluorotelomer acid (5:3 FTCA), perfluorohexanoic acid (PFHxA) and perfluoropentanoic acid (PFPeA) were detected as later degradation products (after 168 h). This work provides biochemical and metabolic insights into 6:2 FTS biodegradation by the Actinobacterium D. aurantiaca J3, informing the fate of PFAS in the environment.

The present investigation is an attempt to assess the contamination of heavy metals in the ground and surface water of the Singrauli industrial belt area. Pollution indices like heavy metal index (HPI), contamination index (CD) and heavy metal evaluation index (HEI) are used for the evaluation of heavy metal pollution (arsenic As, mercury Hg, cadmium Cd, and lead Pb). Contour maps are constructed to interpret metal spatial distribution. Further, the land-use/land-cover (LULC) maps for the year 2000, 2010 and 2016 are prepared using Landsat satellite data. A total of 48 water samples (Groundwater (27), Surface water (21)) are analysed for heavy metal concentration. Eighty-eight percent of groundwater and 90% of surface water samples are contaminated with Hg. Similarly, high concentrations of Pb and Cd were found in the samples. Surprisingly, all the water samples have As concentration above the WHO permissible limit of 10 ppb. Further, 95% of the samples have an HPI value greater than 100 indicating high heavy metal contamination. CD value denotes contamination of 89% of the samples with heavy metals (As, Hg, Cd, Pb). Through spatial distribution, it can be interpreted that most of the contaminated samples lie near thermal power plants, ash ponds, and coal mines. LULC (land use/land cover) study shows a significant decrease in water bodies by (108 km2), agricultural land by (54 km2) and bare/fallow land by (51 km2) from 2000 to 2016. During these 16 years, there has been a fourfold increase in the overburden, a threefold increase in dumping yards, a 2.5 times increase in urban areas, and a twofold increase in mining areas. Both the environment and the water quality are deteriorating at an alarming rate. Such scientific investigations are relevant for risk management studies of potable water. The knowledge acquired from such assessment shall be considered with utmost priority by concerned authority considering degrading water quality in the study area. Hence, this study is applicable for designing action plans and control measures to reduce water resource pollution.
