RESUMO

Carbon foot prints (CFs) studies based on life cycle assessment between sheep farming systems and green house gases (GHG) emissions is one of the best indicators to quantify the amount of GHG emissions per kg of product. Therefore, a life cycle assessment (LCA) study was conducted for three different sheep farming systems i.e. intensive system (stall fed only), semi-intensive (grazing with supplementation) and extensive system (grazing only) under semiarid region of India to assess the carbon cost of sheep rearing. The total CFs were estimated to be 16.9, 15.8 and 17.1 kg CO2-eq in intensive, semi-intensive and extensive system of grazing indicating semi-intensive system to be most carbon (C) efficient. For 1kg mutton production in semi-intensive and intensive system, around 30% and 24% CFs were contributed from enteric fermentation and feed respectively, whereas, in extensive system, the contribution of enteric fermentation increased up to 50%. The carbon foot prints analysis gives an insight of carbon inputs used but the amount of CO2 sequestered in soil making LCA a holistic approach for estimating GHG emissions from livestock.

RESUMO

A feeding trial was conducted for a period of 60 days during extreme summer months to observe the effect of supplementation of Moringa oleifera leaves containing concentrate pellets on nutrient utilization, antioxidant status, and reproductive performance in Avishaan ewes reared under semi-arid condition. Forty adult non-pregnant cyclic ewes (2-3 years, 31.8 ± 0.81 kg body weight) were selected and randomly allocated into 2 groups of 20 animals each, viz., G-I (control) and G-II (treatment). The ewes were grazed on natural pasture for 8 h, offered ad libitum Cenchrus ciliaris hay after grazing and concentrate pellets @ 300 g/animal/day. The ewes in G-I were offered conventional concentrate pellets, whereas G-II ewes were offered concentrate pellets containing 15% Moringa leaves. The mean temperature humidity index during the period of study was 27.5 ± 0.3 and 34.6 ± 0.4 at 0700 h and 1400 h, respectively, indicating severe heat stress. Nutrient intake and utilization were comparable between the two groups. The antioxidant status was higher in G-II ewes as the values of catalase, superoxide dismutase, and total antioxidant capacity were higher (P < 0.05) in G-II ewes compared to G-I. The conception rate was higher (100%) in G-II ewes than G-I ewes (70%). Multiple birth percentage was 77.8% in G-II ewes, and it was comparable with the herd average of Avishaan (74.7%). However, ewes in G-I group exhibited a marked decline in multiple birth percentage (28.6%) than the normal herd average. Hence, it can be concluded that inclusion of Moringa oleifera leaves in feeding of prolific Avishaan ewes improved their antioxidant status resulting in optimum reproductive performance during stressful summer months.

RESUMO

Sixty Chinchilla rabbits (28 days old) were divided into three equal groups (n = 20). Rabbits in MOL0 (control) were fed pellets containing 700 g cowpea hay/kg pellet as forage source, whereas rabbits in MOL700 and MOL950 were fed pellets containing 700 and 950 g moringa leaves/kg feed pellets, respectively. Average daily gain and feed conversion ratio was comparable in MOL700 and MOL0, however, it was higher in MOL950. Among the carcass traits, dressing percent was higher while, chilling loss was lower in MOL700 treatment. The Longissimus thoracis muscle of rabbits fed moringa leaves containing pellets (MOL700 and MOL950) had lower saturated fatty acid content, higher C18:3n-3 and total n-3 fatty acids along with lower thrombogenic index value. Hence, incorporating moringa leaves at 70% level is beneficial in terms of improved growth performance and functional attributes of meat than diet- containing sole moringa leaves.

RESUMO

Background Good governance and leadership are essential to improve healthy life expectancy particularly in low and middle-income countries (LMICs). This study aimed to epitomize the challenges and opportunities for leadership and good governance for the health system to address non-communicable diseases particularly cardiovascular diseases (CVD) in Nepal. Objective The objective of this study was to understand and document CVD programs and policy formulation processes and to identify the government capacity to engage stakeholders for planning and implementation purposes. Method A national-level task force was formed to coordinate and steer the overall need assessment process. A qualitative study design was adopted using "The Health System Assessment Approach". Eighteen indicators under six topical areas in leadership and governance in cardiovascular health were assessed using desk review and key informant interviews. Result Voice and accountability exist in planning for health from the local level. The government has shown a strong willingness and has a strategy to work together with the private and non-government sectors in health however, the coordination has not been effective. There are strong rules in place for regulatory quality, control of corruption, and maintaining financial transparency. The government frequently relies on evidence generated from large-scale surveys for health policy formulation and planning but research in cardiovascular health has been minimum. There is a scarcity of cardiovascular disease-specific protocols. Conclusion Despite plenty of opportunities, much homework is needed to improve leadership and governance in cardiovascular health in Nepal. The government needs to designate a workforce for specific programs to help monitor the enforcement of health sector regulations, allocate enough funding to encourage CVD research, and work towards developing CVD-specific guidelines, protocols, and capacity building. RESUMO

BACKGROUND & OBJECTIVES: Universal coverage of population with long-lasting insecticidal nets (LLINs) living in endemic areas inhabited by ethnic tribal communities or in difficult to reach areas was found effective for disease control where other interventions such as indoor residual spraying have limited success because of operational issues. Evaluation of different LLINs with varied insecticides and fabrics are being evaluated to meet the demand of new products. This study was undertaken on two brands of LLINs, DuraNet© and Interceptor® that varied in fabric and manufacturing technologies to assess the usability in field conditions for atleast three years. METHODS: In large-scale field trials DuraNet©, alpha cypermethrin incorporated polyethylene net, was evaluated in Odisha state while, Interceptor® net, alpha cypermethrin coated polyester net was evaluated in Chhattisgarh and Gujarat states for a period of three years following WHO guidelines. Durability, usage pattern and washing behavior were monitored through periodic surveys and physical examination of nets. RESULTS: Survivorship of both the nets was 84-100% and every night usage rate was >62% in all seasons. Washing frequency was largely within the prescribed limits. The proportion of DuraNet© with holes was 26.7% in year one 74% in year three. In Gujarat, proportion of Interceptor® nets with holes at six months was 33.3% increased to 87% in year three and in Chhattisgarh, 6.7% after six months to 93.3% after three years of use respectively. INTERPRETATION & CONCLUSION: Both the nets revealed a useful life of three years under the field condition.

RESUMO

Utilization of low-input feed resources rich in plant bioactive compounds is a promising strategy for modulating the fatty acid profile in ruminant products. They manipulate microbes involved in rumen biohydrogenation and increase the accumulation of desirable fatty acids at the tissue level. Therefore, the present study was undertaken to assess the effect of dietary supplementation of aniseed straw and eucalyptus leaves on growth performance, carcass traits and fatty acid profile of finisher lambs. Thirty-six Malpura hogget were divided into three treatment groups of 12 each, reared individually in pen (1.6 m × 1.1 m) and fed ad libitum complete feed blocks made up of 55 parts concentrate, 5 parts molasses and 40 parts roughage. Roughage in control (Con) was 20 parts each of ardu (Ailanthus excelsa) leaves and oat (Avena sativa) straw. In test diets, that is, Con-as and Con-el, 10% aniseed (Pimpinella anisum) straw and Eucalyptus rudis leaves, respectively, were added by replacing 5% each of oat straw and eucalyptus leaves. The lambs were weighed weekly; and at the end of 3 months of feeding trial, the lambs were slaughtered to study the carcass traits, composition and product evaluation. Average daily gain (ADG) and DM intake (DMI) was higher (P < 0.05) in Con-as compared to Con and Con-el, while ADG and feed conversion ratio decreased (P < 0.05) by 29.4% and 36.4%, respectively, in Con-el compared to Con. Carcass traits showed lower (P < 0.05) loin eye area and chilling loss in the Con-el group compared to the Con-as and Con, and the total carcass fat compared to Con-as. However, the keeping quality of meat improved in both Con-as and Con-el which was reflected by lower (P < 0.05) thiobarbituric acid-reactive substances values. Nuggets prepared from Con and Con-as meat had superior (P < 0.05) sensory attributes with an overall palatability. Fatty acid profile of longissimus thoracis muscle showed lower (P < 0.05) atherogenic and thrombogenic indices in Con-as and higher (P < 0.05) in Con-el group. Moreover, in Con-as group, the proportion of C16:0 was lower (P < 0.05) and C18:3n-3 was higher (P < 0.05), but no effect was observed on the amount of conjugated linoleic acid (CLA; C18:2 c9t11). In case of adipose tissue, the content of CLA was higher (P < 0.05), and the ratio of n-6:n-3 was more nearer to desirable levels in Con-as group. Therefore, it can be concluded that aniseed straw is a promising feed supplement compared to eucalyptus leaves for improving meat quality and fatty acid profile in lambs.

RESUMO

INTRODUCTION: Recurrent pregnancy losses (RPLs) are seen in up to 15-20% of all clinically recognized pregnancies, 1-2% of women in general population. Repeated losses are seen in 5-10% of women. The prevalence of chromosomal rearrangements is 6.65% in couples with repeated pregnancy losses. Two to 4% of RPL are associated with parental balanced reciprocal and Robertsonian translocations. MATERIALS AND METHODS: The study was conducted at a tertiary care hospital in New Delhi, and in total, 204 couples with RPL enrolled in the study. RESULTS: In total, 4490 couples presented to the obstetric clinic, of which 204 (4.5%) couples had repeated pregnancy losses. Cytogenetic analysis was done in 198 couples. Out of total 198 patients, 14 patients (7.1%) had cytogenetic alterations. Most common aberrations observed were structural rearrangements, of which reciprocal translocations were more common. In our study cohort, all the couples had maternal age of ≤ 35 years and all the alterations were seen either in mother or in both parents. DISCUSSION: Our study highlights that cytogenetic alterations not only are common in first trimester miscarriages, but are an important event in miscarriages presenting at later period of gestation and in young mothers as well.

RESUMO

We study the quantum Hall plateau transition on rectangular tori. As the aspect ratio of the torus is increased, the two-dimensional critical behavior, characterized by a subthermodynamic number of topological states in a vanishing energy window around a critical energy, changes drastically. In the thin-torus limit, the entire spectrum is Anderson localized; however, an extensive number of states retain a Chern number C≠0. We resolve this apparent paradox by mapping the thin-torus quantum Hall system onto a disordered Thouless pump, where the Chern number corresponds to the winding number of an electron's path in real space during a pump cycle. We then characterize quantitatively the crossover between the one- and two-dimensional regimes for finite torus thickness, where the average Thouless conductance also shows anomalous scaling.

RESUMO

Background Androgen deficiency is diagnosed on the basis of clinical symptoms and laboratory assessment of testosterone level. Different screening tools have been developed to evaluate the sign and symptoms. Objective In this study, we examine the validity and reliability of the Nepali version of the quantitative Androgen Deficiency in Aging Male (qADAM) questionnaire to screen androgen deficiency in Nepali male. Method English dialectal quantitative Androgen Deficiency in Aging Male questionnaire was forward translated to Nepali version and backward translated. This version was reviewed by a panel of an endocrinologist, a clinical psychiatrist, a physician, and a clinical biochemist. A final Nepali version of qADAM was developed. Thirty-one healthy male aged 31-70 years were administered with the questionnaire in two separate occasions two weeks apart. Cronbach's alpha and test-retest reliability were calculated to identify validity and reliability, respectively. Result In the Nepali translated questionnaire, Cronbach's alpha for internal consistency from ten items is good (0.68). The Cronbach's alpha for internal consistency from nine items without item 7 is 0.706. Seven out of ten items had an R-value of > 0.7. In the total sample, Standard Error Mean (SEM) ranged from 0.00-0.44 for qADAM. SEM% are low for all variables (0.00-11.20%). MDC95 ranged 0.00-1.234. MDC95% ranged 0.00 - 31.05% and was < 30% for majority of variables (90%). Conclusion The final translated Nepali questionnaire seems reliable and valid. A future study measuring the Nepali questionnaire with testosterone level and another biochemical test in control and androgen deficiency patients will help validate the questionnaire.

RESUMO

BACKGROUND: Neuroimaging studies have identified obesity-related differences in the brain's resting state activity. An imbalance between homeostatic and reward aspects of ingestive behaviour may contribute to obesity and food addiction. The interactions between early life adversity (ELA), the reward network and food addiction were investigated to identify obesity and sex-related differences, which may drive obesity and food addiction. METHODS: Functional resting state magnetic resonance imaging was acquired in 186 participants (high body mass index [BMI]: ≥25: 53 women and 54 men; normal BMI: 18.50-24.99: 49 women and 30 men). Participants completed questionnaires to assess ELA (Early Traumatic Inventory) and food addiction (Yale Food Addiction Scale). A tripartite network analysis based on graph theory was used to investigate the interaction between ELA, brain connectivity and food addiction. Interactions were determined by computing Spearman rank correlations, thresholded at q < 0.05 corrected for multiple comparisons. RESULTS: Participants with high BMI demonstrate an association between ELA and food addiction, with reward regions playing a role in this interaction. Among women with high BMI, increased ELA was associated with increased centrality of reward and emotion regulation regions. Men with high BMI showed associations between ELA and food addiction with somatosensory regions playing a role in this interaction. CONCLUSIONS: The findings suggest that ELA may alter brain networks, leading to increased vulnerability for food addiction and obesity later in life. These alterations are sex specific and involve brain regions influenced by dopaminergic or serotonergic signalling.

RESUMO

Background Thyroid dysfunction (TD) is one of the major public health concerns in Nepal. This study intends to identify factors in association with thyroid dysfunction and its hospital based prevalence. Objective To determine hospital-based prevalence of thyroid dysfunction and to identify and analyze factors in association with thyroid dysfunction. Method A retroprospective cross-sectional study was conducted among patients attending Dhulikhel Hospital-Kathmandu University Hospital (DH,KUH) from January to December, 2015 with prospective part carried out from July to December 2015 in the patients visiting Dhulikhel hospital for health check-up and requested for Thyroid Function Tests (TFTs). Thyroid Function Tests of 1530 subjects were performed in the biochemistry laboratory and semi-structured questionnaire were introduced to 312 participants with convenient sampling method and their anthropometric measurements were taken. Chi-square test, Pearson's correlation and student-t test were used as a measure to analyze factors. Result Out of total 1530 subjects, 35.3% were having thyroid dysfunction with the prevalence of overt hyperthyroidism, subclinical hyperthyroidism, overt hypothyroidism and subclinical hypothyroidism to be 2.5%, 2.4%, 5.6% and 24.8% respectively. The distribution of thyroid dysfunction was preponderant among females and among older age. The prevalence of thyroid dysfunction was highest among Terai/Madhesi (40.0%) and lowest among Newars (28.5%) with higher fraction of hypothyroidism in each ethnic group. There were significant correlation between Thyroid Stimulating Hormone (TSH) with waist-hip ratio (p<0.05) and weight (kg) with free thyroxine (fT4) (p<0.05). Direct pesticide exposure (p<0.05) was significantly associated with hypothyroidism. The serum fT4 was significantly different (p<0.05) among vegetarian and non-vegetarian whereas TSH and free triiodothyronine (fT3) were not significantly different. Smoking, alcoholism, dietary food habits, diabetes and hypertension were not significantly associated with thyroid dysfunction. Conclusion Thyroid dysfunction was highly prevalent among participants with higher proportion of subclinical hypothyroidism. Besides, direct pesticide exposure found to be factor in association with hypothyroidism, this study could not find significance relation with other established risk factors.

RESUMO

BACKGROUND: This experiment aimed at assessing polyphenol-rich plant biomass to use in complete feed making for the feeding of ruminants. METHODS: An in vitro ruminal evaluation of complete blocks (CFB) with (Acacia nilotica, Ziziphus nummularia leaves) and without (Vigna sinensis hay) polyphenol rich plant leaves was conducted by applying Menke's in vitro gas production (IVGP) technique. A total of six substrates, viz. three forages and three CFBs were subjected to in vitro ruminal fermentation in glass syringes to assess gas and methane production, substrate degradability, and rumen fermentation metabolites. RESULTS: Total polyphenol content (g/Kg) was 163 in A. nilotica compared to 52.5 in Z. nummularia with a contrasting difference in tannin fractions, higher hydrolysable tannins (HT) in the former (140.1 vs 2.8) and higher condensed (CT) tannins in the later (28.3 vs 7.9). The potential gas production was lower with a higher lag phase (L) in CT containing Z. nummularia and the component feed block. A. nilotica alone and as a constituent of CFB produced higher total gas but with lower methane while the partitioning factor (PF) was higher in Z. nummularia and its CFB. Substrate digestibility (both DM and OM) was lower (P < 0.001) in Z. nummularia compared to other forages and CFBs. The fermentation metabolites showed a different pattern for forages and their CFBs. The forages showed higher TCA precipitable N and lower acetate: propionate ratio in Z. nummularia while the related trend was found in CFB with V. sinensis. Total volatile fatty acid concentration was higher (P < 0.001) in A. nilotica leaves than V. sinensis hay and Z. nummularia leaves. It has implication on widening the forage resources and providing opportunity to use forage biomass rich in polyphenolic constituents in judicious proportion for reducing methane and enhancing green livestock production. CONCLUSION: Above all, higher substrate degradability, propionate production, lower methanogenesis in CFB with A. nilotica leaves may be considered useful. Nevertheless, CFB with Z. nummularia also proved its usefulness with higher TCA precipitable N and PF. It has implication on widening the forage resources and providing opportunity to use polyphenol-rich forage biomass for reducing methane and enhancing green livestock production.

RESUMO

Amanitatullossiana, a new species of Amanita [subgenus Lepidella] section Lepidella from India is described. The species is characterised by its ash grey to brownish-grey pileus covered with dark grey to greyish-black universal veil remnants, the upper part of its rooting stipe base covered by several rows of recurved scales, broadly ellipsoid to ellipsoid basidiospores, absence of basidial clamp connections and pileal remnants of universal veil comprising abundant, disordered inflated cells intermixed with scattered filamentous hyphae. Molecular phylogenetic analysis and morphology both support the association of A.tullossiana with species of Bas' stirps Cinereoconia - A.cinereoconia and A.griseoverrucosa. Two species, A.griseoverrucosa and A.virgineoides are reported here as new records for India.

RESUMO

Banana is a non- model crop plant, and one of the most important crops in the tropics and sub tropics. Heat stress is the major abiotic stress affecting banana crop production because of its long growth period and is likely to become a threat due to global warming. To understand an acquired thermotolerance phenomenon at the molecular level, the RNA-seq approach was employed by adapting TIR method. A total of 136.38 million high quality reads were assembled. Differentially expressed genes under induction (I) was 3936, I + L was 2268 and lethal stress was 907 compared to control. Gene ontology and DGE analysis showed that genes related to heat shock factors, heat shock proteins, stress associated proteins, ROS scavenging, fatty acid metabolism, protein modification were significantly up regulated during induction, thus preparing the organism or tissue at molecular and cellular level for acquired thermotolerance. KEGG pathway analysis revealed the significant enrichment of pathways involved in protein processing, MAPK signaling and HSPs which indicates that these processes are conserved and involved in thermo tolerance. Thus, this study provides insights into the acquired thermotolerance phenomena in plants especially banana.

RESUMO

We propose a spatiotemporal quench protocol that allows for the fast preparation of ground states of gapless models with Lorentz invariance. Assuming the system initially resides in the ground state of a corresponding massive model, we show that a superluminally moving "front" that locally quenches the mass, leaves behind it (in space) a state arbitrarily close to the ground state of the gapless model. Importantly, our protocol takes time O(L) to produce the ground state of a system of size ∼L^{d} (d spatial dimensions), while a fully adiabatic protocol requires time ∼O(L^{2}) to produce a state with exponential accuracy in L. The physics of the dynamical problem can be understood in terms of relativistic rarefaction of excitations generated by the mass front. We provide proof of concept by solving the proposed quench exactly for a system of free bosons in arbitrary dimensions, and for free fermions in d=1. We discuss the role of interactions and UV effects on the free-theory idealization, before numerically illustrating the usefulness of the approach via simulations on the quantum Heisenberg spin chain.

RESUMO

BACKGROUND/OBJECTIVES: The brain has a central role in regulating ingestive behavior in obesity. Analogous to addiction behaviors, an imbalance in the processing of rewarding and salient stimuli results in maladaptive eating behaviors that override homeostatic needs. We performed network analysis based on graph theory to examine the association between body mass index (BMI) and network measures of integrity, information flow and global communication (centrality) in reward, salience and sensorimotor regions and to identify sex-related differences in these parameters. SUBJECTS/METHODS: Structural and diffusion tensor imaging were obtained in a sample of 124 individuals (61 males and 63 females). Graph theory was applied to calculate anatomical network properties (centrality) for regions of the reward, salience and sensorimotor networks. General linear models with linear contrasts were performed to test for BMI and sex-related differences in measures of centrality, while controlling for age. RESULTS: In both males and females, individuals with high BMI (obese and overweight) had greater anatomical centrality (greater connectivity) of reward (putamen) and salience (anterior insula) network regions. Sex differences were observed both in individuals with normal and elevated BMI. In individuals with high BMI, females compared to males showed greater centrality in reward (amygdala, hippocampus and nucleus accumbens) and salience (anterior mid-cingulate cortex) regions, while males compared to females had greater centrality in reward (putamen) and sensorimotor (posterior insula) regions. CONCLUSIONS: In individuals with increased BMI, reward, salience and sensorimotor network regions are susceptible to topological restructuring in a sex-related manner. These findings highlight the influence of these regions on integrative processing of food-related stimuli and increased ingestive behavior in obesity, or in the influence of hedonic ingestion on brain topological restructuring. The observed sex differences emphasize the importance of considering sex differences in obesity pathophysiology.

RESUMO

OBJECTIVE: Chhattisgarh in India is a malaria-endemic state with seven southern districts that contributes approximately 50-60% of the reported malaria cases in the state every year. The problem is further complicated due to asymptomatic malaria cases which are largely responsible for persistent transmission. This study was undertaken in one of the forested villages of the Keshkal subdistrict in Kondagaon district to ascertain the proportion of the population harbouring subclinical malarial infections. STUDY DESIGN: Community-based cross-sectional study. METHODS: Mass blood surveys were undertaken of the entire population of the village in the post-monsoon seasons of 2013 and 2014. Fingerprick blood smears were prepared from individuals of all ages to detect malaria infections in their blood. Individuals with fever at the time of the survey were tested with rapid diagnostic tests, and parasitaemia in thick blood smears was confirmed by microscopy. Malaria-positive cases were treated with anti-malarials in accordance with the national drug policy. RESULTS: Peripheral blood smears of 134 and 159 individuals, including children, were screened for malaria infection in 2013 and 2014, respectively. Overall, the malaria slide positivity rates were 27.6% and 27.7% in 2013 and 2014, respectively, and the prevalence rates of asymptomatic malaria were 20% and 22.8%. This study showed that, for two consecutive years, the prevalence of asymptomatic malaria infection was significantly higher among children aged ≤14 years (34.4% and 34.1% for 2013 and 2014, respectively) compared with adults (15.2% and 18.2% for 2013 and 2014, respectively; P = 0.023 and 0.04, respectively). CONCLUSION: The number of asymptomatic malaria cases, especially Plasmodium falciparum, is significant, reinforcing the underlying challenge facing the malaria elimination programme in India.

Single crystals of YVO4 with different doping concentrations of Yb (1.5, 3.0, 8.0, and 15.0 at. %) and with good crystalline quality (FWHM ∼43-55 arc sec of rocking curve) were grown by the optical floating zone technique. Refractive index measurements were carried out at four wavelengths as a function of temperature. The measurements show that as the doping concentration of Yb is increased, the refractive index varies marginally for ne whereas there is a significant change in the value of no. The thermo-optic coefficient (dn/dT) was found to be positive with a value ∼10-5/°C, which is 1 order higher than that for the undoped YVO4 crystal. The thermo-optic coefficient is higher for ne compared to that of no. Also, a set of relations describing the wavelength dependence of the thermo-optic coefficient were established that are useful for calculating the thermo-optic coefficient at any temperature in the range 30°C-150°C and at any wavelength in the range 532-1551 nm.

RESUMO

Nutrient utilization, body condition and carcass traits of cull ewes were studied in three dietary regimens based on complete feed block (CFB) feeding to control (C) with rumen protected protein (RPP), CU [RPP + urea (6 g/kg)] and CUF [RPP + urea + rumen protected fat (RPF; 40 g/kg)]. The RPP component (g/kg) in C had 1% formaldehyde-treated soy flakes 50, mustard cake 50 and sesame cake 30. The mustard and sesame cakes were replaced with urea on equivalent N basis in CU and CUF. The ewes were offered ad libitum CFB composed (g/kg) of concentrate 650, roughage 300 and molasses 50. The digestibility of OM and EE was higher (p < 0.05) in CUF than in CU and C, while that of NDF and ADF was lower (p < 0.05). The CP digestibility was higher in both CU and CUF. Utilization of N and Ca was higher (p < 0.05) in CUF compared with C and CU. Urinary purine derivatives analysis and microbial N synthesis were similar in all the groups. Blood glucose concentration improved at 90 day compared with 0 day. Ruminal attributes showed a higher (p < 0.05) pH, total N, TCA precipitable N, entodiniomorphs and total ciliate population in CU and CUF than in the control. The CUF had higher (p < 0.01) DM, digestible OM and ME intake, which resulted in higher (p < 0.05) weight gain and better feed efficiency. Ewes in all the groups showed an improvement in carcass traits at 90 day. The pre-slaughter weight was higher (p < 0.05) and dissected and KOH bone content was lower in CUF. Thus, feeding of CFB with urea as a cheaper N source and RPF to enrich with energy supported the improved performance as evidenced from higher nutrient input, 'utilization and efficiency with enhanced carcass traits' for better marketability and returns from cull ewes.

RESUMO

In 2017, there is no adjuvant systemic therapy proven to increase overall survival in non-metastatic renal cell carcinoma (RCC). The anti-PD-1 antibody nivolumab improves overall survival in metastatic treatment refractory RCC and is generally tolerable. Mouse solid tumor models have revealed a benefit with a short course of neoadjuvant PD-1 blockade compared to adjuvant therapy. Two ongoing phase 2 studies of perioperative nivolumab in RCC patients have shown preliminary feasibility and safety with no surgical delays or complications. The recently opened PROSPER RCC trial (A Phase 3 RandOmized Study Comparing PERioperative Nivolumab vs. Observation in Patients with Localized Renal Cell Carcinoma Undergoing Nephrectomy; EA8143) will examine if the addition of perioperative nivolumab to radical or partial nephrectomy can improve clinical outcomes in patients with high risk localized and locally advanced RCC. With the goal of increasing cure and recurrence-free survival (RFS) rates in non-metastatic RCC, we are executing a three-pronged, multidisciplinary approach of presurgical priming with nivolumab followed by resection and adjuvant PD-1 blockade. We plan to enroll 766 patients with clinical stage ≥T2 or node positive M0 RCC of any histology in this global, randomized, unblinded, phase 3 National Clinical Trials Network study. The investigational arm will receive two doses of nivolumab 240 mg IV prior to surgery followed by adjuvant nivolumab for 9 months. The control arm will undergo the current standard of care: surgical resection followed by observation. Patients are stratified by clinical T stage, node positivity, and histology. The trial is powered to detect a 14.4% absolute benefit in the primary endpoint of RFS from the ASSURE historical control of 55.8% to 70.2% at 5 years (HR = 0.70). The study is also powered to detect a significant overall survival benefit (HR 0.67). Key safety, feasibility, and quality of life endpoints are incorporated. PROSPER RCC exemplifies team science with a host of planned correlative work to investigate the impact of the baseline immune milieu and changes after neoadjuvant priming on clinical outcomes.
