RESUMO

Reference genes (RGs) with uniform expression are used for normalization of reverse transcription quantitative PCR (RT-qPCR) data. Their optimization for a specific biological context, e.g. a specific tissue, has been increasingly considered. In this article, we compare RGs identified by expression data meta-analysis restricted to the context tissue, the jejunum of Mus musculus domesticus, i) to traditional RGs, ii) to expressed interspersed repeated DNA elements, and iii) to RGs identified by meta-analysis of expression data from diverse tissues and conditions. To select the set of candidate RGs, we developed a novel protocol for the cross-platform meta-analysis of microarray data. The expression stability of twenty-four putative RGs was analysed by RT-qPCR in at least 14 jejunum samples of the mouse strains C57Bl/6N, CD1, and OF1. Across strains, the levels of expression of the novel RGs Plekha7, Zfx, and Ube2v1 as well as of Oaz1 varied less than two-fold irrespective of genotype, sex or their combination. The gene set consisting of Plekha7 and Oaz1 showed superior expression stability analysed with the tool RefFinder. The novel RGs are functionally diverse. This facilitates expression studies over a wide range of conditions. The highly uniform expression of the optimized RGs in the jejunum points towards their involvement in tightly regulated pathways in this tissue. We also applied our novel protocol of cross-microarray platform meta-analysis to the identification of RGs in the duodenum, the ileum and the entire small intestine. The selection of RGs with improved expression stability in a specific biological context can reduce the number of RGs for the normalization step of RT-qPCR expression analysis, thus reducing the number of samples and experimental costs.

Fatty acids are essential for immune cell function. Maternal dietary fatty acid supply influences body fat composition of their offspring. As a first step to study immunonutritional interactions at an early age of pigs, four sows were fed a diet containing sunflower oil or oil from seal blubber during pregnancy and lactation. Corresponding piglets were sacrificed at three consecutive time points in the suckling period and their mesenteric lymph nodes and spleen were analysed by gas chromatography for levels of fatty acid. At the same time mononuclear cells of these organs and of the intestinal lymphoid tissue from the jejunum were isolated and subpopulations characterised by flow cytometry. Levels of fatty acids from the lymphatic organs of the piglets were significantly influenced by the maternal diet. The concentration of the fatty acids 20:5n-3, 22:5n-3 and 22:6n-3 were higher in the spleen and mesenteric lymph node of piglets suckled to sows of the test diet. Additionally, suckling time affected the levels of some long chain polyunsaturated fatty acids. Dietary effects were seen on some subpopulations including CD4-CD8alpha+ lymphocytes of the mesenteric lymph nodes and CD4+CD8alpha+ lymphocytes of the lamina propria, which were higher in the group fed seal blubber oil. The levels of CD21+ B-cells were higher in the group fed sunflower oil. The results indicate that the maternal diet and suckling time affect the fatty acid status of the investigated lymphatic tissues of piglets, but may have minor effects on the investigated lymphocyte subpopulations.

Infected breeder pigs and contaminated feed represent potential sources of Salmonella introduction to fattening pig herds and may thereby cause human infections acquired via consumption of contaminated pork. Modelling approaches such as quantitative microbial risk assessment could improve the design of strategies for control and tracing of Salmonella in the feed chain. However, the construction of such models requires a thorough understanding of the dynamics of the feed chain, including production processes, microbial processes and transport logistics. The present article illustrates a conceptual model of Salmonella in the pig feed chain and explores the possibilities for quantitative modelling including identifying major gaps in data. Information was collected from peer-reviewed scientific journals, official documents and reports and by means of interviews with experts from authorities and the feed industry. Data on prevalence of Salmonella in different parts of the feed chain are difficult to compare as observed prevalence may be biased by variations in sampling procedures as well as limitations of the detection methods. There are almost no data on numbers of Salmonella in commodities of the feed chain, which often makes it difficult to evaluate risks, intervention strategies and sampling plans in a quantitative manner. Tracing the source of Salmonella contamination is hampered by the risk of cross-contamination as well as various mixing and partitioning events along the supply chain, which sometimes makes it impossible to trace the origin of a lot back to a batch or producer. Available information points to contaminated feed materials, animal vectors and persistent contamination of production environments as important sources of Salmonella in feed production. Technological procedures such as hydrothermal or acid treatment can be used to control Salmonella in feed production. However, a large fraction of pig feed is produced without decontamination procedures. Prevention of recontamination and control of moisture throughout the chain are thus critical factors for controlling Salmonella in feed production. To verify successful control it is necessary to have monitoring strategies able to detect low levels of Salmonella heterogeneously distributed in large volumes of feed and feed material in bulk. Experience from monitoring programs and research investigations indicates that sampling of dust and sweepings from control points along the production line is an efficient strategy to gain an indication of Salmonella contamination.

The maternal-fetal transfer and subsequent uptake of sow milk enriched with n-6- or n-3-polyunsaturated fatty acids may not only influence neonatal body fat but may also have an impact on the immune function of newborn piglets. Sows were fed a diet containing sunflower oil as n-6-source or oil from seal blubber with long chain polyunsaturated n-3-fatty acids during pregnancy and lactation. Sow serum was investigated during pregnancy and serum and milk during lactation; piglet serum and liver were investigated in the suckling period until day 19. Piglet leukocyte subpopulations were characterised by flow cytometry and leukocyte proliferation was tested after stimulation with mitogens. No differences were noted in performance. The serum and milk fatty acid status of the sows was markedly influenced by the diet. Eicosapentaenoic acid (20:5n-3), 22:5n-3 and 22:6n-3 were higher (p < 0.001) in serum and liver of piglets delivered from sows fed the seal blubber oil. Piglets at birth had lower lymphocyte counts (p < 0.01) than piglets 19 days after birth. However, no influence of feeding the different oil sources was noted on lymphocyte phenotyping and leukocyte proliferation test. The results of the present study show that the maternal diet affected the fatty acid status of neonates, but much more in the sucking period. Immunological traits were not affected, probably as the mononuclear cell lineage is too immature around birth. Effects of PUFA n-3 might only be seen at a later time point or in the polymorphonuclear cell lineage as they were dominating right after birth.
