RESUMO

Global potent greenhouse gas nitrous oxide (N2O) emissions from soil are accelerating, with increases in the proportion of reactive nitrogen emitted as N2O, i.e., N2O emission factor (EF). Yet, the primary controls and underlying mechanisms of EFs remain unresolved. Based on two independent but complementary global syntheses, and three field studies determining effects of acidity on N2O EFs and soil denitrifying microorganisms, we show that soil pH predominantly controls N2O EFs and emissions by affecting the denitrifier community composition. Analysis of 5438 paired data points of N2O emission fluxes revealed a hump-shaped relationship between soil pH and EFs, with the highest EFs occurring in moderately acidic soils that favored N2O-producing over N2O-consuming microorganisms, and induced high N2O emissions. Our results illustrate that soil pH has a unimodal relationship with soil denitrifiers and EFs, and the net N2O emission depends on both the N2O/(N2O + N2) ratio and overall denitrification rate. These findings can inform strategies to predict and mitigate soil N2O emissions under future nitrogen input scenarios.

Experiments conducted to understand how arbuscular mycorrhizal (AM) inoculation or biochar application affect plant growth and heavy metal uptake have thus far looked at single applications of either soil amendment. There is little evidence of their synergistic effects, in particular for plants grown in cadmium (Cd) contaminated soil. We conducted a mesocosm experiment to investigate the effect of AM inoculation (Glomus intraradices BEG 141) and/or wheat-straw biochar amendment on maize (Zea mays L. cv. Hongdan No. 897) growth, antioxidant enzymatic activities, and Cd uptake, as well as soil Cd speciation under applications of 0, 3, 6 mg Cd per kg soil. Applying either AM inoculant or biochar alone significantly increased maize growth and reduced Cd uptake. Furthermore, solo AM inoculation alleviating Cd stress more fully than biochar, in turn facilitating maize growth and decreasing soil Cd translocation into plant tissue. Still, solo biochar amendment was more effective at inducing soil alkalinization and contributing to Cd immobilization. Adding biochar together with AM inoculant significantly promoted fungal populations compared to a control. Amending soil with AM inoculant and biochar together produced the largest increase in maize growth and decrease in tissue Cd concentrations. This effect was additive, with 79.1% greater biomass, 51.42%, 82.91%, 43.96% higher activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and 50.06%, 67.19%, 58.04% and 76.19% lower Cd concentrations in roots, stems, leaves, and ears, respectively, at a 6 mg kg-1 Cd contamination rate. The combined treatment also had a synergistic effect on inducing soil alkalinization and causing Cd immobilization, and decreasing Cd phytoavailability and post-harvest transfer risks. These results suggest that AM inoculation in combination with biochar application may be applicable not only for maize production but also for phytostabilization of Cd-contaminated soil.

Nitric oxide (NO) and polyamines (PAs) are two kinds of important signal in mediating plant tolerance to abiotic stress. In this study, we observed that both NO and PAs decreased alkaline stress in tomato plants, which may be a result of their role in regulating nutrient balance and reactive oxygen species (ROS), thereby protecting the photosynthetic system from damage. Further investigation indicated that NO and PAs induced accumulation of each other. Furthermore, the function of PAs could be removed by a NO scavenger, cPTIO. On the other hand, application of MGBG, a PA synthesis inhibitor, did little to abolish the function of NO. To further elucidate the mechanism by which NO and PAs alleviate alkaline stress, the expression of several genes associated with abiotic stress was analyzed by qRT-PCR. NO and PAs significantly upregulated ion transporters such as the plasma membrane Na(+)/H(+) antiporter (SlSOS1), vacuolar Na(+)/H(+) exchanger (SlNHX1 and SlNHX2), and Na(+) transporter and signal components including ROS, MAPK, and Ca(2+) signal pathways, as well as several transcription factors. All of these play important roles in plant adaptation to stress conditions.
