RESUMO

Silicon (Si) application could significantly alleviate the toxic effects of cadmium (Cd) on the growth and development of rice. Here, we examined the regulatory effects of Si on Cd accumulation and stress response in rice seedlings through a hydroponic root separation test. The results showed that the biomass of rice seedlings decreased significantly under Cd stress, while the addition of Si could alleviate such negative effect. The uptake, transfer, and accumulation of Cd in rice seedling were significantly affected by Si addition under Cd stress. Si application under the unilateral Cd stress (Si-Cd+Si, Si-Cd) increased Cd-retention coefficient of root by 83.3%-83.6%, which restricted the transfer of Cd from root to aboveground. However, the treatment with Si added to the non-stressed side (Si-Cd) elevated the uptake and accumulation of Cd in rice seedling, with the accumulation in root being increased by 48.2% when compared to the treatment under the unilateral Cd stress without the addition of Si (CK-Cd). The treatment with Si added in two sides (Si-Cd+Si) decreased the uptake of Cd both in root and aboveground parts by 36.7% and 54.9%, respectively. The addition of Si under bilateral Cd stress (Cd-Cd+Si) significantly reduced the Cd uptake of both the root and aboveground parts by 57.8% and 46.5%, respectively, compared to the treatment of bilateral Cd stress (Cd-Cd). Higher Si concentration in rice seedling was found under the Cd stress. More Si was accumulated in rice seedling to resist the Cd stress when Si was added. The addition of Si affected the absorption of other metal elements in rice seedlings, including calcium (Ca), magnesium (Mg) and manganese (Mn). The concentrations of Ca and Mg in root and aboveground parts were significantly increased by Si addition under bilateral Cd-stress (Cd-Cd+Si), but Mn concentration was changed with the stress degree of Cd. The activities of superoxide dismutase (SOD) and peroxidase (POD) in root were affected by Si under Cd stress, especially for the Si-Cd treatment. The activity of POD in the root of the Cd-stress side and that of SOD in non-stress side were significantly increased, which benefit to scavenging the free radicals induced by Cd stress. In conclusion, Si could regulate the growth of rice seedlings, the uptake of elements such as Cd and Si, and the antioxidant reaction of the root system under the Cd stress. High Si concentration in plant is conducive to enhancing Cd tolerance.

RESUMO

Aim To investigate the effects of aralosdie C (SMC)isolated from aralosides on contractile func-tion and calcium transients in ischemia / reperfusion(I/R)-induced rat myocardial injury and the role of CaMK II. Methods The cardiomyocytes were divided into control group,I/ R group,I/ R + SMC group,KN-93+ I/ R + SMC group. The effect of SMC on the con-tractile function and calcium transient of I/ R cells was measured by cell shrinkage and ion concentration sim-ultaneous detection system. Results SMC(8 μmol· L - 1 )improved the contractile function of I/ R cardio-myocytes and the calcium transients, and SMC in-creased the rate of calcium transient and [Ca2 +]i up /down regulation significantly. While CaMK II was blocked by KN-93,the effect of SMC on contractile function and calcium transients was weakened. Con-clusions SMC can significantly improve the systolic function and calcium transients of cardiomyocytes in I/R rats, and the protective effect of SMC on cardiomyo-cytes may be related to CaMK II.

RESUMO

Objective To investigate the effect of testosterone on mitotic orientation in rat prostate epithelial cells and the relative differential gene expression. Methods Twenty SPF male SD rats were divided into 2 groups at random and then subjected to castration. One group of rats was administrated with testosterone 3.7 mg daily for 30 days and the control group was only injected with olive oil.Microscopic analysis was performed using immunohistochemistry.Differential gene expression analysis was conducted by gene microarray and RT-PCR techniques. Results In the testosterone-adminis-trated group, there was a significant mitosis orientation parallel to the basement membrane. But in the control group, mito-sis orientation was oriented perpendicular to the basement membrane.Using the gene microarray and RT-PCR techniques, the cell proliferation genes such as Ran, Tgm4 and Wnt2 in Wnt signal pathway were up-regulated in the testosterone group.Conversely, suppressor cell proliferation genes such as Dkk3 and Fas were down-regulated.Conclusions Mitotic orientation of prostate epithelia cells is changed after testosterone administration.Wnt signal pathway and AR singling path-way also have an influence on the mitosis orientation and cell proliferation.

RESUMO

OBJECTIVE: To investigate the effect of pH value and fluoride ions on the corrosion resistance of pure Ti and Ni-Cr-Ti alloy in the artificial saliva. METHODS: Electrochemical technique was used to measure the electric potential of corrosion (Ecorr), current density of corrosion (Icorr) and polarization resistance (Rp) of pure titanium and Ti-Ni-Cr alloy in the artificial saliva with different pH value and fluoride concentrations. After electrochemical analysis, microstructure and phase diffraction were examined by FSEM. RESULTS: With the lower pH value, the Ecorr and Icorr of pure titanium and Ti-Ni-Cr alloy increased, the Rp decreased, there was a significant difference (P<0.05). The Ecorr and Icorr increased markedly, the Rp significantly reduced in the artificial saliva containing 0.2% NaF (P<0.01). FSEM showed that pure titanium and Ti-Ni-Cr alloy surface corrosion, pure titanium in the artificial saliva containing 0.2% NaF was most serious. CONCLUSION: Lower pH value decreases the corrosion resistance of pure titanium and Ti-Ni-Cr alloy and the artificial saliva containing fluoride ions decreases the corrosion resistance of pure titanium.

RESUMO

OBJECTIVES: To explore the role of androgen-estrogen balance in benign prostatic hyperplasia (BPH) induced by varying doses of estradiol/testosterone propionate (E(2)/TP) in castrated rats. MATERIALS AND METHODS: A total of 222 rats were divided into 37 groups at random, including 35 groups of different E(2)/TP, one control, and one castrated group. All 37 groups except the control group were castrated, for eliminating endogenesis of testosterone in rats. The treated groups were administered testosterone propionate (TP; at the dosages of 0.15, 0.74, 3.7, 18.5, and 92.6 mg/kg), combined with estradiol (E(2); at the dosage of 0, 0.4, 2, 10, 50, 250, and 1250 µg/kg) diluted in vegetable oil for 30 days, respectively, whereas the control groups received only vegetable oil. All prostate specimens were removed under anesthesia, then fixed and embedded in paraffin, for measuring the organ quotient, volume, area of prostate glandular cavity, and the height of prostate epithelia. RESULTS: When the dosages of TP were 0.15, 3.7, 18.5, and 92.6 mg/kg, the degree of prostatic hyperplasia had no obvious dose-effect relationship with E(2). When TP was 0.74 mg/kg, with the increase of the dosage of E(2), the volume and quotient of prostate were increasing. However, when the dosage of E(2)exceeded 50 µg/kg, E(2)/TP was 5/74, the prostatic volume did not increase obviously. CONCLUSION: The proper levels of E2/TP play an important role in the pathogenesis of BPH. In rats, the balance point of E(2)/TP is 5/74.

RESUMO

<p><b>BACKGROUND</b>Adrenocorticotrophin (ACTH)-secreting pituitary adenomas account for approximately 7% - 14% of all pituitary adenomas, but its pathogenesis is still enigmatic. This study aimed to explore mechanisms underlying the pathogenesis of ACTH-secreting pituitary adenomas.</p><p><b>METHODS</b>We used fiber-optic beadarray to examine gene expression in three ACTH-secreting adenomas compared with three normal pituitaries. Four differentially expressed genes from the three ACTH-secreting adenomas and three normal pituitaries were chosen randomly for validation by reverse transcriptase-real time quantitative polymerase chain reaction (RT-qPCR). We then analyzed the differentially expressed gene profile with Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway.</p><p><b>RESULTS</b>Fiber-optic beadarray analysis showed that the expression of 28 genes and 8 expressed sequence tags (ESTs) were significantly increased and the expression of 412 genes and 31 ESTs were significantly decreased. Bioinformatic and pathway analysis showed that the genes HIGD1B, EPS8, HPGD, DAPK2, and IGFBP3 and the transforming growth factor (TGF)-β signaling pathway and extracellular matrix (ECM)-receptor interaction pathway may play important roles in tumorigenesis and progression of ACTH-secreting pituitary adenomas.</p><p><b>CONCLUSIONS</b>Our data suggest that numerous aberrantly expressed genes and several pathways are involved in the pathogenesis of ACTH-secreting pituitary adenomas. Fiber-optic beadarray combined with pathway analysis of differential gene expression appears to be a valid method of investigating tumour pathogenesis.</p>

RESUMO

To evaluate the efficacy and toxicity of low-dose cytarabine and aclarubicin in combination with granulocyte colony-stimulating factor (G-CSF) protocol for patients with relapsed acute myeloid leukemia (AML). A total of fifty relapsed patients have been enrolled, including 13 early relapsed and 37 late relapsed. 24 patients were male and 26 were female, with age ranging from 15 to 69 (median 47) years. Out of them, 7 patients relapsed after allogeneic peripheral blood stem cell transplantation (allo-PBSCT), 3 patients relapsed after autologous peripheral blood stem cell transplantation (auto-PBSCT), 25 patients relapsed after received regimens including high dose cytarabine and 15 patients relapsed after CR or stopping chemical therapy themself in course of consolidatory therapy. 30 relapsed patients received CAG regimen, and 20 patients (control group) received an anthracycline in combination with cytarabine. The results indicated that after one course, the complete remission (CR) rate was 46.7% (14/30), the CR rate after allo-PBSCT was 50% (3/6), the early death rate was 3.3% in CAG group; and CR rate was 30% (6/20) and the early death rate was 15% in control group. Myelosuppression was mild to moderate, and no severe nonhematologic toxicity was observed in two groups. The overall median times in CAG group and control group were 22 and 19 months respectively. In conclusion, CAG regimen as the induction therapy is effective and well tolerable with low side effects for relapsed patients who had received high dose cytarabine, auto-PBSCT or allo-PBSCT.

RESUMO

PURPOSE: To investigate the correlation between Streptococcus mutans' adhesion and surface corrosion of the casting titanium and Ti alloy with different kinds of polishing. METHODS: The casting titanium and Ti alloy were respectively processed into size of 30 mm x 8mm x 1mm in tablets. Each material was prepared for 32 specimens, randomly divided into two groups, dealt with mechanical polishing and electrochemistry polishing, and then randomly divided into four groups (blank control, media control, inoculated media control for three months and six months).Four specimens were used in each group under hypoxia conditions. The labeled specimen surfaces were then aseptically placed in the test tube with one transfer per week. The specimens were removed and sterilized. The surface roughness of specimens was measured by FTS i120 contourgraph and the alteration of the surface was observed under SIRION scanning electron microscope. One-way ANOVA was performed with SPSS11.O software package. RESULTS: In casting titanium, blank control media control, inoculated media control for three months and six months showed no influence on Ra value(P>0.05).In Ti alloy, the Ra value of the inoculated media control group was significantly different from those of blank control group(P<0.05) and metal control group(P<0.05). All the specimen surfaces had pits in disorder and in different size. CONCLUSIONS: Streptococcus mutans' adhesion causes surface corrosion of the casting titanium and Ti alloy, and mechanical polishing and electrochemistry polishing have no influence on corrosion.

RESUMO

<p><b>AIM</b>To investigate the effect of diazoxide preconditioning and the role of ERK and JNK in cellular signaling during diazoxide preconditioning protection in isolated spontaneous hypertension rat (SHR) hearts.</p><p><b>METHODS</b>Hearts were isolated from male SHR rats, and perfused on a Langendorff apparatus. Five groups were considered (n = 6). Con: after 40 min perfusion the hearts were submitted to 25 min ischemia followed by 30 min reperfusion. IP: the hearts were preconditioned with 2 periods of 5 min ischemia and 10 min reperfusion prior to 25 min ischemia. DP: the hearts were preconditioned with 2 periods of 10 min K-H solution with 50 micromol x L(-1) diazoxide and 5 min K-H solution reperfusion prior to 25 min ischemia. 5-HD: perfuse with 100 micromol x L(-1) 5-HD (a special mitochondrial ATP sensitive potassium channel blocker) for 10 min followed by 30 min K-H solution perfusion before 25 min ischemia. 5-HD + DP: 100 micromol x L(-1) 5-HD was given for 10 min before diazoxide preconditioning.</p><p><b>RESULTS</b>During reperfusion, comparing with Con group, the recoveries of left ventricle developed pressure (LVDP), + dP/dt(max), - dP/dt(max) and left ventricle end diastolic pressure (LVEDP) were improved in IP and DP groups (P < 0.01 vs Con). At the end of reperfusion, compared with Con group, the expression of ERK in myocardium were higher in IP and DP groups (P < 0.01 vs Con), there was no significance between 5-HD and Con group, but 5-HD couldn't inhibit the expression of ERK induced by diazoxide preconditioning. The expression of JNK in IP and DP groups were decreased (P < 0.05 vs Con), this effect could been inhibited by 5-HD.</p><p><b>CONCLUSION</b>These results indicated that diazoxide preconditioning could mimic ischemic preconditioning, the activation of ERK expression and the declining of JNK expression involved in diazoxide preconditioning in isolated SHR hearts.</p>

RESUMO

OBJECTIVE: To introduce a method for repairing and restoring hand function of the patients suffering from large surface area burn with scarce normal skin for reconstruction. METHODS: Seven patients (ten hands) were treated in our department from April 1994 to February 2001. The TBSA involved with second- or third-degree burns was 85%-96%. All the hands had severe scar contracture on the dorsum and lost most of their function. A scarred skin flap, based on the ulnar border of the hand was elevated integrally on the dorsum. A random-pattern abdominal flap at a less-scarred area was designed and elevated to cover the defect of the hand while the scarred skin flap of the hand was transferred to the donor site of the abdominal flap. RESULTS: All patients were followed for 0.5 to 4 years postoperatively. The range of motion of the metacarpophalangeal joint and the space capacity of the first web were greatly improved. All patients regained self-care ability. CONCLUSIONS: This method is simple and has satisfactory results. Under the circumstances where normal skin was not available for reconstruction, the function of the burned hand could be greatly improved by this method.

RESUMO

<p><b>OBJECTIVE</b>To introduce a method for repairing and restoring hand function of the patients suffering from large surface area burn with scarce normal skin for reconstruction.</p><p><b>METHODS</b>Seven patients (ten hands) were treated in our department from April 1994 to February 2001. The TBSA involved with second- or third-degree burns was 85%-96%. All the hands had severe scar contracture on the dorsum and lost most of their function. A scarred skin flap, based on the ulnar border of the hand was elevated integrally on the dorsum. A random-pattern abdominal flap at a less-scarred area was designed and elevated to cover the defect of the hand while the scarred skin flap of the hand was transferred to the donor site of the abdominal flap.</p><p><b>RESULTS</b>All patients were followed for 0.5 to 4 years postoperatively. The range of motion of the metacarpophalangeal joint and the space capacity of the first web were greatly improved. All patients regained self-care ability.</p><p><b>CONCLUSIONS</b>This method is simple and has satisfactory results. Under the circumstances where normal skin was not available for reconstruction, the function of the burned hand could be greatly improved by this method.</p>
