RESUMO

Jasmines are commercially grown for their fragrant flowers and essential oil. The present study investigates the composition of the volatile compounds from flowers of Jasminum sambac cv. Ramanthapuram Gundumalli and its variants that were evolved through colchicine. GC-MS analysis revealed that the flowers possessed major terpenes and sesquiterpenes such as Linalool, α-farnesene, germacrene-D, geranyl Linalool and D-Limonene as well as benzenoids (including benzyl acetate, benzyl alcohol and (Z)-Cinnamyl benzoate). The relative abundance of these volatile compounds in the variants have shown higher percentages than their wild-type (parent) which indicates that the variants possessed enhanced volatile composition. The new variations generated in floral volatile composition of J. sambac through polyploidisation are likely to have significant impact on the loose flower and perfume industries. Besides, the identified unique compounds can also be used as metabolic signatures to characterise the novel variants.

Nimbolide, a tetranortriterpenoid (limonoid) compound isolated from the leaves of Azadirachta indica, was screened both in vitro and in silico for its antimicrobial activity against Fusarium oxysporum f. sp. cubense, Macrophomina phaseolina, Pythium aphanidermatum, Xanthomonas oryzae pv. oryzae, and insecticidal activity against Plutella xylostella. Nimbolide exhibited a concentration-dependent, broad spectrum of antimicrobial and insecticidal activity. P. aphanidermatum (82.77%) was more highly inhibited than F. oxysporum f. sp. cubense (64.46%) and M. phaseolina (43.33%). The bacterium X. oryzae pv. oryzae forms an inhibition zone of about 20.20 mm, and P. xylostella showed about 66.66% mortality against nimbolide. The affinity of nimbolide for different protein targets in bacteria, fungi, and insects was validated by in silico approaches. The 3D structure of chosen protein molecules was built by homology modelling in the SWISS-MODEL server, and molecular docking was performed with the SwissDock server. Docking of homology-modelled protein structures shows most of the chosen target proteins have a higher affinity for the furan ring of nimbolide. Additionally, the stability of the best-docked protein-ligand complex was confirmed using molecular dynamic simulation. Thus, the present in vitro and in silico studies confirm the bioactivity of nimbolide and provide a strong basis for the formulation of nimbolide-based biological pesticides. Cotton-melon aphid, Aphis gossypii Glover (Hemiptera: Aphididae), is emerging as a potential threat to cotton cultivation worldwide. The resistance categories in Gossypium arboreum to A. gossypii still need to be explored. We screened 87 G. arboreum and 20 Gossypium hirsutum genotypes against aphids under natural field conditions. Twenty-six selected genotypes from these 2 species were tested under glasshouse conditions for resistance categories (antixenosis, antibiosis, and tolerance). Resistance categories were assessed by no-choice antibiosis assay, free-choice aphid settling assay, cumulative aphid days using population buildup tests, chlorophyl loss index, and damage ratings. No-choice antibiosis experiment revealed that the G. arboreum genotypes GAM156, PA785, CNA1008, DSV1202, FDX235, AKA2009-6, DAS1032, DHH05-1, GAM532, and GAM216 had a significant adverse effect on aphid development time, longevity, and fecundity. Gossypium arboreum genotypes CISA111 and AKA2008-7 expressed a low level of antixenosis but possessed antibiosis and tolerance. Aphid resistance persisted uniformly at different plant developmental stages studied. The chlorophyl loss percentage and damage rating scores were lower in G. arboreum than in G. hirsutum genotypes, indicating the existence of tolerance in G. arboreum to aphids. Logical relations analysis of resistance contributing factors depicted the presence of antixenosis, antibiosis, and tolerance in the G. arboreum genotypes PA785, CNA1008, DSV1202, and FDX235, indicating their utility for evaluating the mechanisms of resistance and aphid resistance introgression breeding into G. hirsutum to develop commercially cultivated cotton lines.

Nucleotide Binding Site - Leucine Rich Repeat (NBS-LRR) genes play a significant role in plant defense against biotic stresses and are an integral part of signal transduction pathways. Vat gene has been well reported for their role in resistance to Aphis gossypii and viruses transmitted by them. Despite their importance, Vat like NBS-LRR resistance genes have not yet been identified and studied in cotton species. This study report hundreds of orthologous Vat like NBS-LRR genes from the genomes of 18 cotton species through homology searches and the distribution of those identified genes were tend to be clustered on different chromosome. Especially, in a majority of the cases, Vat like genes were located on chromosome number 13 and they all shared two conserved NBS-LRR domains, one disease resistant domain and several repeats of LRR on the investigated cotton Vat like proteins. Gene ontology study on Vat like NBS-LRR genes revealed the molecular functions viz., ADP and protein binding. Phylogenetic analysis also revealed that Vat like sequences of two diploid species, viz., G. arboreum and G. anomalum, were closely related to the sequences of the tetraploids than all other diploids. The Vat like genes of G. aridum and G. schwendimanii were distantly related among diploids and tetraploids species. Various hormones and defense related cis-acting regulatory elements were identified from the 2 kb upstream sequences of the Vat like genes implying their defensive response towards the biotic stresses. Interestingly, G. arboreum and G. trilobum were found to have more regulatory elements than larger genomes of tetraploid cotton species. Thus, the present study provides the evidence for the evolution of Vat like genes in defense mechanisms against aphids infestation in cotton genomes and allows further characterization of candidate genes for developing aphid and aphid transmitted viruses resistant crops through cotton breeding.

A recombinant inbred line mapping population of intra-species upland cotton was generated from a cross between the drought-tolerant female parent (AS2) and the susceptible male parent (MCU13). A linkage map was constructed deploying 1,116 GBS-based SNPs and public domain-based 782 SSRs spanning a total genetic distance of 28,083.03 cM with an average chromosomal span length of 1,080.12 cM with inter-marker distance of 10.19 cM.A total of 19 quantitative trait loci (QTLs) were identified in nine chromosomes for field drought tolerance traits. Chromosomes 3 and 8 harbored important drought tolerant QTLs for chlorophyll stability index trait while for relative water content trait, three QTLs on chromosome 8 and one QTL each on chromosome 4, 12 were identified. One QTL on each chromosome 8, 5, and 7, and two QTLs on chromosome 15 linking to proline content were identified. For the nitrate reductase activity trait, two QTLs were identified on chromosome 3 and one on each chromosome 8, 13, and 26. To complement our QTL study, a meta-analysis was conducted along with the public domain database and resulted in a consensus map for chromosome 8. Under field drought stress, chromosome 8 harbored a drought tolerance QTL hotspot with two in-house QTLs for chlorophyll stability index (qCSI01, qCSI02) and three public domain QTLs (qLP.FDT_1, qLP.FDT_2, qCC.ST_3). Identified QTL hotspot on chromosome 8 could play a crucial role in exploring abiotic stress-associated genes/alleles for drought trait improvement. RESUMO

The dawdling development in genetic improvement of cotton with conventional breeding program is chiefly due to lack of complete knowledge on and precise manipulation of fiber productivity and quality. Naturally available cotton continues to be a resource for the upcoming breeding program, and contemporary technologies to exploit the available natural variation are outlined in this paper for further improvement of fiber. Particularly emphasis is given to application, obstacles, and perspectives of marker-assisted breeding since it appears to be more promising in manipulating novel genes that are available in the cotton germplasm. Deployment of system quantitative genetics in marker-assisted breeding program would be essential to realize its role in cotton. At the same time, role of genetic engineering and in vitro mutagenesis cannot be ruled out in genetic improvement of cotton.
