One male and one female giant panda (Ailuropoda melanoleuca) from a Belgian zoo were anesthetized on eight different occasions over a course of 4 yr for electro-ejaculation (n = 3) or artificial insemination (n = 5). Medetomidine (0.03-0.04 mg/kg) and ketamine (2.5-3 mg/kg) were administered by intramuscular remote injection. Animals gained sternal recumbency with the loss of response to external stimuli after 4.9 ± 1.6 min (mean ± SD). The trachea was intubated with a 14-mm-internal diameter endotracheal tube; anesthesia was maintained with isoflurane in oxygen adjusted according to the required depth of anesthesia with a small-animal circle system. Physiological variables (heart rate, respiratory rate, oxygenation, end tidal carbon dioxide partial pressure and non-invasive blood pressure) were measured and remained within an acceptable range throughout anesthesia. Atipamezole (0.17-0.25 mg/kg) was administered intramuscularly after anesthesia. Recoveries were rapid and uneventful. Medetomidine 0.03 mg/kg and ketamine 2.5 mg/kg IM appeared to be the preferred doses for giant pandas.

Giant pandas feed almost exclusively on bamboo but miss lignocellulose-degrading genes. Their gut microbiome may contribute to their nutrition; however, the limited access to pandas makes experimentation difficult. In vitro incubation of dung samples is used to infer gut microbiome activity. In pandas, such tests indicated that green leaves are largely fermented to ethanol at neutral pH and yellow pith to lactate at acidic pH. Pandas may feed on either green leaves or yellow pith within the same day, and it is unclear how pH, dung sample, fermentation products and supplied bamboo relate to one another. Additionally, the gut microbiome contribution to solid bamboo digestion must be appropriately assessed. Here, gut microbiomes derived from dung samples with mixed colors were used to ferment green leaves, also by artificially adjusting the initial pH. Gut microbiomes digestion of solid lignocellulose accounted for 30-40% of the detected final fermentation products. At pH 6.5, mixed-color dung samples had the same fermentation profile as green dung samples (mainly alcohols), while adjusting the initial pH to 4.5 resulted in the profile of yellow dung samples (mainly lactate). Metaproteomics confirmed that gut microbiomes attacked hemicellulose, and that the panda's alpha amylase was the predominant enzyme (up to 75%).

Broad-spectrum beta-lactamase (BSBL)-producing Enterobacteriaceae impose public health threats. With increased popularity of zoos, exotic animals are brought in close proximity of humans, making them important BSBL reservoirs. However, not much is known on the presence of BSBLs in zoos in Western Europe. Fecal carriage of BSBL-producing Enterobacteriaceae was investigated in 38 zoo mammals from two Belgian zoos. Presence of bla-genes was investigated using PCR, followed by whole-genome sequencing and Fourier-transform infrared spectroscopy to cluster acquired resistance encoding genes and clonality of BSBL-producing isolates. Thirty-five putatively ceftiofur-resistant isolates were obtained from 52.6% of the zoo mammals. Most isolates were identified as E. coli (25/35), of which 64.0% showed multidrug resistance (MDR). Most frequently detected bla-genes were CTX-M-1 (17/25) and TEM-1 (4/25). Phylogenetic trees confirmed clustering of almost all E. coli isolates obtained from the same animal species. Clustering of five isolates from an Amur tiger, an Amur leopard, and a spectacled bear was observed in Zoo 1, as well as for five isolates from a spotted hyena and an African lion in Zoo 2. This might indicate clonal expansion of an E. coli strain in both zoos. In conclusion, MDR BSBL-producing bacteria were shown to be present in the fecal microbiota of zoo mammals in two zoos in Belgium. Further research is necessary to investigate if these bacteria pose zoonotic and health risks.

In captive rhinoceros stillbirth and perinatal death are recorded at a rate of 6-17% in the various species. At the same time there is a substantial lack of knowledge on rhinoceros parturition. Yet, predicting parameters for birth and progress of parturition are fundamental for the recognition of dystocia and perinatal problems. Therefore, we here intended to pay close attention to the Achilles heel of the 1.5-2.5 year reproduction cycle in rhinoceros, the parturition. For the prediction of parturition we recorded timelines for pre-birth udder development, genital swelling, milk production, behavioral unrest, and decrease of serum progesterone concentration and the gestational length in 19 white rhinoceros. First, second and third labour stage, foetal presentation and events in perinatal period were recorded to describe normal parturition and establish a guideline for better birth management in rhinoceros. Udder development and genital swelling were observed 3 and 2 weeks prior birth, respectively. Milk production was observed to start up to 3 weeks prior birth and increased significantly in the last week with most significant increase one day before parturition to 50.6 ± 45.4 mL (p < 0.006). Serum progesterone concentration started to decrease 7 days prior parturition and more significantly 48 h before parturition. While behavioral unrest and first stage labour was not observed reliably in all females the break of foetal waters and thus the start of second stage labour was unmistakably observed. Second stage labour, when foetal membranes had ruptured until the foetus was born, took 1:50 ± 0:20 h:min. Eighty-four percent of fetuses were born in anterior presentation (n = 16/19) and the final expulsion took <25 min suggesting that this is the normal presentation in white rhinoceros. In the less frequent posterior presentation final expulsion took up to 47 min. Overall, 95% of calves were born alive. Calves were standing and nursing in 0:55 ± 0:12 min and 3:32 ± 0:53 h:min, respectively. In 10.5% of births (n = 2/19) in anterior presentation perinatal complications occurred. Stillbirth occurred once (5.3% n = 1/19) when the foetus was born in posterior presentation. The recorded gestational length was 506 ± 2d. Delivering live offspring is of key importance to establish a new generation and secure long-term survival of a species. Various pre-birth changes, significant decrease of serum progesterone 48 h prior birth, different labour stages, foetal presentation and perinatal events described here add substantial knowledge on the understanding of normal rhinoceros parturition and may help diagnose dystocia and perinatal complications.

BACKGROUND: Echocardiography is the most frequently used non -invasive diagnostic tool to evaluate cardiac anatomy and function in domestic species but increasingly also in non -domestic species, especially since cardiac disease is being recognized as an important cause of death in captive primates. The purpose of this cross -sectional study was to investigate the feasibility of transthoracic echocardiography in healthy squirrel monkeys as well as to provide species specific normal values for standard echocardiographic measurements. A secondary aim was to determine plasma and serum levels of the cardiac biomarkers, N -terminal pro -brain natriuretic peptide (NT -proBNP) and cardiac troponin T (cTnT). Furthermore, a commercial, non -invasive, smartphone -based ECG (AliveCor Vet TM) monitoring device was used to evaluate the heart rate and rhythm and to diagnose possible arrhythmias. RESULTS: In this study, transthoracic echocardiography of 14 squirrel monkeys was performed in right and left lateral recumbency. Similar standard right parasternal and left apical images were obtained as in dogs and cats and normal values for routine two -dimensional, time motion mode and Doppler mode measurements were generated. Thirteen animals were considered healthy and one squirrel monkey was identified with significant aortic dilation and regurgitation and consequently values obtained from this animal were not used when species specific normal values were calculated. NT -ProBNP and cTnT concentrations were available for 7 of the 13 healthy monkeys with NT -proBNP concentrations below detection limit in all animals and a mean cTnT concentration of 0.049 ng/mL. Electrocardiography was performed in all squirrel monkeys. The mean heart rate was 172 bpm. Frequent supraventricular premature beats were diagnosed in the squirrel monkey suffering from significant aortic dilation and regurgitation. CONCLUSION: This study presents echocardiographic normal values and additional cardiovascular data in anaesthetised Saimiri monkeys, fundamental from both the perspective of zoo animal health care as well as scientific research, since the squirrel monkey is often used as an animal model for human disease.

Cardiac disease has been recognized as a major cause of death in captive nonhuman primates, which necessitates diagnostic (imaging) techniques to screen for and diagnose preclinical and clinical stages of possible cardiac conditions. Echocardiography is currently the most commonly used diagnostic tool for evaluation of cardiac anatomy and function. Complete with thoracic radiography and blood levels of two cardiac biomarkers, N-terminal probrain natriuretic peptide (NT-proBNP) and cardiac troponin T (cTnT), it gives an extensive examination of the cardiorespiratory system. The purpose of this cross-sectional cohort study is to describe normal thoracic anatomy using thoracic radiography, and to provide normal values for echocardiographic measurements in 20 ring-tailed lemurs (Lemur catta). Additionally, cardiac biomarkers were determined. Three radiographic projections of the thoracic cavity and a complete transthoracic echocardiography were performed in 20 clinically healthy ring-tailed lemurs during their annual health examinations. Similar standard right parasternal and left apical echocardiographic images were obtained as described in dogs and cats and normal values for routine two-dimensional (2D-), time-motion (M-) and Doppler mode measurements were generated. Furthermore, a noninvasive smartphone base ECG recording and blood concentrations of cardiac biomarkers were obtained. Other radiographic measurements are provided for the skeletal and respiratory systems such as the trachea to inlet ratio and tracheal inclination. Knowledge of the normal radiographic thoracic and echocardiographic anatomy and function are fundamental for the diagnosis and follow-up of cardiac disease in affected individuals and for species screening, and will be of added value in future research in and conservation of this endangered species.

The giant panda is known worldwide for having successfully moved to a diet almost exclusively based on bamboo. Provided that no lignocellulose-degrading enzyme was detected in panda's genome, bamboo digestion is believed to depend on its gut microbiome. However, pandas retain the digestive system of a carnivore, with retention times of maximum 12 h. Cultivation of their unique gut microbiome under controlled laboratory conditions may be a valid tool to understand giant pandas' dietary habits, and provide valuable insights about what component of lignocellulose may be metabolized. Here, we collected gut microbiomes from fresh fecal samples of a giant panda (either entirely green or yellow stools) and supplied them with green leaves or yellow pith (i.e., the peeled stem). Microbial community composition was substrate dependent, and resulted in markedly different fermentation profiles, with yellow pith fermented to lactate and green leaves to lactate, acetate and ethanol, the latter to strikingly high concentrations (∼3%, v:v, within 3.5 h). Microbial metaproteins pointed to hemicellulose rather than cellulose degradation. The alpha-amylase from the giant panda (E.C. 3.2.1.1) was the predominant identified metaprotein, particularly in reactors inoculated with pellets derived from fecal samples (up to 60%). Gut microbiomes assemblage was most prominently impacted by the change in substrate (either leaf or pith). Removal of soluble organics from inocula to force lignocellulose degradation significantly enriched Bacteroides (in green leaf) and Escherichia/Shigella (in yellow pith). Overall, different substrates (either leaf or pith) markedly shaped gut microbiome assemblies and fermentation profiles. The biochemical profile of fermentation products may be an underestimated factor contributing to explain the peculiar dietary behavior of giant pandas, and should be implemented in large scale studies together with short-term lab-scale cultivation of gut microbiomes.

Pseudopregnancy is a physiological occurrence in mammals which have copulation induced ovulation, but is rarely described in spontaneous ovulating species. In this study, three cases of prolonged luteal lifespan are reported in non-pregnant Asian elephants (Elephas maximus). Case 1 was a 25-year-old female that had produced three calves previously; Case 2 was a nulliparous and 32-year-old at the start of the pseudopregnancy episode; and Case 3 occurred in a 49-year-old nulliparous elephant. Serum progesterone metabolite concentrations remained elevated for 10 months in Case 1. Urinary progestagens were high for >16 months in Case 2 and for five months in Case 3. In Case 1, multiple persistent corpora lutea were visualized monthly by ultrasonography. In all three cases, uterine leiomyoma were present and progestagen concentrations decreased spontaneously. In Case 1, the elephant became pregnant 3 years later, whilst with Case 2, the female resumed estrous cycling normally, and for the Case 3 female, there was continuation with another prolonged luteal phase before ovarian function was purposely suppressed. These examples indicate that persistently elevated progestagen concentrations may not always be indicative of pregnancy in elephants. The reasons for prolonged luteal lifespan are not understood, although serum prolactin concentrations quantified in the Case 1 female were elevated compared to values from previous reports and two other herd mates. Furthermore, all three elephants had varying degrees of uterine pathologies. It is believed that the resulting damage to the endometrium may have led to a reaction similar to implantation, which includes prolactin secretion. Prolactin may exert luteotropic properties and is thought to initiate luteal rescue during pregnancy in elephants.

The purpose of this prospective study was to describe the normal anatomy and provide reference ranges for measurements of thoracic radiography on Squirrel monkeys (n = 13). Thoracic radiography is a common non-invasive diagnostic tool for both cardiac and non-cardiac thoracic structures. Furthermore cardiac disease is a common condition in captive primates. In this study, left-right lateral, right-left lateral and dorsoventral projections of 13 healthy Squirrel monkeys were reviewed during their annual health examinations. The mean Vertebral Heart Score on the left-right and right-left lateral projections were 8,98 ± 0,25 and 8,85 ± 0,35 respectively. The cardio-thoracic ratio on the dorsoventral projection was 0,68 ± 0,03. The trachea to inlet ratio was 0,33 ± 0,04. Other measurements are provided for the skeletal, cardiac and respiratory systems. Knowledge of the normal radiographic thoracic anatomy is fundamental in clinical as well as research settings for accurate diagnosis of diseases.

Reproductive monitoring for captive breeding in giant pandas is based on behavioural observation and non-invasive hormone analysis. In urine, interpretation of results requires normalisation due to an animal's changing hydration. Correction of urinary concentrations based on creatinine is the gold standard. In this study, a largely unexplored, easy-to-perform normalisation technique, based on urinary specific gravity (USpG), was examined and compared to creatinine. To this extent, six cycles from two female pandas (SB741(1) and SB569(5)) were monitored through urine analysis for oestrogen, progesterone, ceruloplasmin and 13,14-dihydro-15-keto-PGF2a (PGFM). The Pearson's correlation between creatinine and USpG was high (r = 0.805-0.894; p < 0.01), indicative for a similar performance of both normalisation methods. However, generally lower values were observed during pro-oestrus and primary (progesterone) rise. This could be associated with huge shifts in appetite, monitored by faecal output (kg) with an averaged > 50% decrease during oestrus and >50% increase during primary progesterone rise. In parallel, respectively highest and lowest creatinine and USpG levels, were measured, with creatinine obviously more affected as a result of linkage with muscle tissue metabolism affected by reproductive hormones. As a consequence, metabolite levels were significantly different between both corrected datasets with significantly higher oestrogen peak levels during oestrus ranging from 2.13-86.93 and 31.61-306.45 ng/mL (USpG correction) versus 2.33-31.20 and 36.36-249.05 ng/mL Cr (creatinine correction) for SB569 and SB741 respectively, and significant lower progesterone levels during primary progesterone rise ranging from 0.35-3.21 and 0.85-6.80 ng/mL (USpG correction) versus 0.52-10.31 and 2.10-272.74 ng/mL Cr (creatinine correction) for SB569 and SB741 respectively. Consequently, USpG correction rendered unbiased profiles, less subject to variation and metabolic artefacts and therefore allowed a more straightforward identification of peak oestrogen and onset of secondary progesterone rise, being potentially advantageous for future studies unravelling key giant panda reproductive events, including (delayed) implantation. The alternative application of USpG as a normalisation factor was further supported by its easy application and environmental and technical robustness.

General anesthesia poses risks for larger zoo species, like cardiorespiratory depression, myopathy, and hyperthermia. In ruminants, ruminal bloat and regurgitation of rumen contents with potential aspiration pneumonia are added risks. Thus, the use of sedation to perform minor procedures is justified in zoo animals. A combination of detomidine and butorphanol has been routinely used in domestic animals. This drug combination, administered by remote intramuscular injection, can also be applied for standing sedation in a range of zoo animals, allowing a number of minor procedures. The combination was successfully administered in five species of nondomesticated equids (Przewalski horse [ Equus ferus przewalskii; n = 1], onager [ Equus hemionus onager; n = 4], kiang [ Equus kiang ; n = 3], Grevy's zebra [ Equus grevyi ; n = 4], and Somali wild ass [ Equus africanus somaliensis; n = 7]), with a mean dose range of 0.10-0.17 mg/kg detomidine and 0.07-0.13 mg/kg butorphanol; the white ( Ceratotherium simum simum; n = 12) and greater one-horned rhinoceros ( Rhinoceros unicornis ; n = 4), with a mean dose of 0.015 mg/kg of both detomidine and butorphanol; and Asiatic elephant bulls ( Elephas maximus ; n = 2), with a mean dose of 0.018 mg/kg of both detomidine and butorphanol. In addition, the combination was successfully used for standing sedation in six species of artiodactylids: giraffe ( Giraffa camelopardalis reticulata; n = 3), western bongo ( Tragelaphus eurycerus eurycerus; n = 2), wisent ( Bison bonasus ; n = 5), yak ( Bos grunniens ; n = 1), water buffalo ( Bubalus bubalis ; n = 4) and Bactrian camel ( Camelus bactrianus ; n = 5). The mean dose range for artiodactylid species except bongo was 0.04-0.06 mg/kg detomidine and 0.03-0.06 mg/kg butorphanol. The dose in bongo, 0.15-0.20 mg/kg detomidine and 0.13-0.15 mg/kg butorphanol, was considerably higher. Times to first effect, approach, and recovery after antidote were short. The use of detomidine and butorphanol has been demonstrated to be a reliable, safe alternative to general anesthesia for a number of large ungulate species.

The Spix's macaw (Cyanopsitta spixii) is the rarest parrot on earth. The remaining captive population consists of 79 individuals. Captive propagation is ongoing to increase the number of individuals for future reintroduction back into the wild. Unfortunately, from 2004 to 2012, only 33 chicks hatched from 331 eggs. Semen evaluation and assisted reproduction might help to overcome this problem. Therefore, a recently developed electro-stimulated semen collection technique was used in Spix's macaws. Semen collection was successful in 39 of 78 attempts in 10 out of 17 males. Examination of the semen included evaluation of volume, color, consistency, contaminations and pH, as well as determination of motility, viability, morphology, concentration, and total count of spermatozoa. The median volume of semen samples was 5.6 µl. On average, 34.7 ± 21.9% (median 30%) of the sperm were motile and 23.1 ± 22.1% (median 16.5%) were progressively motile. In addition to spermatozoa, round cells were detected in the samples. Median sperm concentration was 15,500/µl (range 500-97,500/µl) and median viability was 50% (range 5-87%). Morphological examination revealed in 26.5% normal spermatozoa, high numbers of malformations of the head (50.2%) and tail region (20.5%), with 29% of all sperm showing multiple abnormalities. Artificial insemination was performed in three females; two eggs laid after artificial insemination had spermatozoa present on the perivitelline layer, suggesting the possible success of the insemination technique. Although no fertilization could be demonstrated, these preliminary results are promising, as they indicate that assisted reproduction might be a tool for species conservation in the Spix's macaw.

Basal metabolic rates in mammals are mainly determined by body mass, but also by ecological factors. Some mammalian species inhabiting hot, dry environments were found to have lower metabolic rates compared to temperate species. We studied energy metabolism in Phillip's dikdik (Madoqua saltiana phillipsi), a small antelope inhabiting xeric shrubland habitats in the Eastern 'horn' of Africa, and compared results to literature data. We measured body mass (BM) changes and digestibility in 12 adults kept on different food intake levels to determine, by extrapolation to zero BM change, maintenance energy requirements (MEm) for metabolizable energy (ME). The MEm averaged at 404±20kJMEkgBM(-0.75)d(-1). In addition we conducted 24h-chamber respirometry with seven fed (non-fasted) individuals. Their mean metabolic rate as calculated from oxygen consumption was 403±51kJkgBM(-0.75)d(-1), corroborating the results of the feeding experiments. Selecting the 20 lowest values of the respiration measurement period to estimate resting metabolic rate (RMR) resulted in a mean RMR of 244±39kJkgBM(-0.75)d(-1), which was not significantly lower than the expected basal metabolic rate of 293kJkgBM(-0.75)d(-1). Therefore, resting metabolism was similar to the expected average basal metabolism of a mammal of this size, which suggests a comparatively low metabolic rate in dikdiks. Compared to literature data Phillip's dikdiks have a MEm similar to measurements reported for small domestic ruminants, but considerably lower than those reported for other wild ruminant species inhabiting temperate and cold climates.

A highly lethal hemorrhagic disease associated with infection by elephant endotheliotropic herpesvirus (EEHV) poses a severe threat to Asian elephant husbandry. We have used high-throughput methods to sequence the genomes of the two genotypes that are involved in most fatalities, namely, EEHV1A and EEHV1B (species Elephantid herpesvirus 1, genus Proboscivirus, subfamily Betaherpesvirinae, family Herpesviridae). The sequences were determined from postmortem tissue samples, despite the data containing tiny proportions of viral reads among reads from a host for which the genome sequence was not available. The EEHV1A genome is 180,421 bp in size and consists of a unique sequence (174,601 bp) flanked by a terminal direct repeat (2,910 bp). The genome contains 116 predicted protein-coding genes, of which six are fragmented, and seven paralogous gene families are present. The EEHV1B genome is very similar to that of EEHV1A in structure, size, and gene layout. Half of the EEHV1A genes lack orthologs in other members of subfamily Betaherpesvirinae, such as human cytomegalovirus (genus Cytomegalovirus) and human herpesvirus 6A (genus Roseolovirus). Notable among these are 23 genes encoding type 3 membrane proteins containing seven transmembrane domains (the 7TM family) and seven genes encoding related type 2 membrane proteins (the EE50 family). The EE50 family appears to be under intense evolutionary selection, as it is highly diverged between the two genotypes, exhibits evidence of sequence duplications or deletions, and contains several fragmented genes. The availability of the genome sequences will facilitate future research on the epidemiology, pathogenesis, diagnosis, and treatment of EEHV-associated disease.

RESUMO

Mycobacterium liflandii has been responsible for an emerging infection reported in the international trade of Western clawed frogs (Silurana tropicalis). This study shows that this mycolactone-producing Mycobacterium (MPM) has expanded its distribution range to France. The results of this study suggest that the use of in vitro fertilization to maintain genetic lines could be a temporary solution for valuable S. tropicalis propagation.

Pre-determining fetal sex is against the random and equal opportunity that both conceptus sexes have by nature. Yet, under a wide variety of circumstances, populations shift their birth sex ratio from the expected unity. Here we show, using fluorescence in situ hybridization, that in a population of pygmy hippopotamus (Choeropsis liberiensis) with 42.5% male offspring, males bias the ratio of X- and Y-chromosome-bearing spermatozoa in their ejaculates, resulting in a 0.4337±0.0094 (mean±s.d.) proportion of Y-chromosome-bearing spermatozoa. Three alternative hypotheses for the shifted population sex ratio were compared: female counteract male, female indifferent, or male and female in agreement. We conclude that there appears little or no antagonistic sexual conflict, unexpected by prevailing theories. Our results indicate that males possess a mechanism to adjust the ratio of X- and Y-chromosome-bearing spermatozoa in the ejaculate, thereby substantially expanding currently known male options in sexual conflict.

HISTORY: Medical knowledge of pygmy hippopotami is limited. Anaesthesia has been considered a challenge because of the anatomy, semi-aquatic life style and aggressive behaviour. Polycystic kidney disease (PKD) has been described and can contribute to active kidney disease potentially affecting anaesthesia. PHYSICAL EXAMINATION AND MANAGEMENT: Fourteen pygmy hippopotami were anaesthetized for general health assessment and reproductive procedures. Animals (estimated bodyweight 250 kg) were darted intramuscularly with 0.08 mg kg(-1) medetomidine and 1.2 mg kg(-1) ketamine. After endotracheal intubation, anaesthesia was maintained with isoflurane delivered either by circle system (100% oxygen) or by Triservice apparatus (air or air/oxygen admixture). Heart rate (HR) respiratory rate (f(R) ), oxygen saturation (SpO(2)) and end tidal CO(2) were recorded at 5-minute intervals. Atipamezole was administered intramuscularly (0.4 mg kg(-1)) at the end of the procedure. Statistical analysis was performed using anova (p < 0.05). Most animals rapidly became recumbent although five hippopotami needed additional drugs to assure acceptable immobilization. There were no statistical differences in mean HR between animals with or without PKD (PKD: 34 ± 8 beats minutes(-1); no PKD: 33 ± 6 beats minutes(-1)), f(R) (PKD: 15 ± 7 breaths minutes(-1); no PKD; 12 ± 5 breaths minutes(-1)) and end tidal CO(2) (PKD: 7.1 ± 1.3 kPa; no PKD: 7.8 ± 1.4 kPa). SpO(2) was higher in animals receiving 100% oxygen or air with oxygen (92 ± 8% and 91 ± 9% respectively) compared with animals receiving air only (77 ± 5%) (p = 0.003). Recovery was uneventful after atipamezole administration. FOLLOW-UP: There were no apparent adverse effects after anaesthesia during a 24-hour follow-up period. DISCUSSION AND CONCLUSIONS: Medetomidine-ketamine-isoflurane induced satisfactory anaesthesia in this species. Incremental induction doses were related to remote injection and the animals' thick skin. There were no differences in anaesthetic parameters in animals with or without PKD. Supplemental oxygen should be mandatory during anaesthesia in this species.

Twenty-six adult semi-free-ranging Bennett's wallabies were anesthetized. Animals in group MA received medetomidine 0.1 mg/kg and alfaxalone 4 mg/kg i.m. in a 5-ml dart, whereas those in group MK received medetomidine 0.1 mg/kg and ketamine 5 mg/kg i.m. in a 3-ml dart. Dosages were based on estimated body weights. The wallabies were allowed to recover spontaneously or, if still nonresponsive at the end of the procedure, were given atipamezole 0.5 mg/kg (half the dose via i.m. and the other half via i.v.). Heart rate and respiratory rate were monitored at 5-min intervals, temperature at 10-min intervals, and two arterial blood samples were taken for blood gas analysis. Statistical analysis was performed by using analysis of variance (P < 0.05). The use of 5-ml darts in group MA compared with 3-ml darts in group MK could potentially increase the risk of iatrogenic trauma and should be considered. Induction and maintenance of anesthesia were satisfactory in both groups. There were no significant differences between the groups in mean time to first effect, recumbency, and approach, or to time to sternal recumbency and standing after reversal with atipamezole. Although bradycardia was present in both groups, no statistical differences were calculated for respiratory rate and heart rate, whereas the mean cloacal temperature was significantly lower in group MA (P = 0.01). Mixed acid-base disturbances occurred in both groups. All but one animal in group MK needed atipamezole at the end of the procedure. No adverse effects were observed after recovery.

OBJECTIVE: To investigate physiological and sedative/immobilization effects of medetomidine or dexmedetomidine combined with ketamine in free-ranging Chinese water deer (CWD). STUDY DESIGN: Prospective clinical trial. ANIMALS: 10 free-ranging adult Chinese water deer (11.0±2.6 kg). METHODS: Animals were darted intramuscularly with 0.08±0.004 mg kg(-1) medetomidine and 3.2±0.2 mg kg(-1) ketamine (MK) or 0.04±0.01 mg kg(-1) dexmedetomidine and 2.9±0.1 mg kg(-1) ketamine (DMK). If the animal was still laterally recumbent after 60 minutes of immobilization, atipamezole was administered intravenously (MK: 0.4±0.02 mg kg(-1), DMK: 0.2±0.03 mg kg(-1)). Heart rate (HR) respiratory rate (f(R)) and temperature were recorded at 5-minute intervals. Arterial blood was taken 15 and 45 minutes after initial injection. Statistical analysis was performed using Student's t-test or ANOVA. p<0.05 was considered significant. RESULTS: Animals became recumbent rapidly in both groups. Most had involuntary ear twitches, but there was no response to external stimuli. There were no statistical differences in mean HR (MK: 75±14 beats minute(-1); DMK: 85±21 beats minute(-1)), f(R) (MK: 51±35 breaths minute(-1); DMK; 36±9 breaths minute(-1)), temperature (MK: 38.1±0.7 °C; DMK: 38.4±0.5 °C), blood gas values (MK: PaO(2) 63±6 mmHg, PaCO(2) 49.6±2.6 mmHg, HCO(3)(-) 30.8±4.5 mmol L(-1); DMK: PaO(2) 77±35 mmHg, PaCO(2) 45.9±11.5 mmHg, HCO(3)(-) 31.0±4.5 mmol L(-1)) and biochemical values between groups but temperature decreased in both groups. All animals needed antagonism of immobilization after 60 minutes. Recovery was quick and uneventful. There were no adverse effects after recovery. CONCLUSION AND CLINICAL RELEVANCE: Both anaesthetic protocols provided satisfactory immobilisation. There was no clear preference for either protocol and both appear suitable for CWD.

Bilateral transpalpebral ultrasonography was performed on 53 captive Asian elephants (Elephas maximus) in order to describe the normal ultrasonographic appearance and measurements of the Asian elephant eye. Transpalpebral ultrasonography was performed using a portable ultrasound unit and a 4-7-MHz broadband curvilinear transducer on animals housed at seven institutions in the United Kingdom and in Sri Lanka. Both males and females were included in the study and ages ranged from 14 mo to 65 yr. Ultrasonic examinations were conducted on unsedated animals, without the use of topical or local anesthesia. The ultrasonographic appearance of the globe and intraocular structures of the Asian elephant eye is similar to that in other species. Biometry measurements recorded for adult (n=41) and juvenile (n=10) Asian elephants were: axial length, 3.44 +/- 0.21 cm and 3.18 +/- 0.19 cm (mean +/- SD); equatorial diameter, 3.88 +/- 0.32 cm and 3.60 +/- 0.24 cm; corneal thickness, 0.17 +/- 0.02 cm and 0.16 +/- 0.02 cm; anterior segment depth, 0.45 +/- 0.08 cm and 0.36 +/- 0.07 cm; lens diameter, 1.90 +/- 0.14 cm and 1.75 +/- 0.19 cm; lens depth, 1.01 +/- 0.12 cm and 0.94 +/- 0.10 cm, and posterior segment depth, 1.82 +/- 0.17 cm and 1.72 +/- 0.15 cm, respectively. Multiple linear regression analysis indicated a significant relationship between the explanatory variables (age, sex, and height) and the dependent variables (axial length, equatorial diameter, corneal thickness, anterior segment depth, lens diameter, and lens depth). The main finding of this statistical test was that the globe increases in size as the animal ages. Transpalpebral ultrasonography was found to be an effective and practical imaging modality in the evaluation of the Asian elephant eye, without the need for chemical restraint.
