OBJECTIVES: This study aimed to evaluate the optical and mechanical performance of two single-shade resin-based composites (RBCs) compared to those of a conventional RBC for restoring posterior teeth. METHODS: Two single-shade RBCs, Omnichroma (Tukoyama) and Vittra Unique APS (FGM), and a conventional RBC, Filtek Z350XT shade A2 (3M Oral Care), were evaluated in this study. The optical shade-matching performance was measured using a spectrophotometer. The light emitted by VALO Grand (Ultradent) and transmitted through the 2.0-mm RBC specimens was evaluated using beam profiling. Knoop hardness (KH, N/mm2), degree of conversion (DC, %) at the top and bottom, flexural strength (FS, MPa), elastic modulus (E, GPa), postgel shrinkage (Shr, %), and shrinkage stress (MPa) were evaluated. Scanning electron microscopy (SEM) was used to characterize the filler. Data for FS, E, and Shr were analyzed by one-way analysis of variance (ANOVA) and KH and DC by repeated one-way ANOVA measurement followed by Tukey test (α=0.05). The modified von Mises stress values, light transmission, and SEM images were analyzed qualitatively. RESULTS: All single-shade RBCs exhibited higher chromatic adaptation than the Filtek Z350XT (p<0.001). Omnichroma exhibited less color difference than Vittra Unique APS, irrespective of the shade. The light transmitted through Omnichroma and Vittra Unique APS increased during polymerization. KH and DC values were significantly reduced from the top to the bottom of the specimens for all RBCs (p<0.001); however, the ratio values were always higher than 80%. In general, all RBCs demonstrated similar mechanical properties. All RBCs exhibited a similar FS (p=0.083) and Shr value (p=0.144). Filtek Z350XT exhibited significantly higher E (p<0.001) than both single-shade RBCs. All RBCs exhibited similar shrinkage stress during restoration and similar residual stress during occlusal loading. CONCLUSIONS: Single-shade Omnichroma and Vittra Unique APS increased light transmission during light-activation, demonstrating better chromatic adaptation than conventional Filtek Z350XT. In general, Omnichroma and Vittra Unique APS exhibited similar mechanical properties and shrinkage stress distributions as Filtek Z350XT during light-activation and occlusal loading.

OBJECTIVE: This study aimed to evaluate the effect of activated charcoal toothpaste on the color stability of teeth subjected to tooth bleaching and pigmenting agents. METHODS: A total of 120 bovine crowns were randomly divided into 12 groups (n=10) according to two study factors: staining solutions (three levels): saliva (control), coffee, and red wine; and toothpaste (four levels): BPC, Bianco Pro Clinical (Bianco Oral Care) (Control); BIW, Black is White (Curaprox); BCA, Bianco Carbon (Bianco Oral Care); and NAT, Natural Suavetex (Suavetex). The samples were subjected to office bleaching with a 35% hydrogen peroxide-based gel (Whiteness HP Blue, FGM), followed by immersion in the solution for 45 minutes per day and daily toothbrushing for 7 days. The color (ΔE) and luminosity changes (ΔL*) were measured using reflectance spectroscopy (Vita EasyShade). The CIE values (L*, a*, b*) were measured at baseline after bleaching (T0) and immediately after immersion in solution each day (Ti1-Ti7) and after all toothbrushing cycles (Tb1-Tb7). ΔE and ΔL were analyzed using a two-way analysis of variance and Tukey's test (α=0.05). The clinically unacceptable level of ΔE > 3.3 was used to evaluate the color change. RESULTS: The color change was significantly influenced by the staining solutions and toothpastes (p<0.001). The color change (ΔE) was significantly higher when immersed in wine than in coffee, and lower ΔE values were observed for artificial saliva (control), irrespective of the toothpaste used. In artificial saliva, BPC, BIW, and BCA resulted in significantly lower ΔE values than NAT, which presented a clinically unacceptable level of dental color change (ΔE>3.3). Coffee resulted in a lower (L*) reduction than wine, irrespective of the toothpaste used. CONCLUSION: Charcoal toothpastes resulted in a color change on the surface of the tooth enamel (ΔE). The bleaching effect of the charcoal toothpastes and control evaluated in this study partially reduced the color changes on the surface of the tooth enamel caused by staining solutions but was unable to reestablish the measured values to the baseline. For teeth immersed in artificial saliva, the color change was not noticeable in BCA, BIW, and control-BPC (ΔE≥3.3), except for NAT, which showed a significant color change.

OBJECTIVE: This study was designed to evaluate the effects of charcoal toothpaste on the surface roughness, color stability, and marginal staining of resin composite restorations. METHODS: A total of 100 bovine incisors was collected. The crowns were sectioned and randomly divided into 10 groups (n=10) according to two study factors: toothpaste groups and nanoparticle resin composite groups. Five toothpastes-Bianco Pro Clinical (Bianco Oral Care, Uberlândia, MG, Brazil) - Control group; Bianco Carbon (Bianco Oral Care); NAT, Natural Suavetex Carvão Ativado (Suavetex, Uberlândia, MG, Brazil); Nano Action Black Be Emotion (Polishop, Jundiaí, SP, Brazil); and BIW, Black is White (Curaprox, Curaden AG, Kriens, Switzerland)-and two resin composites-Z350XT (Filtek Z350XT, 3M Oral Care) and Vittra (Vittra APS FGM, Joinville, SC, Brazil)-were used. Circular cavities with a diameter of 4 mm and a depth of 1 mm were prepared on the buccal face of the tooth crowns and restored with resin composites. The specimens were subjected to three months of simulated toothbrushing. The surface roughness (right angle [Ra], in micrometers [µm]) of the resin composites was measured before and after toothbrushing in five areas per specimen. The resin composite color and luminosity changes (ΔE and ΔL, respectively) were measured using reflectance spectroscopy (Vita EasyShade). Macro photographs were taken before and after toothbrushing to qualitatively analyze the marginal staining (MSt) of the resin composite restorations. Scanning electron microscopy (SEM) was performed before and after the simulated toothbrushing. Ra data were analyzed using two-way analysis of variance with repeated measures and the Tukey HSD test; MSt was analyzed using Kruskal-Wallis and Dunn tests (α=0.05), and the resin composite color change was analyzed using the clinically unacceptable level of ΔE > 3.3. RESULTS: Simulated brushing increased Ra irrespective of the resin composite or toothpaste used. No significant differences were found in Ra between the control group and all groups on which the charcoal toothpastes were tested. A clinically unacceptable level of resin composite color change (ΔE>3.3) was found after the use of most charcoal toothpastes. Use of Bianco Carbon resulted in marginal staining similar to that of the control group and was lower than that of the other charcoal toothpastes. Vittra brushed with black toothpaste showed the highest marginal staining. CONCLUSION: Use of charcoal toothpaste resulted in Ra values of resin composites similar to those found with conventional toothpastes. Charcoal toothpaste generally resulted in clinical resin composite color changes (ΔE). All charcoal toothpastes, except Bianco Carbon, caused marginal staining of the resin composite restorations.

The present study determined i) the presence of proteins (oviduct-specific glycoprotein, OVGP1; heat shock protein-70A, HSPA1A; heat shock protein-A8, HSPA8; annexin A1, ANXA1; annexin A5, ANXA5; and myosin-9, MYH9) known to be involved in early reproduction in the oviduct fluid (OF) of anestrous goats; and ii) the functional effect of during IVF on polyspermy modulation and embryonic development. In vitro-matured oocytes were co-cultured with spermatozoa (1.0, 2.0, or 4.0 x 106 cells/mL) for 18 h in SOF medium supplemented with 5 µg/mL of heparin, 4 µg/mL gentamicin, and 10% estrus sheep serum (CTRL1, CTRL2, and CTRL4 groups) or the same medium plus 10% OF (OF1, OF2, and OF4 groups) obtained from anestrus goats. The analysis of OF by western blotting confirmed the presence of the six proteins tested for. The increase in sperm concentration had no effect (P > 0.05) on the penetration rate in any group; however, monospermy rate decreased as sperm concentration was increased in both OF and CTRL. Regardless of the concentration used, when data were pooled, OF supplementation improved (P < 0.05) monospermy and tended (P = 0.057) to enhance IVF efficiency. Additionally, IVF efficiency was higher (P < 0.05) in OF1 than in OF4 [60 ± 13 vs 37 ± 5%). The development capacity was not affected (P > 0.05) by the sperm concentration and OF treatment, and the average values were cleavage (72 ± 2.6%), blastocyst (37 ± 3.0%), blastocyst in relation to the cleaved (51 ± 4.8%), hatched (62 ± 1.2%), and number of cells per blastocyst (174 ± 1.8%). In conclusion, the six proteins analyzed are present in the OF of anestrous goats, and the supplementation of this OF during IVF may modulate the polyspermy incidence and enhance IVF efficiency, especially when 1x106 sperm per mL is used.

This study evaluated the role of progesterone (P4) and medroxyprogesterone acetate (MAP) on the molecular status of immature cumulus-oocyte complexes (COCs) and the implications for oocyte quality in sheep. The number of viable COCs per ewe and the rate of COCs screened for developmental competence by brilliant cresyl blue positive (BCB+) were similar (P > 0.05), respectively, across treatments (P4: 7.7 ± 0.7 and 4.7 ± 1.2; MAP: 5.7 ± 1.0 and 3.5 ± 2.3; and control: 5.7 ± 1.1 and 3.6 ± 2.4). The COCs' gene expression was altered by exogenous progestogens compared with the control group: markers of steroidogenic pathway (FSH receptor [FSHr], LH receptor [LHr], and estradiol receptor α) and of quality (zygote arrest 1, growth differentiation factor 9, and B-cell lymphoma 2) were in abundance in P4 (P < 0.05). In addition, reelin protein (RELN) was downregulated, and Bcl-2 was upregulated in MAP (P < 0.05). In the P4 vs MAP comparison, FSHr, LHr, and RELN genes were upregulated (P < 0.05) in the P4 group. In conclusion, P4 and MAP promoted dissimilar effects on transcriptome profiling of immature BCB-selected COCs, possibly due to the differences in the chemical structure of progestogens and concentrations of serum P4. Exogenous P4 impacted positively on the profile of genes related to oocyte quality.

The intriguing questions concerning gall development refer to the processes of the remodelling of the host plant organ. Such processes involve the restructuring of cell walls and can be influenced by phenolics, indole-3-acetic acid (IAA) and reactive oxygen species (ROS). Alterations in cell walls demand the interference in the coupling of cellulose fibrils and hemicelluloses (xyloglucans) at specific stages of gall development. In addition to cell wall remodelling, hemicelluloses, such as the, xyloglucans and heteromannans can act as reserve carbohydrates, while xylans provide rigidity to the secondary cell walls. Developmental traits of the lenticular, fusiform and globoid galls on Inga ingoides (Fabaceae) were analysed using anatomical, cytometric, histochemical and immunocytochemical tools. Phenolics, IAA and ROS accumulated in similar gall tissue compartments, and may have influenced the restructuring of hemicelluloses and pectins. Contrary to expectations, cell wall flexibility regarding the dynamics of xyloglucans and cellulose fibrils does not relate to a temporal scale. The detection of xyloglucans in nutritive cell walls relate to carbohydrate nutritional resources to the galling insect, while xylans were associated to the lignified cell walls. Heteromanans were not detected, either in non-galled or galled tissues. The patterns of cell expansion during gall development relied on the relationship among phenolics, ROS and IAA with the hemicelluloses (xyloglucans and xylans) and cellulose fibrils. Although cell wall dynamics is specific to each gall morphotype in I. ingoides, the xyloglucans function as carbohydrate reserve to the gall inducers, which constitutes a functional trait common to the three morphotypes.

CLINICAL RELEVANCE: The clinician should consider the polymerization shrinkage stress when selecting a composite resin for posterior restorations. The use of post-gel shrinkage values should guide the selection of a composite resin for posterior teeth. SUMMARY: Objectives: The objective of this study was to evaluate the effect of the method used for calculation of polymerization shrinkage, total or post-gel, on the shrinkage stress of conventional and bulk-fill composite resins for restoring endodontically treated teeth using finite element analysis.Methods and Materials: Four composite resins were tested for post-gel shrinkage (P-Shr) by the strain-gauge test and total shrinkage (TShr) using an optical method (n=10). Two conventional composite resins, Filtek Z350 XT (3M-ESPE; Z350) and TPH3 Spectrum (Dents-ply; TPH3) and two bulk-fill composite resins. Filtek Bulk-Fill Posterior (3M-ESPE; POST) SureFil SDR flow (Dentsply; SDR) were tested. Elastic modulus (E), diametral tensile strength (DTS), and compressive strength (CS) were also determined (n=10). The residual shrinkage stress was evaluated by finite element analysis with four restorative techniques: incremental with Z350 and TPH3; SDR/TPH3 (two bulk increments of 4 mm and two occlusal increments); and two bulk increments of 5 mm for POST. Data for P-Shr, T-Shr, E, DTS, and CS were analyzed by analysis of variance and Tukey's test (α=0.05), and residual shrinkage was analyzed quantitatively and qualitatively by the modified von Mises criteria.Results: SDR had the lowest CS values, POST and TPH3 had similar and intermediate values, and Z350 had the highest CS. TPH3 and Z350 had similar DTS values and values higher than SDR. Z350 and POST had higher P-Shr, and SDR had lower T-Shr. T-Shr resulted in higher shrinkage stress than P-Shr values. SDR/TPH3 resulted in higher shrinkage stress when using T-Shr and lower values when using the P-Shr value.Conclusion: T-Shr resulted in higher stress in the enamel and in root dentin close to the pulp chamber than P-Shr values. The selection of the T-Shr or P-Shr changed the ranking of the shrinkage stress of the tested composite resin.

The effect of short-term administration of medroxyprogesterone acetate (MPA) or natural progesterone (P4) during ovarian stimulation with FSH on oocyte recovery was investigated in Santa Inês ewes. Ewes were treated with an intravaginal sponge containing MPA for 6 d; GnRH was applied 36 h after sponge removal and FSH was given in 3 injections (40, 24, and 16 mg, respectively) every 12 h after (D0, approximate ovulation time). At the first FSH dose, the ewes received either a new MPA sponge (n = 10) or a controlled device for internal release impregnated with P4 (n = 10) or did not receive any device (n = 10). Ovarian dynamics were assessed every 12 h by transrectal ultrasonography from D-3 to D2. Oocytes were recovered by laparoscopic ovum pick-up (LOPU) on D2 and graded by morphologic quality. The number of small, medium, and large follicles at D0 and D2 (ultrasound examinations), number of both follicles aspirated and oocytes recovered at LOPU, recovery rate, and oocyte grade did not differ (P > 0.05) among treatments. Thus, the short-term use of MPA or P4 during ovarian stimulation did not affect the first-wave follicle population or morphologic quality of oocytes. We would suggest that, in this protocol, the use of exogenous progestin is unnecessary.

The galls induced by Ditylenchus gallaeformans (Nematoda) on leaves of Miconia albicans have unique features when compared to other galls. The nematode colonies are surrounded by nutritive tissues with promeristematic cells, capable of originating new emergences facing the larval chamber, and providing indeterminate growth to these galls. Considering enzyme activity as essential for the translocation of energetic molecules from the common storage tissue (CST) to the typical nutritive tissue (TNT), and the major occurrence of carbohydrates in nematode galls, it was expected that hormones would mediate sink strength relationships by activating enzymes in indeterminate growth regions of the galls. Histochemical, immunocytochemical and quantitative analyses were made in order to demonstrate sites of enzyme activity and hormones, and comparative levels of total soluble sugars, water soluble polysaccharides and starch. The source-sink status, via carbohydrate metabolism, is controlled by the major accumulation of cytokinins in totipotent nutritive cells and new emergences. Thus, reducing sugars, such as glucose and fructose, accumulate in the TNT, where they supply the energy for successive cycles of cell division and for nematode feeding. The histochemical detection of phosphorylase and invertase activities indicates the occurrence of starch catabolism and sucrose transformation into reducing sugars, respectively, in the establishment of a gradient from the CST towards the TNT. Reducing sugars in the TNT are important for the production of new cell walls during the indeterminate growth of the galls, which have increased levels of water-soluble polysaccharides that corroborate such a hypothesis. Functional relationship between plant hormone accumulation, carbohydrate metabolism and cell differentiation in D. gallaeformans-induced galls is attested, providing new insights on cell development and plant metabolism.

We report a new method for histochemical localization of cytokinins (CKs) in plant tissues based on bromophenol blue/silver nitrate staining. The method was validated by immunohistochemistry using anti-trans-zeatin riboside antibody. Indole-3-acetic acid (auxin, IAA) was localized by anti-IAA antibody in plant tissues as a proof for IAA histolocalization. We used root sections, because they are major sites of CKs synthesis, and insect galls of Piptadenia gonoacantha that accumulate IAA. Immunostaining confirmed the presence of zeatin and sites of accumulation of IAA indicated by histochemistry. The colors developed by histochemical reactions in free-hand sections of plant tissues were similar to those obtained by thin layer chromatography (TLC), which reinforced the reactive sites of zeatin. The histochemical method for detecting CKs is useful for galls and roots, whereas IAA detection is more efficient for gall tissues. Therefore, galls constitute a useful model for validating histochemical techniques due to their rapid cell cycles and relatively high accumulation of plant hormones.

This study evaluated colour-Doppler ultrasound imaging (UI) as a substitute for laparoscopy to count the corpora lutea (CL) in superovulated sheep. Twenty-five Santa Ines ewes were superovulated three times at 21-day intervals. Corpora lutea were counted by colour-Doppler UI (CLDOPPLER ) 6 days after each superovulation and confirmed by laparoscopy (CLLAP ) 12 hr later. The mean number of CL was similar for both techniques (2.1 ± 2.5 vs. 2.1 ± 2.7 for CLDOPPLER and CLLAP , respectively) with a significant positive correlation (r = .94; r2 =.89). Colour-Doppler UI effectively evaluated the ovarian response in superovulated ewes and efficiently identified animals that did not respond to superovulation.

Objetivou-se avaliar a influência da somatotropina recombinante bovina (rbST) sobre a produção e os constituintes do leite de búfalas entre 63 e 154 dias em lactação. Foram utilizadas 22 búfalas, distribuídas em dois grupos experimentais: grupo rbST - aplicação de 500 mg de rbST a cada 14 dias; grupo controle - sem aplicação de rbST. A cada sete dias, foi aferida a produção de leite de todas as búfalas e coletada uma amostra para análise físico-química. As variáveis produtivas e as oriundas de análises laboratoriais foram avaliadas como medidas repetidas no tempo, utilizando-se o comando Repeated gerado pelo procedimento GLM do SAS. A média dos parâmetros estudados para os grupos rbST e controle foram, respectivamente: produção de leite - 6,54 vs. 6,68 kg; gordura - 6,31 vs. 6,34%; proteína 3,86 vs. 3,81%; lactose - 4,96 vs. 5,02%; sólidos totais - 16,05 vs. 16,03%; extrato seco desengordurado - 9,75 vs. 9,74%; contagem de células somáticas - 329,90 vs. 171,68 (x 1000/mL); e elecondutividade - 2,87 vs. 2,81mS/cm. This study aimed to evaluate the effect of recombinant bovine somatotropin (rbST) on milk yield and the proportion of buffalo milk components during lactation. Twenty-two buffaloes randomly distributed in two experimental groups were used: Group rbST - application of 500mg rbST every 14 days, between 63 and 154 days in milk (DIM); Control Group - without treatment. Weekly, the milk yield of buffaloes was measured and a sample was collected for physicochemical analysis. The response variables were evaluated as repeated measures, using the Repeated procedure through the GLM procedure of SAS. Means of each variable after rbST and Control were: Milk yield - 6,54 vs. 6,68 kg; Fat - 6,31 vs. 6,34%; Protein - 3,86 vs. 3,81%; Lactose - 4,96 vs. 5,02%; Milk solids - 16,05 vs. 16,03%; Defatted dry matter - 9,75 vs. 9,74%; Somatic Cells Count - 329,90 vs. 171,68 (x 1000/mL); and electrical conductivity- 2,87 vs. 2,81mS/cm. The use of 500mg of rbST administered every two weeks, between 63 and 154 DIM did not affect milk yield, proportion of milk constituents and SCC of dairy buffaloes.(AU)

OBJECTIVES: To compare bulk-fill with incremental filling techniques for restoring large mesio-occlusal-distal (MOD) restorations. METHODS: Seventy-five molars with MOD preparations were divided into five groups: Z350XT, incrementally filled with Filtek Z350XT and four bulk-fills-FBF/Z350XT, Filtek Bulk Fill/Filtek Z350XT; VBF/CHA, Venus Bulk Fill/Charisma Diamond; SDR/EST-X, SDR/Esthet-X HD; TEC, TetricEvoCeram Bulk Fill. Cuspal strains were measured using strain-gauges (n=10): CSt-Re, during restorative procedure; CSt-100N, during 100N occlusal loading; CSt-Fr, at fracture load. Before fracture load, teeth were load-cycled. Fracture resistance, fracture mode, and enamel cracks were recorded. The other five teeth were used for Elastic modulus (E) and Vickers hardness (VH). Post-gel shrinkage (Shr), diametral tensile strength (DTS) and compressive strength (CS) were determined (n=10). Shrinkage stresses were analyzed using finite element analysis. RESULTS: SDR had similar CS values as TEC, lower than all other composites. CHA had similar DTS values as Z350XT, higher than all other composites. Z350XT had the highest mean Shr and SDR the lowest Shr. New enamel cracks and propagation was observed after the restoration, regardless of filling technique. Z350XT had lower fracture resistance than bulk-fill composite techniques. No significant differences in failure modes were found. E and VH were constant through the depth for all techniques. Bulk-filling techniques had lower stresses compared to Z350XT. CONCLUSIONS: Flowable bulk-fill composites had lower mechanical properties than paste bulk-fill and conventional composites. All bulk-fill composites had lower post-gel shrinkage than conventional composite. Bulk-fill filling techniques resulted in lower cusp strain, shrinkage stress and higher fracture resistance. CLINICAL SIGNIFICANCE: Using bulk-fill composites cause lower CSt wich indicates lower stress in restored tooth. Furthermore, bulk-fill composites have a higher fracture resistance. Therefore, clinicians may choose the bulk-fill composite to decrease undesirable effects of restoration while simplifying filling procedure.

OBJECTIVE: The aim of this paper was to evaluate our results of radiofrequency catheter ablation (RFCA) of accessory pathways in patients with WPW syndrome. STUDY PATIENTS: We studied 100 consecutive patients with WPW syndrome, 52 men and 48 women, mean age 37 +/- 15 years who underwent RFCA. All patients were symptomatic, with documented episodes of supraventricular tachycardia and 9% of patients had underlying cardiac disease. METHODS: The RFCA was performed without antiarrhythmic drugs in the same session of the electrophysiologic diagnosis. The location of the accessory pathway site was obtained by catheter mapping, based on the premature and/or the presence of Kent potentials. According to the location of the accessory pathway, the ablation catheter was introduced either by the femoral vein or artery with mapping of the tricuspid or mitral ring. In the first cases performed energy application was manually controlled and thereafter was temperature guided with an upper temperature limit of 70 degrees C. We considered primary success criteria the disappearance of the delta wave in the surface ECG and the absence of ventricular preexcitation under atrial pacing and after adenosine injection. Clinical success was defined as the absence of clinical recurrence of tachycardia during the follow-up period. RESULTS: The primary success rate achieved was 88%; 91% in the left free wall pathways, 100% in the right free wall and 85% in the septal pathways (antero-septal-83%; right postero-septal-76.5%; left postero-septal-92%). A second ablation procedure was performed in seven of the twelve patients with primary unsuccess obtaining a final success rate of 93% (left free wall-94.5%; septal pathways-91.6%). After a mean follow-up period of 8 +/- 7 months clinical recurrence occurred in 9% (eight patients), five of which are under anti-arrhythmic therapy (62.5%). Clinical success rate at the end of the follow-up period was 88%. CONCLUSIONS: In our experience RFCA has shown to be safe and with a high success rate in patients with symptomatic pre-excitation. In this group of patients it was an effective therapy.
