RESUMO

Under complete denervation of m. gastrocnemius by crushing of sciatic nerve it was established that the intensity of oxygen free radicals formation increased 3 times, lipid peroxidation intensity 2 times, and the activity of antioxidative enzymes (glutathione reductase, superoxide dismutase) increased by 30-50%. Under these conditions the content of sarcolemmal neutral lipids increases and that of phospholipids decreases, proteins disintegrate, muscle atrophy increases. The use of the substances stimulating growth of tissues (neurobolil and thyroxin) 3-5 times increases the activity of enzymes of antioxidant protection of glutathione oxidase and superoxide dismutase. Considerable decrease of lipids peroxidation is observed, and the level of the muscle atrophy decreases.

RESUMO

In different time after application of 12.5 microM colchicine-inhibitor of exoplasmic current to the ischiadicus the quantity of amino acid residues of methionine and cysteine in the sarcoplasmic reticulum decreases, its protein content changes. The Ca(2+)-ATPase activity increases on the 3d day after colchicine application and decreases on the 14th day. Analogical changes are observed in the capacity of the sarcoplasmic reticulum to transport calcium ions, the Michaelis constant in this case being stable. Passive outflow of Ca2+ from the sarcoplasmic reticulum vesicules changes in such a way: the Michaelis constant decreases with a simultaneous increase in the maximal velocity after 14 and 28 days of colchicine application.

RESUMO

The ability of sarcoplasmic reticulum to regulate Ca(2+)-metabolism was studied at different terms after surgical denervation. The increase of Ca(2+)-ATP-ase activity, intensification of active Ca(2+)-accumulation under insignificant change of passive Ca(2+)-outflow from sarcoplasmic reticulum vesicles were observed 3-days after denervation. At more remote terms 14 days and 28 days after denervation all these parameters decreased except for Km: Ca(2+)-ATP activity and active Ca2+ accumulation decreased below normal level.

RESUMO

It is shown in the experiments on rats that the atrophy of skeletal muscles which has developed as a result of disturbances of neurotrophic control leads to the changes in protein spectrum of sarcolemma, growth of activity of membrane-associated enzymes (Na+, K(+)-ATPase, Ca(2+)-ATPase), intensive functioning of the prooxidative and antioxidative system of the muscle fibers.

RESUMO

It is demonstrated by experiments with rabbits that the Ca2+-ATP-ase activity is stabilized when using combined anesthetics (diacetylcholine + halothane + N2O) as distinct from application of halothane. A decrease in the cholinesterase activity is less pronounced than under the halothane action but more than with the diacetylcholine application. A decrease in the Na+, K+-ATP-ase activity is observed with all types of anesthesia. A considerable inhibition of creatine kinase under the action of combined anesthesia and halothane and an increase of the lactate dehydrogenase activity under diacetylcholine application in mitochondria are shown. Reliable differences in the succinic dehydrogenase activity are not detected.

RESUMO

Ca2+-ATPase of rats and rabbits skeletal muscle sarcolemma was studied as affected by relaxants: tubocurarine, myorelaxin and dioxonium, and by anesthetic fluothane in experiments in vitro and in vivo with lung artificial ventilation during curarization. The musclular relaxant, myorelaxin and anesthetic fluothane both in the experiments in vitro and in vivo inhibited the activity of the enzyme, dioxonium under these conditions caused no significant changes. Tubocurarin in vitro activated the enzyme and in vivo, vice versa, induced inhibition of the enzymic activity. The latter evidences for a many-sided character of the tubocurarine metabolic effect in the living organism. Combination of tubocurarine with fluothane had a less pronounced inhibitory effect on the activity of Ca2+-ATPase than the components used separately.
