RESUMO

This experiment compared the effects of 2 chronic heat stress (HS) models, constant (coHS), and cyclic (cyHS), on broiler performance, carcass characteristics, and meat quality. A total of 720 male chicks from a Cobb 500 line were placed in 12 environmentally controlled chambers divided into 2 pens of 30 birds. Before the experimental HS models were applied, chamber temperatures were gradually decreased from 32°C at placement to 24°C on d 20. From 20 to 41 d, 4 chambers were set to 35°C (coHS), and 4 chambers were set to 35°C for 12 h and 24°C for the next 12 h (cyHS). Four thermoneutral chambers were maintained at 24°C with half of the birds pair-fed to equalize feed intake (FI) with coHS birds (TN-coPF) and half fed ad-libitum (TN-al). From 20 to 41 d, FI and BW gain (BWG) of cyHS, coHS and TN-coPF birds were decreased (P < 0.001), whereas feed conversion ratio (FCR) was increased (P < 0.001) for coHS and TN-coPF birds compared with TN-al birds. The overall BWG and FCR of coHS birds were lower (P < 0.001) than TN-coPF birds. Both HS models reduced (P < 0.001) carcass weight, pectoralis major yield, total breast meat yield, and increased (P < 0.001) wing yield relative to TN-al birds, with each of these measurements more impacted by coHS than by cyHS. Pair-fed birds had lower (P < 0.001) fat pad and a higher total breast meat yield than coHS birds. They also had the lowest (P < 0.001) pectoralis major ultimate pH and yellowness, and these parameters were lower (P < 0.001) for coHS birds than for TN-al birds. Both HS models reduced (P < 0.001) the incidence of woody breast and white striping. Thus, these data indicate that the detrimental effects of HS cannot be entirely explained by reduced FI and that HS per se affects metabolic pathways associated with muscle and lipid accretion in broilers.

This study aimed to determine the effects of supplementing pregnant heifers with the organic selenium (Se) source 2-hydroxy-4-methylselenobutanoic acid (HMSeBA) during the last 8 wk of pregnancy on dam and calf Se status. A total of 42 in-calf heifers were recruited to the study and randomly allocated to 1 of 3 treatments; a negative control (Con), sodium selenite (NaSe), or HMSeBA. Animals were blocked by body weight, body condition score, and expected calving date before treatment allocation. Following enrollment, all animals underwent a 7-wk wash-out period, after which they received their respective supplements, top-dressed daily onto a basal diet for the last 8 wk of pregnancy. Heifer blood samples were taken at weekly intervals from enrollment until 2 wk before expected calving date and as soon as possible after calving for determination of whole-blood glutathione peroxidase activity (GSH-Px) and plasma Se and malondialdehyde (MDA) concentrations. Selenized AA were determined in plasma samples taken at 3 wk precalving. A colostrum sample was taken as close to parturition as possible for determination of colostrum total Se, selenized AA, and IgG concentration. Calves were blood sampled as close to birth as possible for determination of whole-blood GSH-Px activity and plasma Se and MDA concentrations. Differences in whole-blood GSH-Px activity did not become apparent until calving; GSH-Px activity was lowest in Con heifers but similar between NaSe and HMSeBA heifers. Plasma Se was lowest in unsupplemented heifers and greatest in those supplemented with HMSeBA; this was attributable to greater selenomethionine concentrations in the plasma of HMSeBA heifers. Colostrum Se was lowest in Con heifers and greatest in HMSeBA heifers. The greater Se concentration of HMSeBA heifers was attributable to a greater proportion of total Se comprising selenocysteine; the reason for this is not known. There was no effect of supplementation on colostrum IgG concentration. Plasma Se was lowest in calves born to Con heifers and greatest in those born to HMSeBA heifers. There were no effects of treatment on calf whole-blood GSH-Px activity or plasma MDA concentration. The enhanced Se status associated with HMSeBA supplementation is likely a consequence of selenomethionine supply and may confer benefits to both the dam and her calf postpartum.

The aim of this study was to compare the efficacy of a new organic Se (2-hydroxy-4-methylselenobutanoic acid [HMSeBA]) source (SO) with sodium selenite (SS) and selenized yeast (SY) at various dietary levels for growth performance and tissue Se deposition in growing pigs. A total of 112 crossbred (Pietrain × [Large White × Landrace]) gilts were allotted at an average body weight of 26.73 kg to 7 dietary treatments with 8 replicate pens of 2 pigs per pen. Pigs were fed basal diets unsupplemented or supplemented either with SS, SY, or SO each at 0.1 or 0.3 mg Se/kg of diet for 32 d. Feed intake and BW were recorded during the experimental period. At the end of the experiment, blood, liver, and psoas major muscle of all gilts were collected for total Se and relative bioavailability determination. No differences were observed on final BW, ADG, ADFI, and G:F among dietary treatments. All Se-supplemented groups exhibited greater total Se contents in plasma (P < 0.01) and liver (P < 0.01) compared with unsupplemented control group. However, Se retention in psoas major muscle was improved only when organic Se source (SY or SO) was added to diets (P < 0.01). Regardless the Se level, the Se deposition in muscle was greater (P < 0.01) in pigs supplemented with SO than those supplemented with SY. Slope ratio assay confirmed the greater bioavailability of Se from organic compared with inorganic Se and also revealed that the relative bioavailability of Se from HMSeBA for plasma, liver, and muscle Se response was 170, 141, and 162%, respectively, for SY. This study shows a potential advantage of HMSeBA supplementation in the increase of Se contents in pig tissues, indicating that this new organic Se source could be an alternative source of Se in swine nutrition.

The aim of this study was to compare the effects of a new organic Se [2-hydroxy-4-methylselenobutanoic acid (HMSeBA)] with routinely used mineral and organic Se sources (sodium selenite and selenized yeast) on chosen performance criteria and Se deposition in egg and muscle of laying hens. A total of 240 laying hens (40 wk of age) were randomly assigned to 6 treatments for 56 d with 8 replicates of 5 hens per replicate. The 6 treatments were as follows: control group received basal diet without Se supplementation; the second, fourth, and sixth experimental groups (SS-0.2, SY-0.2, and HMSeBA-0.2, respectively) were fed basal diet supplemented with Se at 0.2 mg/kg from sodium selenite, selenized yeast, and HMSeBA, respectively; and the third and fifth experimental groups (SY-0.1, and HMSeBA-0.1, respectively) were fed basal diet supplemented with Se at 0.1 mg/kg from selenized yeast and HMSeBA, respectively. No difference was observed among dietary treatments on feed intake, egg weight, and laying rate. All hens fed the Se-supplemented diets exhibited greater total Se contents in their eggs compared with control hens (P < 0.01). The egg Se concentrations were greater in hens fed organic Se (HMSeBA-0.2, P < 0.01, and SY-0.2, P < 0.01) than those fed the SS-0.2. In addition, hens fed the diet with HMSeBA-0.2 accumulated more Se in their eggs (+28.78%; P < 0.01) and muscles (+28%; P < 0.01) than those fed the diet supplemented with SY-0.2. These results showed the greater ability of HMSeBA to increase Se deposition in eggs and breast muscle of laying hens, which can subsequently lead to greater supply of Se for humans.

The uncommon amino acid 3,4-dihydroxyphenylalanine (DOPA) was determined colorimetrically using isonicotinic acid hydrazide (IAH) in the crude extracts obtained from fresh material of Vicia faba minor. DOPA was found to accumulate at concentrations inconsistent with its solubility in water. Other orthodiphenols do not interfere in DOPA determination. The accumulated DOPA was only of the L-form. The DOPA content in the dry material of Vicia faba was significantly lower than that from the corresponding fresh material, suggesting some kind of DOPA oxidation during drying. Subsequent analyses designed to evaluate the actual potential of V. faba minor in the accumulation of DOPA were performed with fresh material. It was observed that the DOPA concentration in the tissues of V. faba minor is reproducible when expressed on a fresh matter weight basis. In young seedlings L-DOPA production is significantly enhanced by light via an increased fresh matter yield. The DOPA content was found to be increased in mature plants grown on media enriched with low concentrations of NaCl (up to 25 mM) that showed a positive effect on their growth. NaCl induced an overproduction of L-DOPA even when growth was restricted by ion excess. On the contrary, the DOPA level dropped in mature plants submitted to water stress. Nodulated plants were found to contain higher content of L-DOPA than the non-nodulated ones grown with NO(3)(-) and NH(4)(+). It is suggested that DOPA hyperproduction by Vicia faba minor might be optimized by growing plants in mild saline conditions and by inoculating them with efficient salt tolerant strains of R. leguminosarum.
