RESUMO

A previous method for determination of the radiochemical purity (RCP) value of [125I]sodium iothalamate uses two paper strips and solvents (total developing time is approximately 2.5 h). To simplify and shorten the RCP testing procedure, our laboratory has developed a single-strip chromatography method that not only distributes free 125I and [125I]sodium iothalamate to different relative front (Rf) locations, but is also faster and easier to perform. RCP of [125I]sodium iothalamate was determined with the use of a 10-cm instant thin-layer chromatography strip impregnated with polysilicic acid gel (ITLC-SA) as the solid phase, and a mobile phase of 2-butanol:acetic acid:water (140:2.5:70, v/v). By using autoradiography and counting the strip segments in a gamma counter, our results indicated that free 125I migrated to Rf = 0.89-1.00 while the [125I]sodium iothalamate moved to Rf = 0.44-0.67. The total developing time for the single-strip ITLC-SA system was approximately 1 h.

Adult female mink (Mustela vison) were fed diets that contained Fusarium moniliforme culture material that provided low- or high-dose dietary concentrations of 86 or 200 ppm fumonisin B1, 22 or 42 ppm fumonisin B2, and 7 or 12 ppm fumonisin B3, respectively, from approximately two weeks prior to breeding through gestation and lactation. Breeding performance of the females was not affected by consumption of the fumonisin diets. However, 58% of the mated females fed the high-dose diet (254 ppm total fumonisins) whelped compared to 100% of those fed the control and low-dose diets (115 ppm fumonisins). There was a statistically significant, dose-dependent decrease in kit (young mink) body weights at birth and a notable, but non-significant, decrease in litter size. The percentage of stillborn kits was directly proportional to the concentration of fumonisins in the dams' diets. Fumonisin concentrations in milk collected from those fed the high-dose diets were approximately 0.7% of the dietary fumonisin concentrations. Lactational exposure to fumonisins did not significantly decrease kit survival from birth through three weeks of age. Hepatic cell vacuolation was present in 25% of the control and 80% of the high-dose adults. No treatment-related gross or histologic lesions were observed in the kit mink. Numerous differences in hematologic and serum chemical parameters were noted between the control and fumonisin-exposed mink.

Thirty-six male mink were fed diets that contained 0, 1, 2 or 4% supplemental salt (sodium chloride) and were given drinking water ad libitum for 7 d. Three mink on each diet were then placed on ad libitum, 50% ad libitum or 25% ad libitum drinking water for the next 14 d. Ad libitum water consumption was directly proportional to the salt content of the diets. Feed consumption was inversely related to the level of dietary salt, although water restriction had a greater effect in reducing feed consumption than did the supplemental salt. The clinical signs of salt toxicity-water restriction observed were increased thirst, mild dehydration, decreased feed consumption, decreased body weight, rough coat, crusty nose and eyes, irritability in the early stage, and lethargy in the later stages. In general, serum and urinary sodium and chloride ion concentrations increased with increasing dietary salt concentrations. Expressed as a percent of brain weight, liver, spleen, kidney and heart weights of mink fed supplemental salt were less than the control weights. Adrenal gland weights increased in response to water restriction. Brain sodium concentrations were not affected by salt supplementation when drinking water was provided ad libitum. However, restricting drinking water generally resulted in increased brain sodium concentrations. Mild to moderate micro- or macrovesicular vacuolar changes were observed in the livers of some mink fed each level of dietary salt, but were especially prominent in the mink restricted in drinking water.

RESUMO

A case of splenic arteriovenous fistula leading to portal hypertension and ascites is presented. The recognition of this vascular lesion is important. When portal hypertension is secondary to a splenic arteriovenous fistula, the complications of portal hypertension can be avoided by the resection of the lesion. The diagnosis can be suspected by clinical findings and confirmed by angiographic delineation of the fistula. Surgical resection of splenic arteriovenous fistulae must include the fistula and all dilated venous tributaries to prevent thrombosis and recurrent portal hypertension.
