RESUMO

The efficiency of bone tissue regeneration by decellularized tooth matrix, demineralized tooth matrix, and commercial xenograft Bio-Oss Spongiosa was compared on the model of a critical-size circular defect in the alveolar bone of the upper jaw of adult Wistar rats. The defect healing dynamics was assessed using histological, histomorphometrical, and immunohistochemical methods on days 30 and 60. In contrast to demineralized matrix and commercial xenograft, decellularized matrix induces the formation of the new bone tissue by day 60. Decellularized matrix can be considered as a biomaterial for cell-free tissue engineering for alveolar bone restoration in dentistry and maxillofacial surgery.

We analyzed the suitability of various collagen-based scaffolds for culturing and osteogenic differentiation of mesenchymal stromal cells (MSC). Decellularized and lyophilized swine intestinal submucosa (SIS) and porous collagen sponge made from reconstructed bovine derma (PCS) were the most effective in promoting MSC adhesion, survival, and growth. MSC from rat and mouse bone marrow and rat adipose tissue successfully adhered to the scaffold surface and penetrated into its deep layers. These scaffolds were also the most effective in inducing osteogenesis. These results indicate that microarchitectonics of PCS and SIS is optimal for support of MSC growth and osteogenic differentiation.

We studied the effect of bovine brain gangliosides, individual ganglioside GM1, and melatonin on the rate of wound closure under in vitro conditions and the effect of melatonin on the rate of wound healing under in vivo conditions. It was shown that bovine brain gangliosides and melatonin reliably increased cell migration in the experimental wound model. This effect was detected when the cell cultures were treated with the test preparations after wound infliction and when the cultures of human keratinocytes were pretreated before wounding. Analysis of the effect of melatonin on the rate of wound healing in vivo showed that melatonin accelerated this process, especially at the middle stages corresponding to the proliferation phase (days 3-6 after surgery). Histological analysis revealed intensification of epidermal cell proliferation at the edges of the wound starting from day 4 after surgery.

We studied the effect of mesenchymal stromal cells and conditioned media on healing of full-thickness skin wound in rats. Cell transplantation to the wound bed did not accelerate wound closing and had no effect on the severity of inflammation, but increased vasculariza - tion of the granulation tissue in 14 days after injury. After injection of conditioned medium to the wound, less pronounced inflammation or enhanced epithelialization was observed. The angiogenic effect was observed only after repeated administration of conditioned medium and was associated with slower regeneration, probably due to skin traumatization by repeated injections. At the same time, fetal skin fibroblasts stimulated angiogenesis only after transplantation in high doses and the medium conditioned by these fibroblasts had no effect on wound healing.

The kinetics of protein synthesis was investigated in primary cultures of hepatocytes from old rats in serum-free medium. The rats were fed mixed fodder supplemented with glutamic acid and then transferred to a regular mixed fodder. The amplitude of protein synthesis rhythm in hepatocytes isolated from these rats increased on average 2-fold in comparison with the rats not receiving glutamic acid supplement. Based on this indicator reflecting the degree of cell-cell interactions, the cells from old rats were not different from those of young rats. The effect was preserved for 3-4 days. These results are discussed in connection with our previous data on preservation of the effect of single administration of gangliosides, noradrenaline, serotonin, and other synchronizers on various cell populations. In contrast to the other investigated factors, glutamic acid is capable of penetrating the blood-brain barrier, which makes its effect possible not only in the case of hepatocytes and other non-brain cells, but also in neurons.

Dense cultures of hepatocytes from old rats (~2 years old, body weight 530-610 g) are different from similar cultures of hepatocytes from young rats by the low amplitude of protein synthesis rhythm. Addition of glutamic acid (0.2, 0.4, or 0.6 mg/ml) into the culture medium with hepatocytes of old rats resulted in increase in the oscillation amplitudes of the protein synthesis rhythm to the level of young rats. A similar action of glutamic acid on the protein synthesis kinetics was observed in vivo after feeding old rats with glutamic acid. Inhibition of metabotropic receptors of glutamic acid with α-methyl-4-carboxyphenylglycine (0.01 mg/ml) abolished the effect of glutamic acid. The amplitude of oscillation of the protein synthesis rhythm in a cell population characterizes synchronization of individual oscillations caused by direct cell-cell communications. Hence, glutamic acid, acting as a receptor-dependent transmitter, enhanced direct cell-cell communications of hepatocytes that were decreased with aging. As differentiated from other known membrane signaling factors (gangliosides, norepinephrine, serotonin, dopamine), glutamic acid can penetrate into the brain and thus influence the communications and protein synthesis kinetics that are disturbed with aging not only in hepatocytes, but also in neurons.

RESUMO

It is shown that the effect of 5-fluorouracil (5-FU) on mesenchymal stromal cells (MSCs) of the bone marrow and fetal liver of the rat increases the population of cells with a reduced proliferative potential. It is shown that, in the course of passaging, the 5-FU-resistant MSCs of the bone marrow and fetal liver lose their ability to differentiate in the osteogenic direction more and less rapidly, respectively, than the 5-FU-sensitive MSCs.

This article provides information about the depositories of parasitic worms in the scientific institutes and museums in the United States, Japan, and Europe (the total number of samples and the availability of types of helminths from various classes), as well as information on the availability of electronic catalogues of the collections in the continental, national, and regional centers for collective use. The extent of this material has determined the necessity of creating digital collections and libraries that would represent a new form of storing, displaying, and exchanging information for scientific research. An analysis was performed of the current state of approaches and methods of development of the specialized information retrieval system (IRS) and databases (DBs) on the parasitic worms in Russia on the basis of a common conceptual data model, taking into account their local use (as desktop systems of database management) and access by scientists worldwide via the Internet.

The study was performed on 30 male rats of Wistar line (weight 330-360 g, age 3.5 months).In an experimental model of damage to the femur bone in the hip joint studied the effect of low frequency electrical stimulation of the damaged area on the rate of regeneration of bone. The animals were divided into two groups. Control (15 rats) and experienced (15 rats). In the experimental animals underwent stimulation of the injury site for 5 min daily for 7 days, 14 days and 21 days. Stimulation was carried out using a device "Osteon-1" generating a mixed signal of two voltage pulse of varying duty cycle, one of which is modulated to a higher frequency. Signals were not synchronized with respect to each other, unipolar with varying frequencies and amplitudes. The obtained results show the effectiveness of the electrical stimulation currents of low frequency in the restoration of bone tissue after damage. Morphological studies showed that electrical stimulation to accelerate the regeneration of damaged bone at all stages of the study (7, 14, 21 day), causes a more pronounced integration of newly formed bone with the old intact bone and promote the formation of more powerful periosteal calluses in comparison with the control.

The annual production of waste in health care institutions (HCI) tends to increase because of the growth of health care provision for population. Among the many criteria for selecting the optimal treatment technologies HCI is important to provide epidemiological and chemical safety of the final products. Environmentally friendly method of thermal disinfection of medical waste may be sterilizators of medical wastes intended for hospitals, medical centers, laboratories and other health care facilities that have small and medium volume of processing of all types of waste Class B and C. The most optimal method of centralized disposal of medical waste is a thermal processing method of the collected material.

Comparison of mesenchymal stromal cells of embryonic and adult rat spleen showed that splenic cells from 20-day rat fetuses exhibit the capacity for clonal growth, express surface antigens CD73, CD90, and CD106, and have weak osteogenic and adipogenic potencies, while splenic cells from adult animals are characterized by lower cloning efficiency, rapid decrease of proliferative activity during passaging, the absence of CD73 and CD90 expression, and are incapable of osteogenesis The observed changes are probably related to extinction of myelopoiesis in the spleen during the postnatal ontogeny.

RESUMO

A comparative analysis of Russian and European legislation concerning to the waste management has been performed. There were revealed principal differences in Russian and European legislation in methodology of the waste classification. In Europe, there is no methodology for breaking up waste into hazard classes, and for the denomination of the danger there are used hazard lists which fail to give information about the extent of their danger. Medical waste in the European legislation are not selected into the separate category as being included in terms of articles and lists in the annexes to the directives or other legal acts. There are considered requirements of the Russian and European legislation in the area of the landfill waste burial. In the frameworks of the proposals for the implementation of international experience in the waste management there was drafted the project of Sanitary rules on hygiene requirements to the arrangement and the contents of landfills for residential solid waste, which includes requirements concerning not only residential solid waste, but also medical waste.

Promising application of nanoparticles and nanomaterials is the creation of sanitary hygienic means of new generation used for disinfection of water and indoor surfaces of mass use, furniture, sanitary technical equipment by virtue of modifying traditional materials to bring them effective biocidal properties, and for the development of methods in vitro for assessment their toxicity In this paper the possibility of the use various forms of silver, copper and aluminum as disinfectant for bacterial test organisms in the aquatic environment and assess their toxicity on biological models of continuous culture of BGM cells (a stable line of African green monkey kidney cells) and HEp-2 (Human epithelial type 2 (HEp-2) cells, derived from a human laryngeal carcinoma) is considered.

Fetal liver is known as a source of multipotent mesenchymal stromal cells. These cells are routinely isolated by adhesion to plastic, but thus prepared culture is contaminated by other cells. For instance, primary cell culture of from rat fetal liver, apart from fibroblasts with phenotypic characteristics of mesenchymal stromal cells, contained skeletal muscle precursors, myofibroblasts, and epitheliocytes expressing cytokeratin-19 (the latter was also detected in some fibroblast-like cells probably undergoing epithelio-mesenchymal transition). During passaging, fibroblasts become practically the only cell type in the culture.

RESUMO

Water samples and surface swabs from 15 Moscow public indoor swimming pools (SP), where disinfection was carried with the use of chlorine reagents, were studied and excess of bacteriological indicators standards was revealed, infestants of fungous skin and nails diseases were allocated. The research results showed both the absence of epidemic significant information value of bacteriological indicators in relation to infestants of fungous diseases and discrepancy of efficiency of existing recommendations for disinfection actions to requirements on providing sanitary-and-epidemiologic safety of SP visitors.

RESUMO

The possibility of using 12 heterogeneous sensitizers (HS) based on phthalocyanines covalently grafted to aminopropyl silicagel for disinfection of water from bacteria has been studied. For reliable water quality control the technique of performing bacteriological analysis in the presence of HS beads in the sample has been elaborated. The conditions increasing the efficiency of photo disinfection in the presence of HS were studied. Algorithm for estimation of photo disinfectant effect of HS against bacteria was substantiated. Obtained data confirm the perspective of further studies on the substantiation of the possibility of the application of HS for water disinfection.

RESUMO

The paper gives data on the use of techniques to detect and register Salmonella in the water objects, by applying a new liquid nutrient medium. Experimental and field studies have shown its advantage over the accumulation media widely used in practical healthcare. It has been ascertained that the nutrient medium not only accumulates biomass, but also provides the restoration of the biological properties of uncultivated Salmonella species. The use of the nutrient medium at practical laboratories makes it possible to unify guidelines for the examination of water objects with varying degrees of biological pollution and to obtain the comparable results of analyses.
