RESUMO

The possibility of vilosen usage for the immune system damage liquidation was studied. Rats obtained discrete rentgen irradiation during 1 month in the total dose of 4 Gr. Mice obtained internal 131I irradiation in a dose of 9.25 kBk/g. It was established that thymus and spleen masses, quantity of their cells, blood leukocytes and antibody production decreased by as external and internal irradiation. Irradiated animals treated with vilosen restored their immune system functional state partly or completely. The preparation was assumed to be used for the correction of immune system radiation damage.

RESUMO

The effects of splenoside on the reactions of delayed-type hypersensitivity and graft versus host were investigated. Even single splenoside injection (50 mg/10 g b.w.) increased graft versus host reaction in newborn mice-hybrids F1. Splenoside influences the delayed-type hypersensitivity reaction in dose-depending manner: the dose of 50 mg/10 g increases the reaction and 100 mg/10 g decreases it.

RESUMO

Ten-month-old male rats thymectomized in early post-puberty were ascertained to have lower plasma luteinizing and testicular hormones and higher testicular steroid dehydrogenase activity. Following 24 hours of administration of a thymocyte membrane structural agent, pituitary gonadotropic and gonadal androgenic functions became normal. There was no agreement between the hormonal parameters and the levels of nucleic acids in the testes and prostate.

RESUMO

The influence of the thymus on the growth hormone (GH) level in the mouse hypophysis has been studied. It has been found, that syngeneic transfer of thymocytes of young mice into thymectomized mice causes on increase of the growth hormone level in the hypophysis. Administration of hydrocortisone simultaneously with transfer of thymocytes accelerated the GH level increase. The administration of only hydrocortisone into thymectomized mice does not lead to changes in the GH level. The increased GH level was observed when thymocytes were replaced by some structures shedding from thymocytes after their stimulation with short peptide thymic hormone. It is supposed that thymocytes can stimulate GH accumulation in the hypophysis.

RESUMO

The supernatant obtained from the mouse thymocytes incubated with oligopeptide thymic factor can partially restore autorosette formation in heated at 45 degrees C and washed thymocytes which have lost the receptors for autologous erythrocytes. It is supposed that decreased level of autorosette formation after treatment of thymocytes with thymic factor is caused by shedding of receptors for autologous erythrocytes.

RESUMO

The photooxidation of rabbit and human IgG in the presence of methylene blue leads to loss of antibody and complement fixation activity. Under these conditions 46% of tryptophan, 30% of tyrosine and 42% of phenylalanine are destroyed. The IgG incubation with methylene blue in darkness causes no changes in the content of aromatic amino acid residues or in the antibody activity, but leads to deterioration in their complement fixation properties.

RESUMO

A receptor structure that has an affinity to autologous and xenogenic red blood cells was isolated from rat thymocytes by means of adsorption on immobilized rabbit IgG. This structure restores the rosette formation on addition to thymocytes deprived of their receptors for autologous red blood cells and having no capacity for rosette formation. The substance affinity for red blood cells and rabbit IgG is caused by its glycoprotein with a molecular weight of 12 000 daltons, that displays the properties inherent in thymocyte Fc-receptors. The purified glycoprotein can also restore rosette formation in thymocytes deprived of their receptors for autologous red blood cells. A suggestion is made that at least one of Fc-receptors of the T-lymphocyte membrane is capable of reacting with red blood cells.

RESUMO

Some properties of glycoprotein isolated from rat thymocytes were studied. The glycoprotein (its molecular weight is found to be about 12000 Daltons) has an affinity for autologous and xenogenous red blood cells as well as for Fc portion of IgG. It is shown that the glycoprotein belongs to the thymocyte plasma membrane receptors responsible for interaction with erythrocytes and possesses the Fe receptor properties. The data give reason to conclude that at least one of the Fc receptors has the affinity for SRBC, ARBC and Fe portion of IgG.

RESUMO

A substance reacting with sheep red blood cells and rabbit IgG was isolated from the rat thymocytes and calf thymus as a homogeneous preparation. Some chemical properties of this substance were studied. The molecular weight of the agglutinating substance was found to be 12300 according to the SDS-gel electrophoresis data. The infrared spectra show that this substance contains peptide bonds and a carbohydrate component. The amide II absorption band observed in the infrared spectrum disappears after treatment of the agglutinating substance with immobilized pronase. The treatment with immobilized pronase leads to degradation of this substance into two components detected by thin layer chromatography and to the loss of its agglutinating properties. Hexose was determined in the substance with an anthrone reagent. This substance is supposed to be a low-molecular glycoprotein.

RESUMO

Photooxidation of anti-HSA antibodies, isolated by affinity chromatography, was carried out using methylene blue as a photosensitizing agent. Activity of anti-HSA antibody as well as an absorption maximum at 280 nM pH 7.0 disappeared due to photooxidation. Incubation of antibodies with methylene blue in the darkness was ineffective. Antigenic properties of human IgG were unaltered after photooxidation under the same conditions but the absorption maximum of the immunoglobulin at 280 nM disappeared.

RESUMO

A factor reacting with SRBC and rabbit IgG was obtained under mild conditions from rat thymus and spleen. The isolation procedure includes incubation of thymocytes or splenocytes with IgG-cellulose adsorbent, destruction of cells, washing the adsorbent and elution of an adsorbed material at pH 2. This preparation as well as the purified substance previously obtained by affinity chromatography on IgG-cellulose columns were found to agglutinate both SRBC and autologous erythrocytes. Preincubation in 1% SDS leads to dissociation of the preparation into several components separated by gel electrophoresis. A probable relation of this structure to the rosette forming capacity of T-lymphocytes is discussed.

RESUMO

A fraction was isolated from the sulphate extract of rats and calfs thymus. This fraction reacts with sheep erythrocytes and rabbits IgG in the agglutination test. An identical fraction from the rats spleen and liver is not active in the same concentration. Active substance was purified by affinity chromatography on IgG cellulose column. The absorption spectrum of the purified substance in 220-300 nm region was studied at pH 7. This substance has not the absorption maxima in 280 nm. The sedimentation constant of the crude preparation is approximately 1.7 s. It supposed, that this substance is a specifical receptor of thymic cells.
