RESUMO

Our previous work showed that a proliferation-inducing ligand (APRIL) was involved in the development of acute lymphoblastic leukemia (ALL) in children. However, the precise role of APRIL in ALL remains unknown. To investigate this issue, we silenced and overexpressed APRIL in Nalm-6 ALL cells using short hairpin RNA targeting the APRIL gene and recombinant human APRIL, respectively, and evaluated the effects on cell proliferation, apoptosis, and migration. APRIL mRNA and APRIL and matrix metalloproteinase-2 protein levels were evaluated by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) and western blott, respectively. We found that APRIL expression was reduced by shRNA-mediated knockdown in Nalm-6 cells; this was associated with a decrease in cell proliferation (P<0.05). APRIL knockdown increased apoptosis (P<0.01) but suppressed cell migration along with matrix metalloproteinase-2 protein level. Overexpressing recombinant human APRIL had the opposite effects in each case (P<0.05). These results demonstrate a link between APRIL expression and ALL development and suggest that APRIL is a potential therapeutic target for ALL treatment.

The EphA5 receptor is well established as an axon guidance molecule during neural system development and plays an important role in dendritic spine formation and synaptogenesis. Our previous study has showed that EphA5 is decreased in the developing brain of congenital hypothyroidism (CH) and the EphA5 promoter methylation modification participates in its decrease. c-Fos, a well-kown transcription factor, has been considered in association with brain development. Bioinformatics analysis showed that the EphA5 promoter region contained five putative c-fos binding sites. The chromatin immunoprecipitation (ChIP) assays were used to assess the direct binding of c-fos to the EphA5 promoter. Furthermore, dual-luciferase assays showed that these three c-fos protein binding sites were positive regulatory elements for EphA5 expression in PC12 cells. Moreover, We verified c-fos positively regulation for EphA5 expression in CH model. Q-PCR and Western blot showed that c-fos overexpression could upregulate EphA5 expression in hippocampal neurons of rats with CH. Our results suggest that c-fos positively regulates EphA5 expression in CH rat model.

We conducted a prospective, randomized, and controlled trial to assess the optimal dose for GnRH antagonist, cetrorelix, for Chinese women during the course of ovarian stimulation. The patients were randomly divided into two groups, in which 48 patients were advised to inject 0.25 mg Cetrorelix daily (the 0.25 mg group), while 39 patients were instructed to receive a daily dose of 0.125 mg cetrorelix (the 0.125 mg group). In general, a daily dose of 0.125 mg cetrorelix could be more optimal for Chinese women as manifested by the lower cancellation rate, higher implantation rate and clinical pregnancy rate. Specifically, daily administration of 0.125 mg cetrorelix for patients under 35 years old is associated with a 3-fold higher implantation rate and a 5-fold higher clinical pregnancy rate as compared with that of those patients ≥ 35 years old. On the contrary, higher rates for implantation and clinical pregnancy were noted by daily injection of 0.25 mg cetrorelix in elder patients (≥ 35 years old) as compared with that of young patients (< 35 years old). Together, our data suggest that a daily dose of 0.125 mg cetrorelix could be more optimal for patients < 35 years old, while 0.25 mg/day of cetrorelix are likely conducive to higher implantation and clinical pregnancy rate for those patients ≥ 35 years old. These data could be important for preventing LH surge while maintaining optimal LH levels necessary for embryo implantation for Chinese women during the course of IVF-ET treatment.

The knowledge of the relationship between sequence characteristics of insecticidal crystal proteins (ICP) and their inhibitory against Plutella xylostella provided helpful information for the rational design of ICP with desirable activity against Plutella xylostella. The four key loops of ICP with determined activities against Plutella xylostella were selected to study the quantitative relationship between sequence characteristics and insecticidal activity. The first principle components' score vectors for 20 amino acids were assigned to converting amino acids into data. The six key sites X3, X9, X12, X13, X14 and X19 were predicted by stepwise regression method. The amino acids L/ X3, S/ X9, S/ X12, T/ X13, A/ X14 and G/ X19 found by partial least squares regression and second order polynomial models were predicted to increase the activity of ICP against Plutella xylostella.

RESUMO

The principal component analysis (PCA) was applied to the data processing in training sets, the new principal components were then used as input data for support vector machine model. A prediction model for optimum pH of chitinase was established based on uniform design. When The regularized constant C, epsilon and Gamma were 10, 0.7 and 0.5 respectively, the calculated pHs fitted the reported optimum pHs of chitinase very well and the MAPEs (Mean Absolute Percent Error) was 3.76%. At the same time, the predicted pHs fitted the reported optimum pHs well and the MAE (Mean Absolute Error) was 0.42 pH unit. It was superior in fittings and predictions compared to the model based on back propagation (BP) neural network.

A quantitative structure-property relationship (QSPR) model in terms of amino acid composition and the activity of Bacillus thuringiensis insecticidal crystal proteins was established. Support vector machine (SVM) is a novel general machine-learning tool based on the structural risk minimization principle that exhibits good generalization when fault samples are few; it is especially suitable for classification, forecasting, and estimation in cases where small amounts of samples are involved such as fault diagnosis; however, some parameters of SVM are selected based on the experience of the operator, which has led to decreased efficiency of SVM in practical application. The uniform design (UD) method was applied to optimize the running parameters of SVM. It was found that the average accuracy rate approached 73% when the penalty factor was 0.01, the epsilon 0.2, the gamma 0.05, and the range 0.5. The results indicated that UD might be used an effective method to optimize the parameters of SVM and SVM and could be used as an alternative powerful modeling tool for QSPR studies of the activity of Bacillus thuringiensis (Bt) insecticidal crystal proteins. Therefore, a novel method for predicting the insecticidal activity of Bt insecticidal crystal proteins was proposed by the authors of this study.
