RESUMO

The research on video analytics especially in the area of human behavior recognition has become increasingly popular recently. It is widely applied in virtual reality, video surveillance, and video retrieval. With the advancement of deep learning algorithms and computer hardware, the conventional two-dimensional convolution technique for training video models has been replaced by three-dimensional convolution, which enables the extraction of spatio-temporal features. Specifically, the use of 3D convolution in human behavior recognition has been the subject of growing interest. However, the increased dimensionality has led to challenges such as the dramatic increase in the number of parameters, increased time complexity, and a strong dependence on GPUs for effective spatio-temporal feature extraction. The training speed can be considerably slow without the support of powerful GPU hardware. To address these issues, this study proposes an Adaptive Time Compression (ATC) module. Functioning as an independent component, ATC can be seamlessly integrated into existing architectures and achieves data compression by eliminating redundant frames within video data. The ATC module effectively reduces GPU computing load and time complexity with negligible loss of accuracy, thereby facilitating real-time human behavior recognition.

Previous studies revealed a controversial role of mechanistic target of rapamycin complex 1 (mTORC1) and mTORC1-regulated macroautophagy in isoproterenol (ISO)-induced cardiac injury. Here we investigated the role of mTORC1 and potential underlying mechanisms in ISO-induced cardiomyocyte necrosis. Two consecutive daily injections of ISO (85 mg/kg, s.c.) or vehicle control (CTL) were administered to C57BL/6J mice with or without rapamycin (RAP, 5 mg/kg, i.p.) pretreatment. Western blot analyses showed that myocardial mTORC1 signaling and the RIPK1-RIPK3-MLKL necroptotic pathway were activated, mRNA expression analyses revealed downregulation of representative TFEB target genes, and Evan's blue dye uptake assays detected increased cardiomyocyte necrosis in ISO-treated mice. However, RAP pretreatment prevented or significantly attenuated the ISO-induced cardiomyocyte necrosis, myocardial inflammation, downregulation of TFEB target genes, and activation of the RIPK1-RIPK3-MLKL pathway. LC3-II flux assays confirmed the impairment of myocardial autophagic flux in the ISO-treated mice. In cultured neonatal rat cardiomyocytes, mTORC1 signaling was also activated by ISO, and inhibition of mTORC1 by RAP attenuated ISO-induced cytotoxicity. These findings suggest that mTORC1 hyperactivation and resultant suppression of macroautophagy play a major role in the induction of cardiomyocyte necroptosis by catecholamine surges, identifying mTORC1 inhibition as a potential strategy to treat heart diseases with catecholamine surges.

CONTEXT: Fructus Ligustri Lucidi (FLL), a commonly used herb of traditional Chinese medicine (TCM), is the fruit of Ligustrum lucidum Ait. (Oleaceae). The ethanol extract of FLL is a potential candidate for preventing and treating postmenopausal osteoporosis (PMOP) by nourishing the liver and kidneys. OBJECTIVE: This study determines whether an ethanol extract of FLL has anti-osteoporotic effects in ovariectomized (OVX) mice and explores the underlying mechanism. MATERIALS AND METHODS: The OVX model of eight-week-old C57BL/6J female mice was taken, and ovariectomy was used as PMOP. Mice were divided into five groups: sham-operated group (n = 10), OVX group (n = 10), OVX + E2 group (n = 10; 0.039 mg/kg), OVX + FLL group (n = 10; 2 g/kg) and OVX + FLL group (n = 10; 4 g/kg). Mice were treated by gavage with FLL or CMCNa once daily for 8 weeks. We harvested uteri, femur, and tibias from mice; bone mineral density (BMD) and bone microstructure were obtained by X-ray absorptiometry and micro-CT. Furthermore, the effect of FLL on the balance of osteoblast and adipocyte differentiation was investigated using bone marrow mesenchymal stem cells (BMMSCs). RESULTS: The results indicated that FLL did not affect OVX-induced estradiol reduction. Compared with OVX mice, FLL significantly increased BMD (63.54 vs. 61.96), Conn. D (86.46 vs. 57.00), and left tibial strength (13.91 vs. 11.27), decreased Tb. Sp (0.38 vs. 0.44) and body fat content (4.19% vs. 11.24%). FLL decreased osteoclast activity and enhanced RUNX2 expression; inhibited perilipin peroxisome proliferator-activated receptor gamma (PPARγ) expression and adipocyte differentiation from BMMSCs. CONCLUSIONS: FLL prevented additional bone loss and improved bone microstructure in OVX mice by modulating bone and fat balance, suggesting that FLL might be a therapeutic agent for PMOP.

BACKGROUND: Chronic inflammatory disorders in atrial fibrillation (AF) contribute to the onset of ischemic stroke. Systemic immune inflammation index (SIII) and system inflammation response index (SIRI) are the two novel and convenient measurements that are positively associated with body inflammation. However, little is known regarding the association between SIII/SIRI with the presence of AF among the patients with ischemic stroke. METHODS: A total of 526 ischemic stroke patients (173 with AF and 353 without AF) were consecutively enrolled in our study from January 2017 to June 2019. SIII and SIRI were measured in both groups. Logistic regression analysis was used to analyse the potential association between SIII/SIRI and the presence of AF. Finally, the correlation between hospitalization expenses, changes in the National Institutes of Health Stroke Scale (NIHSS) scores and SIII/SIRI values were measured. RESULTS: In patients with ischemic stroke, SIII and SIRI values were significantly higher in AF patients than in non-AF patients (all p < 0.001). Moreover, with increasing quartiles of SIII and SIRI in all patients, the proportion of patients with AF was higher than that of non-AF patients gradually. Logistic regression analyses demonstrated that log-transformed SIII and log-transformed SIRI were independently associated with the presence of AF in patients with ischemic stroke (log-transformed SIII: odds ratio [OR]: 1.047, 95% confidence interval CI = 0.322-1.105, p = 0.047; log-transformed SIRI: OR: 6.197, 95% CI = 2.196-17.484, p = 0.001). Finally, a positive correlation between hospitalization expenses, changes in the NIHSS scores and SIII/SIRI were found, which were more significant in patients with AF (all p < 0.05). CONCLUSIONS: Our study suggests SIII and SIRI are convenient and effective measurements for predicting the presence of AF in patients with ischemic stroke. Moreover, they were correlated with increased financial burden and poor short-term prognosis in AF patients presenting with ischemic stroke.

Ubiquitin carboxyl-terminal hydrolase L1 (UCHL1) is a deubiquitinase known to play essential roles in the nervous tissue. Myocardial upregulation of UCHL1 was observed in human dilated cardiomyopathy and several animal models of heart disease, but the (patho)physiological significance of UCHL1 in cardiomyocytes remains undefined. Hence, we conducted this study to fill this critical gap. We produced cardiomyocyte-restricted Uchl1 knockout (CKO) by coupling the Uchl1-floxed allele with transgenic Myh6-Cre in C57B/6J inbred mice. Mice transgenic for Myh6-Cre were used as controls (CTL). Myocardial Uchl1 proteins were markedly reduced in CKO mice but they did not display discernible abnormal phenotype. Ten-week old CTL or CKO mice were subjected to left anterior descending artery ligation (myocardial infarction, MI) or sham surgery (Sham) and characterized at 7- and 28-day after surgery. Compared with Sham mice, significant increases in myocardial UCHL1 proteins were detected in CTL MI but not in CKO MI mice. MI-induced left ventricular (LV) chamber dilation, reduction of ejection fraction (EF) and fractional shortening (FS), and LV anterior wall thinning detected by echocardiography were comparable between the CTL MI and CKO MI groups 7-day post-MI. However, by 28-day post-MI, MI-induced LV chamber dilatation, EF and FS reduction, increases of myocardial ubiquitin conjugates, and increases in the heart weight to body weight ratio and the ventricular weight to body weight ratio were significantly more pronounced in CKO MI than CTL MI mice. As further revealed by LV pressure-volume relationship analyses, CKO MI mice but not CTL MI mice displayed significant decreases in stroke volume, cardiac output, and the maximum rates of LV pressure rising or declining and of LV volume declining, as well as significant increases in LV end-diastolic pressure and Tau, compared with their respective Sham controls. LC3-II flux assays reveal that autophagic flux is decreased in CKO mouse myocardium as well as in cultured Uchl1-deficient cardiomyocytes. In conclusion, UCHL1 of cardiomyocytes is dispensable for development but promotes macroautophagy in cardiomyocytes. Upregulation of UCHL1 in post-MI hearts occurs primarily in the cardiomyocytes and protects against post-MI cardiac remodeling and malfunction likely through supporting autophagic flux and proteostasis during a stress condition.

Background: Catecholamine surges and resultant excessive ß-adrenergic stimulation occur in a broad spectrum of diseases. Excessive ß-adrenergic stimulation causes cardiomyocyte necrosis, but the underlying mechanism remains obscure. Necroptosis, a major form of regulated necrosis mediated by RIPK3-centered pathways, is implicated in heart failure; however, it remains unknown whether excessive ß-adrenergic stimulation-induced cardiac injury involves necroptosis. Hence, we conducted the present study to address these critical gaps. Methods and Results: Two consecutive daily injections of isoproterenol (ISO; 85 mg/kg, s.c.) or saline were administered to adult mixed-sex mice. At 24 h after the second ISO injection, cardiac area with Evans blue dye (EBD) uptake and myocardial protein levels of CD45, RIPK1, Ser166-phosphorylated RIPK1, RIPK3, and Ser345-phosphorylated MLKL (p-MLKL) were significantly greater, while Ser321-phosphorylated RIPK1 was significantly lower, in the ISO-treated than in saline-treated wild-type (WT) mice. The ISO-induced increase of EBD uptake was markedly less in RIPK3 -/- mice compared with WT mice (p = 0.016). Pretreatment with the RIPK1-selective inhibitor necrostatin-1 diminished ISO-induced increases in RIPK3 and p-MLKL in WT mice and significantly attenuated ISO-induced increases of EBD uptake in WT but not RIPK3-/- mice. Conclusions: A large proportion of cardiomyocyte necrosis induced by excessive ß-adrenergic stimulation belongs to necroptosis and is mediated by a RIPK1-RIPK3-dependent pathway, identifying RIPK1 and RIPK3 as potential therapeutic targets for catecholamine surges.

This study compared the community structure of macrobenthos in bare tidal areas and vegetated regions under two reclamation patterns, fully enclosed and opened reclamation, in Yangtze wetland, China. Compared with fully enclosed reclamation areas, opened reclamation areas had higher water salinity, higher number of species and abundance, and less disturbed habitat health status. Compared with bare tidal areas, vegetated regions were more suitable for the survival of macrobenthos and its habitat health status was more stable. Salinity, pH and larger soil particle diameter were positively associated with macrobenthic diversity, while total nitrogen and total phosphorus were negatively associated. Overall, compared with fully enclosed reclamation and vegetated regions, opened reclamation and bare tidal areas had less negative effects upon macrobenthic community and habitat health. Opened reclamation is preferred for wetland reclamation, and planting indigenous plants and releasing species adapted to fresh water is beneficial to the restoration of macrobenthic communities.

Lipids play an important role in protecting poikilotherms from cold stress, but relatively little is known about the regulation of lipid metabolism under cold stress, especially in crustaceans. In the present study, red-clawed crayfish Cherax quadricarinatus was employed as a model organism. Animals were divided into four temperature groups (25, 20, 15 and 9 °C) and treated for 4 weeks, with the 25 °C group serving as a control. The total lipid content in the hepatopancreas as well as the triglyceride, cholesterol and free fatty acid levels in the hemolymph were determined. Lipids stored in the hepatopancreas and hemolymph decreased with decreasing temperature, with changes in the 9 °C group most pronounced, indicating that lipids are the main energy source for crayfish at low temperatures. Furthermore, enzyme activity of lipase, fatty acid synthase, acetyl-CoA carboxylase, and lipoprotein esterase, and gene expression analysis of fatty acid synthase gene, acetyl-CoA carboxylase gene and carnitine palmitoyltransferase gene showed that the digestion, synthesis and oxidation of lipids in the hepatopancreas were inhibited under low temperature stress, but expression of sphingolipid delta-4 desaturase (DEGS) was increased, indicating an increase in the demand for highly unsaturated fatty acids at low temperatures. Analysis of the expression of genes related to the AMP-activated protein kinase (AMPK) signalling pathway revealed that the adiponectin receptor gene was rapidly upregulated at low temperatures, which may in turn activate the expression of the downstream AMPKα gene, thereby inhibiting lipid anabolism.

Previous research has identified microplastics as new environmental pollutants that are widely distributed in a variety of environments, including aquaculture environments. However, the potential hazard of microplastics to aquaculture animals, especially toward lipid metabolism involved with the survival and growth of aquatic animal, has not yet been investigated. In the present study, redclaw crayfish (Cherax quadricarinatus) were exposed to different concentrations of 200 nm-sized polystyrene microspheres (0, 0.5, and 5 mg/L) for 21 days, to investigate the effects of microplastics on lipid metabolism. After ingestion, the microplastics were distributed in the intestines and hepatopancreas, and appeared to inhibit the growth of Cherax quadricarinatus. Subsequently, the lipid levels in the hepatopancreas and hemolymph was detected, and found that after 21 days of exposure, the lipid content and free fatty acids in the hepatopancreas and hemolymph decreased significantly, and total cholesterol and triglycerides levels increased significantly in the hemolymph. This might have been caused by insufficient intake of exogenous fat. A significant decrease in lipase activity also supported this view. The activity of lipoprotein lipase related to lipolysis in the hepatopancreas increased significantly, while the activity of fatty acid synthase related to fat synthesis increased, and the activity of acetyl-CoA carboxylase decreased. These results indicated disturbed lipid metabolism in the hepatopancreas. The significant increase of lipid transport-related low-density lipoprotein indicated that the lipolytic capacity was higher than the lipid synthesis capacity. The expression levels of fatty acid metabolism-related genes FAD6 and FABP decreased significantly, indicating that the fatty acid utilization ability of hepatopancreas cells was inhibited, which was consistent with the results of enzyme activities. Thus, microplastics represent a potential hazard to redclaw crayfish, at least on lipid metabolism. This study provided basic data on the ecotoxicological effects of microplastics on crustaceans.

Two isoforms of Glutathione S-Transferase (GST) genes, belonging to mu (Dp-GSTm1 and Dp-GSTm2) and sigma (Dp-GSTs1 and Dp-GSTs2) classes, were cloned and characterised in the freshwater Daphnia pulex. No signal peptide was found in any of the four GST proteins, indicating that they were cytosolic GST. A highly conserved glutathione (GSH) binding site (G-site) occurred in the N-terminal sequence, and a substrate binding site (H-site), interacting non-specifically with the second hydrophobic substrate, was present in the C-terminal. A Tyr residue, for the stabilization of GSH, was found to be conserved in the analysed sequences. The secondary and tertiary structures indicated that these genes possess the typical cytosolic GST structure, including a conserved N-terminal domain with a ßαßαßßα motif. The µ loop (NVGPAPDYDR and NFIGAEWDR in Dp-GSTm1 and Dp-GSTm2, respectively) was identified between the ßαß (ß1α1ß2) and αßßα motifs (α2ß3ß4α3) in the N-terminal domain. The expressions of Dp-GSTs1, Dp-GSTs2, and Dp-GSTm1 were higher in other age groups compared to the newly-born neonates (1 d); however, the expression of Dp-GSTm2 first increased and then decreased with age. Gene expression was significantly reduced by high concentration (1 and 2 mg/L) of 75 nm polystyrene nanoplastic. However, nanoplastic exposure at the predicted environmental concentration (1 µg/L) had a low effect. Exposure of mothers to nanoplastic (1 µg/L) elevated the Dp-GSTs2 level in their neonates. These results improve our understanding on the response of different types of Daphnid GST to environmental contaminants, especially nanoplastic.

Recently, research on the biological effects of nanoplastics has grown exponentially. However, studies on the effects of nanoplastics on freshwater organisms and the mechanisms of the biological effects of nanoplastics are limited. In this study, the content of reactive oxygen species (ROS), gene and protein expression in the MAPK-HIF-1/NFkB pathway, and antioxidant gene expressions and enzyme activities were measured in Daphnia pulex exposed to polystyrene nanoplastic. In addition, the full-length extracellular signal-regulated kinases (ERK) gene, which plays an important role in the MAPK pathway, was cloned in D. pulex, and the amino acid sequence, function domain, and phylogenetic tree were analyzed. The results show that nanoplastic caused the overproduction of ROS along with other dose-dependent effects. Low nanoplastic concentrations (0.1 and/or 0.5 mg/L) significantly increased the expressions of genes of the MAPK pathway (ERK; p38 mitogen-activated protein kinases, p38; c-Jun amino-terminal kinases, JNK; and protein kinase B, AKT), HIF-1 pathway (prolyl hydroxylasedomain, PHD; vascular endothelial growth factor, VEGF; glucose transporter, GLUT; pyruvate kinase M, PKM; hypoxia-inducible factor 1, HIF1), and CuZn superoxide dismutase (SOD) along with the activity of glutathione-S-transferase. As the nanoplastic concentration increased, these indicators were significantly suppressed. The protein expression ratio of ERK, JNK, AKT, HIF1α, and NFkBp65 (nuclear transcription factor-kB p65) as well as the phosphorylation of ERK and NFkBp65 were increased in a dose-dependent manner. The activities of other antioxidant enzymes (catalase, total SOD, and CuZn SOD) were significantly decreased upon exposure to nanoplastic. Combined with our previous work, these results suggest that polystyrene nanoplastic causes the overproduction of ROS and activates the downstream pathway, resulting in inhibited growth, development, and reproduction. The present study fosters a better understanding of the biological effects of nanoplastics on zooplankton.

Background: The effect of transient electromagnetic fields on the function of pacemakers is not well-evaluated. There is a lack of effective methods for clinicians to reduce electromagnetic susceptibility (EMS) during implantation of pacemakers. This study aimed to evaluate whether a novel method of handling the excess leads in the pocket can lower the EMS of pacemakers and consequently reduce the effect of electromagnetic interference caused by transient electromagnetic fields on pacemakers. Methods: An in vitro chest model was established to simulate the clinical condition of dual-chamber pacemaker implantation. Three different intertwining patterns of excess leads were examined: parallel, twisted once, and multiple twisted-pair. Oscillated currents were injected into a copper electrical wire set horizontally above the model to create a radiated magnetic field to simulate the transient daily electromagnetic exposure of pacemakers. The electromagnetic induction of current was measured. The occurrence of EMS-related adverse events was evaluated when the induced pulsed voltage was applied. Results: Transient electromagnetic fields can induce electromagnetic noise in the pacing loop and inhibit the release of pacing pulses. The multiple twisted-pair intertwining pattern of excess leads was associated with a lower induced voltage amplitude than both the parallel and once-twisted patterns (P < 0.001). Even once twisted could significantly reduce induced voltage amplitude compared to not twisted (P < 0.001). A lower incidence of pacing inhibition was also observed in the multiple twisted-pair group than in the other two groups (P < 0.001). Conclusions: Transient electromagnetic fields can cause pacing inhibition. Twisting the excess leads for multiple turns in the pocket is an effective method to reduce the EMS of the dual-chamber pacemaker.

Background: Atrial fibrillation (AF) is increasingly considered an age-related degenerative disease, whose process is associated with the development of impaired left atrial (LA) performance. However, the subtle dynamic changes of LA performance in AF during aging have yet to be fully elucidated. Atrial fibrosis is a key substrate for the development of AF, but the progression of fibrosis during aging and its relationship with LA dysfunction need to be further explored. Methods: A total of 132 control individuals and 117 persistent AF patients were prospectively studied. Subjects were further stratified into three age groups (age group 1: younger than 65 years, age group 2: between 65 and 79 years old, and age group 3: older than 80 years). The two-dimensional speckle tracking imaging was carried out for analyzing the alterations in LA function underlying LA remodeling, whereas electroanatomic mapping was performed to investigate LA fibrosis burden. In animal study, aged mice and young mice served as research subjects. Echocardiography and histological staining were used to assess LA performance and fibrosis burden, respectively. Results: Echocardiography showed progressive increases in LA dimension and LA stiffness index, and progressive decreases in LA global longitudinal strain and LA strain rates with advancing age in both AF and control cohorts, which was more prominent in AF cohort. Electroanatomic mapping showed progressive decrease in mean LA voltage and progressive increases in LA surface area, low-voltage area %, and LA volume with advancing age, whereas more significant alterations were observed in AF patients. Moreover, left atrial global longitudinal strain was positively correlated with mean LA voltage, whereas LA stiffness index was negatively related to mean LA voltage. In animal experiment, increased LA size and pulmonary artery dimension as well as longer P-wave duration and more prominent LA fibrosis were found in aged mice. Conclusions: This study provides new evidence of subtle changes in structure and performance of left atrium and their association with atrial fibrosis in both AF and non-AF subjects during physiological aging. In addition, our study also provides normal values for LA structure and performance in both AF and non-AF conditions during aging. These measurements may provide an early marker for onset of AF and LA adverse remodeling.

Cladocera are small freshwater crustaceans that have attracted considerable attention in recent years. They are commonly used for studying senescence. In this study, we used LC-MS/MS with eight-plex iTRAQ to perform a comparative proteomic analysis of senescence in Daphnia pulex. Of 3076 primordial proteins, 2325 were credible (the remaining were low-confidence proteins) and 247 significantly differentially expressed proteins (DEPs). Of the latter, 87, 91, and 69 DEPs were identified in the Day 15 vs. Day 5, Day 20 vs. Day 5, and Day 25 vs. Day 5 groups, respectively. Gene ontology enrichment analysis showed that oxidative damage may be the main cause of senescence in D. pulex. Using Kyoto Encyclopedia of Genes and Genomes pathway analysis, we found that the peroxisome pathway played an important role in aging. Our results suggest that D. pulex alleviates excessive oxidative damage by altering key enzymes involved in carbohydrate and protein metabolism.

Rhythmic changes in reactive oxygen species (ROS) levels with time have been previously reported, but circadian rhythmic changes in ROS levels in Daphnia pulex remain unexplored. Here, in light-dark (L/D 16:8, L/D 8:16), continuous light (LL), and continuous dark (DD) conditions were set up to analyze the accumulation of ROS and activity of antioxidant enzymes in D. pulex at different time points. We observed that ROS levels significantly varied from day to night. By fluorescence probe labeling, we noted that ROS majorly accumulated in the thoracic limbs, brood chamber, and rectum in D. pulex. In addition, in the L/D 16:8 and 8:16 groups, ROS level was negatively correlated to the gene expression level of catalase, glutathione S-transferase, and glutathione peroxidase. In the LL and DD groups, food supply emerged as a zeitgeber, driving D. pulex to maintain a specific biological rhythm. The ability to scavenge hydroxyl radicals and activity of antioxidant enzymes were also affected by L/D cycles. To summarize, we indicate that different L/D cycles can induce different oxidation-antioxidant system responses in D. pulex and that Daphnia can adapt to the living environment by circadian rhythm regulation of the oxidation-antioxidant balance.

The biological effects of nanoplastics are a growing concern. However, most studies have focused on exposure to high concentrations or short-term exposure. The potential effects of exposure to low environmental nanoplastic concentrations over the long-term and across multiple generations remain unclear. In the present study, Daphnia pulex was exposed over three 21-day generations to a typical environmental nanoplastic concentration (1 µg/L) and the effects were investigated at physiological (growth and reproduction), gene transcription and enzyme activity levels. Chronic exposure did not affect the survival or body length of D. pulex, whereas the growth rate and reproduction were influenced in the F2 generation. Molecular responses indicated that environmental nanoplastic concentrations can modulate the response of antioxidant defenses, vitellogenin synthesis, development, and energy production in the F0-F1 generations, and prolongation resulted in inhibitory effects on antioxidant responses in F2 individuals. Some recovery was observed in the recovery group, but reproduction and stress defenses were significantly induced. Taken together, these results suggest that D. pulex recovery from chronic exposure to nanoplastic may take several generations, and that nanoplastics have potent long-term toxic effects on D. pulex. The findings highlight the importance of multigenerational and chronic biological evaluations to assess risks of emerging pollution.

Cytochrome P450 (CYP) enzymes are one of the largest protein families, and they metabolise a wide range of lipophilic organic endogenous and exogenous compounds. Many cytochrome P450 genes have been cloned and characterised, and they are frequently used as biomarkers in environmental toxicology studies because of their sensitivity and inducibility. In the present study, the full-length cDNAs of DpCYP370B and DpCYP4 were cloned from Daphnia pulex for the first time. The sequence of DpCYP370B consisted of an ORF of 1515 bp that encoded a 504 amino acid polypeptide, while the sequence of DpCYP4 comprised an ORF of 1527 bp that encoded a 508 amino acid polypeptide. Homologous alignments revealed the presence of a conserved cysteine haeme-iron ligand signature, FxxGxxxCxG, located in the C-terminal portion. Both the proteins contained a sequence for a transmembrane region that was deduced to be located in the endoplasmic reticulum. Subsequently, the expression levels of DpCYP370B and DpCYP4, as well as those of CYP4AN1, CYP4C33, and CYP4C34, were investigated using quantitative real-time PCR after exposure to five polystyrene nanoplastic concentrations: 0 (control), 0.1, 0.5, 1, and 2 mg/L for 21 days. Except for DpCYP4, the highest mRNA expression was observed at 0.5 mg/L nanoplastics; next, the expression of three of the enzymes (DpCYP370B, CYP4AN1, CYP4C34,) decreased to that of the control level at 1 and 2 mg/L doses of nanoplastics. The expression of DpCYP4 did not significantly change compared with that of the control group. These results indicated that CYP genes might play an important role in protecting D. pulex against nanoplastic pollutants.

To assess the ecological health of the Chongming Dongtan Nature Reserve (CDNR), univariate and multimetric indices-AZTI's Marine Biotic Index (AMBI), multivariate AMBI (M-AMBI), abundance-biomass comparison curves, taxonomic diversity, Shannon-Wiener index (H') (log2), and Benthic Opportunistic Polychaetes Amphipods index (BOPA) - were used to translate the macrobenthic community into classifications of ecological health in 2016 and 2017. Based on the results of the various indicators, the overall habitat status of the CDNR ranged from good to high status. Sites 4 and 5, which fell in the remaining tidal flats following reclamation activities, were classified as disturbed, whereas the degree of disturbance at site 3 was low. The results indicate that the CDNR is under some form of human disturbance, which includes reclamation and animal (mainly cattle) grazing. H' (log2) and M-AMBI index may be more suitable for assessing ecological quality in intertidal zones, including the CDNR.

The effects of microplastic exposure on the non-specific immune responses and intestinal microflora remain unclear. In this study, juveniles of the Chinese mitten crab (Eriocheir sinensis) were exposed to different concentrations of microplastics (0, 0.04, 0.4, 4, and 40 mg/L) for 7, 14, and 21 days to explore their effects. Under microplastic-induced stress, the contents or activities of most immune-related factors [haemocyanin (Hc), alkaline phosphatase (AKP), phenoloxidase (PO), lysozyme (LSZ), and acid phosphatase (ACP)] decreased after an initial increase in the low-dose or short exposure times in the haemolymph and hepatopancreas. The trends in Hc and LSZ gene expression were consistent with the corresponding changes in enzyme activities. Moreover, the haemocyte expression of caspase and MyD88 in the groups with microplastic-induced stress was higher than that in the control group, whereas the expression levels in the hepatopancreas were first increased and then decreased. Furthermore, the relative abundance of Firmicutes and Bacteroidetes decreased following exposure to 40 mg/L microplastics, whereas that of Fusobacteria and Proteobacteria increased. These results indicate that microplastics affect immune enzyme activity and immune-related gene expression and change the diversity and composition of the intestinal microflora in E. sinensis.

The water flea Daphnia pulex, a small crustacean that lives in freshwater ponds, undergoes parthenogenesis and gamogenesis according to environmental conditions. In D. pulex, different morphological characteristics can be observed using a microscope during aging. In this study, we recorded the growth, reproduction, and survival indicators of parthenogenetic D. pulex females to analyze the morphological changes that occur during senescence. We observed the growth and development of the parthenogenetic females on the 7th, 14th, 21st, and 28th days by using scanning electron microscopy and ultrastructure of the gonads on the 7th, 14th and 21st days with transmission electron microscopy. We found that body length and reproductive capacity in the parthenogenetic individuals first increased and then decreased with aging, and the survival rate decreased gradually. The depression of the brood pouch on the back of the parthenogenetic females implied a decline in gonadal function and gradual aging. During senescence, the number of lipid droplets in the gonads of the parthenogenetic females decreased, and the integrity of the mitochondria and peroxisome was destroyed. The results of our study revealed the morphological characteristics of senescence in D. pulex.
