RESUMO

Traditionally, monitoring approaches to survey marine caves have been constrained by equipment limitations and strict safety protocols. Nowadays, the rise of new approaches opens new possibilities to describe these peculiar ecosystems. The current study aimed to explore the potential of Structure from Motion (SfM) photogrammetry to assess the abundance and spatial distribution of the sessile benthic assemblages inside a semi-submerged marine cave. Additionally, since impacts of recent date mussel Lithophaga lithophaga illegal fishing were recorded, a special emphasis was paid to its distribution and densities. The results of SfM were compared with a more "traditional approach", by simulating photo-quadrats deployments over the produced orthomosaics. A total of 22 sessile taxa were identified, with Porifera representing the dominant taxa within the cave, and L. lithophaga presenting a density of 88.3 holes/m2. SfM and photo-quadrats obtained comparable results regarding species richness, percentage cover of identified taxa and most of the seascape metrics, while, in terms of taxa density estimations, photo-quadrats highly overestimated their values. SfM resulted in a suitable non-invasive technique to record marine cave assemblages. Seascape indexes proved to be a comprehensive way to describe the spatial pattern of distribution of benthic organisms, establishing a useful baseline to assess future community shifts.

Microbial communities inhabiting sponges are known to take part in many metabolic pathways, including nutrient cycles, and possibly also in the bioaccumulation of trace elements (TEs). Here, we used high-throughput, Illumina sequencing of 16S rRNA genes to characterize the prokaryotic communities present in the cortex and choanosome, respectively the external and internal body region of Chondrosia reniformis, and in the surrounding seawater. Furthermore, we estimated the total mercury content (THg) in these body regions of the sponge and in the corresponding microbial cell pellets. Fifteen prokaryotic phyla were detected in association with C. reniformis, 13 belonging to the domain Bacteria and two to the Archaea. No significant differences between the prokaryotic community composition of the two regions were found. Three lineages of ammonium-oxidizing organisms (Cenarchaeum symbiosum, Nitrosopumilus maritimus, and Nitrosococcus sp.) co-dominated the prokaryotic community, suggesting ammonium oxidation/nitrification as a key metabolic pathway within the microbiome of C. reniformis. In the sponge fractions, higher THg levels were found in the choanosome compared to the cortex. In contrast, comparable THg levels found in the microbial pellets obtained from both regions were significantly lower than those observed in the corresponding sponge fractions. Our work provides new insights into the prokaryotic communities and TEs distribution in different body parts of a model organism relevant for marine conservation and biotechnology. In this sense, this study paves the way for scientists to deepen the possible application of sponges not only as bioindicators, but also as bioremediation tools of metal polluted environments.

RESUMO

DNA barcoding is a versatile approach that has revolutionized taxonomy and other akin topics in biology and ecology, due to its simplicity and relatively costless procedures. The method consists in the production of one or a few amplicons from informative genetic regions via Sanger sequencing. These markers are selected because they tend to evolve at a similar pace as speciation, allowing to discriminate organismal species. The applicability of this technique is here portrayed for the taxonomical identification of marine sponges (phylum: Porifera) as an exemplification.

Thanks to several European directives and conventions there is a general increase of awareness regarding the key ecological role of coralligenous habitats in the Mediterranean Sea, addressing several research projects to standardize protocols for the description of its integrity. Here we surveyed 13 stations along the Italian coasts of the Western Mediterranean Sea, using video-transects technique, comparing the biological structure of coralligenous assemblages and testing the importance of their three-dimensional complexity as a proxy to define their health conditions. We considered the diversity of taxa, fishing impacts and the entity of damage on gorgonian's choenenchyme due to thermal stress, to evidence a gradient in the coralligenous health conditions. Here we developed a method to evaluate coralligenous complexity, selecting categories of taxa particularly sensitive to multiple stressors, named Structural Descriptors to describe the three-dimensional structure of the bioconcretions and to assess a unique Index of 3D - Structural Complexity.

The sponge fauna has been largely overlooked in the Archipelago of Hawai'i, notwithstanding the paramount role of this taxon in marine ecosystems. The lack of knowledge about Porifera populations inhabiting the Hawai'ian reefs limits the development of ecological studies aimed at understanding the functioning of these marine systems. Consequently, this project addresses this gap by describing the most representative sponge species in the shallow waters of the enigmatic bay of Kane'ohe Bay, in O'ahu Island. A total of 30 species (28 demosponges and two calcareous sponges) living associated to the reef structures are here reported. Six of these species are new records to the Hawai'ian Porifera catalogue and are suspected to be recent introductions to these islands. Morphological descriptions of the voucher specimens are provided, along with sequencing data of two partitions involving the mitochondrial cytochrome oxidase subunit 1 (COI) marker and a fragment covering partial (18S and 28S) and full (ITS-1, 5.8S and ITS-2) nuclear ribosomal genes. Species delimitations based on genetic distances were calculated to valitate how taxonomic assignments from DNA barcoding aligned with morphological identifications. Of the 60 sequences submitted to GenBank ~88% are the first sequencing records for the corresponding species and genetic marker. This work compiles the first catalogue combining morphological characters with DNA barcoding of Hawai'ian sponges, and contributes to the repository of public databases through the Sponge Barcoding Project initiative.

Sponges are key components of the benthic assemblages and play an important functional role in many ecosystems, especially in coral reefs. The Indonesian coral reefs, located within the so-called "coral triangle", are among the richest in the world. However, the knowledge of the diversity of sponges and several other marine taxa is far from being complete in the area. In spite of this great biodiversity, most of the information on Indonesian sponges is scattered in old and fragmented literature and comprehensive data about their diversity are still lacking. In this paper, we report the presence of 94 species recorded during different research campaigns mainly from the Marine Park of Bunaken, North Sulawesi. Six species are new for science and seven represent new records for the area. Several others are very poorly known species, sometimes recorded for the second time after their description. For most species, besides field data and detailed descriptions, pictures in vivo are included. Moreover, two new symbiotic sponge associations are described. This work aims to increase the basic knowledge of Indonesian sponge diversity as a prerequisite for monitoring and conservation of this valuable taxon.

Pronounced differences with respect to the extent of infestation and the degree of Lithophaga lithophaga shell damage inflicted by euendolithic taxa at two sites in the Adriatic Sea representing different productivity conditions, are described. Shells collected from the eastern part of Kastela Bay, which is characterized by higher primary productivity, have significantly more shell damage then the shell collected from a site on the outer coast of the island of Ciovo exposed to the oligotrophic Adriatic Sea. The presence of endoliths and their perforations were detected in different layers of the shell, including solidly mineralized parts of the skeleton and within the organic lamellae incorporated into the shell. Phototrophic endoliths were not observed in the specimens. The most serious damage to L. lithophaga shells was the boring clionaid sponge Pione vastifica, which was more common in shells collected from Kastela.

Temperate reefs, built by multilayers of encrusting algae accumulated during hundreds to thousands of years, represent one of the most important habitats of the Mediterranean Sea. These bioconstructions are known as "coralligenous" and their spatial complexity allows the formation of heterogeneous microhabitats offering opportunities for a large number of small cryptic species hardly ever considered. Although sponges are the dominant animal taxon in the coralligenous rims with both insinuating and perforating species, this group is until now poorly known. Aim of this work is to develop a reference baseline about the taxonomic knowledge of sponges and, considering their high level of phenotypic plasticity, evaluate the importance of coralligenous accretions as a pocket for biodiversity conservation. Collecting samples in four sites along the coast of the Ligurian Sea, we recorded 133 sponge taxa (115 of them identified at species level and 18 at genus level). One species, Eurypon gracilis is new for science; three species, Paratimea oxeata, Clathria (Microciona) haplotoxa and Eurypon denisae are new records for the Italian sponge fauna, eleven species are new findings for the Ligurian Sea. Moreover, seventeen species have not been recorded before from the coralligenous community. The obtained data, together with an extensive review of the existing literature, increase to 273 the number of sponge species associated with the coralligenous concretions and confirm that this habitat is an extraordinary reservoir of biodiversity still largely unexplored, not only taxonomically, but also as to peculiar adaptations and life histories.

Chemical analysis of the organic extract of Theonella swinhoei yielded two new tridecadepsipeptides of the theonellapeptolide family, namely sulfinyltheonellapeptolide, characterized by a methylsulfinylacetyl group at the N-terminus, and theonellapeptolide If, the first member of this class of compounds to show four valine residues. The structures of the compounds, isolated along with the known theonellapeptolide Id, were determined by extensive 2D NMR and MS/MS analyses followed by application of Marfey's method. The isolated peptides exhibited moderate antiproliferative activity against HepG2 cells, a hepatic carcinoma cell line.

Chemical analysis of the Indonesian sponge Plakortis cfr. lita afforded two new analogues of the potent trypanocidal agent manadoperoxide B (1), namely 12-isomanadoperoxide B (2) and manadoperoxidic acid B (3). These compounds were isolated along with a new short chain dicarboxylate monoester (4), bearing some interesting relationships with the polyketide endoperoxides found in this sponge. Some semi-synthetic analogues of manadoperoxide B (6-8) were prepared and evaluated for antitrypanosomal activity and cytotoxicity. These studies revealed crucial structure-activity relationships that should be taken into account in the design of optimized and simplified endoperoxyketal trypanocidal agents.

Chemical investigation of an Indonesian specimen of Theonella swinhoei afforded the new dimeric macrolides isoswinholide B (5) and swinholide K (6), along with the known swinholides A (1), B (2) and D (3) and isoswinholide A (4). Isoswinholide B showed an unprecedented 21/19' lactonization pattern, while swinholide K included an sp(2) methylene attached at C-4 and an additional oxymethine group at C-5, whose configuration has been determined through application of J-based configuration analysis. The isolated swinholides (1-6), with the exception of isoswinholide B, showed a cytotoxic activity on HepG2 (hepatocarcinoma cell line) in the nanomolar range.

Sponges are characterised by high levels of phenotypic plasticity, thus allowing the same species to live in different habitats by taking different shapes. Here we describe 28 sponge species associated with the octocorals Carijoa riisei, Paratelesto rosea and Alertigorgia hoeksemai in Indonesia, Hawai'i and Vietnam, including four species that are new to science (Chondropsis subtilis, Hymedesmia (Hymedesmia) spinata, Hymedesmia (Stylopus) perlucida, Mycale (Aegogropila) furcata). Moreover, a large proportion of the described sponge species (21.4%) represent new records for the studied areas (Indonesia and Hawai'i). In total, we have studied 47 colonies of C. riisei associated with 24 sponge species, 5 colonies of P. rosea associated with 4 species and one colony of A. hoeksemai associated with one sponge species. Collectively, these examples of associations highlight the importance of epibiosis as a biodiversity enhancing process.

Leucettamols, bifunctionalized sphingoid-like compounds obtained from a marine sponge Leucetta sp., act as non-electrophilic activators of the TRPA1 channel and potent inhibitors of the icilin-mediated activation of the TRPM8 channel, while they are inactive on CB1, CB2 and TRPV1 receptors. Leucettamols represent the first compounds of marine origin to target TRPA1 and the first class of natural products to inhibit TRPM8 channels. The preparation of a small series of semi-synthetic derivatives revealed interesting details on the structure-activity relationships within this new chemotype of simple acyclic TRP modulators.

Chemical investigation of the marine sponge Plakortis cfr. lita afforded a library of endoperoxyketal polyketides, manadoperoxides B-K (3-5 and 7-13) and peroxyplakoric esters B(3) (6) and C (14). Eight of these metabolites are new compounds and some contain an unprecedented chlorine-bearing THF-type ring in the side chain. The library of endoperoxide derivatives was evaluated for in vitro activity against Trypanosoma brucei rhodesiense and Leishmania donovani. Some compounds, such as manadoperoxide B, exhibited ultrapotent trypanocidal activity (IC(50) = 3 ng mL(-1)) without cytotoxicity. Detailed examination of the antitrypanosomal activity data and comparison with those available in the literature for related dioxane derivatives enabled us to draw a series of structure-activity relationships. Interestingly, it appears that minor structural changes, such as a shift of the methyl group around the dioxane ring, can dramatically affect the antitrypanosomal activity. This information can be valuable to guide the design of optimized antitrypanosomal agents based on the dioxane scaffold.

From the polar organic extract of the Indonesian sponge Dasychalina sp. we have isolated haplosamate A (1), a unique C(28) sterol containing a sulfate group at C-3 and a methyl phosphate at C-15, along with its new desulfo analogue 2, whose structure has been secured by detailed NMR investigation. Compounds 1 and 2, as well as their semi-synthetic analogues 3-5, have been evaluated for interaction with CB(1) and CB(2) receptors through a binding test. Desulfohaplosamate (2) showed a selective affinity for CB(2) receptors in the low µM range, while a semi-synthetic derivative with cleaved ring B showed a complete loss of affinity for both receptors, highlighting the importance of an intact steroid nucleus. To our knowledge, haplosamate derivatives represent the first CB receptor ligands belonging to the class of steroids.

The chemical investigation of an Indonesian specimen of Theonella swinhoei afforded four aurantosides, one of which, aurantoside J (5), is a new compound. The structure of this metabolite, exhibiting the unprecedented N-α-glycosidic linkage between the pentose and the tetramate units, has been determined through detailed spectroscopic analysis. The four obtained aurantosides have been tested against five fungal strains (four Candida and one Fusarium) responsible of invasive infections in immuno-compromised patients. The non-cytotoxic aurantoside I (4) was the single compound to show an excellent potency against all the tested strains, thus providing valuable insights about the antifungal potential of this class of compounds and the structure-activity relationships.

The new endoperoxyketal polyketides manadoperoxides A-D (2-5) have been isolated from the Indonesian sponge Plakortis cfr. simplex and their stereostructures established by means of spectroscopic data and semisynthetic transformations. Manadoperoxides were assayed in vitro against D10 and W2 strains of Plasmodium falciparum and showed moderate antimalarial activity compared to that of plakortin (1) and peroxyplakoric B(3) ester (9), the latter differing from manadoperoxide B only by minor structural details. This unexpected difference in the antimalarial activity has been rationalized on the basis of our recently published model for the interaction of 1,2-dioxanes with heme and production of C-centered radicals toxic to the parasite. For the manadoperoxides, either the endoperoxide linkage is inaccessible to the heme iron or the O1 radical cannot evolve to produce a C-centered radical.

The chlorinated polyene aurantoic acid (1) and the 4-methylene sterol dehydroconicasterol (2) were isolated from the Indonesian sponge Theonella swinhoei, and their structures were elucidated by interpretation of spectroscopic data. Aurantoic acid is a unique member in the class of naturally occurring conjugated polyene derivatives, while dehydroconicasterol is the likely biogenetic precursor of the major Theonella 4-methylene sterols.

RESUMO

Two new steroidal derivatives, named clionastatins A and B, have been isolated from the burrowing sponge Cliona nigricans. These molecules are tri-and tetrachlorinated androstane derivatives, respectively, and they represent the first polyhalogenated steroids found in a natural organism, either marine or terrestrial, and the first examples of halogenated androstanes in nature. Both clionastatins proved to be potently cytotoxic.
