RESUMO

Removal and recovery of methyl chloride (CH3Cl) from exhaust gas of organic silicon industry is highly important from the perspective of environment and economy. For the first time, a tailor-made microporous coordination polymer (Mn-BDC-TPA) was synthesized and applied to the efficient capture and recovery of CH3Cl from related gas mixtures. The high adsorption capacity of CH3Cl (163.4 cm3/g) and high adsorption selectivity of CH3Cl over other impurity gases (1965 for N2, 65 for CH4, and 16 for C2H6) were achieved at 298 K and 100 kPa due to the dual-cage pore system and larger polarizability of CH3Cl. Dynamic breakthrough experiments demonstrate the excellent CH3Cl recovery performance (capacity of >98 cm3/g and purity of >95%) in one adsorption-desorption cycle from the CH3Cl-involved binary, ternary, or quaternary gas mixture.

Purification of ethylene (C2H4) as the most extensive and output chemical, from complex multi-components is of great significance but highly challenging. Herein we demonstrate that precise pore structure tuning by controlling the network hydrogen bonds in two highly-related porous coordination networks can shift the efficient C2H4 separation function from C2H2/C2H4/C2H6 ternary mixture to CO2/C2H2/C2H4/C2H6 quaternary mixture system. Single-crystal X-ray diffraction revealed that the different amino groups on the triazolate ligands resulted in the change of the hydrogen bonding in the host network, which led to changes in the pore shape and pore chemistry. Gas adsorption isotherms, adsorption kinetics and gas-loaded crystal structure analysis indicated that the coordination network Zn-fa-atz (2) weakened the affinity for three C2 hydrocarbons synchronously including C2H4 but enhanced the CO2 adsorption due to the optimized CO2-host interaction and the faster CO2 diffusion, leading to effective C2H4 production from the CO2/C2H2/C2H4/C2H6 mixture in one step based on the experimental and simulated breakthrough data. Moreover, it can be shaped into spherical pellets with maintained porosity and separation performance.

An ultramicroporous metal-organic framework (MOF) constructed from dysprosium(III) and oxalate, termed Dy-F-oxa, is carefully studied for inverse separation of CO2 from C2H2. Adsorption experiments and modeling studies reveal that the high CO2 adsorption is attributed to the preferential sites for CO2 by coordinated water. After the equimolar gas mixture breakthrough experiment, C2H2 can be directly produced as a pure effluent.

BACKGROUND: Streptococcus dysgalactiae subsp. dysgalactiae has been identified as an animal pathogen that is thought to occur only in animal populations. Between 2009 and 2022, humans infected with SDSD were reported rarely. There is a lack of details on the natural history, clinical features, and management of disease caused by this pathogen. This case outlines a human SDSD with muscle aches and progressive loss of muscle strength leading to immobility and multi-organ dysfunction syndrome. CASE PRESENTATION: She presented with muscle pain and weakness, and later developed a sore throat, headache and fever with a maximum temperature of 40.5 °C. The muscle strength of the extremities gradually decreased to grade 1 and the patient was unable to move on his own. Next-generation blood sequencing and multi-culture confirmed the presence of Streptococcus dysgalactiae and Streptococcus dysgalactiae subsp. Dysgalactiae, respectively. A Sequential Organ Failure Assessment score of 6 indicated septicemia, and therapeutic antibiotics were prescribed empirically. After 19 days of inpatient treatment, the patient's condition greatly improved and completely recovered within a month. CONCLUSION: Symptoms of Streptococcus dysgalactiae subsp. dysgalactiae presenting with progressive limb weakness resemble polymyositis, so a precise differential diagnosis is essential. Multidisciplinary consultation is helpful when polymyositis cannot be ruled out and facilitates the choice of an optimal treatment protocol. In the context of this case, penicillin is an effective antibiotic for Streptococcus dysgalactiae subsp. dysgalactiae infection.

BACKGROUND: Laparoscopic partial nephrectomy has been widely used in renal cell carcinoma treatment. The efficacy of GreenLight laser on Laparoscopic partial nephrectomy is still unknown. AIM: To present the first series of laparoscopic partial nephrectomy (LPN) by GreenLight laser enucleation without renal artery clamping. Due to the excellent coagulation and hemostatic properties of the laser, laser-assisted LPN (LLPN) makes it possible to perform a "zero ischemia" resection. METHODS: Fifteen patients with T1a exogenous renal tumors who received high-power GreenLight laser non-ischemic LPN in our hospital were retrospectively analyzed. All clinical information, surgical and post-operative data, complications, pathological and functional outcomes were analyzed. RESULTS: Surgery was successfully completed in all patients, and no open or radical nephrectomy was performed. The renal artery was not clamped, leading to no ischemic time. No blood transfusions were required, the average hemoglobin level ranged from 96.0 to 132.0 g/L and no postoperative complications occurred. The mean operation time was 104.3 ± 8.2 min. The postoperative removal of negative pressure drainage time ranged from 5.0 to 7.0 d, and the mean postoperative hospital stay was 6.5 ± 0.7 d. No serious complications occurred. Postoperative pathological results showed clear cell carcinoma in 12 patients, papillary renal cell carcinoma in 2 patients, and hamartoma in 1 patient. The mean creatinine level was 75.0 ± 0.8 µmol/L (range 61.0-90.4 µmol/L) at 1 mo after surgery, and there were no statistically significant differences compared with pre-operation (P > 0.05). The glomerular filtration rate ranged from 45.1 to 60.8 mL/min, with an average of 54.0 ± 5.0 mL/min, and these levels were not significantly different from those before surgery (P > 0.05). CONCLUSION: GreenLight laser has extraordinary cutting and sealing advantages when used for small renal tumors (exogenous tumors of stage T1a) during LPN. However, use of this technique can lead to the generation of excessive smoke.

Herein, by replacement of the linear terephthalate linker with the bending 2,5-thiophenedicarboxylate (tdc2-) linker in the typical (3,9)-connected metal-organic framework, with a reduced 8-connected hydroxyl-centered trinuclear cluster, a new (3,8)-connected network, [Ni3(µ3-OH)(tdc)3(tpp)] [DZU-1; tpp = 2,4,6-tris(4-pyridyl)pyridine], was synthesized. The modified pore environment enables DZU-1 to selectively adsorb C2H2 over CO2 in an efficient manner.

Herein, we propose the required general pore features for one-step C2H4 production from a C2 hydrocarbon mixture, such as: (i) a non-metallic binding site; (ii) polar pore surface; and (iii) narrow pocket. Four representative MOFs (Ni-BDC-INA, CPM-173, UiO-66, and DMOF-1) and two counterexamples (MOF-74-Co and IISERP-MOF2) were selected to verify this concept, by conducting single-component gas adsorption measurements, multiple-component breakthrough experiments, and molecular simulations. The established pore features are a stepping-stone for the design and synthesis of new MOFs with improved performance for C2H4 separation.

Non-clear renal cell carcinomas (nccRCCs) are less frequent in kidney cancer with histopathological heterogeneity. A better understanding of the tumor biology of nccRCC can provide more effective treatment paradigms for different subtypes. To reveal the heterogeneity of tumor microenvironment (TME) in nccRCC, we performed 10x sing-cell genomics on tumor and normal tissues from patients with papillary renal cell carcinoma (pRCC), chromophobe RCC (chrRCC), collecting duct carcinoma (CDRCC) and sarcomatoid RCC (sarRCC). 15 tissue samples were finally included. 34561 cells were identified as 16 major cell clusters with 34 cell subtypes. Our study presented the sing-cell landscape for four types of nccRCC, and demonstrated that CD8+ T cells exhaustion, tumor-associated macrophages (TAMs) and sarcomatoid process were the pivotal factors in immunosuppression of nccRCC tissues and were closely correlated with poor prognosis. Abnormal metabolic patterns were present in both cancer cells and tumor-infiltrating stromal cells, such as fibroblasts and endothelial cells. Combined with CIBERSORTx tool, the expression data of bulk RNA-seq from TCGA were labeled with cell types of our sing-cell data. Calculation of the relative abundance of cell types revealed that greater proportion of exhausted CD8+ T cells, TAMs and sarRCC derived cells were correlated with poor prognosis in the cohort of 274 nccRCC patients. To the best of our knowledge, this is the first study that provides a more comprehensive sight about the heterogeneity and tumor biology of nccRCC, which may potentially facilitate the development of more effective therapies for nccRCC.

One-step adsorptive purification of ethylene (C2H4) from four-component gas mixtures comprising acetylene (C2H2), ethylene (C2H4), ethane (C2H6) and carbon dioxide (CO2) is an unmet challenge in the area of commodity purification. Herein, we report that the ultramicroporous sorbent Zn-atz-oba (H2oba = 4,4-dicarboxyl diphenyl ether; Hatz = 3-amino-1,2,4-triazole) enables selective adsorption of C2H2, C2H6 and CO2 over C2H4 thanks to the binding sites that lie in its undulating pores. Molecular simulations provide insight into the binding sites in Zn-atz-oba that are responsible for coadsorption of C2H2, C2H6 and CO2 over C2H4. Dynamic breakthrough experiments demonstrate that the selective binding exhibited by Zn-atz-oba can produce polymer-grade purity (>99.95%) C2H4 from binary (1:1 for C2H4/C2H6), ternary (1:1:1 for C2H2/C2H4/C2H6) and quaternary (1:1:1:1 for C2H2/C2H4/C2H6/CO2) gas mixtures in a single step.

Low-concentration ethane capture is crucial for environmental protection and natural gas purification. The ideal physisorbent with strong C2 H6 interaction and large C2 H6 uptake at low-concentration level has rarely been reported, due to the large pKa value and small quadrupole moment of C2 H6 . Herein, we demonstrate the perfectly size matching between the ultramicropore (pore size of 4.6 Å) and ethane (kinetic diameter of 4.4 Å) in a nickel pyridine-4-carboxylate metal-organic framework (IISERP-MOF2), which enables the record-breaking performance for low concentration C2 H6 capture. IISERP-MOF2 exhibits the large C2 H6 adsorption enthalpy of 56.7 kJ/mol, and record-high C2 H6 uptake at low pressure of 0.01-0.1 bar and 298 K (1.8 mmol/g at 0.01 bar). Molecule simulations and C2 H6 -loading crystal structure analysis revealed that the maximized interaction sites in IISERP-MOF2 with ethane molecule originates the strong C2 H6 adsorption. The dynamic breakthrough experiments for gas mixtures of C2 H6 /N2 (1/999, v/v) and C2 H6 /CH4 (5/95, v/v) proved the excellent low-concentration C2 H6 capture performance.

Rigid molecular sieving materials are the ideal candidates for gas separation (e. g., C2 H2 /C2 H4 ) due to their ultrahigh adsorption selectivity and the absence of gas co-adsorption. However, the absolute molecular sieving effect for C2 H2 /C2 H4 separation has rarely been realized because of their similar physicochemical properties. Herein, we demonstrate the absolute molecular sieving of C2 H2 from C2 H4 by a rigid ultra-microporous metal-organic framework (F-PYMO-Cu) with 1D regular channels (pore size of ca. 3.4 Å). F-PYMO-Cu exhibited moderate acetylene uptake (35.5 cm3 /cm3 ), but very low ethylene uptake (0.55 cm3 /cm3 ) at 298 K and 1 bar, yielding the second highest C2 H2 /C2 H4 uptake ratio of 63.6 up to now. One-step C2 H4 production from a binary mixture of C2 H2 /C2 H4 and a ternary mixture of C2 H2 /CO2 /C2 H4 at 298 K was achieved and verified by dynamic breakthrough experiments. Coupled with excellent thermal and water stability, F-PYMO-Cu could be a promising candidate for industrial C2 separation tasks.

BACKGROUND: Renal cell carcinoma (RCC) is one of the most common malignant tumors originating from the renal parenchymal urinary epithelial system. Tripartite motif 47 (TRIM47) is a member of the TRIM family proteins, which has E3 ligase activity and has been demonstrated to be involved in the occurrence and prognosis of many tumors. The main purpose of this study is to explore the role and potential mechanism of TRIM47 in promoting malignant biological behavior of RCC. MATERIALS AND METHODS: TRIM47 mRNA and protein levels in human renal cancer and paired normal adjacent tissues were detected by qRT-PCR and Western blot. The effects of TRIM47 knockdown and overexpression in renal cell carcinoma cells on cell proliferation, invasion and xenograft tumor growth in nude mice were analyzed. The molecular mechanism was explored by mass spectrometric exploration,Western blot and immunoprecipitation assays. RESULTS: TRIM47 promoted RCC cell proliferation in vitro and in vivo as an oncogene. Mechanistically, TRIM47 exerted an E3 ligase activity by interacting with P53 protein to increase its ubiquitination and degradation, which further promoted the malignant biological behavior of RCC. CONCLUSIONS: Our study demonstrated that the TRIM47-P53 axis played a functional role in RCC progression and suggested a potential therapeutic target for RCC.

Ethylene production from C2 hydrocarbon mixtures through one separation step is desirable but challenging because of the similar size and physical properties of acetylene, ethylene, and ethane. Herein, we report three new isostructural porous coordination networks (NPU-1, NPU-2, NPU-3; NPU represents Northwestern Polytechnical University) that are sustained by 9-connected nodes based upon a hexanuclear metal cluster of composition [Mn6(µ3-O)2(CH3COO)3]6+. NPU-1/2/3 exhibit a dual cage structure that was systematically fine-tuned in terms of cage size to realize selective adsorption of C2H2 and C2H6 over C2H4. Dynamic breakthrough experiments demonstrated that NPU-1 produces ethylene in >99.9% purity from a three-component gas mixture (1:1:1 C2H2/C2H4/C2H6). Molecular modeling studies revealed that the dual adsorption preference for C2H2 and C2H6 over C2H4 originates from (a) strong hydrogen-bonding interactions between electronegative carboxylate O atoms and C2H2 molecules in one cage and (b) multiple non-covalent interactions between the organic linkers of the host network and C2H6 molecules in the second cage.

Background: Cancer stem cells (CSCs) are biologically characterized by self-renewal, multi-directional differentiation and infinite proliferation, inducing anti-tumor drug resistance and metastasis. In the present study, we attempted to depict the baseline landscape of CSC-mediated biological properties, knowing that it is vital for tumor evolution, anti-tumor drug selection and drug resistance against fatal malignancy. Methods: We performed single-cell RNA sequencing (scRNA-seq) analysis in 15208 cells from a pair of primary and metastatic sites of collecting duct renal cell carcinoma (CDRCC). Cell subpopulations were identified and characterized by t-SNE, RNA velocity, monocle and other computational methods. Statistical analysis of all single-cell sequencing data was performed in R and Python. Results: A CSC population of 1068 cells was identified and characterized, showing excellent differentiation and self-renewal properties. These CSCs positioned as a center of the differentiation process and transformed into CDRCC primary and metastatic cells in spatial and temporal order, and played a pivotal role in promoting the bone destruction process with a positive feedback loop in the bone metastasis microenvironment. In addition, CSC-specific marker genes BIRC5, PTTG1, CENPF and CDKN3 were observed to be correlated with poor prognosis of CDRCC. Finally, we pinpointed that PARP, PIGF, HDAC2, and FGFR inhibitors for effectively targeting CSCs may be the potential therapeutic strategies for CDRCC. Conclusion: The results of the present study may shed new light on the identification of CSCs, and help further understand the mechanism underlying drug resistance, differentiation and metastasis in human CDRCC.

The pore-size effect on ethane adsorption and ethane/ethylene separation in three isostructural metal azolate frameworks (MAF-123-Mn/Zn/Cu) were thoroughly investigated. MAF-123-Mn/Zn/Cu were synthesized by the solvothermal method on a gram scale. Decreasing the pore size from 6.1 to 4.9 Å leads to an increase in the ethane adsorption energy from 23 to 27.5 kJ mol-1 and further ethane/ethylene separation efficiency. Molecule simulations revealed that a shorter ethane-framework interaction distance in MAF-123-Zn than that in MAF-123-Mn is responsible for the increased adsorption energy. Dynamic breakthrough experiments manifest that these metal azolate frameworks can effectively produce high-purity ethylene from ethane in one adsorption step.

BACKGROUND: To identify the association between the width of the gastric conduit and the benign anastomotic stricture (BAS) after esophagectomy with end-to-side cervical anastomosis for esophageal cancer. METHODS: Patients with esophageal cancer who underwent esophagectomy between July 2013 and July 2014 were included in this study. The gastric conduit was used for reconstruction in all patients and end-to-side cervical anastomosis were performed using a circular stapler. The patients were divided into a narrow group (3-5 cm) and a wide group (>5 cm) based on the gastric conduit width. Univariate and multivariate logistic regressions were used to analyze the possible factors (patients' age, gender, preoperative comorbidities, neoadjuvant chemotherapy, gastric conduit width, anastomotic leakage) that could affect the incidence of BAS. RESULTS: Two-hundred and one patients were included in this study. The median follow-up period was 29 months (17-58 months). Seven cases (3.5%) showed anastomotic leakage in the postoperative period and 38 patients (18.9%) developed BAS; all within the first year of follow-up. In univariate analysis, the width of the gastric conduit was the only risk factor for the development of BAS (odds ratio [OR] = 3.36, P = .005). In multivariate logistic regression analysis, the wide group was an independent significant risk factor for the development of BAS developing compared with the narrow group (OR = 2.84, P = .02). CONCLUSIONS: A wide gastric conduit width (>5 cm) is an independent risk factor for the development of BAS after esophagectomy and stapled cervical end-to-side anastomosis for esophageal cancer.

Bladder cancer is one of the most commonly diagnosed urological malignancies. Acquired resistance to chemotherapy is a great barrier for achieving successful treatment of bladder cancer. In the present study, we investigated the effect and mechanisms of icaritin, a flavonol glycoside derived from genus Epimedium, against human bladder cancer cells. It was found that despite the low cytotoxicity in normal human HEK293 cells, icaritin significantly inhibited the proliferation and colony formation of BT5637 and T24 bladder cancer cells time- and dose-dependently compared to the DMSO vehicle control. Moreover, cell viability monitored through mitochondrial membrane potential was inhibited markedly after icaritin treatment. Further investigation indicated that icaritin may inhibit epirubicin (EPI)-induced autophagy, and acted synergistically with EPI to suppress the proliferation of BT5637 and T24 cells. These findings suggest that icaritin may prove to be a novel potent therapeutic agent for the treatment of bladder cancer.
