RESUMO

BACKGROUND: The veterinary pharmaceutical industry has shown significant growth in recent decades. Several factors contribute to this increase as the demand for the improvement of the quality of life of both domestic and wild animals, together with the need to improve the quality, productivity, and safety of foodstuffs of animal origin. METHODS: The goal of this work was to identify the most suitable medicines for animals that focus on drug delivery routes as those for humans, although they may have different devices, such as collars and ear tags. RESULTS: Recent advances in drug delivery systems for veterinary use are discussed, both from academic research and the global market. The administration routes commonly used for veterinary medicines are also explored, while special attention is given to the latest technological trends to improve the drug performance, reducing the number of doses, animal stress, and side effects. CONCLUSION: Drug delivery system in veterinary decreased the number of doses, side effects, and animal stress that are a small fraction of the benefits of veterinary drug delivery systems and represent a significant increase in profit for the industry; also, it demands investments in research regarding the quality, safety, and efficacy of the drug and the drug delivery systems.

RESUMO

Objectives This study aimed to analyze the efficacy of platelet-rich plasma obtained from the peripheral, autologous blood of the patients in pain complaints reduction and functional improvement of knee osteoarthritis compared with the standard treatment with injectable corticosteroid, such as triamcinolone. Methods The patients were followed-up clinically at the preinfiltrative visit, with quantitative evaluation using the Knee Society Score (KSS), the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) score, and the Kellgren and Lawrence scales. In addition, they were reevaluated with the same scales after 1 month and 6 months of intervention with 2.5 mL of triamcinolone acetate or 5 mL of platelet-rich plasma. The study was performed on 50 patients with knee osteoarthritis treated at the Medical Specialty Center and randomly divided into equivalent samples for each therapy. Results The present study verified the reduction of pain scores, such as the WOMAC score, and elevations of functional scales, such as the KSS, evidenced in 180 days when using platelet-rich plasma, a therapy that uses the autologous blood of the patient and has fewer side effects. Conclusion Although both platelet-rich plasma and corticosteroid therapies have been shown to be effective in the reduction pain complaints and functional recovery, there was a statistically significant difference between them at 180 days. According to the results obtained, platelet-rich plasma presented longer-lasting effects within 180 days in the treatment of knee osteoarthritis.

RESUMO

Objective This research aims to study the effectiveness of platelet-rich plasma (PRP) in the treatment of patients with rotator cuff impingement syndrome compared with the treatment with subacromial injection of corticosteroids. Methods This is a double-blind, randomized, comparative clinical trial. The patients were clinically evaluated with the use of the Disabilities of the Arm, Shoulder and Hand (DASH) outcome measure, University of California Los Angeles (UCLA) shoulder rating scale and Constant-Murley shoulder outcome score (CMS) on the day of application, and then again after 1, 3 and 6 months. Results No statistically significant differences were found ( p < 0.05) when comparing the results of the DASH outcome measure, UCLA shoulder rating scale and CMS of the two groups at baseline and after 1, 3, and 6 months of treatment with subacromial injection. After the treatment, both groups showed a significant improvement in the DASH and UCLA scores ( p < 0.05) when compared with the baseline. However, the CMS at 6 months after treatment with steroids was lower than the baseline. Concusions These findings suggest that PRP is a safe treatment and can be a useful tool in the therapeutic arsenal against of the rotator cuff diseases, for there was no significant difference between the subacromial use of PRP and corticosteroids.

RESUMO

The objective of this work was to characterize the toxicological profile of a newly developed sunscreen formulation based on polymeric nanocapsules (NCs) loading benzophenone-3 (BZP3). NCs composed of poly(ε-caprolactone) carrot oil and Pluronic® F68 were produced by emulsification-diffusion method. Their mean particle size (Z-Ave) ranged from 280 to 420 nm, polydispersity index (PDI) was below 0.37, while zeta potential (ZP) reached about |+11 mV|. No cytotoxic effects were observed in L929 fibroblast cell line for the blank (i.e., non-loaded) NCs and BZP3-loaded NCs (BZP3-NCs). The semi-solid sunscreen formulation was stable over time (centrifugation testing) and exhibited non-Newtonian pseudoplastic behavior, which is typical of products for topical application onto the skin. The sun protection factor (SPF) value reached 8.84, when incorporating BZP3-NCs (SPF of 8.64) into the semi-solid formulation. A synergistic effect was also observed when combining the formulation ingredients of nanocapsules, i.e., SPF of carrot oil was 6.82, blank NCs was 6.84, and BZP3-loaded NCs was 8.64. From the hen's egg-chorioallantoic membrane test (HET-CAM) test, the non-irritation profile of the developed formulations could also be confirmed. The obtained results show a promising use of poly(ε-caprolactone) nanocapsules to be loaded with lipophilic sunscreens as benzophenone-3.

RESUMO

Hydrogels are three-dimensional, hydrophilic networks, comprising polymeric chains linked through physical or chemical bonds. In the area of food, hydrogels have great potential to be used in food packaging systems or as carriers of bioactive components. This paper reviews the nature of hydrogels, their 3D network conformation, their functional properties, and their potential applications in food packaging systems. Regarding their potential food packaging applications, hydrogels can present a conformation which allows their use as part of a packaging system to control the humidity generated by food products with high water content. Moreover, the incorporation of nanoparticles into hydrogels may grant them antimicrobial activity. Finally, although the current research in this field is still limited, the results obtained so far are promising for innovative and potential applications in the food field, which also include their integration into intelligent food packaging systems and their direct incorporation into food matrices as a flavor carrier system.

RESUMO

PURPOSE:: To evaluate the effect of aqueous extract of Baccharis trimera leaves on the proliferative capacity of the liver after partial hepatectomy (PH) in rats. METHODS:: Twenty Wistar rats weighing between 300 and 450g were divided into two groups: control (HP) and test (HP100-rats that received the aqueous extract of Baccharis trimera for four days at a dose of 100 mg / kg / day). On the fifth day, animals from both groups underwent resection of 70% of the liver. Twenty-four hours later, they were sacrificed and the remnant liver was removed and prepared for studied through PCNA immunohistochemistry. Data analysis for comparison between the two groups was made through the non-parametric statistical test Mann-Whitney test. RESULTS:: In all the animals studied was found most abundant nuclear immunostaining positive hepatocytes interlobular located in regions of the liver. Quantitative analysis of PCNA-positive cells revealed positivity rate significantly higher mean (p = 0.02) in HP100 group (77.1 ± 13.6) compared to the HP group (45.8 ± 12.9). CONCLUSION:: DAdministration of aqueous extract of the leaves of Baccharis trimera 100 mg/kg of animal has a significant positive effect on liver regeneration in rats, 24 hours after hepatectomy (70%).

RESUMO

Ulcerative colitis (UC) is a common intestinal inflammatory disease with an etiology that is not well understood. Although the anti-inflammatory and anti-oxidant effects of the hydroalcoholic extract of Brazilian red propolis (HERP) have been reported in various experimental models, its protective effect in models of UC have not been evaluated. The purpose of this study was to investigate the chemopreventive effect of hydroalcoholic extract of Brazilian red propolis (HERP) in acetic acid-induced colitis (AAIC) using a rodent model. The HERP was chemically characterised by HPLC/DAD analyses. Male rats were randomly assigned into four groups: sham, vehicle (with AAIC, treated with vehicle), P10 (with AAIC, treated with 10mg/kg HERP), and P100 (with AAIC, treated with 100mg/kg HERP). Treatments were performed for 7days, and colitis was induced on day seven. Animals were euthanized 24h after colitis induction and body weight, colon length, gross and histological scores, malondialdehyde (MDA) and myeloperoxidase (MPO) concentrations in colon tissue, and the immunohistochemical expression of inducible nitric oxide synthase (iNOS) were assessed. The major compounds found in HERP were liquiritigenin (68.8mg/g), formononetin (54.29mg/g), biochanin A (30.97mg/g), and daidzein (19.90mg/g). Rats treated with 10mg/kg HERP demonstrated significant decreases in MPO concentrations, gross and histological scores of tissue damage, and iNOS expression (p<0.05). Similarly, rats treated with 100mg/kg HERP demonstrated significant decreases in MPO levels (p<0.05) and histological scores of tissue damage (p<0.05). The results of this study indicate that oral administration of HERP attenuates AAIC in rats, which may be due to anti-inflammatory effects related to iNOS inhibition.

RESUMO

There are a range of products available which claim to accelerate the healing of burns; these include topical agents, interactive dressings and biomembranes. The aim of this study was to assess the effect of a gelatin-based membrane containing usnic acid/liposomes on the healing of burns in comparison to silver sulfadiazine ointment and duoDerme® dressing, as well as examining its quantification by high performance liquid chromatography. The quantification of the usnic acid/liposomes was examined using high performance liquid chromatography (HPLC) by performing separate in vitro studies of the efficiency of the biomembranes in terms of encapsulation, drug release and transdermal absorption. Then, second-degree 5cm2 burn wounds were created on the dorsum of nine male pigs, assigned into three groups (n=3): SDZ - animals treated with silver sulfadiazine ointment; GDU - animals treated with duoDerme®; UAL - animals treated with a gelatin-based membrane containing usnic acid/liposomes. These groups were treated for 8, 18 and 30days. In the average rate of contraction, there was no difference among the groups (p>0.05). The results of the quantification showed that biomembranes containing usnic acid/liposomes were controlled released systems capable of transdermal absorption by skin layers. A macroscopic assay did not observe any clinical signs of secondary infections. Microscopy after 8days showed hydropic degeneration of the epithelium, with intense neutrophilic infiltration in all three groups. At 18days, although epidermal neo-formation was only partial in all three groups, it was most incipient in the SDZ group. Granulation tissue was more exuberant and cellularized in the UAL and GDU groups. At 30days, observed restricted granulation tissue in the region below the epithelium in the GDU and UAL groups was observed. In the analysis of collagen though picrosirius, the UAL group showed greater collagen density. Therefore, the UAL group displayed development and maturation of granulation tissue and scar repair that was comparable to that produced by duoDerme®, and better than that produced by treatment with sulfadiazine silver ointment In addition, the UAL group showed increased collagen deposition compared to the other two groups.

RESUMO

CONTEXT: Peripheral axon injury and degeneration are often mediated by oxidative stress and inflammation. The hydroalcoholic extract of the red propolis (HERP) has attracted great attention because of its antioxidant and anti-inflammatory activities. OBJECTIVE: The objective of this work is to study the effect of HERP on nerve repair and functional recovery after sciatic nerve injury (SNI) in rats. MATERIALS AND METHODS: The chemical markers in HERP were identified using high-resolution mass spectroscopy. After axonotmesis of sciatic nerve, ibuprofen (IBP) and HERP treatments were orally administered for 28 d. Behavioural tests were performed weekly after SNI. The myelinated axon number was counted using morphometric analysis. RESULTS: The compounds found in HERP were pinocembrin, formononetin, vestitol, and biochanin A. The animals that underwent SNI showed a significant decrease in motor function based on the Basso, Beattie and Bresnahan scale and sciatic functional index compared with sham animals until 7 d after the surgery (p < 0.05). After 14 and 21 d, the SNI groups treated with either HERP or IBP showed significant improvement (p < 0.01), and the SNI group treated with HERP 10 mg/kg showed accelerated motor recovery compared with the other groups (p < 0.01). SNI caused also a reduction in the myelinated axon counts, and treatment with HERP 10 mg/kg induced a significant increase in the number of myelinated fibres compared with all other groups. CONCLUSION: HERP promoted regenerative responses and accelerated functional recovery after sciatic nerve crush. Thus, it can be considered to be a new strategy or complementary therapy for treating nerve injuries.

Introduction: Propolis is a complex resinous substance produced by bees that has therapeutic properties, such as antimicrobial activity, anti-inflammatory, healing. Few studies exist on the red variety of propolis, found in the state of Sergipe. Objective: Evaluation of the antimicrobial action of the extract of propolis red collected in the northeastern state of Sergipe, against strains of Enterococcus faecalis. Material and method: The red propolis samples were collected in Brejo Grande/SE - Brazil and identified according to their sensory characteristics, granulometry and physical chemical requirements. The content of flavonoids in dried extract was determined. Solutions of red propolis (EEP) were prepared at concentrations of 1%; 2.5%; 5% and 7.5%. The bacterial strain used was Enterococcus faecalis - ATCC 29212. The antibacterial activity was verified by in vitro tests (disk diffusion test and determination of minimum bactericidal concentration - CBM) and ex vivo (using human extracted teeth). The test, ex vivo contaminated teeth were divided into three groups with 10 teeth each. Group 1 was treated with propolis to 7.5% (concentration determined in CBM test), the Group 2 was treated as positive control with sodium hypochlorite solution 2.5% and Group 3 was used as a negative control and was treated only with sterile saline. Result: The extract of propolis promoted inhibition zone compared to results from solution of sodium hypochlorite to 2.5%, showing values between 12 and 16 mm. There was no bacterial growth after root conduct irrigation with EEP to 7.5%. Conclusion: Propolis collected showed medium content of flavonoids (1.8%) and physical chemical characteristics consistent to those required by the Brazilian Government. At 7.5% of propolis extract, we observed a higher antibacterial potential than others groups. .

RESUMO

The aim of this study was to assess the effect of oral administration of Hydroalcoholic Extract of Green Propolis (HEGP) on dermal carcinogenesis in rodent model. For the biological assay, we used 36 mice, assigned into 6 groups (n=6): CTR (treated with 100 mg/kg HEGP and no tumor induction), TUM (treated with water and tumor induction), GP10 (treated with 10 mg/kg HEGP and tumor induction), GP50 (treated with 50 mg/kg HEGP and tumor induction) and GP100 (treated with 100 mg/kg HEGP and tumor induction). Cancer induction was performed in the back of the mice by topical application of DMBA. After 16 weeks, mice were euthanized and their backs were submitted to post-mortem histological analysis. The mean number of lesions developed in TUM (4.14±0.89) was significantly higher than in GP10 (2.05±1.02), GP50 (1.8±1.92) and GP100 (2.5±1.73) (p<0.05). The tumors formed in HEGP-treated groups were histologically more differentiated, but only in PV100 in situ lesions were evidenced. Infiltration of anatomical noble structures was less frequent in HEGP-treated groups (p<0.05). Our data suggest that oral administration of HEGP provided partial inhibition of DMBA-induced dermal carcinogenesis, as well as appeared to modulate the differentiation and infiltrative potential of the carcinomas in rodent model.

PURPOSE: To evaluate modulatory effects of a hydroalcoholic extract of Brazilian red propolis (HERP) on dermal carcinogenesis using a murine model. METHODS: The HERP was used at concentrations of 10, 50 and 100 mg/kg (PROP10, PROP50 and PROP100, respectively) to modulate dermal carcinogenesis induced by the application of 9,10-dimetil-1,2-benzatraceno (DMBA) on the backs of animals. RESULTS: The chemical compounds identified in HERP included propyl gallate, catechin, epicatechin and formononetin. PROP100 treatment resulted in significantly decreased tumor multiplicity throughout the five weeks of tumor promotion (p<0.05), and this concentration also resulted in the highest frequency of verrucous tumors (p<0.05). All of the tumors that developed in DMBA-treated animals were regarded as squamous cell carcinomas and were either diagnosed as non-invasive verrucous carcinomas or invasive squamous cell carcinomas (SCCs). The average score for malignancy was significantly lower in the PROP100-treated group than the non-treated group (p<0.05), but there was no difference between the other groups (p>0.05). CONCLUSION: The oral administration of hydroalcoholic extract of Brazilian red propolis at a dose of 100 mg/kg had a significant modulatory effect on the formation, differentiation and progression of chemically induced squamous cell carcinoma in a murine experimental model.

RESUMO

PURPOSE: To evaluate the effects of fatty acids-incorporated collagen-based dressing films on wound healing in rodents. METHODS: Therefore, surgical wounds were performed in the back of 80 Wistar rats, and dressed with collgane-based films (COL), and collagen-based films containing fatty acids (AGEF50 and AGEF100). Undressed wounds were regarded as controls (CTR). The animals were euthanized after three, seven, 14 and 21 days, and the macroscopic wound contraction rates (WRC) were assessed. The wounded area was also analyzed by conventional and polarized light microscope. RESULTS: No sign of abscess or hypertrophic scar formation was observed in none of the groups. At seven days, the WRR of AGEF50 was significantly higher than CTR (p<0.01), whereas at 14 days, both AGE 50 and AGE100 showed a significant increase of the WRR compared to CTR (p<0.001) and COL (p<0.01). Both films promoted increased influx of neutrophils at three days (p<0.01), but reduced significantly the mononuclear infiltrate at 14 days (p<0.05). It was also observed earlier maturation of the granulation tissue, full epithelization and cutaneous appendages development, as well as better collagenization, in AGEF50 and AGEF100. CONCLUSION: The application of AGEF50/100 as wound dressing improved wound healing in rodents.

RESUMO

Achatina fulica's secretion has been recently related to antibacterial, antifungal, and cicatricial properties, and it is influenced by the kind of food offered. Therefore, this study investigated the healing potential of dressing films based on mucous secretion of Achatina fulica. Thus, surgical wounds performed in black wistar rats were dressed with films based on collagen, and on mucous secretion of A. fulica fed with Lactuca sativa; undresses worked as wounds control. After 3, 7, 14 and 21 days the animals were euthanized, and the wounds were microscopically evaluated. On the 3rd and 7th days dressing films based on mucus provided acceleration of the formation maturation of granulation tissue, better epithelization rates, and more rapid replacement of type III for type I collagen fibers. On the 14th and 21st days, these dressings induced more intense deposition and better architectural disposition of type I collagen fibers, and hastened the regeneration of cutaneous phaneros. Dressings obtained from A. fulica fed with Lactuca sativa provided the most expressive results. This study suggests that films produced with mucous secretion of A. fulica can be successfully employed as wound dressing, particularly if snails are fed with Lactuca sativa.

UNLABELLED: Studies have demonstrated that flavonoid compounds of green propolis have antitumoral activity. STUDY DESIGN: Experimental study. AIMS: To evaluate the effect of a hydroalcoholic extract of green propolis (EPV) on chemically induced epithelial dysplasias in rat tongues. METHODS AND MATERIALS: DMBA was brushed on the lingual dorsum of rats 3x/week on alternate days--100 (PROP1), 200 (PROP2) and 300 mg/kg (PROP3) EPV was administered orally for 20 weeks. EPV or DMBA were replaced by their vehicles and applied as positive (TUM1 and TUM2) and negative controls (CTR1 and CTR2), respectively. The lingual epithelium was histologically analyzed and graded according a binary system and the WHO classification; the data were compared using ANOVA (*p<0.05). RESULTS: The EPV yield was 41% and the flavonoid yield was 0.95±0.44%. According to the Binary System, TUM1, TUM2 and PROP1 were considered high risk lesions, with significantly higher morphological alteration rates compared to the other groups (p<0.05), which were considered low risk lesions. Based on the WHO classification, moderate dysplasia was TUM1 and TUM2, mild dysplasia was PROP1, PROP2 and PROP3, and non-dysplastic epithelium was CTR1 and CTR2. CONCLUSION: EPV seems to play an important protective role against chemically-induced lingual carcinogenesis in rats.
