RESUMO

Control algorithms have been proposed based on knowledge related to nature-inspired mechanisms, including those based on the behavior of living beings. This paper presents a review focused on major breakthroughs carried out in the scope of applied control inspired by the gravitational attraction between bodies. A control approach focused on Artificial Potential Fields was identified, as well as four optimization metaheuristics: Gravitational Search Algorithm, Black-Hole algorithm, Multi-Verse Optimizer, and Galactic Swarm Optimization. A thorough analysis of ninety-one relevant papers was carried out to highlight their performance and to identify the gravitational and attraction foundations, as well as the universe laws supporting them. Included are their standard formulations, as well as their improved, modified, hybrid, cascade, fuzzy, chaotic and adaptive versions. Moreover, this review also deeply delves into the impact of universe-inspired algorithms on control problems of dynamic systems, providing an extensive list of control-related applications, and their inherent advantages and limitations. Strong evidence suggests that gravitation-inspired and black-hole dynamic-driven algorithms can outperform other well-known algorithms in control engineering, even though they have not been designed according to realistic astrophysical phenomena and formulated according to astrophysics laws. Even so, they support future research directions towards the development of high-sophisticated control laws inspired by Newtonian/Einsteinian physics, such that effective control-astrophysics bridges can be established and applied in a wide range of applications.

INTRODUCTION: Artificial intelligence (AI) shows immense potential in medicine and Chat generative pretrained transformer (ChatGPT) has been used for different purposes in the field. However, it may not match the complexity and nuance of certain medical scenarios. This study evaluates the accuracy of ChatGPT 3.5 and 4 in providing recommendations regarding the management of postprostatectomy urinary incontinence (PPUI), considering The Incontinence After Prostate Treatment: AUA/SUFU Guideline as the best practice benchmark. MATERIALS AND METHODS: A set of questions based on the AUA/SUFU Guideline was prepared. Queries included 10 conceptual questions and 10 case-based questions. All questions were open and entered into the ChatGPT with a recommendation to limit the answer to 200 words, for greater objectivity. Responses were graded as correct (1 point); partially correct (0.5 point), or incorrect (0 point). Performances of versions 3.5 and 4 of ChatGPT were analyzed overall and separately for the conceptual and the case-based questions. RESULTS: ChatGPT 3.5 scored 11.5 out of 20 points (57.5% accuracy), while ChatGPT 4 scored 18 (90.0%; p = 0.031). In the conceptual questions, ChatGPT 3.5 provided accurate answers to six questions along with one partially correct response and three incorrect answers, with a final score of 6.5. In contrast, ChatGPT 4 provided correct answers to eight questions and partially correct answers to two questions, scoring 9.0. In the case-based questions, ChatGPT 3.5 scored 5.0, while ChatGPT 4 scored 9.0. The domains where ChatGPT performed worst were evaluation, treatment options, surgical complications, and special situations. CONCLUSION: ChatGPT 4 demonstrated superior performance compared to ChatGPT 3.5 in providing recommendations for the management of PPUI, using the AUA/SUFU Guideline as a benchmark. Continuous monitoring is essential for evaluating the development and precision of AI-generated medical information.

The antidiabetic drug metformin and antiepileptic drug lamotrigine are contaminants of emerging concern that have been detected in biowaste-derived amendments and in the environment, and their fate must be carefully studied. This work aimed to evaluate their sorption behaviour on soil upon digestate application. Experiments were conducted on soil and digestate-amended soil as a function of time to study kinetic processes, and at equilibrium also regarding the influence of trace metals (Pb, Ni, Cr, Co, Cu, Zn) at ratio pharmaceutical/metal 1/1, 1/10, and 1/100. Pharmaceutical desorption experiments were also conducted to assess their potential mobility to groundwater. Results revealed that digestate amendment increased metformin and lamotrigine adsorbed amounts by 210% and 240%, respectively, increasing organic matter content. Metformin adsorption kinetics were best described by Langmuir model and those of lamotrigine by Elovich and intraparticle diffusion models. Trace metals did not significantly affect the adsorption of metformin in amended soil while significantly decreased that of lamotrigine by 12-39%, with exception for Cu2+ that increased both pharmaceuticals adsorbed amounts by 5 - 8%. This study highlighted the influence of digestate amendment on pharmaceutical adsorption and fate in soil, which must be considered in the circular economy scenario of waste-to-resource.

General relativity minimally coupled to a massive, free, complex scalar field, is shown to allow asymptotically flat solutions, nonsingular on and outside the event horizon, describing two spinning black holes (2sBHs) in equilibrium, with coaxial, aligned angular momenta. The 2sBHs configurations bifurcate from solutions describing dipolar spinning boson stars. The BHs emerge at equilibrium points diagnosed by a test particle analysis and illustrated by a Newtonian analog. The individual BH "charges" are mass and angular momentum only. Equilibrium is due to the scalar environment, acting as a (compact) dipolar field, providing a lift against their mutual attraction, making the 2sBHs (h)airborne. We explore the 2sBHs domain of solutions and its main features.

ABSTRACT Purpose: Sling as a therapeutic option for male stress urinary incontinence (SUI) has been reviewed in the last two decades, as it is a relatively simpliest surgery compared to artificial urinary sphincter and has the ability to modulate urethral compression. This study aims to evaluate the efficacy, rate of complications, quality of life and the effects on bladder emptying of the Argus T® compressive and ajustable sling in moderate and severe male SUI treatment. Materials and Methods: Men eligible for stress urinary incontinence treatment after radical prostatectomy were recruited and prospectively evaluated, from March 2010 to November 2016. It was selected outpatient men with moderate and severe SUI, after 12 months of radical prostatectomy, who have failed conservative treatment. All patients had a complete clinical and urodynamic pre and post treatment evaluation, by means of clinical history, physical examination, urine culture, 1-hour pad test and ICIq-SF questionnaire. The UDS was performed after 12, 18 and 24 months postoperatively. Results: Thirty-seven men underwent sling surgery, 19 patients (51.4%) with moderate and 18 (48.6%) with severe SUI. The minimum follow-up time was 5 years. Overall, we had a success rate of 56.7% at 60 months follow-up. After surgery, we did not observe significant changes in the urodynamic parameters evaluated during the follow-up. No patient had urodynamic bladder outlet obstruction (BOO) after sling implantation. Readjustment of the Argus T® sling was performed in 16 (41%) of the patients and 51% of the patients reported some adverse event. Conclusion: We demonstrate a long-term efficacy and safety of Sling Argus T® as an alternative to moderate and severe male SUI treatment. Furthermore, in our study bulbar urethra compression does not lead to bladder outlet obstruction.

The Feline Leukemia Virus Subgroup C Receptor 1a (FLVCR1a) is a transmembrane heme exporter essential for embryonic vascular development. However, the exact role of FLVCR1a during blood vessel development remains largely undefined. Here, we show that FLVCR1a is highly expressed in angiogenic endothelial cells (ECs) compared to quiescent ECs. Consistently, ECs lacking FLVCR1a give rise to structurally and functionally abnormal vascular networks in multiple models of developmental and pathologic angiogenesis. Firstly, zebrafish embryos without FLVCR1a displayed defective intersegmental vessels formation. Furthermore, endothelial-specific Flvcr1a targeting in mice led to a reduced radial expansion of the retinal vasculature associated to decreased EC proliferation. Moreover, Flvcr1a null retinas showed defective vascular organization and loose attachment of pericytes. Finally, adult neo-angiogenesis is severely affected in murine models of tumor angiogenesis. Tumor blood vessels lacking Flvcr1a were disorganized and dysfunctional. Collectively, our results demonstrate the critical role of FLVCR1a as a regulator of developmental and pathological angiogenesis identifying FLVCR1a as a potential therapeutic target in human diseases characterized by aberrant neovascularization.

Elucidating cellulose-lignin interactions at the molecular and nanometric scales is an important research topic with impacts on several pathways of biomass valorization. Here, the interaction forces between a cellulosic substrate and lignin are investigated. Atomic force microscopy with lignin-coated tips is employed to probe the site-specific adhesion to a cellulose film in liquid water. Over seven thousand force-curves are analyzed by a machine-learning approach to cluster the experimental data into types of cellulose-tip interactions. The molecular mechanisms for distinct types of cellulose-lignin interactions are revealed by molecular dynamics simulations of lignin globules interacting with different cellulose Iß crystal facets. This unique combination of experimental force-curves, data-driven analysis, and molecular simulations opens a new approach of investigation and updates the understanding of cellulose-lignin interactions at the nanoscale.

In this paper, we present the Type I multivariate zero-inflated Conway-Maxwell-Poisson distribution, whose development is based on the extension of the Type I multivariate zero-inflated Poisson distribution. We developed important properties of the distribution and present a regression model. The AIC and BIC criteria are used to select the best fitted model. Two real data sets have been used to illustrate the proposed model. Moreover, we conclude by stating that the Type I multivariate zero-inflated Conway-Maxwell-Poisson distribution produces a better fitted model for multivariate count data with excess of zeros.

The oxidation process of 2,6-dimethoxyphenol (2,6-DMP) by laccase from Botryosphaeria rhodina MAMB-05 and the corresponding enzyme-mediator systems was studied using cyclic voltammetry (CV). The enzyme was classified as a high oxidation potential laccase (> 0.70) V vs. NHE) based on its Redox potential at different pHs. The cyclic voltammograms for 2,6-DMP (- 58.7 mV pH-1) showed that its oxidation potential decreased more significantly compared to the enzyme (- 50.2 mV pH-1) by varying the pH. The 2,2'-azino-bis[3-ethyl-benzothiazoline-6-sulfonic acid] diammonium salt (ABTS) and 2,2,6,6-tetramethylpiperidine 1-oxyl radical (TEMPO) mediators were effectively oxidized by laccase from B. rhodina MAMB-05. The influence of laccase on the comproportionation of ABTS and the ionic step of the oxidation of TEMPO was also studied using CV. A higher potential difference was observed between laccase and the substrate, and correlated with higher enzyme activity. For the laccase-mediator systems, there was no clear correlation of potential difference between laccase and mediators with enzyme activity towards 2,6-DMP. This observation suggests that there are other limiting parameters for enzyme activity despite Redox potential difference, especially during ionic steps of the mechanism.

Fatal infections with Cytauxzoon felis are common in domestic cats, and jaguars (Panthera onca) are believed carriers of this protozoan. Fatal C. felis infections have never been described in jaguars before. Thus, this study describes such an infection in a 5-month-old captive-reared jaguar that presented hyporexia and died at 26 days after the first clinical signs. During necropsy, samples were taken from several tissues, some were fixed in 10% formalin and process for histopathological diagnosis, and some samples were used for DNA extraction, amplification via PCR and sequencing of the potential pathogens. Grossly, marked splenomegaly and icterus were observed. Histologically, numerous structures that are morphologically compatible with Cytauxzoon spp. schizonts obliterated multiple blood vessels in the brain, leptomeninges, spinal cord, lungs, heart, skeletal muscle, adrenal gland, kidneys, spleen, small intestine and pancreas. C. felis was identified by PCR in many organs. Thus, C. felis infection in jaguars can be fatal, and the clinicopathological findings are similar to those of cytauxzoonosis in other wild and domestic felid species.

Since the discovery of polydopamine (PDA), there has been a lot of progress on using this substance to functionalize many different surfaces. However, little attention has been given to prepare functionalized surfaces for the preparation of flexible electrochemical paper-based devices. After fabricating the electrodes on paper substrates, we formed PDA on the surface of the working electrode using a chemical polymerization route. PDA nanofilms on carbon were characterized by contact angle (CA) experiments, X-ray photoelectron spectroscopy, scanning electron microscopy, atomic force microscopy (topography and electrical measurements) and electrochemical techniques. We observed that PDA introduces chemical functionalities (RNH2 and RC═O) that decrease the CA of the electrode. Moreover, PDA nanofilms did not block the heterogeneous electron transfer. In fact, we observed one of the highest standard heterogeneous rate constants (ks ) for electrochemical paper-based electrodes (2.5 ± 0.1) × 10-3  cm s-1 , which is an essential parameter to obtain larger currents. In addition, our results suggest that carbonyl functionalities are ascribed for the redox activity of the nanofilms. As a proof-of-concept, the electrooxidation of nicotinamide adenine dinucleotide showed remarkable features, such as, lower oxidation potential, electrocatalytic peak currents more than 30 times higher when compared to unmodified paper-based electrodes and electrocatalytic rate constant (kobs ) of (8.2 ± 0.6) × 102  L mol-1  s-1 .

It was recently shown that a scalar field suitably coupled to the Gauss-Bonnet invariant G can undergo a spin-induced linear tachyonic instability near a Kerr black hole. This instability appears only once the dimensionless spin j is sufficiently large, that is, j≳0.5. A tachyonic instability is the hallmark of spontaneous scalarization. Focusing, for illustrative purposes, on a class of theories that do exhibit this instability, we show that stationary, rotating black hole solutions do indeed have scalar hair once the spin-induced instability threshold is exceeded, while black holes that lie below the threshold are described by the Kerr solution. Our results provide strong support for spin-induced black hole scalarization.

A photoelectrochemical biosensing strategy for the highly sensitive detection of the flavonoid rutin was developed by synergizing the photoelectrocatalytic properties of hematite (α-Fe2O3) decorated with palladium nanoparticles (PdNPs) and the biocatalysis towards laccase-based reactions. The integration of α-Fe2O3.PdNPs with a polyphenol oxidase as a biorecognition element yields a novel biosensing platform. Under visible light irradiation, the photoactive biocomposite can generate a stable photocurrent, which was found to be directly dependent upon the concentration of rutin. Under the optimal experimental conditions, the cathodic photocurrent, measured at 0.33 V vs. Ag/AgCl, from the square-wave voltammograms presented a linear dependence on the rutin concentration within the range of 0.008-30.0 × 10-8 mol L-1 (sensitivity: 1.7 µA·(× 10-8 M-1)·cm-2), with an experimental detection limit (S/N = 3) of 8.4 × 10-11 mol L-1. The proposed biosensor device presented good selectivity towards rutin in the presence of various organic compounds and inorganic ions, demonstrating the potential application of this biosensing platform in complex matrices. This bioanalytical device also exhibited excellent operational and analytical properties, such as intra-day (standard deviation, SD = 0.21%) and inter-day (SD = 1.30%) repeatability, and long storage stability (SD = 2.80% over 30 days).Graphical abstract.

BACKGROUND AND OBJECTIVE: Rectal cancer is often surgically treated, but it is still associated with morbidity rates. Minimally invasive techniques are increasingly being used to reduce complications, and the use of such techniques can be found in the literature. This study aims to report our experience in a reference oncology center. METHODS: A retrospective cohort study was performed on a prospective database of patients who underwent robotic surgery for rectal cancer using the single-docking technique from September 2014 to April 2018. Clinical and surgical variables, as well as morbidity and mortality rates, were analyzed. RESULTS: One hundred and two patients underwent robotic surgery. Intraoperative complications occurred in six patients (4.9%), and postoperative complications in 24 patients (23.5%), of which anastomotic fistula represented 3.9%. The conversion rate was 1.96%. Two cases (1.9%) faced death within 30 days. The median length of hospitalization was 3 days. The median number of lymph nodes dissected was 15. Clinical and surgical data were correlated with postoperative complications, and no statistically significant differences were found. CONCLUSION: Robotic surgery is a safe and feasible approach to manage rectal cancer. The method presents satisfactory results with regard to the rate of operative complications, conversion rate, oncologic outcomes, and length of hospitalization.

Cell-penetrating peptides (CPPs) are a topical subject potentially exploitable for creating nanotherapeutics for the delivery of bioactive loads. These compounds are often classified into three major categories according to their physicochemical characteristics: cationic, amphiphilic, and hydrophobic. Among them, the group of hydrophobic CPPs has received increasing attention in recent years due to toxicity concerns posed by highly cationic CPPs. The hexapeptide PFVYLI (P, proline; F, phenylalanine; V, valine; Y, tyrosine; L, leucine; and I, isoleucine), a fragment derived from the C-terminal portion of α1-antitrypsin, is a prototypal example of hydrophobic CPP. This sequence shows reduced cytotoxicity and a capacity of nuclear localization, and its small size readily hints at its suitability as a building block to construct nanostructured materials. In this study, we examine the self-assembling properties of PFVYLI and investigate its ability to form noncovalent complexes with nucleic acids. By using a combination of biophysical tools including synchrotron small-angle X-ray scattering and atomic force microscopy-based infrared spectroscopy, we discovered that this CPP self-assembles into discrete nanofibrils with remarkable amyloidogenic features. Over the course of days, these fibrils coalesce into rodlike crystals that easily reach the micrometer range. Despite lacking cationic residues in the composition, PFVYLI forms noncovalent complexes with nucleic acids that retain ß-sheet pairing found in amyloid aggregates. In vitro vectorization experiments performed with double-stranded DNA fragments indicate that complexes promote the internalization of nucleic acids, revealing that tropism toward cell membranes is preserved upon complexation. On the other hand, transfection assays with splice-correction oligonucleotides (SCOs) for luciferase expression show limited bioactivity across a narrow concentration window, suggesting that the propensity to form amyloidogenic aggregates may trigger endosomal entrapment. We anticipate that the findings presented here open perspectives for using this archetypical hydrophobic CPP in the fabrication of nanostructured scaffolds, which potentially integrate properties of amyloids and translocation capabilities of CPPs.

This research was carried out to evaluate the effect of mycotoxins on the performance of horses through physiological parameters, and hematology and serum biochemistry analyses. The essay lasted 40 days, with 12 days for adaptation and 28 days of experimentation. In the experimental stage, the horses were distributed in a completely randomized design, with three treatments with four animals each. The treatments used were 0 (control), 50 ppb and 100 ppb of Aflatoxin B1 (AFB1) added to a concentrate in a basal diet. The basal diet contained mycotoxins from feedstuffs naturally contaminated. The exercise test was performed over the 21th day of the experimental stage. The exercise consisted in an interval training test with a warm-up of 17 mins at a trot followed by three gallops of 450m/min. The heart rate was monitored between the gallops. Before the exercise test and immediately after the third gallop, the physiological and blood parameters were evaluated, and continued up to 48 hours after the exercise. The results of the physiological, hematological and biochemical parameters were submitted to analysis of variance (ANOVA) and compared by the Tukey test at 5% of significance. The presence of AFB1 in the diet influenced the alkaline phosphatase activity, which presented higher values in horses fed diet with inclusion of 100 ppb AFB1, suggesting a hepatotoxic activity associated with the others mycotoxins naturally present in the feedstuffs.(AU)


Esta pesquisa foi conduzida para avaliar o efeito de micotoxinas no desempenho de equinos com avaliações fisiológicas e análises hematológicas e da bioquímica sérica. O ensaio durou 40 dias, com 12 dias de adaptação e 28 dias de experimentação. Na fase experimental, os equinos foram distribuídos em delineamento inteiramente casualizado em três tratamentos, com quatro animais cada. Os tratamentos utilizados foram 0 (controle), 50 ppb e 100 ppb de Aflatoxina B1 (AFB1) adicionada ao concentrado de uma dieta basal. A dieta basal continha alimentos naturalmente contaminados por micotoxinas. O teste de desempenho foi executado no 21º dia da fase experimental por meio de teste intervalado consistindo em aquecimento ao trote por 17 minutos, seguido de três galopes de 450m/min. A frequência cardíaca (FC) foi monitorada entre os galopes. Antes do exercício e imediatamente após o terceiro galope, os parâmetros fisiológicos e sanguíneos foram avaliados e continuaram sendo monitorados até 48 horas após o exercício. Os resultados dos parâmetros fisiológicos, hematológicos e bioquímicos foram submetidos à análise de variância (ANOVA) e comparados pelo teste de Tukey a 5% de significância. A presença de AFB1 na dieta influenciou a atividade da fosfatase alcalina, que apresentou valores mais elevadas na dieta com inclusão de 100 ppb de AFB1, sugerindo uma atividade hepatotóxica associada às outras micotoxinas naturalmente presentes nos alimentos.(AU)

The pursuit of biocompatible, breathable and skin-conformable wearable sensors has predominantly focused on synthetic stretchable hydrophobic polymers. Microbial nanocellulose (MNC) is an exceptional skin-substitute natural polymer routinely used for wound dressing and offers unprecedented potential as substrate for wearable sensors. A versatile strategy for engineering wearable sensing platforms is reported, with sensing units made of screen-printed carbon electrodes (SPCEs) on MNC. As-prepared SPCEs were used to detect the toxic metals cadmium (Cd2+) and lead (Pb2+) with limits of detection of 1.01 and 0.43 µM, respectively, which are sufficient to detect these metal ions in human sweat and urine. SPCEs functionalized through anodic pre-treatments were used for detecting uric acid and 17ß-estradiol in artificial sweat, with detection limits of 1.8 µM and 0.58 µM, respectively. The electrochemical treatment created oxygen groups on the carbon surfaces, thus improving wettability and hydrophilicity. MNC was herein exploited as an adhesive-free, yet highly skin-adherent platform for wearable sensing devices that also benefit from the semi-permeable, non-allergenic, and renewable features that make MNC unique within the pool of materials that have been used for such a purpose. Our findings have clear implications for the developments on greener and more biocompatible but still efficient substrates and may pave the route for combining immunosensing devices with drug delivery therapies.

In this era of precision medicine, there is an increasingly urgent need for highly sensitive tests for detecting tumors such as colon cancer (CC), a silent disease where the first symptoms may take 10-15 years to appear. Mass spectrometry-based lipidomics is an emerging tool for such clinical diagnosis. We used ultra-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry operating in high energy collision spectral acquisition mode (MSE) mode (UPLC-QTOF-MSE) and gas chromatography (GC) to investigate differences between the plasmatic lipidic composition of CC patients and control (CTR) subjects. Key enzymes in lipidic metabolism were investigated using immuno-based detection assays. Our partial least squares discriminant analysis (PLS-DA) resulted in a suitable discrimination between CTR and CC plasma samples. Forty-two statistically significant discriminating lipids were putatively identified. Ether lipids showed a prominent presence and accordingly, a decrease in glyceronephosphate O-acyltransferase (GNPAT) enzyme activity was found. A receiver operating characteristic (ROC) curve built for three plasmalogens of phosphatidylserine (PS), named PS(P-36:1), PS(P-38:3) and PS(P-40:5), presented an area under the curve (AUC) of 0.998, and sensitivity and specificity of 100 and 85.7% respectively. These results show significant differences in CC patients' plasma lipid composition that may be useful in discriminating them from CTR individuals with a special role for plasmalogens.

The ringdown and shadow of the astrophysically significant Kerr black hole (BH) are both intimately connected to a special set of bound null orbits known as light rings (LRs). Does it hold that a generic equilibrium BH must possess such orbits? In this Letter we prove the following theorem. A stationary, axisymmetric, asymptotically flat black hole spacetime in 1+3 dimensions, with a nonextremal, topologically spherical, Killing horizon admits, at least, one standard LR outside the horizon for each rotation sense. The proof relies on a topological argument and assumes C^{2} smoothness and circularity, but makes no use of the field equations. The argument is also adapted to recover a previous theorem establishing that a horizonless ultracompact object must admit an even number of nondegenerate LRs, one of which is stable.
