In this study, we undertook the genomic characterization of 54 pseudorabies virus (PRV) strains isolated in Italy during 1984-2010. The characterization was based on partial sequencing of the UL44 (gC) and US8 (gE) genes; 44 strains (38 for gene gE and 36 for gC) were isolated on pig farms; 9 originated from dogs and 1 from cattle. These porcine PRV strains, which were closely related to those isolated in Europe and America in the last 20 years, and the bovine strain bovine/It/2441/1992 belong to cluster B in both phylogenetic trees. Six porcine strains that do not belong to cluster B are related in both gE and gC phylogenetic trees to the 'old' porcine PRV strains isolated in the 1970s and 1980s. In the last two decades, the presence of these strains in domestic pig populations has been reduced drastically, whereas they are prevalent in wild boar. The two remaining strains have an interesting genomic profile, characterized by the gC gene being closely related to the old porcine PRV strains, and the gE gene being similar to that of recently isolated strains. Three strains originating from working dogs on pig farms are located in cluster B in both phylogenetic trees. Five strains isolated from hunting dogs have a high degree of correlation with PRV strains circulating in wild boar. The last isolate has a gC gene similar to that in the two porcine strains mentioned previously, and the gE gene is correlated with the strains isolated from hunting dogs. These results provide interesting insight into the genomic characterization of PRV strains and reveal a clear differentiation between the strains isolated from hunting dogs that are related to the wild boar strains and those originating from domestic pigs.

Among the several fish species comercially exploited at the Pantanal of Mato Grosso do Sul, the "pacu" (Piaractus mesopotamicus) stands as one of the most important. Information regarding its exploitation level is necessary for the proper management of its stocks. Between 1996 and 1997 data on total length of the pacu were collected on a monthly basis from specimens caught by professional and sport fishers in the municipality of Corumbá. These data were used to estimate growth parameters and to assess the exploitation level for this species, applying the Beverton and Holt yield per recruit model. Length frequency analysis, carried out with the software FISAT (ELEFAN), was used to estimate growth parameters: 1996: L(infinity) = 87.20 cm; K = 0.34 year(-1); phi(')=3.41; C = 0.74; WP = 0.81; Longevity = 8.40 years; and 1997: L(infinity) = 86.50 cm; K = 0.34 year(-1); phi(')=3.40; C = 0.60; WP = 0.80; Longevity = 8.40 years. The value for t(0) is -0.363 years for mean values of L(infinity) and k. The weight-length relationship, calculated from data derived from experimental fisheries carried out in 1999 and 2000, is described by the equation: W = 0.048LF(2.835). Estimated mortalities and survival rates were: 1996: Z = 1.51 year(-1); M = 0.62 year(-1); F = 0.89 year(-1); S = 21.9%; and 1997: Z = 1.65 year(-1); M = 0.63 year(-1); F = 1.02 year(-1); S = 19.1%. The yield per recruit analysis showed the following values: F(Present) = 0.96 year(-1); F(max) = 0.67 year(-1) ; F(0.1) = 0.51 year(-1) (for L(c) = 26.7 cm). These results suggest that the pacu is overexploited in the area, so that restrictive measures are in need to manage the pacu fisheries.

Growth parameters were estimated for Moenkhausia dichroura (Kner, 1858) (Characiformes, Characidae), a small-sized and very abundant fish of the Pantanal lentic habitats commonly known as "pequira ". A method based on the length frequencies distribution and the ELEFAN I routine from the FISAT program were used. The fish were collected in the Baia da Onça, an oxbowlake of the sub-region Pantanal of Aquidauana, Mato Grosso do Sul, Brazil, from June to December 1988. The standard length of the captured fishes ranged from 29 to 76 mm with an average of 53 mm. The estimated growth parameters were L(infinity) = 81 mm (standard length), k = 0.85 year(-1), C = 0.89, WP = 0.6 (Rn = 0.285). The WP indicated that growth reduction occurred in July, when the lowest temperature of the year was registered. The growth curve showed that captured individuals belonged to three cohorts. The obtained results seem robust and quite compatible with the biology of the fish and its adjustment to the environment. M. dichroura, in spite of not being a direct fishing interest, is an important species in terms of its ecological aspects, due to its abundance and high growth rate, and as a great food source for aquatic organisms and specially for larger fish of economic value. Considering the information gap about small fish, the parameters estimated for pequira constitute a comparison base for other growth studies of small-sized fish species of tropical environments.

O pacu (Piaractus mesopotamicus) é uma das espécies mais capturadas no Pantanal de Mato Grosso do Sul, e para manejá-lo adequadamente são necessárias informações a respeito do seu nível de explotação. Nos anos de 1996 e 1997 foram coletadas amostras mensais de comprimentos de pacu capturados por pescadores profissionais e esportivos desembarcados em Corumbá, Pantanal de Mato Grosso do Sul (MS), com o objetivo de estimar os parâmetros de crescimento e o seu nível de explotação. Para a determinação do nível de explotação foi utilizado o modelo de rendimento por recruta de Beverton e Holt. Através da análise de freqüência de comprimento, pelo pacote FISAT (ELEFAN), foram estimados os parâmetros de crescimento, com os seguintes valores: 1996: L¥ = 87,20 cm; K = 0,34 ano-1; fi´= 3,41; C = 0,74; WP = 0,81; Longevidade = 8,40 anos; e 1997: L¥ = 86,50 cm; K = 0,34 ano-1; fi´= 3,40; C = 0,60; WP = 0,80; Longevidade = 8,40 anos. O valor de t0 é -0,363 para os valores médios de L¥ e k. A relação peso-comprimento foi determinada através de dados de pescarias experimentais em 1999 e 2000, e foi a seguinte: W = 0,048 LF2.835. As taxas de mortalidade estimadas e a taxa de sobrevivência foram: 1996: Z = 1,51 ano-1; M = 0,62 ano-1; F = 0,89 ano-1; S = 21,9 por cento; e 1997: Z = 1,65 ano-1; M = 0,63 ano-1; F = 1,02 ano-1; S = 19,1 por cento. A análise de rendimento por recruta apresentou os seguintes resultados: Fatual = 0,96 ano-1; Fmáx= 0,67 ano-1 ; F0,1 = 0,51 ano-1 (quando Lc = 26,7 cm). Este resultado indica que o pacu está sobreexplotado, e assim há necessidade de se estabelecer medidas restritivas para sua pescaria.

Neste estudo foram estimados os parâmetros de crescimento para Moenkhausia dichroura (Kner, 1858) (Characiformes, Characidae), um peixe de pequeno porte e muito abundante nos ambientes lênticos do Pantanal, onde é conhecido, vulgarmente, como "pequira". Para a estimativa dos parâmetros, foi utilizada uma metodologia baseada na distribuição mensal das freqüências de comprimento por meio da rotina ELEFAN I do programa FISAT. Os peixes foram coletados na Baía da Onça, uma lagoa do Pantanal do rio Aquidauana, Mato Grosso do Sul - Brasil, entre junho e dezembro de 1988. O comprimento dos exemplares capturados variou de 29 a 76 mm com média igual a 53 mm. Os melhores ajustes obtidos para os parâmetros de crescimento foram L¥ = 81 mm (comprimento padrão), k = 0.85 ano-1, C = 0.89 e WP = 0.6 (Rn = 0.285). O valor de WP indicou que a redução do crescimento ocorreu em julho, quando foram registradas as menores temperaturas do ano. A curva de crescimento obtida indicou que foram capturados indivíduos pertencentes a três cortes. Os resultados obtidos parecem robustos e bastante compatíveis com a biologia do peixe e seu ajuste ao ambiente. M. dichroura, apesar de não ser de interesse direto para a pesca, é uma espécie importante sob o ponto de vista ecológico, em função de sua abundância e elevada taxa de crescimento corporal, constituindo uma grande fonte de alimento para os demais organismos aquáticos e, sobretudo, para os peixes maiores de valor econômico. Em função da carência de informações, os parâmetros aqui estimados para a pequira constituem uma base de comparação para outros estudos sobre o crescimento de espécies de peixes de pequeno porte nos ambientes tropicais.

Prematurely born infants who required assisted ventilation may develop chronic lung disease or bronchopulmonary dysplasia (BPD). The cells involved in the reparative process of the premature lung are not well defined. The repair of injured tissues is a highly standardized process and the most important cells are activated (modulated) fibroblasts (myofibroblasts). A key cytokine in controlling repair is transforming growth factor-beta (TGF-beta). To characterize the cells involved in the repair process of the premature lung, we employed immunocytochemical techniques and examined the lungs of 39 autopsied premature babies who had neonatal respiratory distress syndrome (RDS). All were treated in neonatal intensive care units and required mechanical ventilation and supplemental oxygen; all survived for at least 12 hours. Antibodies were employed against vimentin, alpha-smooth muscle (alpha-SM) actin, total muscle actin, desmin, MAC387, and TGF-beta. Our study indicates that myofibroblasts are normally present along terminal airways in the developing lung. These cells increase in number some days after lung injury, form bundles of cells encircling terminal air spaces, and acquire desmin contractile filaments shortly thereafter. Myofibroblasts do not lose their contractile filaments with time, suggesting a conversion to smooth muscle metaplasia. The proliferation and migration of such myofibroblasts at sites of lung injury is associated with the presence of TGF-beta. These findings suggest that myofibroblasts play an important role in premature lung repair. They may point the way to experimental and clinical trials that will identify drugs antagonistic to TGF-beta (or other cytokines). Such antagonists may protect the neonates who are at high risk of developing BPD.

The present study examines the histological features of the lungs of neonates who died of respiratory distress syndrome or related complications after surfactant therapy. Our aim was to determine whether these lungs showed any unusual histological findings. Complete autopsies were performed 6-12 h after death in 10 surfactant-treated preterm infants and in 30 infants who died before surfactant therapy was available. Representative paraffin sections of all pulmonary lobes, stained with haematoxylin and eosin, were examined microscopically. A few selected slides were also stained with periodic acid-Schiff, Vierhoff-van Gieson, and Mallory trichrome. Hyaline membrane disease and bronchopulmonary dysplasia were present in each group, although there was an increased incidence of intra-alveolar haemorrhage in surfactant-treated babies (in 8 of 10 surfactant-treated as compared with 7 of 30 untreated babies). Amongst those treated with surfactant, we observed the persistence of acute alveolar damage with unresolved hyaline membrane disease in 5 infants who died at the ages of 5, 6, 10, 12, and 13 days, respectively, and histological evidence of pneumocyte type 2 hyperplasia and dysplasia in 2 infants who died at 22 and 41 days of age, respectively. These observations reveal that surfactant-treated infants who fail to respond to therapy have continuing alveolar injury and an increased incidence of intra-alveolar haemorrhage. Since oxygen radicals can induce pneumocyte damage and necrosis and since free radicals provoke alveolar haemorrhage in animal models, we propose that the lesions we observed may stem from a lack, in some preterm babies, of specific mechanisms that detoxify oxygen radicals.

Proliferative vitreo-retinopathy (PVR) and subretinal membrane proliferation are the most common complication and cause of failure in retinal-detachment (RD) surgery. In this study, material withdrawn from 21 patients was observed. The vitreal taps of 16 bulbs affected by PVR and which had undergone vitrectomy, along with 5 bulbs obtained by enucleation, were stained with Hematoxylin Eosin and studied immunohistochemically. The cells involved in this proliferative tissue include macrophages, cellular elements of pigmented epithelium origin, fibroblasts and myofibroblasts. From the examination of enucleated bulbs, we can easily recognize that the cellular components of the membrane are represented by fibroblasts, capillaries, and occasional macrophages; meanwhile, PE cells remain at the base of the newly formed tissue.

Following retinal detachment, subretinal fluid (SRF) fills the neoformed space. Subsequently subretinal and preretinal strands of proliferative tissue begin to form. We have collected the subretinal fluid withdrawn during retinal detachment surgery. We have studied subretinal fluid cytologically to evaluate the number and the type of cells present in the fluid, and by means of transmission electron microscopy. The first cell type to be present in the fluid represented degenerated aspects of pigmented epithelial cells (PECs). Successively, other cell types appeared in the fluid as nerve cells (rods, cones and glial cells), macrophages and well preserved pigmented epithelial cells.
