RESUMO

Herein, we report the simple and inexpensive approach for the large-scale fabrication of uniform bottom-up Surface Enhanced Raman Spectroscopy (SERS) substrate. SERS substrate was fabricated by controlled sputtering of 10 nm thick gold film on self-assembled silica nanoparticles (SiNPs) of ~120 nm on glass substrates. The SERS detection has been firstly demonstrated using Rhodamine B as a Raman probe molecule with a detection limit of 10-10 M on Au sputtered SiNPs (i.e., Au@SiNPs). The experimental Raman enhancement from 0 to 6 was achieved on Au@SiNPs due to the generation of multiple SERS hotspot. To combat blood serum fouling, the zwitterionic modification of l-cysteine was done on Au@SiNPs substrates which lowered blood serum fouling by 48%. Our SERS-based sensor demonstrated high reproducibility for the detection of Doxorubicin in undiluted blood serum with a limit of detection of 20 nM, which greatly exceeded the detection range of available methodologies. We envision that the translation of this SERS substrate for the detection of chemo-drugs like Doxorubicin will assist clinicians in making rapid and/or early decisions in patients undergoing sustained chemotherapy to lower its side-effects or to incorporate other treatment methodologies as an option for Personalized treatment.

Laser-induced fluorescence (LIF) spectra were obtained from Maradol papaya fruits at harvested mature, harvested immature, different ripeness stages, and during the ripening. The chlorophyll fluorescence of papaya fruits showed two maxima; one in the red at 680-690 nm (F690), and the other in far-red region at 730-740 nm (F740). The fruits that were harvested immature showed a definite increase in fluorescence intensity at both maxima within the first six days. The fluorescence emission spectra and fluorescence ratios F690/F740 were analyzed. Results showed that intensity and spectral shape are characteristics of different ripeness stages and during the ripening. The values obtained from F690/F740 showed a direct relation with the fruit harvested mature, harvested immature, and fruit in the ripening process. These results demonstrated that LIF is a useful tool for nondestructive monitoring of the changes in chlorophyll content and photosynthetic activity caused by the different ripeness stages and during the ripening of papaya fruits.

Halophilic archaea are interesting microorganisms that produce low biomass and metabolites, complicating their quantification. Raman spectroscopy (RS) is a powerful technique, which requires small samples, attractive for using in archaeal research. The objective of this work was the estimation of bacterioruberin content along with Haloarcula marismortui growth and their correlation with biomass concentration. RS was used to detect characteristic bands of bacterioruberin (vibrational modes C═CH, C─C, and C═C) in H. marismortui culture samples. The intensity of Raman spectra in bacterioruberin and the biomass concentration were adequately correlated. The highest production of bacterioruberin occurred at 60 h. RS is revealed as a reliable technique for the estimation of bacterioruberin in the biomass of H. marismortui, which could be considered as a promising qualitative and quantitative technique to assay metabolites in cell cultures.

A solid-phase extraction procedure followed by analysis by high performance liquid chromatography (HPLC) with UV-vis photodiode array detection (DAD) is proposed to simultaneously determine 11 aging markers in tequila. The method showed good intraday (n=5) and interday (n=3) precision, RSD<1.6% in both cases, for each of the identified compounds. The calibration curves were linear at the tested ranges (R(2)>0.999). Good recoveries (84.2-108.5%) were obtained for 10 of the 11 compounds studied; and the LOD and LOQ ranged from 0.62 to 4.09 microg/mL and 1.9-12.4 microg/mL, respectively. The proposed methodology was applied to a set of 15 authentic tequila samples grouped by aging state (blanco, reposado and añejo). An ANOVA analysis combined with discriminant analysis with stepwise backward variable selection was used to differentiate between the various aging groups based on their oak related compounds content.

RESUMO

We demonstrate the effectiveness of laser-induced fluorescence (LIF) for monitoring the development and stress detection of in vitro tissue cultures in a nondestructive and noninvasive way. The changes in LIF spectra caused by the induction of organogenesis, the increase of the F690/F740 ratio as a result of the stress originated in the organogenic explants due to shoot emergence, and the relationship between fluorescence spectra and shoot development were detected by LIF through closed containers of Saintpaulia ionantha.

Greenhouse-grown plants of Agave tequilana Weber var. azul were inoculated with Erwinia carotovora, the causal agent of stem soft rot. We investigated the laser-induced fluorescence (LIF) of agave plants to determine whether LIF can be used as a noninvasive sensing tool for pathological studies. The LIF technique was also investigated as a means of detecting the effect of the polyamine biosynthesis inhibitor beta-hydroxyethylhydrazine as a bactericide against the pathogenic bacterium Erwinia carotovora. A He-Ne laser at 632.8 nm was used as the excitation source, and in vivo fluorescence emission spectra were recorded in the 660-790-range. Fluorescence maxima were at 690 and 740 nm. The infected plants that were untreated with the bactericide showed a definite increase in fluorescence intensity at both maxima within the first three days after infection. Beginning on the fifth day, a steady decrease in fluorescence intensity was observed, with a greater effect at 740 than at 690 nm. After 30 days there was no fluorescence. The infected plants that had been treated with the bactericide showed no significant change in fluorescence compared with that of the uninfected plants. The ratio of fluorescence intensities was determined to be F 690 nm/F 740 nm for all treatments. These studies indicate that LIF measurements of agave plants may be used for the early detection of certain types of disease and for determining the effect of a bactericide on bacteria. The results also showed that fluorescence intensity ratios can be used as a reliable indicator of the progress of disease.
