RESUMO

Cellulose nanocrystals (CNCs) have received increasing attention because of their superior dispersion and thermal stability. In this study, TEMPO-oxidized cellulose nanocrystal (TOCNC) multifunctional antioxidationantioxidation films (TOCNC-GA film) were prepared by the esterification of TOCNC and gallic acid (GA). TOCNC-GAX films, where X represents the ratio of the amount of GA to the amount of TOCNC, were characterized using scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. The films with the GA:TOCNC ratio of 1:1 achieved higher interfacial compatibility than the other films. The mechanical properties and water resistance of the TOCNC-GA films were superior than those of pure TOCNC films. Moreover, the original TOCNC structure changed owing to the presence of GA, which endowed a certain thermoplasticity owing to the formation of ester groups. The antioxidation properties of the TOCNC-GA1 films reached 43.8 % and 71.85 % after 6 and 24 h, respectively, as evaluated by the 2,2-biphenyl-1-picrylhydrazyl method and the free radical scavenging activities of the TOCNC-GA1 films. The innovative development of the functional antioxidation film presented in this paper has great potential for use in antioxidation packaging materials and food preservation.

This study provides a novel strategy for preparing bio-based antibacterial emulsions stabilized by cellulose nanocrystals (CNCs). Antibacterial ε-polylysine (ε-PL) with a positive charge was introduced into the aqueous phase to modulate the interfacial behavior of CNCs via electrostatic interactions. Pickering emulsions containing ε-PL/CNCs (ε-PL 0.07-0.1 g/L) had significantly better stability, larger emulsion ratio, smaller emulsion droplet diameter, and superior antibacterial ability than emulsions stabilized by CNCs alone. This could be attributed to the formation of a CNC-dense layer at the interface in the continuous phase caused by a reduction of electrostatic repulsion after adding ε-PL. This was confirmed by zeta potential measurements, rheological properties, and bio-freezing scanning electron microscopy. In addition, cinnamaldehyde was introduced into the oil phase to further improve the antibacterial properties of the emulsion, thereby avoiding easy evaporation into water. Our findings provide an innovative solution for preparing bio-based antibacterial emulsions stabilized by ε-PL/CNCs, which will benefit the development of food, medicine, and cosmetic lotions.

In this study, we prepared spherical cellulose nanocrystals (S-CNCs) and stabilized n-hexadecane Pickering emulsions in conjunction with graphene oxide (GO), exploring the interaction between S-CNCs and GO in the emulsions. Both S-CNCs and GO are amphiphilic and synergistically stabilize Pickering emulsions by adhering to the surface of oil droplets and within the emulsion space through hydrogen bonding. GO's two-dimensional sheets assemble into a 3D network structure, further improving the stability of Pickering emulsions. Consequently, the stability of Pickering emulsions can be adjusted by altering the S-CNCs/GO ratio, modifying the spatial distribution relationship of stabilizers in the emulsions. At an S-CNCs concentration of 1 g/L and a GO concentration of 3 g/L, the Pickering emulsion demonstrated excellent stability and exhibited no delamination after 31 days of storage. Thus, the S-CNCs/GO combination serves as an effective Pickering emulsion stabilizer, utilizing the synergistic effect between the two components.

The use of bacterial toxins as antitumor agents has received considerable attention. Immunotoxins based on antigen recognition of single-chain antibodies have been widely explored for cancer therapy. Despite their impressive killing effect on tumor cells, immunotoxins still display unspecific toxicity with undesired side effects. High levels of hypoxia-inducible factor 1α (HIF-1α) are well-known indicators of hypoxia in cancer cells. In this study, different linkers were employed to fuse the immunotoxin DAB389-4D5 scFv (DS) with the oxygen-dependent degradation domain (ODDD) of HIF-1α, a domain selectively facilitating the accumulation of HIF-1α under hypoxia, to construct the oxygen-dependent degradable immunotoxin DS-ODDD (DSO). The engineered fusion protein DSO-2 containing a linker (G4S)3 possesses the best killing effect on cancer cells under hypoxia and displayed considerably reduced nonspecific toxicity to normal cells under normoxic conditions. Flow cytometry, immunofluorescence, and immunoblot analyses demonstrated that DSO-2 was degraded via the ubiquitin-proteasome pathway regulated by the oxygen-sensitive mechanism. Western blot analysis indicated that the degradation of DSO-2 significantly decreased the activation of apoptosis-related molecules in normal cells. The engineered immunotoxin with oxygen-sensing properties developed herein is a potential therapeutic agent for cancer treatment.

PURPOSE: The present study was designed to investigate the risk factors affecting the conversion to open surgery in retroperitoneal laparoscopic nephrectomy of nonfunctioning renal tuberculosis (TB). PATIENTS AND METHODS: The records of 144 patients who underwent a retroperitoneal laparoscopic nephrectomy procedure by a single surgeon were retrospectively reviewed. The following factors, including age, sex, body mass index (BMI), diabetes status, hypertension status, side of kidney, size of kidney, degree of calcification, mild perirenal extravasation, contralateral hydronephrosis, the time of anti-TB, and surgeon experience were analyzed. Univariate and multivariate logistic regression analyses were used for statistical assessment. RESULTS: Twenty-three patients were converted to open surgery and the conversion rate was 15.97%. In univariate analysis, BMI ≥35 kg/m2 (p = 0.023), hypertension (p = 0.011), diabetes (p = 0.003), and kidney size (p = 0.032) were the main factors of conversion to open surgery. Sex, age, side, anti-TB time, calcification, mild extravasation, and surgeon experience were not significantly related. In multivariate regression analysis, BMI ≥35 kg/m2, hypertension, diabetes, and enlargement of kidney were the most important factors for conversion to open surgery. CONCLUSIONS: Depending on the results achieved by a single surgeon, BMI ≥30 kg/m2, diabetes, hypertension, and enlargement of kidney significantly increased the conversion risk in retroperitoneal laparoscopic nephrectomy for nonfunctioning renal TB.
