RESUMO

Phytoplankton blooms are common along the Chinese coast in the East China Sea, driven by various nutrient sources including river discharge, bottom water regeneration, and Kuroshio subsurface water intrusion. A notable 2014 summer bloom off the Zhejiang coast, exhibiting a Chl a concentration of 20.1 µg L-1, was significantly influenced by Changjiang River discharge, and high nutrient concentrations are often observed in the region's surface water. During blooms, primary production peaks at 1686.3 mg C m-3 d-1, indicating substantial CO2 absorption, with surface water fCO2 declining to 299.5 µatm, closely linked to plankton activities. Hypoxia often coincides with these frequent bloom occurrences, implicating marine-derived organic matter decomposition as a pivotal factor. Elevated particulate organic carbon concentrations further support this assumption, alongside increased nutrient levels, fCO2, and low pH in hypoxic waters. These findings underscore the intricate interplay between phytoplankton, nutrient cycling, and hypoxia formation, essential for effective coastal ecosystem management.

Biological organic carbon production and consumption play a fundamental role in the understanding of organic carbon cycling in oceans. However, studies on them in the Kuroshio, the western boundary current in the North Pacific Ocean, are scarce. To better understand the variations of plankton community respiration (CR) and particulate organic carbon (POC), eight cruises. which covered four seasons over a 2-year period, were surveyed across the Kuroshio at the KTV1 transect east of Taiwan. Spatially, a coastal uplift of isotherms (i.e., onshore lifting and offshore deepening) was observed along the KTV1 transect. During the uplift, the cold and nutrient-rich deep waters shoal to shallow water and enhance phytoplankton growth, resulting in higher values of phytoplankton, POC, and plankton CR on the onshore side. In this study, phytoplankton was dominated by picophytoplankton including Prochlorococcus, Synechococcus, and picoeukaryotes. Plankton CR was low, and its mean depth-normalized integrated rate (the upper 100 m water depth) ranged from 7.07 to 22.27 mg C m-3 d-1, to which the picophytoplankton and heterotrophic bacteria contributed the most. The mean depth-normalized integrated value of POC ranged from 12.7 to 21.6 µg C L-1. POC is mainly associated with phytoplankton biomass with a mean carbon ratio of chlorophyll a/POC ≈ 1.03. All results suggest that plankton CR and POC variations may be associated with picoplankton dynamics in the Kuroshio.

Using molecular beam epitaxy, we prepared seven p-type AlGaN samples of ~25% in Al content, including six samples with Mg-doped/un-doped AlGaN alternating-layer structures of different layer-thickness combinations, for comparing their p-type performances. Lower sheet resistance and higher effective hole mobility are obtained in a layer-structured sample, when compared with the reference sample of uniform Mg doping. The improved p-type performance in a layer-structured sample is attributed to the diffusion of holes generated in an Mg-doped layer into the neighboring un-doped layers, in which hole mobility is significantly higher because of weak ionized impurity scattering. Among the layer-structured samples, that of 6/4 nm in Mg-doped/un-doped thickness results in the lowest sheet resistance (the highest effective hole mobility), which is 4.83 times lower (4.57 times higher) when compared with the sample of uniform doping. The effects of the Mg-doped/un-doped layer structure on p-type performance in AlGaN and GaN are compared.

This study evaluates the rarely observed phenomenon of the simultaneous occurrences of phytoplankton blooms, hypoxia, and upwelling along the Zhejiang coast in the East China Sea. Results show that the upwelling uplifted bottom water to 5-10 m below the surface. In the upwelling region, phytoplankton blooms (Chl a = 10.9 µg L-1) occurred and hypoxia or low-oxygen appeared below the surface water. High concentrations of nitrate and phosphate were regenerated in the hypoxic regions, corresponding with mean values (± SD) of 16.9 (± 1.5) and 0.90 (± 0.14) µM, respectively. The upwelling expanded the region of hypoxic water, which nearly reached the surface, thereby increasing the threat to marine life. In addition to fluvial nutrients, the upwelling of water with high nutrient levels, especially phosphates, can enhance phytoplankton blooms. The results suggest that hypoxia can become more severe due to further decomposition of bloom-derived organic matter after blooms crash.

Many studies indicate that variation of marine bacterial beta diversity in the horizontal dimension is mainly attributable to environmental and spatial effects. However, whether and how these two effects drive bacterial beta diversity in the vertical dimension remains unclear, especially when considering seasonal variation in the strength of water stratification. Here, we used 78 paired bacterioplankton community samples from surface and deep chlorophyll maximum (DCM) layers along a transect in the Kuroshio region east of Taiwan across multiple seasons. Variance partitioning was used to evaluate the mechanisms driving the vertical beta diversity between surface-DCM bacterioplankton communities during weak stratification periods (i.e., spring and fall) versus strong stratification periods (i.e., summer). During strong periods of stratification, vertical beta diversity was shaped by both environmental and spatial effects; notably, the strength of stratification played an important role in enhancing environmental dissimilarity and creating a barrier to dispersal. In contrast, during periods of weak stratification, environmental effects dominate, with a non-significant spatial effect due to mixing. Variation of vertical beta diversity for bacterioplankton communities in the Kuroshio region east of Taiwan was structured by different mechanisms across seasons, and was further dependent on stratification strength of the water column.

Hypoxia (O2 ≤ 2 mg L-1) can severely threaten the survival of marine life and alter the biogeochemical cycles of coastal ecosystems. Its impacts are dependent on its duration. In the present study, hypoxia was observed in autumn at the end of October 2011. It may be one of the latest recorded annual hypoxic events in the East China Sea (ECS). In the hypoxic regions, a large amount of nutrients and dissolved inorganic carbon were observed to regenerate. Also, acidification (low pH) was observed. On the other hand, hypoxic dissipation may be due to the destratification caused by the upwelling of the hypoxic regions in the ECS. These results suggest that hypoxia may occur for longer periods of time than expected and, accordingly, the effects of hypoxia on the ECS ecosystems should be reconsidered and further evaluated.

In 2009, the container ship Colombo Queen and the oil tanker W-O BUDMO grounded off Jialeshui and Houwan, respectively, in southern Taiwan. Water quality was monitored at each site to evaluate the environmental impact caused by the resulting oil spills. The results show that the PAHs, turbidity, and other nutrients increased shortly after oil spill, however levels of these parameters eventually returned to baseline levels. On the other hand, DO saturation, pH and chl. a decreased initially, reached maxima after 10days, and returned to the baseline levels after 14days. The chl. a concentration, pH and DO saturation fluctuated in a similar pattern at both sites during the oil spills, likely driven by algal blooms. In this study, we documented a full environmental recovery at coastal areas before, during and after the oil spills.

To evaluate the effect of rainfall intensity on phytoplankton blooms, a continuous monitoring system was deployed during 2015 in a hyper-eutrophic lagoon in Taiwan. Intensive rainfall occurs during the wet summer months, from May to September. Salinity in the lagoon was found to decrease with increasing intensity of rainfall. The magnitude of phytoplankton blooms also increased linearly with increasing rainfall intensity. The chlorophyll a concentration rose by an order of magnitude during the heaviest rainfall. Blooms may be fueled by nutrient enrichment caused by drainage or run-off water from surrounding areas that is channeled into the lagoon during rainfall events. During bloom periods, the rates of net primary production and ecosystem respiration were high. However, this ecosystem was autotrophic for most of the year. As extreme rainfall is predicted to increase, the results of this study imply that the frequency and magnitude of phytoplankton blooms may increase in the future.

This study demonstrated the potential effects of internal waves (IWs) on heterotrophic bacterial activities for the first time. Nine anchored studies were conducted from 2009-2012 in the South China Sea areas with different physical conditions, i.e. areas subjected to elevation IWs, to depression IWs, and to weak/no IWs. The latter two areas were treated as the Control sites. Field survey results indicated that within the euphotic zone, the minima of the depth-averaged bacterial production (IBP; ∼1.0 mgC m-3 d-1 ) and growth rate (IBµ; ∼0.1 d-1 ) at all sites were similar. Except for one case, the maxima of IBP (6-12 mgC m-3 d-1 ) and IBµ (0.55-1.13 d-1 ) of the elevation IWs areas were ∼fivefolds higher than those of the Control sites (IBP 1.7-2.1 mgC m-3 d-1 ; IBµ 0.13-0.24 d-1 ). Replicate surveys conducted at the north-western area of the Dongsha atoll during spring-to-neap (NW1 survey) and neap-to-spring (NW2 survey) tide periods showed a great contrast to each other. Low variation and averages of IBµ in NW1 survey were similar to those of the Control sites, while those in NW2 were similar to the other elevation IWs sites with larger variation and higher averages of IBµ. This finding suggests that bacterial activities may be a function of the lunar fortnightly (14-day) cycle. Enrichment experiments suggested more directly that the limiting inorganic nutrients introduced by the elevation waves (EIWs) may contribute a higher IBµ within the euphotic zone.

Plantaginaceae, a popular traditional Chinese medicine, has long been used for treating various diseases from common cold to cancer. Linalool is one of the biologically active compounds that can be isolated from Plantaginaceae. Most of the commonly used cytotoxic anticancer drugs have been shown to induce apoptosis in susceptible tumor cells. However, the signaling pathway for apoptosis remains undefined. In this study, the cytotoxic effect of linalool on human cancer cell lines was investigated. Water-soluble tetrazolium salts (WST-1) based colorimetric cellular cytotoxicity assay, was used to test the cytotoxic ability of linalool against U937 and HeLa cells, and flow cytometry (FCM) and genechip analysis were used to investigate the possible mechanism of apoptosis. These results demonstrated that linalool exhibited a good cytotoxic effect on U937 and HeLa cells, with the IC50 value of 2.59 and 11.02 µM, respectively, compared with 5-FU with values of 4.86 and 12.31 µM, respectively. After treating U937 cells with linalool for 6 h, we found an increased sub-G1 peak and a dose-dependent phenomenon, whereby these cells were arrested at the G0/G1 phase. Furthermore, by using genechip analysis, we observed that linalool can promote p53, p21, p27, p16, and p18 gene expression. Therefore, this study verified that linalool can arrest the cell cycle of U937 cells at the G0/G1 phase and can arrest the cell cycle of HeLa cells at the G2/M phase. Its mechanism facilitates the expression of the cyclin-dependent kinases inhibitors (CDKIs) p53, p21, p27, p16, and p18, as well as the non-expression of cyclin-dependent kinases (CDKs) activity.

The coral reef ecosystems of Nanwan Bay, Southern Taiwan are undergoing degradation due to anthropogenic impacts, and as such have resulted in a decline in coral cover. As a first step in preventing the continual degradation of these coral reef environments, it is important to understand how changes in water quality affect these ecosystems on a fine-tuned timescale. To this end, a real-time water quality monitoring system was implemented in Nanwan Bay in 2010. We found that natural events, such as cold water intrusion due to upwelling, tended to elicit temporal shifts in coral spawning between 2010 and 2011. In addition, Degree Heating Weeks (DHWs), a commonly utilized predictor of coral bleaching, were 0.92 and 0.59 in summer 2010 and 2011, respectively. Though this quantity of DHW was below the presumed stress-inducing value for these reefs, a rise in DHWs in the future may stress the resident corals.

BACKGROUND: The melanoma-associated antigen (MAGE) gene family consists of different expression patterns in various tumor types. They are considered tumor-specific antigens and are ideal targets for cancer immunotherapy. The purpose of this study is to identify the expression profiles of the MAGE family genes in Taiwanese colorectal cancer patients. METHODS: In this study, a well-constructed chip array platform was used to analyze the expression of the MAGE family genes of 100 colorectal cancer tissues. Statistical analysis of the experimental results and patients' clinical manifestations were also conducted. RESULTS: The results showed MAGE-A2 (87%), -A7 (83%), -A8 (75%), -A12 (71%), -B2 (75%), -B3 (79%), -D2 (75%), -F1 (79%), and -H1 (70%) were significantly overexpressed genes in colorectal cancer tissues. MAGE-A2 was the most highly overexpressed gene among the MAGE family. MAGE-B3 gene expression is statistically correlated with tumor size, lymph node, and UICC stage. In addition, the overexpression of MAGE-D2 and -H1 genes are statistically correlated to the tumor size and depth, respectively (P < 0.05). CONCLUSIONS: This is the first comprehensive report to clarify the differential expression profile of whole MAGE family in CRCs, and it might provide some crucial information about the carcinogenesis and progression in Taiwanese patients with CRC.

Epidermal growth factor receptor (EGFR)-directed tyrosine kinase inhibitors (TKIs) have been established as a treatment option in patients with advanced non-small cell lung cancer (NSCLC). Clinically, PCR and RFLP are commonly used to evaluate the efficacy of TKIs, and these methods require cancer tissues to proceed. In the event a peripheral blood test is able to replace current evaluation methods, a greater clinical application advantage may be achieved. Therefore, in this study, we selected 30 EGFR pathway-related genes and constructed activated EGFR chips to identify overexpression of EGFR pathway-related genes from the peripheral blood of 72 NSCLC patients and 100 normal subjects. According to ROC curve analysis, the best chip interpretation cutoff value was 11 genes. Correlation analysis showed high significance among EGFR mutations, overexpression and the overexpression of EGFR pathway-related genes (p<0.0001). The potential application of this new technique may provide an accurate, instantaneous and convenient drug evaluation tool.

Several coral reefs of Nanwan Bay, Taiwan have recently undergone shifts to macroalgal or sea anemone dominance. Thus, a mass-balance trophic model was constructed to analyze the structure and functioning of the food web. The fringing reef model was comprised of 18 compartments, with the highest trophic level of 3.45 for piscivorous fish. Comparative analyses with other reef models demonstrated that Nanwan Bay was similar to reefs with high fishery catches. While coral biomass was not lower, fish biomass was lower than those of reefs with high catches. Consequently, the sums of consumption and respiratory flows and total system throughput were also decreased. The Nanwan Bay model potentially suggests an overfished status in which the mean trophic level of the catch, matter cycling, and trophic transfer efficiency are extremely reduced.

The mechanisms of photo-catalytic reduction of Cr(VI) were investigated in acidic solutions with and without Fe(III). In a system without Fe(III), no Cr(VI) reduction was observed in dark conditions; conversely, under light conditions, the reduction reaction rate increased to 0.011 and 0.020microM min(-1) at pH 2 and pH 1, respectively, indicating the occurrence of Cr(VI) photo-reduction. The Cr(VI) photo-reduction reaction was induced by the photolysis of water molecules, leading to O(2) production. Upon the addition of Fe(III), the photo-reduction rate of Cr(VI) was significantly enhanced due to the formation of Fe(II), which is the photolytic product of FeCl(2)(+) and the electron donor for Cr(VI) reduction. However, with the same concentration of FeCl complexes, a strong inhibition of Cr(VI) reduction at pH 2 was observed, compared with pH 1. A possible explanation is that FeOH(2+) becomes predominant with increasing pH and that its photolytic product, the OH free radical, is an oxidant for Fe(II) and Cr(III) and can compromise Cr(VI) reduction. The kinetic result of each photo-reduction reaction pathway shows zero-order kinetics, suggesting that the photolysis reaction of H(2)O or FeCl(2+) is the rate-determining step in each pathway. The results also show the potential of developing a homogeneous photo-catalytic method to treat Cr(VI)-containing water.

Before 2001, the ecological protection area in the Kenting National Park (KTNP), southern Taiwan, was poorly described. In this study, a set of four-year data (2001-2004) of seawater qualities at 19 sampling sites around the Nanwan Bay in the KTNP was used to explore anthropogenic impacts to ecological environment, especially coral reefs. The parameters of water quality were analyzed immediately after collection. The results showed that higher values of nutrients and suspended solids were attributed to the higher run-off around Nanwan Bay. The fluxes of nutrients and suspended solids were consistently correlated to rainfall. Hence, equations were developed to calculate nutrient fluxes and suspended solids by using only rainfall data. Our results show that suspended solids and ammonia were the dominant factors leading to the drop in coral coverage. In summary, the water quality in the intertidal zone of Nanwan Bay has been degraded and required greater attention.

Photo-reduction of Cr(VI) in a solution with single or multi-inorganic anions was evaluated. The results show that 38.5 microM Cr(VI) is photo-reduced in the presence of NO(3)(-) at pH 1. The photolysis of NO(3)(-), producing NO(2)(-) or H(2)O(2), may contribute to Cr(VI) reduction. The addition of 0.001-0.1 M chlorite to NO(3)(-) enhanced Cr(VI) photo-reduction when 35.8 microM Fe(III) was present. This enhancement was the combinative result of photolysis of NO(3)(-) and Fe-Cl complexes, leading to the formation of NO(2)(-) and Fe(II), respectively, for Cr(VI) reduction. On the contrary, a significant decrease in Cr(VI) photo-reduction was observed with the addition of PO(4)(3-) and SO(4)(2-). This decrease was due to their strong competition with Fe(III) from Cl(-), resulting in a marked decrease in the concentrations of Fe-Cl complexes. The results suggest that a direct irradiation of acidic wastewaters containing Cl(-), NO(3)(-), and Fe(III) is a feasible strategy for eliminating Cr(VI).

OBJECTIVE: To investigate the association between family history of cardiovascular disease (CVD) and cholesteryl ester transfer protein (CETP) TaqIB polymorphism in Taiwanese subjects. SUBJECTS AND METHODS: In this cross-sectional study, 240 subjects (115 men and 125 women) were divided into two groups based on whether or not they had a parental history of CVD. Polymerase chain reaction/restriction fragment length polymorphism was used to analyze the genotype of the subjects for the TaqIB polymorphism of CETP in intron 1. RESULTS: The frequency of the B1B1 genotype was significantly higher in Taiwanese subjects with a family history of CVD than in those without it (31.2 vs. 18.8%, odds ratio = 1.97, 95% confidence interval = 1.084-3.579, p = 0.035). Siblings with the B1B1 genotype had lower levels of serum high-density lipoprotein cholesterol (HDL-C) than siblings with either B1B2 (46.7 +/- 11.0 vs. 52.5 +/- 11.1 mg/dl, p = 0.034) or B2B2 genotypes (46.7 +/- 11.0 vs. 55.2 +/- 9.6 mg/dl, p = 0.01). CONCLUSION: CETP TaqIB polymorphism is associated with plasma HDL-C levels. The CETP B1B1 genotype may influence the susceptibility to CVD in Taiwan.

A model experiment monitoring the fate of total residue oxidant (TRO) in water at a constant temperature and salinity indicated that it decayed exponentially with time, and with TRO decaying faster in seawater than in distilled water. The reduction of TRO by temperature ( degrees K) was found to fit a curvilinear relationship in distilled water (r(2) = 0.997) and a linear relationship in seawater (r(2) = 0.996). Based on the decay rate, flow rate, and the length of cooling water flowing through at a given temperature, the TRO level in the cooling water of a power plant could be estimated using the equation developed in this study. This predictive model would provide a benchmark for power plant operators to adjust the addition of chlorine to levels necessary to control bio-fouling of cooling water intake pipelines, but without irritating ambient marine organisms.

Anoxia and hypoxia have been widely observed in estuarine and coastal regions over the past few decades; however, few reports have focused on the East China Sea (ECS). In June and August 2003, two cruises sampled at stations covering almost the entire shelf of the ECS to examine hypoxic events and their potential causes. In August, DO concentrations <2-3 mg l(-1) covered an area estimated at greater than 12,000 km(2) (or 432 km(3) volume). In contrast, water column DO concentrations exceeded 4 mg l(-1) throughout most of the shelf region. A sharp density gradient was observed under the mixed layer in August, restricting vertical re-aeration across this strong pycnocline. Oxygen depletion events, such as that described here for the ECS shelf, are fueled by decomposition of newly produced marine and river-borne biogenic substances (as well as older residual organic matter) deposited to the bottom waters.
