RESUMO

Constructed wetlands (CWs) have been used for wastewater treatment for decades, but research on microorganisms involved, especially long-term changes, is still limited. In this study, we evaluated changes in the substrate microbial community in a pilot-scale horizontal subsurface-flow constructed mangrove wetland during 10-years operation. In the 3rd year of operation, microbial biomass carbon and phospholipid fatty acids (PLFAs) reached peak values in two vegetated belts planted with Aegiceras corniculatum (Ac) and Kandelia obovata (Ko), respectively, then stabilized or declined in the 9th and 10th years of operation. PLFA profiles reflecting microbial community compositions varied significantly in the Ac belt during the operation period. Principal component (PCA) and redundancy analyses (RDA) revealed that microbial community compositions were significantly correlated with organic matter content, especially in the 9th and 10th years of operation, implying that the substrate microbial community in constructed mangrove wetland is sensitive to substrate characteristics and can be used as an indicator for long-term performance of CWs.

The aim of the present study was to elucidate the microbial community metabolic profiles in saturated constructed wetland (CW) mesocosms planted with five different wetland plant species fed with water individually spiked with 100 µg L-1 ibuprofen or iohexol. Community-level physiological profiling (CLPP) using Biolog Ecoplates was performed and coupled with the assessment of water quality parameters (water temperature, pH, DO and TOC, TN, NH4-N, PO4-P removal efficiency). The microbial community metabolic profiles (microbial activity, richness, and carbon source utilization), as well as the water quality parameters revealed similar trends among the control mesocosms and the mesocosms fed with water spiked with iohexol and ibuprofen. Significant differences were observed between the planted and unplanted mesocosms and between seasons (summer and winter) within each of the feeding lines (control, iohexol or ibuprofen). The microbial community metabolic profiles in the saturated CW were shaped by plant presence and plant species, while no negative impact of iohexol and ibuprofen presence was noticed at the 100 µg L-1. In addition, the microbial activity and richness were generally higher in planted mesocosms than in the unplanted systems in the summer. For the first time, a positive correlation between iohexol removal and the microbial community metabolic profiles (activity, richness and amines and amides utilization in summer, and carbohydrates utilization in winter) in the saturated mesocosms was observed. Putrescine utilization in the summer and d-cellobiose, d,l-alpha-glycerol phosphate in winter were linked with the metabolic processing of iohexol, while glycogen in summer and l-phenylalanine, Glycyl-l-glutamic acid in winter were linked with ibuprofen removal efficiency in the saturated CW.

Constructed mangrove wetland has been used for wastewater treatment but its long-term performance has not been reported. One-year monitoring of a 10-years old horizontal subsurface-flow constructed mangrove wetland consisting of three belts, two with mangrove plants and one without, revealed that the system maintained high and stable removal percentages of organic matter and nutrients, and planted belts performed better than unplanted control. Substrates in belts planted with Aegiceras corniculatum or Kandelia obovata had higher abundance of ammonifiers, nitrifiers and denitrifiers but lower total heterotrophic bacteria than unplanted substrate. Denaturing gradient gel electrophoresis showed that microbial diversity in planted substrate was significantly lower than that in unplanted one. The bacteria in substrates, irrespective to belts, were phylogenetically related to Proteobacteria (most dominant), Acidobacteria, Firmicutes, Nitrospirae, Gemmatimonadetes, Chloroflexi and Cyanobacteria. The steady performance of this 10-year old constructed mangrove wetland was affected by the abundance and diversity of bacterial community in substrate.

While the production of polybrominated diphenyl ethers (PBDEs) was gradually phased out in the last decade, they may still pose hidden danger to the environment due to their toxicity and persistence. On the other hand, alternative halogenated flame retardants (AHFRs) have been increasingly used as substitutes for PBDEs and may further worsen environmental health. To determine the environmental impact of PBDEs and AHFRs, we examined the historical trends and ecological risks of PBDEs and AHFRs in a typical industrialized city in South China by measuring their concentrations in mangrove sediment. Results showed that the concentrations of PBDEs increased abruptly from 1997 to 2009 due to the use of commercial deca-BDE mixture, but were stabilized in recent years. The concentrations of AHFRs, mainly contributed by decabromodiphenyl ethane (DBDPE), kept increasing from 1997 onwards. Based on the temporal trends, DBDPE is predicted to be predominant over BDE 209 in future. Despite the observed similar concentration between BDE 209 and DBDPE, the former posed a high ecological risk, while the ecological risk of the latter was negligible. Therefore, more attention is required to manage the contamination of BDE 209 in the environment.

Surface sediments were collected from three mangrove wetlands (Yifeng Xi, Shuanghan, and Su'ai Wan) in Shantou coastal zone of South China to investigate spatial distributions of polybrominated diphenyl ethers (PBDEs). The results demonstrate that PBDEs were detected in all the samples, indicating their widespread occurrence in coastal sediments of the studied area. Σ9PBDEs (defined as the sum of nine targeted PBDE congeners except BDE-209) and BDE-209 are in the range of 2.3 to 11.5 and 16.7 to 58.2 ng/g, respectively. BDE-209 is the dominant PBDE congener in all sediment samples. The sediment concentrations of ∑9PBDEs and BDE-209 among the three wetlands decrease in the order of Su'ai Wan > Shuanghan > Yifeng Xi. The concentrations of ∑9PBDEs are higher in mangrove sediments than in mudflats, but no obvious regularity can be found on the correlation between mangrove species and PBDE levels in sediments. The contents of total organic carbon are moderately correlated with BDE-209 concentrations in sediments but not with ∑9PBDE concentrations. The samples collected from different locations show slightly different composition profiles except BDE-209, with BDE-100 and BDE-47 being the pre-dominated congeners. The mudflats exhibit higher abundances of tri- to hexa-substituted congeners than the mangrove sediments. Ecological risk assessment demonstrates that the surface sediments from Shantou may pose a potential ecological risk of exposure to sediment-dwelling organisms.

Bacteria play crucial roles in the combined system of substrate addition and C/N control, which has been demonstrated to improve aquaculture production. However, the complexity of surface-attached bacteria on substrates and suspended bacteria in the water column hamper further application of this system. This study firstly applied this combined system into the culture of grass carp, and then explored the relationship between microbial complexes from surface-attached and suspended bacteria in this system and the production of grass carp. In addition, this study investigated bacterial community structures as affected by four C/N ratios using Illumina sequencing technology. The results demonstrated that the weight gain rate and specific growth rate of grass carp in the CN20 group (C/N ratio 20:1) were the highest (P < 0.05), and dietary supplementation of the microbial complex had positive effects on the growth of grass carp (P < 0.05). Sequencing data revealed that, (1) the proportions of Verrucomicrobiae and Rhodobacter (surface-attached), sediminibacterium (suspended), and emticicia (surface-attached and suspended) were much higher in the CN20 group compared with those in the other groups (P < 0.05); (2) Rhodobacter, Flavobacterium, Acinetobacter, Pseudomonas, Planctomyces, and Cloacibacterium might be important for the microbial colonization on substrates; (3) as the C/N ratio increased, proportions of Hydrogenophaga (surface-attached and suspended), Zoogloea, and Flectobacillus (suspended) increased, but proportions of Bacillus, Clavibacter, and Cellvibro (surface-attached and suspended) decreased. In summary, a combined system of substrate addition and C/N control increased the production of grass carp, and Verrucomicrobiae and Rhodobacter in the surface-attached bacterial community were potential probiotic bacteria that contributed to the enhanced growth of grass carp.

Microbes are believed to be at the core of the wastewater treatment processes in constructed wetlands (CWs). The aim of this study was to assess the microbial biomass carbon (MBC) and Shannon's diversity index (SDI) in the substrate of CWs planted with Phragmites australis, Hymenocallis littoralis, Canna indica and Cyperus flabelliformis, and to relate MBC and SDI to the pollutant removal in the systems. Significant higher MBC was observed in CWs with H. littoralis and C. indica than in CWs with P. australis, and the MBC differed with season and substrate depth. The microbial community in the wetlands included four phyla: Cyanobacteria, Proteobacteria, Chloroflexi, and Acidobacteria, with a more diverse community structure in wetlands with C. flabelliformis. The MBC in the substrate and the SDI of the 15-20 cm depth correlated with the removal of biochemical oxygen demand, NH4-N and NO3-N. Our results indicate that substrate SDI and MBC can both be regarded as bioindicators of the pollutant removal ability in CWs.

We aimed at assessing the effects of four wetland plant species commonly used in constructed wetland systems: Typha, Phragmites, Iris and Juncus for removing ibuprofen (IBU) and iohexol (IOH) from spiked culture solution and exploring the mechanisms responsible for the removal. IBU was nearly completely removed by all plant species during the 24-day experiment, whereas the IOH removal varied between 13 and 80 %. Typha and Phragmites were the most efficient in removing IBU and IOH, respectively, with first-order removal rate constants of 0.38 and 0.06 day(-1), respectively. The pharmaceuticals were taken up by the roots and translocated to the aerial tissues. However, at the end of the experiment, plant accumulation constituted only up to 1.1 and 5.7 % of the amount of IBU and IOH spiked initially. The data suggest that the plants mainly function by facilitating pharmaceutical degradation in the rhizosphere through release of root exudates.

Morphological, structural, and eco-physiological features of roots, nutrient removal, and correlation between the indices were comparatively studied for 35 emergent wetland plants in small-scale wetlands for further investigation into the hypothesis of two types of wetland plant roots (Chen et al., 2004). Significant differences in root morphological, structural, and eco-physiological features were found among the 35 species. They were divided into two types: fibrous-root plants and thick-root plants. The fibrous-root plants had most or all roots of diameter (D) ≤ 1 mm. Roots of D > 1 mm also had many fine and long lateral roots of D ≤ 1 mm. The roots of these plants were long and had a thin epidermis and a low degree of lignification. The roots of the thick-root plants were almost all thicker than 1 mm, and generally had no further fine lateral roots. The roots were short, smooth, and fleshy, and had a thick epidermis. Root porosity of the fibrous-root plants was higher than that of the thick-root plants (p = 0.001). The aerenchyma of the fibrous-root plants was composed of large cavities which were formed from many small cavities, and distributed radially between the exodermis and vascular tissues. The aerenchyma of the thick-root plants had a large number of small cavities which were distributed in the mediopellis. The fibrous-root plants had a significantly larger root biomass of D ≤ 1 mm, of 1 mm < D < 3 mm, above-ground biomass, total biomass, and longer root system, but shorter root longevity than those of the thick-root plants (p = 0.003, 0.018, 0.020, 0.032, 0.042, 0.001). The fibrous-root plants also had significantly higher radial oxygen loss (ROL), root activity, photosynthetic rate, transpiration rate, and removal rates of total nitrogen and total phosphorus than the thick-root plants (p = 0.001, 0.008, 0.010, 0.004, 0.020, 0.002). The results indicate that significantly different root morphological and structural features existed among different wetland plants, and these features had a close relationship to nutrient removal capacity.

Sixteen small-scale wetlands planted with four plant species were constructed for domestic wastewater purification. The objective of this study was to determine the correlations between contaminant removal and soil enzyme activity, root activity, and growth in the constructed wetlands. The results indicated that correlations between contaminant removal efficiency and enzyme activity varied depending on the contaminants. The removal efficiency of NH4+ was significantly correlated with both urease and protease activity in all wetlands, and the removal of total phosphorus and soluble reactive phosphorus was significantly correlated with phosphatase activity in most wetlands, while the removal of total nitrogen, NO3(-) , and chemical oxygen demand (COD) was significantly correlated with enzyme activity only in a few instances. Correlations between soil enzyme activity and root activity varied among species. Activities of all enzymes were significantly correlated with root activity in Vetiveria zizanioides and Phragmites australis wetlands, but not in Hymenocallis littoralis wetlands. Significant correlations between enzyme activity and root biomass and between enzyme activity and root growth were found mainly in Cyperus flabelliformis wetlands. Root activity was significantly correlated with removal efficiencies of all contaminants except NO3(-) and COD in V. zizanioides wetlands. Enzyme activities and root activity showed single-peak seasonal patterns. Activities of phosphatase, urease, and cellulase were significantly higher in the top layer of the substrate than in the deeper layers, and there were generally no significant differences between the deeper layers (deeper than 15 cm).

The aim of the present study is to probe the relation between plant growth and its decontamination effect in constructed wetlands. Four species were studied in the small-scale mono-cultured constructed wetlands, which were fed with domestic wastewater. Plant growth indexes were correlated with contaminant removal performance of the constructed wetlands. Wetlands planted with Cyperus flabelliformis Rottb. showed the highest growth indexes such as shoot growth, biomass, root activity, root biomass increment, and the highest contaminant removal rates, whereas wetlands planted with Vetiveria zizanioides L. Nash had the lowest growth indexes and the lowest removal rates. Above-ground biomass and total biomass were significantly correlated with ammonia nitrogen removal, and below-ground biomass with soluble reactive phosphorus removal. Photosynthetic rate had higher correlation with nitrogen removal in these species. Root activity and root biomass increment was more correlated with 5 d biochemical oxygen demand removal. Chemical oxygen demand removal had lower correlations with plant growth indexes. All four species had higher removal rates in summer and autumn. The results suggest that the effect of plant growth on contaminant removal in constructed wetlands were different specifically in plants and contaminants.

Based on a 2 x 2 contingency table, the Pearson product-moment correlation coefficient and Spearman rank correlation coefficient were used to analyses the interspecies covariation of dominant tree species in different communities of secondary succession series in Heishiding Natural Reserve, Guangdong Province. In early succession stage, 14 pairs of tree species showed a significant interspecies covariation, and 9 pairs of species showed a negative coraviation, indicating that the species pairs needed the same habitats, while five pairs of species showed a positive covariation, which indicated that the species pairs needed different habitats. In the stage of needle broad-leaved mixed forest, only 5 pairs of species showed a significant interspecies covariation, and they were all positive covariation, which indicated the main species needed the same habitats and the interspecies competition were going. In the stage of evergreen broadleaved forest dominated by heliophytes, 4 pairs of species showed a significant interspecies covariation, which was the least one in secondary succession series. Three pairs of them showed a positive covariation. It was the result of interspecies competition that the species pairs needed the same habitats. In the stage of evergreen broadleaved forest dominated by mesophytes, 20 pairs of species showed a significant interspecies covariation, which was the most one in secondary succession series. Nineteen pairs of them showed a positive covariation. It showed positive covariation between species in upper tree layer and in middle or lower tree layer, but dominant species in upper tree layer had no significant interspecies covariation.
