RESUMO

A procedure is developed for isolation of human blood serum fractions, enriched with lipoproteins of definite classes and meant for production of standard human blood sera. Addition of one of these fractions to native human blood serum enabled to increase distinctly selectively the content of main components of corresponding lipoproteins. Lipoprotein-free blood serum might be used as a diluent for a decrease in concentration of lipoproteins and their components in the initial blood serum, while content of the other components was maintained.

RESUMO

An investigation of children with aggravated heredity (coronary disease), and those with vegetovascular dystonia whose parents had no clinically apparent coronary disease demonstrated unidirectional shifts in the lipoprotein system of the two samples as compared to the controls: increased cholesterol levels in low-density lipoproteins and lowered cholesterol in high-density lipoproteins and apoprotein A1. In children with aggravated heredity, these changes are also associated with vegetovascular dystonia, whereas in the absence of the latter they are not apparent or minimal. At the same time, certain differences in cholesterol distribution over lipoprotein fractions and protein/lipid ratios suggest that mechanisms of changes are not identical in children with aggravated heredity and those free from it.

RESUMO

Over the period of one year the authors examined the changes in the parameters of the plasma spectrum of lipoproteins and apolipoproteins in 46 patients with acute macrofocal myocardial infarction and 35 patients with labile angina. In six cases variations in these parameters were studied in the course of unstable angina and in the post-infarction period. It was established that myocardial infarction induced specific alterations in the plasma lipoprotein system not only in the immediate post-infarction period but also at more long-term periods. Myocardial infarction causes deep and prolonged disorders in the apolipoprotein system which may contribute to the progression of atherosclerotic damage to the coronary arteries.

RESUMO

Apoprotein C-II was isolated from human blood plasma by means of column chromatography-gel filtration and DEAE ion exchange chromatography by modified method of Brown et al (1969). Production of antiserum to apoprotein C-II as well as a procedure for rocket immunoelectrophoresis are described for estimation of the apoprotein in blood plasma of men and women of various age, exhibiting different lipid spectra. Content of the apoprotein C-II in blood plasma, estimated by the immunoelectrophoretic procedure, was higher in men as compared with women and correlated with concentration of triglycerides in blood plasma.

RESUMO

Apolipoproteins (apo A-1 and apo B) were measured by rocket immunoelectrophoresis in 53 patients 40-59 years of age with coronary heart disease. Patients with hyperlipoproteinemia and low HDL cholesterol level as well as those with normal lipid levels were compared with age-matched healthy controls. Apo A-1 blood plasma level reduction was found in the former group. The reduction is shown to be due to the decrease of apo A-1 composing HDL3 in type IIa and IIb hyperlipoproteinemia and in normal lipoprotein level. HDL2 apo A-1 reduction is also observed in type IV hyperlipoproteinemia. The rise of apo B level was noted in the blood plasma of patients with coronary heart disease, the changes being mainly connected with the increase in the number of apo B-containing particles. It is shown, that patients with coronary heart disease have an elevated apo B/apo A-1 ratio as compared to the control group, with its value exceeding 1 whatever the lipoprotein level. Changes in the quantitative composition of plasma apolipoproteins are considered to be a factor, ensuring prevalence of cholesterol flow to peripheral tissues over its acceptance and escape.

Content of apolipoprotein (apo A-I) was studied in subfractions of high density lipoproteins (HDL) using quantitative "rocket" immunoelectrophoresis in two groups of men with the level of LPHD cholesterol within 70 mg/dl (27 patients) and above 70 mg/dl (15 patients). An increase in content of cholesterol from HDL2 subfraction and activation of lecithine-cholesterol acyltransferase in hyper-alpha-cholesterolemia (hyper-alpha-lipoproteinemia) were mainly due to elevation in both absolute amount of apo A-I containing in HDL2 of blood plasma and in relative part of apo A-I in HDL2 of blood plasma and in relative part of apo A-I in HDL2. But contrary to the observation, content of apo A-I from HDL3 subfraction was decreased in the impairment. In aggrement with the modern hypothesis on the antiatherogenic nature of hyper-alpha-lipoproteinemia, the impairment is related to intensification of the cholesterol-acceptor function of HDL due to the activating effect of apolipoprotein A-I (content of which is increased in HDL2) on lecithine-cholesterol acyltransferase in vivo.

The content in the blood plasma of the main apolipoproteins of lipoproteins of very low and low density apo-B and tetramethylurea-soluble apo-LP as well as lipoproteins of high density apo-A and the relation to apo-LP cholesterol of these classes of lipoproteins were studied in 34 males with different levels of alpha-LP cholesterol. It is shown that in hypoalphacholesterolemia the plasma apo-A content is reduced with the cholesterol/apo-A ratio being less than in normoalphacholesterolemia; at the same time, the content in the plasma of total triglycerides, cholesterol, and protein of very low density lipoproteins is increased and cholesterol saturation calculated for protein of very low and low density lipoproteins does not change. In hyperalphacholesterolemia the plasma apo-A content is increased both in the absolute amount and in relation to apo-B and TMU-soluble proteins; the content of cholesterol on apo-A in high density lipoproteins is the same as under normal conditions and higher than in hypoalphacholesterolemia. The results obtained are discussed in association with the data in the literature concerning the functions of lipoproteins and apo-LP in relation to the transport of cholesterol into the vascular wall and its drainage.

Some properties of very low density lipoproteins were studied under conditions causing an increase of their concentration in blood plasma. Content of high molecular apolipoprotein B and low molecular lipoproteins were estimated after fractionation using column chromatography and selective precipitation with tetramethylurea. In 216 men (40-59 years old) with hypertriglyceridemia the increase in content of particles of very low density lipoproteins was accompanied by simultaneous alteration in their composition in blood plasma: content of low molecular polypeptides, triglycerides and cholesterol was increased per a weight unit of apoprotein B. At the same time, the absolute concent of apoprotein B and of low molecular apoproteins of very low density lipoproteins was increased in blood plasma. The most distinct alteration in the ratio of apolipoproteins to very low density lipoproteins was observed, if content of testosterone--activator of lipoprotein lipase--was decreased in blood plasma.

Content of 3',5'-cycloAMP was estimated in rabbit blood plasma and urine; activities of adenylcyclase and phosphodiesterase were determined in liver tissue, adipose tissue and adrenal glands within 5, 24 and 48 hrs after hydrocortisone administration. Concentration of 3',5'-cycloAMP was found to be increased 4-6-fold in plasma and urine after the hormone administration. Cortisol was shown to increase also the activity of adenylcyclase and phosphodiesterase in liver tissue within 5 hrs and in adipose tissue--within 24 hrs and 48 hrs after the administration. In adrenal glands hydrocortisone caused an inhibitory effect on the adenylcyclase activity at early steps of the experiment.
