Microminerals are necessary for all bodily functions. In animal species, selenium (Se), copper (Cu) and zinc (Zn) all form part of antioxidant enzymes. Micromineral deficiencies, particularly Se, are well recognized in large animal species in Chile. Glutathion peroxidase (GPx) is a widely used biomarker for Se nutritional status and to diagnose Se deficiency in horses. Superoxide dismutase (SOD) is a Cu and Zn-dependant antioxidant enzyme, although it is not commonly used as a proxy for the nutritional status of these minerals. Ceruloplasmin (CP) is used as a biomarker of Cu nutritional status. This study´s objective was to explore correlation between said minerals and biomarkers in adult horses from southern Chile. Se, Cu, Zn, GPx, SOD, and CP were measured in whole blood of a group of 32 adult horses (5-15 years old). Further, a second group of 14 adult horses (5-15 years old) underwent gluteal muscle biopsies to determine Cu, Zn, GPx, and SOD. Correlations were determined by means of Pearson´s r coefficient. Significant correlations were found for blood GPx and Se (r = 0.79), blood GPx and SOD (r = -0.6), muscular GPx and SOD (r = 0.78), and Cu and CP (r = 0.48). These results confirm previously described strong association between blood GPx and Se in horses, validating the former´s use as a diagnostic proxy of Se deficiency in Chilean horses, and suggest significant interactions between GPx and SOD in blood and muscle tissue.

Initial lameness inflammation leads to chronic lameness and development of chronic pain due to the release of pro-inflammatory mediators such as reactive oxygen species (ROS), which are implicated in the transition from acute to chronic pain, and free radical scavengers countering thiol, substance P (SP), and ß-endorphin (BE). The present study aimed to evaluate the dynamic thiol-disulfide homeostasis, α-tocopherol concentrations and SP and BE concentrations in the spinal cord of chronically lame dairy cows. Ten lame and 10 non-lame cows with a parity range of 2-6 were selected for the study. Lame cows had a history of up to 3 months of lameness. Spinal cord samples were obtained from the L2 to L4 lumbar vertebrae aspect of each animal. A thiol-disulfide homeostasis assay was performed using absorbance, and the α-tocopherol concentration was determined by HPLC. SP and BE concentrations were measured using ELISA kits. The results indicated that SP and BE were significantly higher in the spinal cord of lame cows. In contrast, disulfide levels and α-tocopherol concentrations were significantly lower in the spinal cord of lame cows. In conclusion, disulfide levels and α-tocopherol concentrations indicated a defective antioxidant response in cows with chronic lameness. The results of SP and BE concentrations suggested chronic pain and a defective endogenous analgesic response.

Abstract Background: Barium selenate is an inorganic source of selenium (Se) used in prolonged-release preparations to treat selenium deficiency in bovines. Objective: To evaluate serum concentrations of triiodothyronine (T3) and thyroxine (T4) hormones in newborn calves from mothers supplemented with barium selenate during prepartum. Methods: Six black Frisian pregnant cows were supplemented with barium selenate subcutaneously during the last two months of gestation, until calving. Six cows were used as controls. All cows were subjected to a low Se diet, consisting of hay from natural pasture and commercial concentrate lacking Se. The Se balance was measured through the activity of erythrocyte glutathione peroxidase (GPx). Serum concentration of T3 and T4 in calves was determined by electrochemiluminescence. Results: Se supplementation during prepartum increased GPx activity in cows from day 45 post-supplementation (p<0.05). Calves from supplemented mothers showed higher average serum Se concentration than calves from non-supplemented mothers. The average concentration of T3 in the calves from supplemented mothers was lower in the first hour of life (p<0.05) compared with calves from mothers of the non-supplemented group. A decrease (p<0.05) in T4 serum concentrations was observed in both groups at seven days of age. Conclusions: Administration of barium selenate to cows during prepartum generates a reduction in serum concentration of T3 in the first hour of life of calves.


Resumen Antecedentes: El selenato de bario es una fuente inorgánica de selenio (Se) utilizada en preparaciones de liberación prolongada para corregir el estado de carencia de Se en bovinos. Objetivo: Evaluar las concentraciones séricas de triyodotironina (T3) y tiroxina (T4) en terneros recién nacidos de madres suplementadas durante el preparto con selenato de bario. Métodos: Seis vacas frisón negro con 7 meses de gestación fueron suplementadas vía subcutánea con selenato de bario dos meses previos a la fecha de parto. Otras seis vacas permanecieron como controles. Todas las vacas se mantuvieron con una dieta cuyo aporte de Se fue inferior a los requerimientos y consistió en heno de pradera natural y concentrado comercial sin Se. El balance de Se se midió usando la actividad eritrocitaria de glutatión peroxidasa (GPx) y las concentraciones de T3 y T4 en terneros mediante electroquimioluminiscencia. Resultados: La suplementación con Se aumentó la actividad de GPx en vacas desde el día 45 post suplementación (p<0,05). Los terneros de madres suplementadas mostraron una concentración sérica promedio de Se mayor que los terneros de madres no suplementadas. La concentración promedio de T3 de terneros de madres suplementadas fue menor en la primera hora de vida (p<0,05) que en terneros de madres no suplementadas. A los 7 días de edad hubo una disminución (p<0,05) en las concentraciones séricas de T4 en ambos grupos. Conclusión: La administración de selenato de bario en vacas preparto genera una disminución en la concentración sérica de T3 en la primera hora de vida del ternero.

Objetivo: Avaliar as concentrações séricas de triiodotironina (T3) e tiroxina (T4) em bezerros recém-nascidos de mães suplementadas durante o pré-parto com selenato de bário. Métodos: Seis vacas friesianas negras aos 7 meses de gestação foram suplementadas com selenato de bário por via subcutânea dois meses antes do parto. Seis outras vacas permaneceram como controle. Todas as vacas foram mantidas em uma dieta cuja contribuição de Se foi inferior aos requeridos e consistiram em feno natural da pradaria e concentrado comercial sem Se. O balanço de Se foi medido usando a atividade eritrocitária das concentrações de glutationa peroxidase (GPx) e T3 e T4 em bezerros por eletroquimiluminescência. Resultados: A suplementação com atividade de GPx aumentou em vacas a partir do dia 45 após a suplementação (p<0,05). Os bezerros de mães suplementadas apresentaram uma concentração sérica média de Se maior que os bezerros de mães não suplementadas. A concentração média de T3 dos bezerros das mães suplementadas foi menor na primeira hora de vida (p<0,05) do que nos bezerros das mães não suplementadas. Aos 7 dias de idade houve uma diminuição (p<0,05) nas concentrações séricas de T4 nos dois grupos. Conclusão: A administração de selenato de bário em vacas de parto gera uma diminuição na concentração sérica de T3 na primeira hora de vida do bezerro.

Lameness in dairy cows is a worldwide prevalent disease with a negative impact on animal welfare and herd economy. Oxidative damage and antioxidant system dysfunction are common features of many CNS diseases, including chronic pain. The aim of this study was to evaluate the levels of reactive oxygen species (ROS) and oxidative damage markers in the spinal cord of dairy cows with chronic inflammatory lameness. Locomotion score was performed in order to select cows with chronic lameness. Dorsal horn spinal cord samples were obtained post mortem from lumbar segments (L2-L5), and ROS, malondialdehyde (MDA), and carbonyl groups were measured along with the activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and total antioxidant response (TAR). Lame cows had increased levels of ROS, MDA, and carbonyl groups, while no differences were observed between lame and non-lame cows in SOD, GPx, CAT, and TAR activity. We conclude that painful chronic inflammatory lameness in dairy cows is associated with an increase in ROS, MDA, and carbonyl groups. Nonetheless, an association between ROS generation and dysfunction of the antioxidant system, as previously proposed, could not be established.

RESUMO

Concentrations of calcium, magnesium, copper, zinc, and selenium were measured in plasma from 11 huemul (Hippocamelus bisulcus) from Chilean Patagonia. Except for zinc and copper, concentrations of these minerals were similar to those of other deer species.

RESUMO

Oxygen-derived free radicals are normally generated in many pathways. These radicals can interact with various cellular components and induce cell injury. When free radicals exceed the antioxidant capacity, cell injury causes diverse pathologic changes in the organs. The imbalance between the generation of free radicals and antioxidant defence is known as oxidative stress. The eye can suffer the effect of oxidative damage due to the etiopathogenesis of some pathological changes related to oxidative stress. This paper reviews the role of oxidative stress in the onset and progression of damage in different eye structures, the involvement of the antioxidant network in protecting and maintaining the homeostasis of this organ, and the potential assessment methodologies used in research and in some cases in clinical practice.

Oleic acid (OA) is a nonesterified fatty acid that is released into the blood during lipomobilization at the time of calving in cows, a period where increased risk of infection and acute inflammation is observed. These data suggest potential OA-mediated regulation of innate immune responses. In the present study, we assessed the effects of OA on intracellular calcium release, ERK1/2 phosphorylation, superoxide production, CD11b expression and matrix metalloproteinase-9 (MMP-9) release in bovine neutrophils. Furthermore, the presence of GPR40, an OA receptor, was assessed by RT-PCR, immunoblotting and confocal microscopy. OA induced, in a dose-dependent manner, intracellular calcium mobilization, superoxide production and CD11b expression in bovine neutrophils; these effects were reduced by the intracellular chelating agent BAPTA-AM. OA also induced ERK2 phosphorylation and MMP-9 release. RT-PCR analysis detected mRNA expression of a bovine ortholog of the GPR40 receptor. Using a polyclonal antibody against human GPR40, we detected a protein of 31kDa by immunoblotting that was localized predominately in the plasma membrane. The selective agonist of GPR40, GW9508, induced intracellular calcium mobilization and ERK2 phosphorylation. In conclusion, OA can modulate bovine neutrophil responses in an intracellular calcium-dependent manner; furthermore, these responses could be induced by GPR40 activation.
