RESUMO

In this paper, micro gas sensor was fabricated using indium oxide nanowire for effective gas detection and monitoring system. Indium oxide nanowire was grown using thermal CVD, and their structural properties were examined by the SEM, XRD and TEM. The electric properties for microdropped indium oxide nanowire device were measured, and gas response characteristics were examined for CO gas. Sensors showed high sensitivity and stability for CO gas. And with below 20 mw power consumption, 5 ppm CO could be detected.

Plant cell suspension culture has become the focus of much attention as a tool for the production of secondary metabolites including paclitaxel, a well-known anticancer agent. Recently, it has also been regarded as one of the host systems for the production of recombinant proteins. In order to produce phytochemicals using plant cell cultures, efficient processes must be developed with adequate bioreactor design. Most of the plant secondary metabolites are toxic to cells at the high concentrations required during culture. Therefore, if the product could be removed in situ during culture, productivity might be enhanced due to the alleviation of this toxicity. In situ removal or extractive bioconversion of such products can be performed by in situ extraction with various kinds of organic solvents. In situ adsorption using polymeric resins is another possibility. Using the fact that secondary metabolites are generally hydrophobic, various integrated bioprocessing techniques can be designed not only to lower toxicity, but also to enhance productivity. In this article, in situ extraction, in situ adsorption, utilization of cyclodextrins, and the application of aqueous two-phase systems in plant cell cultures are reviewed.

We have examined specific genes whose expression is altered during apoptosis induced by prostaglandin (PG)A2 and Delta12-PGJ2 in human hepatocellular carcinoma Hep3B cells. Using mRNA differential display, we have identified two genes: one is specifically up-regulated and encodes for human Sox-4 (Sry-HMG box gene) and the other is significantly down-regulated and is the human homolog of yeast Ssf-1, a novel splicing factor. Northern blot analysis confirmed their differential expressions. Interestingly, Sox-4 was highly expressed in subcutaneous tumors grown in nude mice as a xenograft from Hep3B cells. These results suggest that the expression of Sox-4 may be related to the apoptosis pathway leading to cell death as well as to tumorigenesis, and that Ssf-1 gene may serve as a negative regulator of PGA2/Delta12-PGJ2-mediated Hep3B cell apoptosis.

RESUMO

The quadrature sampling technique as a means of detecting the envelope of RF waveform in the baseband is well known. If this technique is applied to a focused ultrasound imaging system using an array transducer, whether it is a synthetic or nonsynthetic focusing system, unwanted phase terms appear in the expressions of the inphase and quadrature components of the baseband signal when an appropriate delay time is introduced to each channel signal for the purpose of focusing. The expressions of the inphase and quadrature components from the point of focusing are derived and analyzed in detail, and a scheme to eliminate the unwanted phase terms is proposed. The resulting phase-error-free quadrature sampling technique is applied to the synthetic focusing system; a system block diagram together with the simulation and experimental results are presented.

A hybrid bioreactor was developed for the production of secondary metabolites from high density cultivation of plant cell suspensions. Some of the advantages of both air-lift and cell-lift by agitation were combined. The addition of a decanting column also made it possible to run a perfusion system for high density culture or to run a two-stage culture efficiently. Cell growth and the production of berberine from Thalictrum rugosum in the hybrid bioreactor are reported in this paper. A cell density up to 31 g/l was obtained by perfusion without any problems in mixing or loss of cell viability and the specific berberine productivity was comparable to that in shake flasks. The maximum berberine concentration was 88 mg/l at 3 weeks of operation and declined thereafter.
