RESUMO

Metamizole (dipyrone) is still a frequently used drug in human and veterinary medicine, especially for pain relief, sometimes also in osteosarcoma treatment. It has a very strong analgesic and antipyretic effect and weaker anti-inflammatory activity. Additionally, it has spasmolytic properties. It is used in many species, including dogs, cats, pigs, cattle, horses, and humans, in Europe, Canada, and South America. The aim of this study was to assess a potential synergism of metamizole as a non-steroidal anti-inflammatory drug with routinely used cytostatics (cisplatin, carboplatin, doxorubicin, and etoposide). In vitro studies were performed on a canine osteosarcoma D-17 cell line and a human U-2 OS cell line. We used the MTT method to assess cell viability, TUNEL staining to assess proapoptotic effects, and propidium iodide to analyse the cell cycle (potential arrest). The obtained results showed that metamizole at 50 µg/mL has potential cytoprotective and anti-apoptotic effects. Metamizole administered simultaneously with cytostatic drugs reduced their cytotoxic effect, which may suggest that such a combination of drugs used in the treatment of osteosarcoma may significantly reduce therapy effectiveness.

Studies of equine stomach prenatal development are very rare, and descriptions usually focus on the processes taking place in the embryonic period. Only general information about gastric organogenesis in the fetal period is available in embryology textbooks on domestic mammals. The material for our study included twenty half-breed horse fetuses divided into three age groups on the basis of known fetal age (verified using the CRL method). Our study consists of the topographical, morphological, and morphometrical description of stomach development between the 4th and 11th months of gestation. Even though the skeletotopy, syntopy, and holotopy of the stomach in the fetal period seems to be relatively unchanged, the organ shape and the proportions between its anatomical parts differed in fetuses from the three age groups. The achieved results were statistically elaborated to estimate the dynamics of the stomach shape. This can be described as changing from medium-wide to wide and from slightly bent to sharply bent. A nonlinear correlation of all metric values with CRL in all age groups was observed. A positive allometric growth rate of different intensity was seen in all metric parameters. All the values increased as the fetal period progressed. Only the parietal surface growth rate gradually changed from strongly positive allometric in the first age group to strongly negative allometric in the third age group. A difference between the non-glandular and glandular mucosa of the stomach was visible in the first group. Development of a well-distinguishable plicated edge margin began in the second age group together with gastric pits and gastric areas. The third age group showed a well-developed gastric groove and angular incisura.

Histological and morphometrical analysis of the stomach wall was performed during the foetal period divided into three age groups (4th-11th month of gestation). The material was taken from non-glandular (the blind ventricular sac) and glandular parts (the plicated edge margin/cardiac part, the body of stomach and the pyloric part) of the stomach. It was preserved and prepared according to the standard protocol. The histological slides were stained (H-E, Masson-Goldner and PAS). The analyses were performed using the light microscope. All measurements were statistically elaborated. The crown-rump length growth rate was estimated as isometric. The blind ventricular sac growth rate was lower than CRL (negative allometric) and the partition of stomach mucosa into non-glandular and glandular part occurred in the 1st age group. The plicated edge margin/cardiac part and the pyloric part shoved similar tendencies. Only the body of stomach demonstrated a higher growth rate than CRL (positive allometric), which can be explained due to the strongest development of fundic glands. Moreover, comparing the adult reference group to the three parts of the foetal period, all metric values were lower than those achieved prenatally. The blind ventricular sac was covered with the multiple plane epithelium. The glandular parts of stomach that formed the superficial concave areas were covered with the simple columnar epithelium in the 1st age group, which developed to the cardiac, fundic, and pyloric glands in the 2rd and 3rd age groups. The propria mucosae was built with the mesenchyme, which differentiated later to the loose connective tissue. The muscular layer of mucosa was not clearly distinguishable in the 1st age group. The muscular layer of the stomach wall was formed with myoblasts in the 1st age group and later in the 2nd and the 3rd age groups built with fusiform myocytes divided into internal and external layers. The non-differentiated cells of glandular epithelium transformed into the parietal and chief cells. The first were visible in the gastric glands of the 2nd age group. Both of them were present in the 3rd age group gastric mucosa. The PAS staining proved a moderate PAS-positive reaction in the 2rd age group, while it was estimated as intense Pas-positive in the gastric glands in the 3rd age group and was comparable to postnatal observation (the adult reference group).

The study discusses in vitro cytotoxicity of a combination of cytostatic drugs (doxorubicin, cisplatin, carboplatin, etoposide) and risedronate sodium against canine and human osteosarcoma (D-17 and U-2 OS). Standard protocols were used for the preparation of cell cultures and evaluation of their viability and apoptosis. MTT assay assessed the culture viability and EC50, while the apoptotic effect of the drugs was checked with a TUNEL assay. Doxorubicin alone showed the strongest cytotoxicity against D-17 (0.056 ± 0.019 µg/mL) and U-2 OS (0.051 ± 0.003 µg/mL), while the lowest cytotoxicity was observed for carboplatin (D-17, 6.45 ± 0.2 µg/mL and U2-OS, 27.5 ± 2.3 µg/mL). Risedronate sodium at 100, 10 and 1 µg/mL lowered viability in OS cell lines by 53.38 ± 1.46 and 49.56 ± 0.7%, 97.08 ± 3.32 and 74.92 ± 4.01%, and 102.67 ± 3.56 and 94.56 ± 3.52%, respectively. In all analyzed drug combinations, risedronate sodium significantly (* p < 0.05) increased the cytotoxicity against tested osteosarcoma cell lines. The decrease in cell viability caused by the studied compound combinations was weaker in canine than in human cell cultures. A combination of doxorubicin (all concentrations), cisplatin (1 µg/mL) and etoposide (1 µg/mL) with 100 µg/mL of risedronate sodium significantly improved the cytotoxicity of the drugs against canine and human osteosarcoma. Administration of carboplatin (1 µg/mL) and risedronate sodium (100 µg/mL), compared to carboplatin per se, produced no significant differences in cytotoxicity against the D-17 cell culture but significantly enhanced cytotoxicity in the U-2 OS line. The strongest apoptosis in both lines was detected for 0.01 µg/mL doxorubicin combined with 100 µg/mL risedronate sodium or 1 µg/mL cisplatin and 100 µg/mL risedronate sodium. In all combinations, the tested compounds revealed a synergistic mechanism of action.

The study consisted of the immunohistochemical analysis of fundic and pyloric mucosa in the equine stomach between the 4th and 11th month of gestation. The accessible material was classified into three age groups using the CRL method. The adult reference group was used to define potential differences between foetal and adult populations of gastric APUD cells. The samples were preserved, prepared, and stained according to the standard protocols. The immunohistochemical reaction was assessed using the semi-quantitative IRS method. The results were documented and statistically analysed. The most significant increase was seen in gastrin (G) cell activity. The activity of other endocrine cells (cholecystokinin (I) cells, somatostatin (D) cells, and somatotropin receptor (SR) cells) was less dynamic. This study proved that the development of APUD cells within the stomach mucosa undergoes quantitative and qualitative changes during stomach development. Our results correspond with the findings described in the accessible literature and prove a strong correlation between morphological changes in the stomach wall and the organ development, growth, and maturation.

The study describes the cytotoxic effect against human and canine osteosarcoma (U-2 OS and D-17) cell lines induced by risedronate sodium and meloxicam per se and in combination. Both cell lines were prepared according to standard procedures for cell cultures studies. The cell viability was estimated in both cell lines treated with chosen concentrations of risedronate sodium and meloxicam. The apoptosis assessment was carried out using TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) assay. EC50 values, computed for risedronate sodium and meloxicam cytotoxicity, showed comparable effects against the canine OS cell line in similar concentration of both drugs. In case of human OS, the stronger cytotoxic effect of risedronate sodium was proved. The EC50 values for meloxicam in both cell lines were, statistically, significantly different (* p < 0.05). Moreover, the cytotoxic effect of a combined administration of meloxicam and risedronate sodium in doses 100 µg/mL, compared with the negative control showed statistically significant differences. The human OS cell line was more resistant to both compounds than the canine OS cell line. The apoptotic effect in canine and human osteosarcoma triggered by risedronate sodium and meloxicam was statistically significant (p < 0.05). The cytotoxic effect induced with 100 µg/mL of risedronate sodium proved statistically significant differences between both tested cell lines compared to negative control. The results obtained with 10 and 100 µg/mL of meloxicam were not statistically significant. The study showed the synergic mechanism of action of risedronate sodium and meloxicam, but the concentrations used in vitro will not be possible to achieve in in vivo. Therefore, our results serve as basis only to design future studies on the tissue level.

In the past, executioners played an important role in the legal system. Besides sentence executions, they also worked as dogcatchers (i.e., eliminating stray animals or cadavers of dead animals from towns), and were responsible for sanitary conditions within their towns and closest neighborhoods. Archaeological explorations of gallows in the towns of Lower Silesia (Poland) provide evidence of such activities, including animal skeletal remains. Archaeozoological analysis of these materials from the towns Kamienna Góra (Landeshut), Zlotoryja (Goldberg), and Jelenia Góra (Hirschberg) are the subjects of this study. Our work also stresses the nature of the executioner's profession in animal health control and town hygiene maintenance before the development of modern veterinary services. The results show significant differences in the frequency of species and distribution of anatomical elements in accessible assemblages compared with animal skeletal remains unearthed in typical waste pits or classical inhumation, allowing the assumption that the animals were anatomically adults, and their health statuses were generally good. The dominant species, equids and dogs, were represented by skeletal remains, with the predominance of less valuable body parts (distal parts of appendices, caudal parts of the vertebral column). The fragmentation of accessible bone assemblages narrows the ability of larger conclusions (i.e., minimum number of individual estimations). The work enlightens the complex role of executioners pertaining to the hygiene of early modern town communities, a role later replaced by professional veterinarians with all of the consequences of the transition process.

This article pertains to the issue of early medieval dogs (10th-mid-13th century) from the territory of Poland and Central Europe. The study is based on dog remains from the Wroclaw Cathedral Island (Ostrów Tumski), one of the most important administrative centres of early medieval Poland, the capital of a secular principality and the seat of diocese authorities. The main morphological and functional types of dogs living in Wroclaw and other parts of Poland were characterized on that basis. It has been concluded that the roles and perceptions of dogs were very ambiguous. On the one hand, they were hunting companionship for the elite and were considered a symbol of devotion and loyalty. On the other hand, dogs symbolised disgrace. In everyday life, these animals were sometimes abused, their skin was sometimes tanned and their bones modified into tools, and in exceptional cases, dogs were even eaten.

In the 2015 excavation season, an east-west oriented burial (2015-Grave-14) built with large dimension stone blocks was unearthed on the south edge of "Area IVi" at the Balatlar Church in Sinop, on the northeastern Black Sea coast of Turkey. In this grave, which is dated between the end of the 6th century AD and the first half of the 7th century AD, a human skeleton was found with the head to the west and a cat skeleton was carefully placed next to the right femur. This study on the burial and the cat skeleton within it shows that, compared to the Roman period, the status of cats reached a higher level during the Byzantine period. It was found that alongside of being a pet, the Balatlar cat was a young healthy female individual that instinctively hunted rodents and birds, given that the remains of a rat and a sparrow were found in the region of the abdominal cavity, corresponding with the stomach location in the living animal. The grave presents the most significant direct archaeological evidence of a pet-human bond recorded at any Byzantine site so far.

The following article concerns the functional use of horse bones in the early Middle Ages (mainly in the period from the mid of the 10th to the 12th/13th century). The authors try to explain how such remains were used and how common it was. It is also discussed whether the special role of the horse in medieval societies somehow restricted its post-mortem usage, or perhaps there was no difference between the skeletal remains of horses and other species in this regard. For this purpose, statistical calculations on the use of the bones of various mammals were made. Only the remains of the species determined during the archaeozoological analysis were taken into account. The specific use of individual parts of a horse skeleton was also noted. In addition, the analysis also encompasses all other types of horse remains that could be used by humans (hide, hair, etc.). The consumption of horse meat was discussed separately: on the basis of the preserved traces, an attempt was made to determine whether it had happened, and if so, how popular it had been. Overall, such comprehensive analysis aims to show the various roles of the horse. It was not only a mount, but also a beast of burden, a source of food and raw material as well. The main purpose of this study is to describe the role of horses in human medieval societies of Ostrów Tumski on the basis of accessible equid remains. The highlighting of the human-horse relationship in the past allows us to understand the importance and value of the horse both as a life companion and the source of food or leather and bone tools.

The present study reports the synthesis, characterization, and crystal structure of a novel bioactive metal-organic framework, [Ag4(µ-PTA)2(µ3-PTA)2(µ4-pma)(H2O)2]n·6nH2O (bioMOF 1), which was assembled from silver(I) oxide, 1,3,5-triaza-7-phosphaadamantane (PTA), and pyromellitic acid (H4pma). This product was isolated as a stable microcrystalline solid and characterized by standard methods, including elemental analysis, 1H and 31P{1H} NMR and FTIR spectroscopy, and single crystal X-ray diffraction. The crystal structure of 1 disclosed a very complex ribbon-pillared 3D metal-organic framework driven by three different types of bridging ligands (µ-PTA, µ3-PTA, and µ4-pma4-). Various bioactivity characteristics of bioMOF 1 were investigated, revealing that this compound acts as a potent antimicrobial against pathogenic strains of standard Gram-negative (Escherichia coli, Pseudomonas aeruginosa) and Gram-positive (Staphylococcus aureus) bacteria, as well as a yeast (Candida albicans). Further, 1 showed significant antiviral activity against human adenovirus 36 (HAdV-36). Finally, bioMOF 1 revealed high cytotoxicity toward an abnormal epithelioid cervix carcinoma (HeLa) cell line with low toxicity toward a normal human dermal fibroblast (NHDF) cell line. This study not only broadens the family of PTA-based coordination polymers but also highlights their promising multifaceted bioactivity.

Fish frequently serve as bioindicators of aquatic environments during their ecological evaluation. Carpathian barbel (Barbus carpathicus, Cyprinidae) is a species common to rivers and lakes of Eurasia and Africa. Seasons of the year can influence its skin morphology and mucus composition. The clinical status of the animal depends on the above-mentioned factors. The aim of this study was a histological, histometrical and electrophoretical analysis of periodical changes in barbel common integument. The accessible material was investigated in histological, cytological and electrophoretic analysis using hematoxylin-eosin staining, histometric morphometry, gel electrophoresis and cytological methods. The results demonstrated significant differences in the investigated parameters for spring-summer and autumn-winter periods. Both skin epithelium morphology (epithelium thickness, number of cell layers, melanophores and mucous cell existence) and mucus composition (proteins, immune system cells, keratinocytes and mucocytes) showed significant differences between investigated seasons. These morphological and physiological changes were more pronounced in the dorsal than ventral regions of common integument. The differences in the physical characteristics of mucus and the histological structure of the skin cannot only serve as a source of useful information about an evaluated ecosystem, but can be also related to additional factors, e.g., microbiological and chemical water contamination.

This report presents canid cranial skeletal pathology from an excavation associated with the Przeworsk culture (III c. BC - V c. AD). The dog skull, an intentional inhumation, was dated to the Roman influence and the Migration period (I - V c. AD. The dog was a relatively large animal with a shoulder height calculated as approximately 60 cm. Massive bone changes localized on the facial surface of the left maxilla required a multistage diagnostic protocol. In addition to traditional macroscopic and morphometric evaluation, we used modern diagnostic imaging techniques such as digital radiography, computed tomography and 3D reconstruction. These, along with histopathological studies, allowed us to identify a primary malignant bone tumor: telangiectatic osteosarcoma.

Partial liver resection is a treatment of choice for liver tumors; the range of parenchyma excision varies from a small part of the tissue surrounding the neoplasm up to 70% of the organ. One of the major concerns during liver resection is blood loss. Thulium lasers which are characterized by the length of emission wave corresponding to a peak absorption of water create a new possibility of cutting tissues efficiently with minimal thermal damage and concurrently providing a good hemostasis control. The aim of our study was to evaluate an impact of liver transection with thulium doped fiber laser on an intraoperative bleeding and histopathological changes during postoperative period in swine model. Ten animals were subjected to open surgery partial liver resection and an incision of liver tissue with an all-fiber, diode-pumped, and continuous-wave Tm3+-doped fiber laser emitting 37.4 W of output power at ~1.94 µm wavelength. The macroscopic and histopathological evaluation was performed intraoperatively as well as 7 and 14 days after surgery. Macroscopically almost no bleeding was observed during surgery and no signs of bleeding were stated after 7 and 14 days. Histopathological analysis of the transection margin revealed a thermal damage area ranging in depth from 620.23 ± 23.82 µm on the day of surgery to 1817.70 ± 211.98 µm after 7 days. In the samples taken intraoperatively and after 7 days a superficial zone of carbonization was visibly separated from the deeper changes. After 14 days one 765.35 µm deep zone characterized by a granulation was present. In conclusion, thulium doped fiber laser is efficacious in cutting with a narrow zone of thermal injury and provides a good hemostasis during liver transection, thus being a potential tool for oncotic liver surgery.

OBJECTIVE: To evaluate efficacy of denervation of the of the hip joint capsule (HJC), as a treatment of hip joint pain. Specifically, we tested the hypothesis that HJC denervation will significantly reduce the number of sensory neurons innervating the capsule. STUDY DESIGN: Denervation of the HJC from a medial or lateral approach was followed by retrograde tracing of sensory neurons innervating the capsule. ANIMALS: Twenty adult male sheep (30-40 kg of body weight; Polish merino breed) were used in the study. METHODS: The hip joint was denervated from medial (n = 5) or lateral (n = 5) surgical approaches. Immediately after denervation, the retrograde neural tract tracer Fast Blue (FB) was injected into the HJC. An additional ten animals (n = 5 for medial and n = 5 for lateral approach) received the same treatment without HJC denervation to provide the appropriate controls. RESULTS: Results of the study revealed that the vast majority of retrogradely labelled sensory neurons innervating the HJC originate from fifth lumbar to second sacral dorsal root ganglia. Both the medial and the lateral denervations significantly reduced the number of sensory neurons innervating the HJC (39.2% and 69.0% reduction respectively). CONCLUSIONS: These results show that denervation of the HJC is an effective surgical procedure for reduction of the sensory neuronal input to the HJC. Moreover, the lateral approach was found to be significantly more effective for reducing sensory innervation as compared to the medial one.

In response to the need for implant materials characterized by high biocompatibility a new type of nanostructured Ti6Al7Nb implants for osseous tissue regeneration have been fabricated. The nanostructured cylindrical implants were manufactured in accordance with 3D CAD data using the Selective Laser Melting (SLM) method. Implants were subjected to chemical polishing using a mixture of nitric acid and fluoride (test group) as well as cleaned in distilled water and isopropyl alcohol (control group). The structural and morphological properties of the obtained samples were determined by using XRD (X-ray powder diffraction), TEM (transmission electron microscopy) and SEM (scanning electron microscopy) techniques. The particle size was verified and calculated by Rietveld method to be in the range of 25-90 nm. In the present study, experimental in vivo tests concerning implants fabricated from a nanostructured Ti6Al7Nb alloy, which may substitute bone tissue, were discussed in detail. The control group and test group were used in the study. The animal model was New Zealand rabbit. The implants were implanted into skull fornix and observed after 1, 2 and 3 months. The results of macroscopic and microscopic analysis proved better osseointegration of chemically modified implants.

Animal remains that are unearthed during archaeological excavations often provide useful information about socio-cultural context, including human habits, beliefs, and ancestral relationships. In this report, we present pathologically altered equine first and second phalanges from an 11th century specimen that was excavated at Wroclaw Cathedral Island, Poland. The results of gross examination, radiography, and computed tomography, indicate osteoarthritis of the proximal interphalangeal joint, with partial ankylosis. Based on comparison with living modern horses undergoing lameness examination, as well as with recent literature, we conclude that the horse likely was lame for at least several months prior to death. The ability of this horse to work probably was reduced, but the degree of compromise during life cannot be stated precisely. Present day medical knowledge indicates that there was little likelihood of successful treatment for this condition during the middle ages. However, modern horses with similar pathology can function reasonably well with appropriate treatment and management, particularly following joint ankylosis. Thus, we approach the cultural question of why such an individual would have been maintained with limitations, for a probably-significant period of time.

Measurements of the root canal during endodontic treatment have a significant influence on the course of the therapeutic process as well as on its final result in both human and veterinary medicine. The apical constriction should be the termination point for the preparation and filling of the root canal. This research was conducted with the use of a Septodont kit consisting of a small chamber filled with the examined solution in which a healthy second incisor was placed. The step back method was applied for the root canal preparation and master apical file of 30 was used. The working length was 22 mm. The examination was conducted with the use of steel as well as nickel titanium hand instruments. Different irrigation solutions and two types of apex locators were used. Measurements of the working length of the root canal showed dependence on the size of the instrument. Examinations carried out in various environments showed that analogical measurements were obtained only for sodium hypochlorite solutions. In other environments the measured sections were shortened. Comparative examinations with the use of steel instruments demonstrated insignificant measurement differences. Compared to these results, the measurements in nickel titanium group were characterized by more considerable deviations.

The excessive temperature fluctuations during dental implant site preparation may affect the process of bone-implant osseointegration. In the presented studies, we aimed to assess the quality of cooling during the use of 3 different dental implant systems (BEGO®, NEO BIOTECH®, and BIOMET 3i®). The swine rib was chosen as a study model. The preparation of dental implant site was performed with the use of 3 different speeds of rotation (800, 1,200, and 1,500 rpm) and three types of cooling: with saline solution at room temperature, with saline solution cooled down to 3°C, and without cooling. A statistically significant difference in temperature fluctuations was observed between BEGO and NEO BIOTECH dental systems when cooling with saline solution at 3°C was used (22.3°C versus 21.8°C). In case of all three evaluated dental implant systems, the highest temperature fluctuations occurred when pilot drills were used for implant site preparation. The critical temperature, defined in the available literature, was exceeded only in case of pilot drills (of all 3 systems) used at rotation speed of 1,500 rpm without cooling.

PURPOSE: Thermographic assessment of temperature distribution within the examined tissues allows a quick, noncontact, noninvasive measurement of their temperature. The aim of the study was to evaluate the usefulness of digital infrared imaging in monitoring experimental inflammation of pleura (PL), lower lip (LL), and left paw (LP) and right paw (RP) of lower limbs in rats. MATERIALS AND METHODS: The inflammatory reaction was induced by injection of 1% carrageenin solution into pleural cavity, lip, or paws. With the use of digital infrared imaging temperature measurement was conducted at 0 to 72 hours of the inflammatory reaction. RESULTS: The temperature decrease was observed at the site of injection directly afterwards. Next, it was gradually increasing and it reached the maximum on the third day of the inflammatory reaction. Statistically significant changes were observed after 48-hour period in PL and LL regions, as well as after 72-hour period in LP and RP regions (P < 0.005). CONCLUSION: It was found that thermographic examination allows for indicating the presence of inflammatory reaction within examined tissues and determining the dynamics of this process. This method could be used as alternative procedure that allows using fewer animals for experiments.
