[Correction of intestinal dysbacteriosis in elderly patients with the use of bifidobacterin introduced by different routes]. / Korrektsiia disbakteriozov kishecknika u pozhilykh bol'nykh s ispol'zovaniem bifidumbakterina pri raznykh sposobakh vvedeniia.

The results of the correction of disturbances in intestinal microflora in elderly patients with the use of bifidumbacterin in the form of powder or suppositories are presented. The effectiveness of the rectal administration of bifidumbacterin (by microinjections through an enema or in suppositories) is shown in comparison with the oral administration of the preparation. The administration of bifidumbacterin in suppositories for 15 days resulted in decreased content of hemolytic Escherichia below the threshold of determination in all examined patients.

A method for estimation of urease activity in gastric mucosa biopsy specimens and Helicobacter pylori cell suspensions.

A method for measuring urease activity in biopsy specimens and Helicobacter pylori cultures from these specimens is proposed. The method is based on measurement (with a portable pH-meter) of the rate of pH changes in a reaction mixture consisting of buffer, substrate (urea), and biopsy specimen or bacterial cells. This method revealed that urease activity of biopsy specimens correlated with that of H. pylori suspension in the same experiment. High urease activity was found in biopsy specimens containing the greatest number of Helicobacter cells; only one of 14 specimens free of H. pylori cells showed no urease activity. Introduction of this method into clinical practice will help to evaluate the contribution of H. pylori to the pathological process.

The anti-Escherichia activity of 188 serum samples obtained from nonimmunized blood donors and 157 batches of the preparation of commercial normal human immunoglobulin were evaluated in the passive hemagglutination test with diagnostica containing E. coli antigens (serovars O2, O14, O26) and Kunin antigen, as well as Re-glycolipid Salmonella minnesota strain R595, used as sensitins. The study demonstrated that from sera of nonimmunized blood donors and from normal human immunoglobulin preparations 5-8% of batches containing anti-Escherichia antibodies to E. coli lipopolysaccharide (serovar O2) in titers of 1:640 and more could be selected. The study revealed that certain anti-Escherichia antibodies of commercial normal human immunoglobulin, to IgG.

The toxicity, immunogenic properties and protective activity of the live culture of E. coli M17 and antigenic preparations obtained from cell suspensions of this strain have been studied under experimental conditions. As revealed in experiments on mice, E. coli M17 live culture has low virulence, moderate toxicity and provides the protection of immunized mice from challenge with homologous and highly virulent E. coli strains. E. coli M17 live culture, when introduced orally or intravenously into rabbits, ensures the synthesis of 02 and H6 antibodies. Blood sera taken from immunized rabbits yield better results than initial sera in experiments on the passive protection of mice. The results of our experiments show the expediency of the clinical trials of Colibacterin as a perspective Escherichia live oral vaccine.

The antigen-binding activity of commercial preparations of immunoglobulin with different molecular composition was studied in the enzyme immunoassay with Escherichia coli lipopolysaccharide and serogroup B Neisseria meningitidis protein antigen. Immunoglobulin preparations for intramuscular injection were found to possess higher specific antigen-binding activity than immunoglobulin preparations for intravenous injection, which was determined by both the presence of the highly active polymer and dimer fractions and greater specific activity of the monomer fraction. A group of intramuscular immunoglobulin preparations with a considerable content of polymers were found to have significantly greater specific activity than a group of preparations containing small amounts of polymers.

The method for the determination of bacterial antibodies to group B meningococci was worked out. The method was used for the determination of antibodies to group B meningococcal vaccine produced in the USSR. The dynamic study of antibodies to protein, polysaccharide and lipopolysaccharide antigens of group B meningococci was made by the method of the enzyme immunoassay (EIA), and the safety of the vaccine was studied by the determination of autoantibodies active against brain tissue antigens. The data thus obtained were indicative of the immunological activity of group B protein-polysaccharide vaccines, manifested by the capacity for stimulating bactericidal antibodies whose level increased 8- to 10-fold after the immunization of monkeys in 2 and 3 injections. Similarity in the dynamics of the formation of bacteriolysins and antibodies to protein antigen, as determined in EIA, was noted. The vaccine was found to stimulate no cytotoxic anticerebral antibodies in the glia migration test, which was indicative of the safety of group B meningococcal vaccine.

To evaluate the specific activity of house-dust allergen, the passive hemagglutination inhibition test was used. Nine commercial batches of house-dust allergen were studied by means of this test. The results of the determination of the activity of house-dust allergen in the passive hemagglutination inhibition test, the skin tests and the indirect mast-cell degranulation test were in good correlation.
