RESUMO

Objective:To evaluate the relationship between blood transfusion factors (massive transfusion, transfusion of stored red blood cells and anhepatic transfusion) and early postoperative outcomes in the patients undergoing orthotopic liver transplantation.Methods:Patients with end-stage liver disease who underwent orthotopic liver transplantation and intraoperative blood transfusion from January 2021 to March 2022 in our hospital were retrospectively selected, and their clinical data were collected. The patients were divided into massive transfusion group (M group, total red blood cell transfusion ≥10 U) and non-massive transfusion group (NM group) according to the amount of intraoperative blood transfused. The patients were divided into fresh blood group (NS group) and stored blood group (S group, red blood cell storage time > 2 weeks) according to the storage time of red blood cells transfused. The patients were divided into transfusion in anhepatic stage group (group T) and nontransfusion in anhepatic stage group (group NT) according to whether blood was transfused in the anhepatic stage. Multivariate logistic regression, generalized linear model and generalized linear mixed model were used to analyze the relationship between blood transfusion factors and primary outcome measures (postoperative pulmonary complications, circulation overload, acute kidney injury, abdominal infection, thrombosis) and between blood transfusion factors and secondary outcome measures (duration of ICU stay, postoperative hospitalization, postoperative temperature ≥38.5 ℃ at any time point after surgery, postoperative liver and kidney function index, coagulation function index, platelet count and preoperative difference).Results:One hundred and six patients were enrolled in this study. The results of multivariate logistic regression analysis: Massive blood transfusion and infusion of stored blood were risk factors for postoperative pulmonary complications, massive blood transfusion was a risk factor for abdominal infection, Child-Turcotte-Pugh score for liver function and time of anhepatic phase were risk factors for postoperative acute kidney injury, Child-Turcotte-Pugh score was a risk factor for circulatory overload, and age and massive blood transfusion were risk factors for thrombosis ( P<0.05). The results of generalized linear model analysis: Intraoperative stored blood transfusion and blood transfusion during the anhepatic phase were risk factors for prolonged duration of ICU stay, massive blood transfusion, intraoperative stored blood transfusion and blood transfusion during the anhepatic phase were risk factors for prolonged hospitalization ( P<0.05), and there was no correlation between massive blood transfusion, intraoperative stored blood transfusion and blood transfusion during the anhepatic phase and temperature ≥38.5 ℃ at any time point after surgery ( P>0.05). The results of generalized linear mixed model analysis: There was statistically significant difference in the difference between preoperative and postoperative liver and kidney function indexes, coagulation function indexes and platelet counts in the patients with or without massive transfusion, stored blood transfusion or blood transfusion during the anhepatic phase ( P<0.05). Conclusions:Massive blood transfusion and infusion of stored blood are the main risk factors for early adverse outcomes after orthotopic liver transplantation in the patients.

The compounding of polyoxometalates (POMs) with structurally well-defined and porous metal-organic frameworks (MOFs) has become a hot research topic. Here, a core-shell type hybrid, {Ag5BW12O40}@[Ag3(µ-Hbtc)(µ-H2btc)]n (called {Ag5BW12O40}@Ag-BTC-2, where BTC = 1,3,5-benzyl carboxylic acid), was successfully prepared via a simple grinding method. IR, XRD, SEM, TEM, and XPS analysis was used to confirm the structure. The specific capacitance is 179.1 F g-1 when the current density is 1 A g-1, using nickel foam as the collector, and the capacitance retention is 97.4% after 5000 cycles. The resulting aqueous-based symmetric supercapacitor has a power density of 496 W kg-1 and an energy density of 12.4 W h kg-1. In addition, the degradation rates using {Ag5BW12O40}@Ag-BTC-2 toward methylene blue (MB), rhodamine B (RhB), and methyl orange (MO) exceeded 90% in 140 min and remained essentially unchanged over five replicate experiments, showing high photocatalytic activity. Meanwhile, when {Ag5BW12O40}@Ag-BTC-2 acts as a H2O2 biosensor, it has a low detection limit (0.19 µM), a wide linear range (0.4 µM-0.27 mM) and high anti-interference properties. This shows that the synthesis of POMOFs via a grinding method is an effective strategy to improve the performance.

Objective To study the relationship between chemotherapeutic resistance to 5-fluorouracil (5-FU) and Golgi phosphoprotein 3 (GOLPH3) expression in human colon cancer cell line HT29.Methods HT29 cells were divided into four groups:control group,siRNA transfection group,experimental group 1 (30 μmol/L 5-FU),and experimental group 2 (siRNA-GOLPH3 + 30 μmol/L 5-FU).The silencing effect of GOLPH3 gene was detected by RT-PCR and Western blot.The tumor proliferation and formation ability of HT29 cell were measured respectively by MTT and plate colony formation assay.The protein expressions of GOLPH3,P-gp and β-catenin in HT29 cell were detected by Western blot.Results Compared with the control group,the expression level of GOLPH3 mRNA (1.000 ± 0.078 vs.0.147 ± 0.021,t =12.296,P < 0.01) and protein(1.003 ± 0.235 vs.0.077 ± 0.399,t =6.723,P < 0.01),the absorbance(A value) (1.000 ± 0.082 vs.0.769 ± 0.086,t =3.885,P < 0.01) and the tumorigenesis (430 ±36 vs.297 ± 21,t =5.492,P < 0.01) in transfection group significantly reduced.The A value (0.803 ± 0.101 vs.1.050 ± 0.140,t =2.855,P < 0.05;0.242 ± 0.091 vs.1.050 ± 0.140,t =9.664,P < 0.001) and the tumorigenesis (73 ± 8 vs.427 ± 29,t =20.363,P < 0.001;305 ± 22 vs.427 ± 29,t =5.840,P < 0.01) in experimental group 1 and experimental group 2 were significantly lower than that in the control group,while the A value (t =8.264,P < 0.001) and the tumorigenesis (t =17.346,P < 0.001) of experimental group 2 significantly decreased than that experimental group 1.Compared to the control group (0.967 ± 0.094,1.001 ± 0.199,1.000 ± 0.101),the expressions of GOLPH3,P-gp and β-catenin in experimental group 1 (3.634 ± 0.574,2.424 ± 0.261,2.324 ± 0.418) significantly upregulated (t =7.944,P < 0.01;t =7.520,P < 0.01;t =5.330,P < 0.01),while the expressions of these proteins in experimental group 2(0.520 ±0.176,0.466 ±0.159,0.933 ±0.049) were significantly lower than that in experimental group 1 (t =8.983,P < 0.01;t =11.106,P < 0.001;t =5.724,P < 0.01).Conclusion Expression of GOLPH3 is associated with resistance to 5-FU in colon cancer HT29 cell by activating Wnt/β-catenin signal pathway.

Objective:To understand the oral health status of urban and rural residents in China,the Fourth National Oral Health Epidemiological Survey (FNOHES) was conducted in 2015.Materials and methods:Based on the principles of multi-stage,stratified,equal capacity,and random sampling,the subjects included five groups of residents aged 3-5,12-15,35-44,55-64,and 65-74 years in all 31 provinces,municipalities,and autonomous regions except Hong Kong,Macao,and Taiwan.The total sample size was 172,425.The survey covered two areas,namely oral health status and knowledge,attitude and oral health behavior.Results:Residents in China had serious caries and poor periodontal condition.The prevalence of caries was above 50% in all age groups,subjects without periodontal disease accounted for <50%,and fewer than 50% of subjects brushed their teeth twice a day.Based on the results of the FNOHES,the following suggestions were proposed for improving the oral health in China in the future:to establish and improve the comprehensive prevention and control system for oral diseases,implement a comprehensive prevention and control strategy for priority groups,strengthen the oral health education for all people to improve oral health literacy,enhance dynamic monitoring to scientifically evaluate oral health status,and coordinate multi-party resources to establish and improve the oral health service security system.Conclusion:The oral health status of Chinese residents and their oral health behavior habits need to be improved.With the support of the administrative departments of the government and unremitting efforts of the oral health staff,all sectors of society must be coordinated to strengthen oral health education for the whole population.Furthermore,comprehensive prevention and control measures must be implemented for priority groups.

Objective To observe the expression dynamics of lens-related transcription factors in human embryonic stem cell (hESC) differentiated into lentoid body(LB).Methods A "three-stage" protocol was used for LB directional differentiation from hESC in vitro.The hESC (D0) and three differentiation stages cells were collected to analyze the expression dynamics of lens development-associated transcription factors by high throughput RNA sequencing technology in hESC-induced LB.Western blot and cell immunofluorescence were used to observe the involved genes at protein level.Results During D0-D6,cells became more round and compact.And during D7-D18,cells morphology gradually changed to spindle.At the end of D35,three-dimensional and transparent structurelentoid body (LB) was obtained.RT-PCR results showed that the stem cell related genes reduced and the lens specific genes increased significantly,and the LB was characterized by the expression of crystallins.According to clustering analysis of high throughput sequencing,a distinct difference in transcription factors gene expression was observed between D0 and D32.Meanwhile,the difference between D6 and D18 was minimum.The expressions of preplacodal genes,including DLX3,DLX5,DLX6,HES1,HES4,OTX2 and EYA1 increased remarkably at the first induction stage and then decreased.Lens-specification gene SOX2 declined gradually and then increased.In addition,the expression of PAX6 increased during all three induction stages.Furthermore,lens-differentiation genes including MAB21L1,CMAF,PROXI and PITX3 had no significant change in the early induction stage,but increased significantly at the third induction stage.Conclusions The expression dynamics of lens development-associated transcription factors in the hESC induced LB corresponded to those in vivo,which indicate that this induction system can recapitulate early lens development well and lay the foundation of studying lens embryonic development andtranscription factor associated congenital lens diseases.

A recombinant peptide Pro-Pro-[Arg11]hPTH(1-34)-Pro-Pro, was designed, biosynthesized, and purified in our lab (generated by substituting Arg for Leu at position 11 and adding -Pro-Pro to the carboxyl terminus of hPTH(1-34)-peptide), and reported to be capable of enhancing serum calcium level in chicken. In this investigation, we studied its impact on the structure, component, and biomechanical properties of rat bones. The pharmacological effect of Pro-Pro-[Arg11]hPTH(1-34)-Pro-Pro was investigated to evaluate its therapeutic potential for the treatment of osteoporosis, one of the most prevalent and rapidly spreading diseases in the world. 3-month-old normal female rats were sham-ovariectomized or ovariectomized, then fed for 14 weeks. Small doses of Pro-Pro-[Arg11]hPTH(1-34)-Pro-Pro were given to the rats afterward (daily subcutaneous injection of 0.4-0.9nmol/100g body weight for 16 weeks). Values of various parameters, including the body weight, the bone mass, the bone geometry, the bone biomechanics, the bone histology, the bone histomorphology, and the serum biochemistry, were collected before or after animal sacrifice. Daily subcutaneous injection of Pro-Pro-[Arg11]hPTH(1-34)-Pro-Pro induced 37.0-42.4% increase in vertebral BMD, 28.5-47.9% increase in femoral BMD, 30.7-43.7% decrease in marrow cavity or increase in trabecular bone area. The peptide also increased calcium, phosphorus, and collagen in bone by 52.1-59.7%, 24.5-34.7%, and 26.3-28.0%, respectively. In terms of mechanic properties, peptide administration elevated bone rigidity by 55.7-84.5%, decreased the deflection by 14.8-26.7%, and improved modulus of elasticity by 28.1-76.4%. These data suggest Pro-Pro-[Arg11]hPTH(1-34)-Pro-Pro has a positive effect on bone mass accumulation and microstructure improvement, fortifies bone strength, and possesses anti-aging capability, which may merit further investigation.

Objective To understand the correlation between self-reported gingival bleeding and type 2 diabetes mellitus (T2DM) in adults in China.Methods The database of China's 2010 Chronic Disease and Risk Factor Surveillance (CCDRFS) survey among people aged ≥ 18 years was used to analyze the demographic characteristics of subjects with self-reporting gingival bleeding and the prevalence of major chronic diseases among adults.Correlation and interaction analyses were conducted on the relationships between frequent gingival bleeding and T2DM,hypertension and dyslipidemia,and interaction of age and gingival bleeding,age and hypertension,age and dyslipidemia,age and gender on the prevalence of T2DM.Results Among 93 647 adults surveyed,87.4% were in Han ethnic group.The incidence of frequent gingival bleeding was higher in females (63.6%) than in males (36.4%).The incidence of frequent gingival bleeding was highest (30.1%) in adults with middle school education level.Among the adults aged 45-60 years,12.8%(2 839/22 179) had T2MD but no gingival bleeding,15.6% (163/1 044) had both frequent gingival bleeding and T2DM,frequent gingival bleeding was correlated with T2DM (OR=1.29,95%CI:1.08-1.54) and the interaction with age had influence on T2DM (P<0.005).In males,frequent gingival bleeding was correlated with T2DM (OR=l.30,95% CI:1.08-1.56,P=0.005).In hypertension group,frequent gingival bleeding was correlated with T2DM (OR=1.25,95% CI:1.07-1.46),and interaction of hypertension and gingival bleeding had influence on T2DM (P<0.05).Conclusions The positive correlation between frequent gingival bleeding and T2DM was observed in adults surveyed,and the interaction of age and hypertension had influence on prevalence of T2DM.Frequent gingival bleeding was correlated with T2DM in males either.

Objective To evaluate the effect of YunKe on bone metabolism of patients with rheumatoid arthritis (RA). Methods A total of 108 RA patients were divided into YunKe group (n = 72), non-YunKe group (n = 36), and 54 controls. The bone mineral densities (BMDs) of the wrists, hips and lumbar spines were measured by dual-energy X-ray absorptiometry. Moreover , the expression of cross-linked N-telopeptides of type I collagen (NTX) in the serum was detected for statistically analyzing the difference between YunKe group and non-YunKe group. Results In RA group, the BMDs of wrist, hip and lumbar spine declined compared with that of control group (P < 0.001, respectively). In YunKe group, the BMDs of wrist, hip and lumbar spine after 6 months all were improved with statistically significance (P < 0.01, respectively); The differences also were statistically significant when compared with those in non-YunKe group (P < 0.01, respectively). In YunKe group, NTX level in the serum declined after 3 months of the treatment. The difference was statistically significant when compared with that in non-YunKe group (P < 0.001), but the difference disappeared 6 months later. Conclusions The BMD of wrist joints is low and declines in RA patients significantly. The treatment withYunKe improves the BMDs of the wrist , hip and lumbar spine and decreases the serum NTX. This study proposed that YunKe could inhibit the chronic loss of bone mass induced by RA.

[ ABSTRACT ] AIM: To investigate the effect of advanced oxidation protein product-human serum albumin ( AOPP-HSA) at different concentrations on the permeability of human umbilical vein endothelial cell ( HUVEC) monolayer and the protective effect of NADPH oxidase inhibitor diphenyleneiodonium ( DPI ) against AOPP-HSA exposure. METHODS: Cultured HUVECs were exposed to 200 mg/L HSA (control) or AOPP-HSA (50, 100 and 200 mg/L).The permeability of the endothelial monolayer was assessed by measuring CMFDA-labeled THP-1 cells across the endothelial cells.The cultured HUVECs were treated with HSA (200 mg/L), AOPP-HSA (200 mg/L), or AOPP-HSA (200 mg/L)+DPI (100 μmol/L), and the activation of NADPH oxidase, endothelial monolayer permeability and cytoskeleton rear-rangement were evaluated.RESULTS: AOPP-HSA increased the permeability of the endothelial cell monolayer, and AOPP-HSA at 200 mg/L significantly increased the phosphorylation level of NADPH oxidase in the cells.Treatment with 100 μmol/L DPI obviously attenuated AOPP-HSA-induced NADPH oxidase activation, the increase in the permeability of the cell monolayer and the cytoskeleton rearrangement.CONCLUSION: AOPP-HSA increases the hyperpermeability of HUVEC monolayer via the phosphorylation of NADPH oxidase, and the NADPH oxidase inhibitor DPI reverses such effects.

Objective To establish a close clinical and easy to operate animal artery bypass grafting model by using vascular anastomosis wheel.Methods 15 rabbits that weighted 2.5-3.5 kg were studied.Each animal underwent an end-toend anastomosis of jugular vein and carotid artery by vascular anastomosis wheel.Carotid ultrasound and flow detection were taken immediately and 2 months after surgery respectively,as well as morphology and pathology were recorded to analyze and evaluate the intimal hyperplasia of vein graft and arteriovenous anastomotic site.Results 14 rabbits were successfully established CABG model,however 1 rabbit died of respiratory inhibition caused of excessive anesthesia.Compared with normal carotid artery,the vein bridge showed significantly lower blood flow [(50.81 ± 1.33) ml/min vs.(70.59 ± 0.68) ml/min,P <0.01,higher PI(2.15 ±0.07vs.1.22 ±0.04,P <0.01)] immediately after surgery.Compared with the vein grafts immediately after surgery,the vein grafts 2 months after surgery showed significantly lower blood flow [(27.46 ± 2.15) ml/min vs.(50.81 ± 1.33) ml/min,P < 0.01].Compared with normal jugular vein,the vein grafts 2 months after surgery showed significantly higher intimal hyperplasia[(160.30 ± 1.78) μm vs.(49.06 ± 2.76) μm,P < 0.01],and higher number of elastic plates(12.36 ± 0.25vs.3.21 ± 0.15,P < 0.01).Conclusion The use of vascular anastomosis wheel to establish an artery bypass graft model can imitate the pathological changes of vein grafts after CABG,which can provide an ideal animal model for various researches on vein grafts.

Objective:To explore the productivity of township health services in Guangzhou. Methods:We use a nonparametric Malmquist index approach to analyze total factor productivity change in Guangzhou’s township health service. The panel data was collected between 2009 and 2013. Results: The average growth rate of Guangzhou’s township health service was 18 . 7% from 2009 to 2013 . This growth was mainly due to improved technical progress but not technical efficiency. Pure technical efficiency and scale efficiency have not developed simultaneously. Conclusion:The change in the total factor productivity was positively affected by technical progress, not by strategic management and business decisions inside township health centers.

<p><b>OBJECTIVE</b>To study the effects of 4 different methods for disinfection of simple breathing vesicles and microbial residue.</p><p><b>METHODS</b>The disinfection tests were divided into 4 groups: G1 group (43 cases) with 500 mg/L chlorine dioxide spray, G2 group (28 cases) with alcohol spray, G3 group (47 cases) with 50 mg/L trichloroisocyanuric acid (TCCA) immersion, and G4 group (46 cases) with 50 mg/L chlorine dioxide solution immersion. After 30 min of disinfection, each group was examined by bacterial culture and colony count. The residual bacteria were identified and typed.</p><p><b>RESULTS</b>The 4 methods showed significant differences in bacterial colony count (P<0.001). The rate of bacterial residue was 0% in G1 group, 53.6% in G2 group, 27.7% in G3 group, and 21.7% in G4 group, showing significant differences between the 4 groups (P<0.001). The residual bacteria included antibiotic-resistant common opportunistic pathogen such as Pseudomonas aeruginosa, Acinetobacter baumannii and Staphylococcus haemolytic.</p><p><b>CONCLUSIONS</b>Disinfection with 500 mg/L chlorine dioxide spray is the best for simple breathing vesicles. Prolonged immersion in TCCA may lead to the growth of drug-resistant pathogens in the breathing vesicles.</p>

<p><b>OBJECTIVE</b>To investigate the changes in the expression of nuclear receptor subfamily1 (Nr1d1) in the lungs of rats with ventilation-induced lung injury and explore the molecular mechanism.</p><p><b>METHODS</b>Seventy-two rats were randomly divided into 4 groups for observation at 4 time points (1:00 AM, 7:00 AM, 1:00 PM and 7:00 PM), and each group was further divided into 3 subgroups (Group FB, Group LV and Group HV). After ventilation for 3 h, the rats were sacrificed and lung tissue samples were obtained for testing the expression of Nr1d1 using gene chip technology and RT-PCR.</p><p><b>RESULTS</b>Nr1d1 gene expression was down-regulated in HV group as shown by gene chip RT-PCR data. Cosine analysis showed that Nr1d1 expression exhibited a rhythmicity in FB group (P<0.05).</p><p><b>CONCLUSION</b>The rhythmicity of Nr1d1 expression disappears as the tidal volume the increase. Diminished Nr1d1 expression is closely correlated with ventilation-induced lung injury in rats.</p>

Objective:To investigate the correlation between the expression of Golgi phosphoprotein 3 (GOLPH3) and the occurrence of apoptosis in colorectal cancer cells (CRC). Methods:Immunohistochemical assays of GOLPH3 and caspase-3 were performed on the paraffin-embedded sections of 62 CRC samples using the standard streptavidin-peroxidase technique. The apoptotic index of the CRCs was examined using the in situ terminal deoxynucleotidyl transferase nick-end labeling technique. The relationship of the GOLPH3 expression, the cell apoptosis, and the clinicopathologic parameters was analyzed. Results:The positive rates of GOLPH3 expression were significantly higher in the CRC tissues (53.2%) than in the normal colorectal mucosa (37.2%;P0.05). The caspase-3 expression and apoptotic index were significantly lower in the GOLPH3-positive CRC tissue than in the GOLPH3-negative tissue (P<0.05). GOLPH3 expression was negatively correlated with the apoptotic index of CRCs based on the Spearman correlation (r=-0.320, P<0.05). Conclusion:GOLPH3 overexpression in CRC tissue is negatively correlated with apoptotic index.

Osteoporosis is a skeletal disease characterized by low bone mass and microarchitectural deterioration of bone tissue, and it is a serious threat to human lives. We previously showed that the N-terminal peptide analog of human parathyroid hormone (Pro-Pro-PTH(1-34)) enhanced plasma calcium concentration. In this paper, we study the impact of PTH N-terminal fragment analog on the structure, component, and mechanical properties of the rat bones. Daily subcutaneous injections of Pro-Pro-hPTH (1-34) induces 26.5-32.8% increase in femur bone mineral density (BMD), 23.0-34.2% decrease the marrow cavity or increase in trabecular bone area. The peptide also increases 16.0-59.5%, 28.8-48.2% and 14.0-17.8% of bone components of calcium, phosphorus and collagen, respectively. In terms of mechanic properties, administration of the peptide elevates the bone rigidity by 45.4-76.6%, decreases the flexibility by 23.0-31.6%, and improves modulus of elasticity by 32.8-63.4%. The results suggest that Pro-Pro-hPTH (1-34) has a positive effect on bone growth and strength, and possesses anti-fracture capability, thus a potential candidate for the application for the treatment of osteoporosis.

ObjectiveTo describe utilizations of maternal health care in migrant maternity in Putuo District and to identify the gap between primipara and multipara.In order to improve utilizations of Maternal Health Care in migrant maternity.MethodsQuestionnaire survey was carried out among lying-in migrant maternity to obtain information on utilization of maternal health care.Database was established the by Epidata 2.1 and Statistical analyzed by SPSS11.0.ResultsThere was significant difference between multipara and primipara in age,household registration and cultural degree(P<0.01 ).Women registered at early stage of the pregnancy with 50.51%,completed full pre-natal health check-up with 82.83%,received postnatal visits with 78.45% in migrant maternity.There was significant difference between multipara and primipara in registered at early stage of the pregnancy,full pre-natal health check-up,received the postpartuu visit,registered at first level hospital and the medical insurance coverage in migrant maternity(P<0.01 ).ConclusionsThe levels of utilizations of maternal health care in migrant maternity in putuo district are significantly lower than local maternity,the levels of utilizations of maternal health care in multipara are lower than primipara in Putuo district.Improve utilizations of maternal health care in multipara and enhance utilizations of maternal health care in migrant maternity in Putuo district.

A recombinant human parathyroid hormone fragment, Pro-Pro-[Arg(11)]hPTH(1-34)-Pro-Pro (MW=4550 Da), was developed by substituting Arg for Leu at position 11 and adding Pro-Pro at the carboxyl terminus. Following a single injection (0.5-13.5 µg/bird) of the rhPTH fragment, the serum calcium level in chickens increased 12-42% (P<0.05) after 1h as determined by the Parson's Chicken Assay. The functional activity of Pro-Pro-[Arg(11)]hPTH(1-34)-Pro-Pro may be due to removal of the N-terminus Pro-Pro- by X-prolyl dipeptidyl peptidase IV (DPPIV) in vivo, increasing its activity compared to Pro-Pro-hPTH(1-34). This artificial rhPTH fragment could be used to increase calcium mobilization and potentially improve bone health.

Objective To study the relationship between the number of examined lymph nodes and the prognosis of colorectal cancer by TNM stage. Methods According to the number of examined lymph nodes, 567 patients of colorectal carcinoma who underwent resection were divided into three groups: ≤ 6,7-11 and ≥ 12, the 5-year overall survival rates of three groups were compared. For each TNM stage ( stage Ⅰ -Ⅳ ) , patients were substratified into two groups basing on the number of examined lymph nodes:0. 05 ) , however, the 5-year survival rates of stage Ⅱ and Ⅲ colorectal cancer in ≥12 group were significantly higher than<12 group(71. 1% vs. 32. 6% ,48. 8% vs. 30. 0% ,P<0. 05) , multivariable analysis revealed that the number of lymph nodes examined was an independent factor of prognosis of stage Ⅱ and Ⅲ colorectal cancer. Conclusions The number of examined lymph nodes significantly influenced the 5-year overall survival rate of TNM stage Ⅱ and Ⅲ colorectal cancer.

A recombinant human parathyroid hormone fragment, Pro-Pro-hPTH(1-34), with molecular weight of 4311.46 was acquired through gene engineering. It was then isolated and purified. The homogeneity of this fragment was characterized by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), high performance liquid chromatography(HPLC), isoelectronic focusing (IEF) electrophoresis and mass spectrometry(MS) methods. Its isoelectric point is 8.0 which was determined by IEF. It was found that the hormone fragment significantly induced calcium increment as compared to the control group (P<0.001) in Parsons's Chicken Assay, an established bioassay for the evaluation of the PTH effect. After the 3-month-old ovariectomized (OVXed) rats, the OVXed rat is one of the two models required by the U.S. Food and Drug Administration for the preclinical assessment of drugs for treating osteoporosis [DeLuca PP, Dani BA. Skeletal effects of parathyroid hormone (1-34) in ovariectomized rats with or without concurrent administration of salmon calcitonin. Am Assoc Pharm Sci 2001;3(4):E27 [1]]. Sprague-Dawley rats were fed for 14 weeks, daily subcutaneous injections of Pro-Pro-hPTH(1-34) for 16 weeks (0.4, 0.6 or 0.9 nmol/100 g body weight), reduced the ovariectomy (OVX)-triggered mass loss of vertebral trabecular bone. The mean Bone Material Density (BMD) increased to 29.2-34.5% in 3-month-old OVXed rats compared to control-vehicle group (P<0.001) and increased to 17.5-22.3% compared to sham-operated groups (P<0.01). In short, A recombinant Pro-Pro-hPTH(1-34) was harvested in purified form and its physico-chemical characterization was determined. It showed significantly enhanced activity upon two typical models for PTH fragments. It can increase the mineral density of vertebral trabecular bone just as synthetic hPTH(1-34), and the functional activity of Pro-Pro-hPTH(1-34) should be due to the removing of Pro-Pro- by Dipeptidyl peptidase IV (DPPIV). This study opened out a simplified method which was cheaper, faster than the conventional one for producing active hPTH fragment, and its applied prospect would be good; Furthermore, it may open up our own path in finding new methods for post-processing of gene-engineering product.
