We aimed to evaluate the impact of nutrient supplementation of beef female calves at pre-weaning on adipogenic determination. Thirty-four female calves were assigned to two experimental treatments: Control (CON, n = 17), where animals were supplemented only with mineral mixture; Supplemented (SUP, n = 17), where animals received energy-protein supplement containing minerals (5 g/kg of BW per day) of their body weight. Animals were supplemented from 100 to 250 days of age, and muscle samples were biopsied at the end of the supplementation period. Regarding the performance variables, there were no differences between treatments for initial body weight (P = 0.75). The final body weight (P = 0.07), average daily gain (P = 0.07), rib eye area (P = 0.03), and rib fat thickness (P = 0.08) were greater in SUP female calves compared with CON treatment. The number of fibro-adipogenic progenitor cells (P = 0.69) did not differ between treatments, while a greater number of intramuscular pre-adipocytes were observed in SUP than CON female calves (P = 0.01). The expression of miRNA-4429 (P = 0.20) did not differ between treatments, while the expression of miRNA-129-5p (P = 0.09) and miRNA-129-2-3p (P = 0.05) was greater in CON than SUP female calves. Our results suggest that nutrient supplementation at early postnatal stages of development enhances the commitment of fibro-adipogenic progenitor cells into the adipogenic lineages allowing to an increase in intramuscular fat deposition potential of the animals later in life.

Elastase-2 (ELA-2) is an angiotensin II-generating enzyme that participates in the cardiovascular system. ELA-2 is involved in hemodynamic and autonomic control and is upregulated in myocardial infarction and hypertension. The inhibition of angiotensin-converting enzyme (ACE) increased ELA-2 expression in the carotid arteries and heart of spontaneously hypertensive rats. In this study, we sought to investigate the role of ACE inhibition in hemodynamic and autonomic balance in elastase-2 knockout (ELA-2 KO) mice. Male ELA-2 KO and C57BL/6 mice were treated with the ACE inhibitor enalapril or saline for 10 days. After treatment, mice underwent surgery for cannulation of the femoral artery and arterial pressure recordings were made five days later in awake animals. The variability of systolic blood pressure (SBP) and pulse interval (PI) was evaluated in the time and frequency domain. Spontaneous baroreflex was assessed by the sequencing method. ACE inhibition caused a significant decrease in mean arterial pressure (117±2.2 vs 100±2.8 mmHg) and an increase in heart rate (570±32 vs 655±15 bpm) in ELA-2 KO mice. Despite a tendency towards reduction in the overall heart rate variability (standard deviation of successive values: 7.6±1.1 vs 4.7±0.6 ms, P=0.08), no changes were found in the root of the mean sum of squares or in the power of the high-frequency band. ACE inhibition did not change the spontaneous baroreflex indices (gain and baroreflex effectiveness index) in ELA-2 KO mice. Altogether, this data suggested that ACE played a role in the maintenance of hemodynamic function in ELA-2 KO mice.

Since nutritional requirements are increased at the end of gestation to meet the demands of the pregnant uterus, pregnant beef cows are susceptible to mobilization of body reserves (mainly fat and amino acids (AAs)) and to alter the metabolism of nutrients in the liver and muscle to support such demands. The objective of this study was to evaluate the effect of CP supplementation on maternal nutrient metabolism in the late gestation of beef cows grazing a low-quality pasture. Forty-three pregnant Nellore cows gestating male fetuses (average age = 6 years; average weight = 544 kg) at 193 ± 30 (mean ± SD) days (d) of gestation were divided into eight groups (experimental units, with four to five cows each). Treatments were (1) control (CON, n = 4): pasture-based (PB) diet without CP supplementation and (2) supplemented (SUP, n = 4): PB diet daily supplemented with 2 g/kg of BW of a 43.5% CP supplement. Liver and skeletal muscle biopsies were performed at 265 days of gestation and samples were collected for mRNA expression. On day 280 of gestation, blood samples were collected to assess plasma levels of AA. The CON-fed cows tended to have greater (P = 0.057) total circulating AA than SUP-fed cows. The circulating glycogenic AA was greater (P = 0.035) in CON than in SUP cows. CON cows was greater for histidine (P = 0.015), methionine (P = 0.007) and alanine (P = 0.036) than SUP cows. The CON- and SUP-fed showed no differences for gluconeogenesis, fatty acid transport and signaling axis markers in the liver. The mRNA expression of markers for skeletal muscle synthesis, p7056k (P = 0.060) and GSK3B (P = 0.096), tended to be greater in cows from CON than SUP group. No differences were found for mRNA expression of markers for skeletal muscle degradation. We conclude that CP supplementation to CP-restricted late-pregnant beef cows reduces the maternal tissue mobilization and changes the profile of plasma circulating AA and the mRNA expression of markers for the synthesis of skeletal muscle tissue.

RESUMO

Objetivo: conhecer a percepção de pacientes colostomizados por causas não oncológicas e seus familiares acerca da forma como aprenderam a cuidar do estoma e da possibilidade de utilização de um vídeo educativo como estratégia de educação em saúde. Métodos: pesquisa qualitativa, realizada por meio de grupo focal com oito pessoas, sendo quatro pacientes e quatro familiares. Os dados foram submetidos à análise de conteúdo temática. Resultados: organizou-se três categorias: Aprendendo a cuidar do estoma; o apoio familiar como potencializador do cuidado e da adaptação; as possibilidades e limitações no uso do vídeo educativo. Considerações finais: o processo de aprender a cuidar do estoma foi marcado pela escassez de orientações de enfermagem e concretizado no aprender fazendo, sendo a colaboração da família importante para o cuidado. O vídeo educativo mostra-se como tecnologia útil para educação em saúde, ao apresentar orientações básicas para o cuidado da colostomia, podendo complementar as orientações de enfermagem.


Objective: to know the perception of colostomized patients due to non-oncological cause and their relatives in front of the use of an educational video as a health education strategy. Methods:qualitative research, performed through a focus group with eight people, four patients and fourrelatives. The data were submitted to the thematic content analysis. Results: three categories were organized: Learning how to take care of and manage the stoma; Acceptance and adaptation: family support as a potentiator of this process; Educational video: possibilities and limitations. Final considerations: the process of learning to take care of the stoma was marked by the scarcity of nursing guidelines and made feasible in learning-doing, with family collaboration being important to care. The educational video is a useful technology in the health education process, presenting basicguidelines for colostomy care, and can to complement nursing orientations.

Para avaliar a substituição do feno Tifton 85 pelo caroço de algodão como fonte de fibra na dieta foram utilizados 24 bezerros da raça Holandesa, distribuídos em dois grupos. O grupo-feno recebeu feno e concentrado separados, fornecidos à vontade, e o grupo- algodão recebeu caroço de algodão incorporado ao concentrado (13,5 por cento), à vontade. O consumo de alimentos, o peso, as alturas da cernelha e do íleo, as circunferências torácica e abdominal, a concentração de glicose sangüínea, as concentrações de acetato, propionato e butirato, o pH do fluido ruminal e os pesos dos compartimentos do estômago foram mensurados. O consumo de alimentos, o desenvolvimento ponderal dos animais, as concentrações de acetato, propionato e butirato, o pH do fluido ruminal, as concentrações de glicose sangüíneas e o peso dos compartimentos do estômago foram semelhantes entre os tratamentos (P>0,05). A necropsia, não foram observadas alterações sugestivas de intoxicação por gossipol. Concluiu-se que o caroço de algodão substituiu, sem prejuízos, o feno como fonte de fibra na dieta de bezerros


Twenty-four Holstein calves divided into two groups were used to evaluate Tifton 85 hay or whole cotton seed as fiber source in their diets. One group received Tifton 85 hay and concentrate ad libitum (hay group) and the other received ad libitum (cotton group) only concentrate with whole cotton seed incorporated (13,5 percent). Feed intake; weight; whither and ileum heights; thoracic and abdominal circumferences; acetate, propionate and butyric concentrations; pH of ruminal content; blood glucose; and rumen, reticulum, omasum and abomasum weights were measured. Feed intake; animal performance; acetate, propionate and butyric concentrations; pH of ruminal content; blood glucose and weight of stomach compartments did not differ between the groups (P>0.05). No clinical evidence of gossypol intoxication was observed. It was concluded that whole cotton seed replaced hay as fiber source in the diets of calves

Utilizaram-se 42 bezerros da raça Holandesa para avaliação do consumo de mistura mineral (MM), até os 60 dias. Os animais foram distribuídos em três grupos conforme o sucedâneo fornecido: grupo soro/leite + MM (SLMM); grupo soro + MM (SMM); e grupo soja/leite + MM (SOMM). Os animais receberam concentrado, feno, MM e água à vontade. A avaliação do ganho de peso foi feita semanalmente. A concentração de glicose plasmática foi medida aos 7, 14 e 21 dias de vida, antes do fornecimento da dieta, e às 3, 6 e 9 horas após a primeira refeição. Realizaram-se hemogramas semanais para avaliações clínicas. Aos 30 e aos 60 dias, colheram-se sangue e urina de cinco animais de cada grupo, os quais foram sacrificados para obtenção de fragmentos do fígado, para análises de minerais. Os animais do grupo SMM apresentaram menores consumos de concentrado e matéria seca total (MST) entre a sexta e oitava semanas (P<0,05), com consumo de concentrado de 674,72; 1.080,02 e 1.223,65g/a/d e consumo de MST de 699,33; 1.134,31 e 1.287,84g/a/d, para os animais dos grupos SMM, SLMM e SOMM, respectivamente. No consumo de feno, houve interação grupos versus semanas (P<0,05). O ganho de peso foi maior nos animais do grupo SOMM, 2,18kg/a/d que nos do grupo SMM, 1,13kg/a/d (P<0,05). O consumo médio da MM no primeiro mês foi de 9,00g/a/d e da quinta a oitava semanas foi 4,62g/a/d. A concentração de glicose plasmática foi diferente entre as idades e os tempos (P<0,05).


Forty-two Holstein calves were used to evaluate the mineral supplement (MS) intake up to 60-day-old. The calves were divided into three groups, according to the milk replacer: whey/milk + MS (WMMS); whey + MS (WMS) and soy/milk + MS (SMMS). Commercial calf starter, hay, MS and water were offered "ad libitum", and the intake was measured daily. Body weight was monitored once a week and blood glucose levels were quantified on 14, 21 and 28-day-old and also before the supply of the diets, 3, 6 and 9 hours after feeding. Blood samples were collected weekly for blood count. On 30 and 60-day-old, five calves of each group had blood and urine samples collected for serum copper determination. After then, the animals were slaughtered and liver fragments were analyzed for minerals contents. It was found that between the sixth and eighth weeks, the calves from WMS group decreased the intake (P<0.05) of starter 674.72g/day and total dry matter (TDM) 699.33 when compared to the calves from WMMS and SMMS groups, which consumed an average of 1,080.02 and 1,223.65g/day of concentrate and 1,134.31 and 1,287.84g/day of TDM respectively. Animals from SMMS group presented higher weight gain (2.18kg/week) than MMS ones (1.13kg/week; P<0.05). The intake mean of MM in the first month was 9.00g/a/d and after the first month was 4.62g/a/d. Blood glucose levels were different among ages and times after feeding (P<0.05).

Quinze bezerros da raça Holandesa, desaleitados aos 30 dias de idade, foram utilizados para avaliar os efeitos da inclusão de fontes protéicas alternativas, em substituição às proteínas do leite integral, na formulação de sucedâneos do leite. Os animais foram divididos em três grupos: o grupo controle (C) - recebeu leite em pó integral reconstituído; o grupo sucedâneo SL (soro/leite) recebeu 41,6 por cento da proteína bruta (PB) vinda do concentrado protéico de soro (CPS), 23,1 por cento do soro de leite e 35,3 por cento do leite em pó integral; e o grupo sucedâneo S (soro) recebeu 68 por cento da PB vinda do CPS e 32 por cento do soro de leite. O consumo de concentrado, feno, sal mineral e água foi mensurado diariamente, enquanto o ganho de peso foi avaliado semanalmente. Não houve diferença (P>0,05) para o consumo de concentrado, sal mineral e água entre os grupos, nos períodos avaliados. O consumo de feno e o ganho de peso foram inferiores nos animais do grupo S (P<0,05). A substituição de 100 por cento do leite integral por CPS e soro de leite influenciou negativamente o desempenho.

A 2(3) factorial design was used in order to evaluate the influence of some adjuvants on the dissolution profile of tablets containing high doses of Maytenus ilicifolia spray-dried extract. Tablets were prepared on a single punch tablet press using 15 mm flat punches by individual direct compression of 650 mg from each formulation containing 375 mg of the spray-dried extract. The factors investigated were disintegrant (croscarmellose sodium or sodium starch glycolate), lubricant (colloidal silicon dioxide or magnesium stearate) and filler/binder (microcrystalline cellulose or lactose). The dissolution profiles were analyzed to determine the dissolution kinetics, the dissolution half-lives (t50%), the similarity factor (f2) and the dissolution efficiency (DE %), which was selected as the response criteria to evaluate the factorial design. The results revealed that in spite of the high content of spray-dried powder in the tablets, the dissolution profiles of the extract did depend on the adjuvant used. The filler/binder had the most important effect on the dissolution efficiency of the tablets.
